Abstract

1396219 Test for sickle cell disease BAXTER LABORATORIES Inc 18 May 1973 [18 May 1972] 23707/73 Heading G1B A reagent kit for testing blood for sickle cell disease comprises an aqueous phosphate buffer solution of molarity from 2 to 2.5 and pH from 7 to 7.2, and haemolyzing agent and a reducing agent preferably each in powdered form, the buffer solution containing from 0.1 to 1<SP>ml</SP> per litre of a non- ionic surfactant. The haemolyzing agent may be saponin, the reducing agent sodium dithionate and the surfactant octyl or nonyl phenoxy polyethoxyethanol. In carrying out the test, a sample of unclotted blood is mixed with a predetermined proportion of a reagent prepared by mixing the kit components, when a turbidity in the mixture indicates the presence of haemoglobins. A microcapillary sample of the turbid mixture is centrifuged and inspected for results characteristic of sickle cell trait or of sickle cell anaemia.
