Abstract
1531974 Enzyme immunoassay of progesterone INSTITUT PASTEUR 14 Oct 1975 [14 Oct 1974] 41912/75 Heading G1B Enzyme immunoassay of progesterone is effected by the step of adding to a sample containing progesterone molecules to be assayed (Ag), progesterone molecules labelled with #-D-galactosidase (Ag-Gz) and an antibody consisting of an antiprogesterone serum (Ab 1 ) wherein the labelled progesterone molecules (Ag-Gz) and the unlabelled progesterone molecules (Ag) compete for binding to antibody and the step of precipitation of the soluble complexes formed in the first step by adding to the reaction mixture a second anti-γ- globulin antibody (Ab 2 ) in an insoluble form, the step of measuring the enzyme activity of the resulting solid phase or of the supernatant, the process being characterized in that the same progesterone derivative, viz. progesterone 11-α-hemisuccinate, is used for coupling the progesterone molecules with #-D-galactosidase (Ag-Gz) and for preparing the anti-progesterone serum (Ab 1 ).
