Abstract
1381168 2-Oxopyridinecarbonitriles; 2- pyridones; cyano derivatives of nucleophiles E R SQUIBB & SONS Inc 26 Jan 1972 [29 Jan 1971 28 May 1971] 3768/72 Heading C2C Novel 2-oxo-pyridinecarbonitriles of the Formula I wherein each R is a hydrogen atom, a C 1-12 alkyl, C 2-12 alkenyl, hydroxyl, C 1-12 alkoxy, nitro, cyano, halo, carboxy, amido or mercapto radical, an aliphatic or aromatic acyl radical of up to 20 carbon atoms, a phenyl radical substituted by one of the foregoing substituents, phenyl or aryloxy of up to 10 carbon atoms or wherein any two adjacent R groups taken together form a benzene ring; are obtained either (a) by treating acetoacetamidopyridine or substituted derivatives thereof with water soluble alkalimetal or alkaline-earth-metal nitrites or (b) by treating alkali-metal salts of 2(1H)-pyridone or substituted derivatives thereof with cyanogen halides. The above carbonitriles may be reacted with nucleophilic reagents to give the corresponding 2-pyridones and cyano-derivatives of the nucleophiles, e.g. compounds of the formula X-C#N, where X is diethylamino, piperidino, pyrrolidino, phenylthio, phenoxy, anilino, pentylamino, phenyl, ethyl, t-butoxy, amino, methylthio, diphenylamino, morpholino, ethoxy, butylthio or di(ethoxycarbonyl)methyl radical. The above pyridone carbonitriles show anticaudidal activity.
