Abstract

The antibiotic 6-demethyltetracycline is produced by cultivating a 6-demethyltetracycline-producing strain of Streptomyces psammoticus especially s. psammoticus ATCC 14125 in an aqueous nutrient medium, e.g. an aqueous medium pH 4-8 containing a water-soluble carbon source, a source of assimilable nitrogen and essential mineral salts, under aerobic submerged conditions until substantial antibiotic activity is imparted to said medium and recovering the antibiotic thereform. Preferably cultivation is carried out at 25 DEG -37 DEG C. for 36-120 hrs. The antibiotic is isolated by acidifying the fermented broth to pH 1.5, filtering, adjusting the filtrate to alkaline pH (pH 8.5-9.0), extracting with a water immiscible aliphatic alcohol of 1-8 carbon atoms, e.g. butanol, extracting the organic phase with a dilute mineral acid, e.g. hydrochloric or sulphuric acid, evaporating the aqueous phase to small volume and adjusting the pH to 5.0 to precipitate 6-demethyltetracycline hydrochloride.
