1292661 Prostanoic acid derivatives AYERST McKENNA & HARRISON Ltd 22 Oct 1969 [22 Oct 1968 28 July 1969] 51866/69 Heading C2C The invention comprises novel prostanoic acid derivatives of the Formula I wherein (a) R<SP>2</SP> is H or C 1-3 alkyl, n is 1 to 6, R is H(OH) and R<SP>1</SP> is O ; (b) wherein R<SP>2</SP> is as defined in (a), R and R<SP>1</SP> are H(OH), and n is 1 to 3, 5 or 6; (c) R<SP>2</SP> and n are as defined in (a) and R and R<SP>1</SP> are both O ; (d) R<SP>2</SP> and n are as defined in (a), R<SP>1</SP> is OH, acetoxy, benzoyloxy, or tetrahydropyran-2-yloxy, R is O, and n is as defined in (b) when R<SP>3</SP> is OH ; and (e) n and R<SP>2</SP> are as defined in (a), R<SP>1</SP> is O, and R is acetoxy, benzoyloxy, benzyloxy or tetrahydro-pyran-2- yloxy, their preparation and the preparation of compounds of the above formula, R<SP>2</SP> is H or C 1 _ 3 alkyl, R and R<SP>1</SP> are H(OH) and n is 4 and R<SP>2</SP> is H or C 1-3 alkyl, R<SP>1</SP> is OH, R is O and n is 4. The compounds of the above formula in which R and R<SP>1</SP> are both O are prepared by treating compounds of the formula with zinc and acetic acid. Reduction of the diazo compounds thus obtained gives compounds of the Formula I above in which R and R<SP>1</SP> are both OH, R is OH and R<SP>1</SP> is O or R is O and R<SP>1</SP> is OH. Any hydroxy groups in the resulting compounds may be esterified or etherified, and any esters obtained hydrolysed. Pharmaceutical compositions contain the above novel compounds (a), (b) or (c) and pharmaceutically acceptable carriers or diluents. The compounds possess hypotensive, antihypotensive, gastric acid secretion inhibiting and/or bonclospasmolytic properties.
