1,195,736. Amide and N-protected peptide production. JELENIOGORSKIE ZAKLADY FARMACEUTYCZNE POLFA. 12 Sept., 1967 [13 Sept., 1966], No. 41667/67. Heading C2C. Amides are prepared by reacting a hydroximic acid chloride with a salt of an organic acid, suitably an N protected aminoacid or peptide, to obtain a hydroxamic ester and subjecting the prepared ester to aminolysis with a primary or secondary amine, optionally containing an ester group, or a salt thereof. The hydroxamic esters may be isolated. Products prepared by this process are PhCO - gly - ONHBz, CBO - gly - ONHC 6 H 11 , CBO - gly - ONHPh, CBO - ala - gly - OEt, CBO - val - gly - OEt, CBO - leu - ONHBz, CBO - phe - gly - OEt, BOC - gly - ONHPh, BOC - gly - phe - OEt, Phthaloyl - gly - ONHBz, Phthaloyl - gly - gly - OEt, HCO - phe - phe - OtBu, C 6 H 5 CONHBz, and C 6 H 5 CH 2 CONHBz. Intermediates isolated are PhCO-gly-O-N = C(OH) - Bz, CBO - gly - O - N = C(OH) - Bz, CBO - ala - O - N = C(OH) - Bz, CBO - val- ON = C(OH) - Bz, CBO - phe - O - N = C(OH)- Bz, BOC - gly - O - N = C(OH) - Bz, Phthaloylgly - O - N = C(OH) - Bz, and HCO - phe - ON=C(OH)-Bz.
