Abstract

1449802 N-(#-Naphthylmethyl)-piperazines SMITHKLINE CORP 3 Feb 1975 [7 Feb 1974] 4536/75 Heading C2C Novel N-(#-naphthylmethyl)-piperazines of the general formula wherein R is a phenyl, C-pyridyl or C-pyrimidinyl ring which is optionally substituted by C 1-5 alkyl, halogen, hydroxy or C 1-5 alkoxy and R 1 is hydrogen, C 1-5 alkyl, halogen, hydroxy or C 1-5 alkoxy, and pharmaceutically acceptable acid addition salts thereof are prepared by reacting a #-chloromethylnaphthalene and a piperazine of the general formulµ or by reacting the said #-chloromethylnaphthalene with piperazine and introducing the desired N<SP>4</SP>-substituent into the resulting N<SP>1</SP>-(R 1 - substituted - # - naphthylmethyl) - piperazine, followed optionally by salification of the product. R 1 - Substituted - # - chloromethylnaphthalenes are prepared by reducing the appropriate #- naphthoic acid with LiAlH 4 and chlorinating the resulting #-naphthalenemethanol. Pharmaceutical compositions having anti- Parkinsonism activity comprises, as active ingredient, a N-(#-naphthylmethyl)-piperazine of the first general formula above or pharmaceutically acceptable acid addition salt thereof, together with a pharmaceutical carrier.
