Abstract

<PICT:1006547/C2/1> The antibiotic Takamycin is obtained by cultivating Streptomyces takataensis or an antibiotic Takanycin - producing mutant thereof in a culture medium containing as essential constituents carbon and nitrogen sources under aerobic conditions and recovering the antibiotic from the mycelium and the culture broth. Preferably the culture medium is liquid, contains inorganic salts and is cultivated at pH 7.0 and 27-30 DEG C. for 3 to 6 days under submerged aerobic conditions. Takamycin is recovered from the mycelium by extraction with methanol, ethanol or acetone, followed by washing with ether or ethyl acetate and recrystallization from aqueous ethanol; and from the culture filtrate by adsorbtion on Japanese acid clay, elution with butanol/pyridine/water, concentrating the eluate, and washing and recrystallizing as above. Takamycin is a yellow crystalline amphoteric substance decomposing at 150 DEG C., soluble in pyridine, dimethylformamide and glacial acetic acid, hardly soluble in diethyl ether; chloroform, petroleum ether and moderately soluble in methanol, ethanol and acetone, particularly at an alkaline pH and insoluble in water at acid and neutral pH; having u.v. maxima at 341, 360, 380 and 405 mm (Img/ 100 ml. of methanol); an I.R. spectrum as shown in Fig. 2; gives negative ninhydrin and o-nitrobenzaldehyde reactions and shows the elemental analysis c = 60.48-59.51%, H = 7.93-7.70% and N = 3.05-2.64%. The antibiotic Takamycin in a vaseline base is used to treat Trycohophyton infections.
