Abstract

A process for the isolation of a fibrinolytic factor from the tissue of animal organs comprises extracting the said tissue with boiling water maintained at a pH from 8 to 9, hydrolysing the extract obtained with a proteinase at a pH from 5 to 6, dialysing the hydrolysed solution, and precipitating material having a fibrinolytic activity with a water-miscible precipitating agent at a pH from 5 to 6. In an example, swine duodenum was suspended in boiling water at pH 8.6, solids were separated after 12 minutes, and the pH of the extract was adjusted to pH 6 before the extract was subjected to lysis with papainase at 65 DEG C. After 24 hours, the reaction mixture was filtered, and the filtrate was concentrated and dialysed in bags of Cellophane (the word "Cellophane" is a registered Trade Mark) against running water. The dialysate was then mixed with acetone at pH 5, and the precipitate which formed was collected and dried. The product may be further purified by treating an aqueous solution thereof with benzyl-dimethyl-2-[2-(p-1,1,3, 3,-tetramethylbutylphenoxy)-ethoxy] ethyl ammonium chloride at pH 12, lowering the pH to 4, and washing the resulting precipitate with deionized water. The liquids collected were dialysed, the dialysate was adjusted to pH 6.5-7 and poured into acetone containing KCNS. The precipitate was washed with acetone to remove all traces of KCNS, before being vacuum-dried and powdered.
