Abstract

Suitable substrates are coated with a monomolecular protein layer consisting of an immunologically reactive first protein and of an inert protein. The molecules of the first protein are moreover separated from each other by the molecules of the inert protein. The substrate with a monomolecular coating is contacted with a solution of an immunologically reactive, second protein which is specific for the first protein. This results in the second protein molecules binding to the first protein molecules to form a bimolecular layer on the substrate. In contrast to known substrates of this type, the second protein molecules show after formation of the bimolecular layer a larger number of active sites for another immunological reaction. In addition, when large first proteins are used there is in this way a significant increase in the contrast between the mono- and the bimolecular parts of the layer. The substrates with a bimolecular coating obtained in this way can be used for the detection and determination of various other immunologically reactive substances.
