Novel 1,2,3,4 - tetrahydro - benzo - [61[1,6] . naphthyridines of the general formula wherein R<SP>1</SP> is hydrogen, a halogen, nitro, lower alkyl or lower alkoxy, R<SP>2</SP> is hydrogen, lower alkyl, phenyl, halophenyl, lower alkyl-phenyl, phenyl-lower alkyl, lower alkanoyl, lower alkylsulphonyl, phenyl-sulphonyl, lower alkyl-carbamoyl, di-lower alkyl-carbamoyl, lower alkoxylower alkyl, di-lower alkylamino-lower alkyl or benzoyl and R<SP>3</SP> and R<SP>4</SP> are each hydrogen, lower alkyl, di-lower alkyl-phenyl, halophenyl, lower alkyl-phenyl, lower alkoxy-phenyl or phenyl-lower alkyl, the term " lower " meaning that the radical referred to contains up to 6 carbon atoms, and pharmaceutically acceptable acid addition salts thereof are prepared by reaction of a 2-aminobenzonitrile and a 4- piperidone of the respective general formulµ followed by optional conversion of the product to a pharmaceutically acceptable acid addition salt thereof. Pharmaceutical compositions having central nervous system depressant and analgesic activity comprises as active ingredient, a 1,2,3,4- tetrahydro - benzo - [6][1,6] - naphthyridine of the first general formula above or a pharmaceutically acceptable acid addition salt thereof, together with a pharmaceutically acceptable carrier, and may be administered orally or parenterally.
