Abstract
Hydroxy cobalamin and Vitamin B12 are obtained by cultivating Propionibacterium Freudenreichii or Propionibacterium Shermanii, inoculating a suitable medium with the culture, thereby producing Vitamin B12 and its analogues and then, at the end of its growth and production cycle, separating the cells from the broth, and suspending these in physiological saline in the presence of 5-6 dimethylbenzimidazole or this compound together with a non-toxic salt capable of releasing cyanide ions such as sodium thiocyanate or potassium thiocyanate so as to convert the analogues to cobalamin and Vitamin B12. In examples, the bacteria are kept in a "stub culture" prepared with peptone, meat extract, yeast, tomato juice and agar and then this culture is grown on a coin steep-dextron medium. The cells are separated as a dense cream and are then suspended in sodium chloride solution and the required precursors added. The reaction is stopped when optimum conversion of the analogues to Vitamin B12 has been effected. The vitamin may be extracted by the method described in Specification 1,007,971.
