[Also in Division C3] A process for the production of a purified aqueous solution of penicillinamidase comprises the steps of: (1) adding to a penicillinamidasecontaining fermentation broth a quaternary ammonium salt and a metal hydroxide or a metal salt having a divalent metallic cation which does not destroy or inhibit penicillinamidase, whereby the penicillinamidase is caused to be affixed to the water-insoluble solids present in the broth, the amount of quaternary ammonium salt being such that the yield of penicillinamidase is greater than that obtained in the absence of the quaternary ammonium salt; (2) separating the water-insoluble solids from the broth; (3) mixing the water-insoluble solids with water, whereby said penicillinamidase is dissolved in the water; (4) separating the penicillinamidase-containing supernatant water layer from the solid materials; (5) adding to the separated water layer a solid absorbing agent incapable of absorbing penicillinamidase; and (6) removing the solid absorbent from the aqueous solution to provide a purified aqueous solution of penicillinamidase. The broth may result from the cultivation of Escherichia Coli in an appropriate nutrient medium. The penicillinamidase may be used to hydrolyse potassium benzyl pencillin to 6-aminopenicillanic acid which may then be used as the starting material for semi-synthetic penicillins.
