Abstract

The invention comprises endolo-indolazine compounds of the general formula <FORM:1003645/C2/1> wherein R is alkyl containing 1 to 5 carbon atoms and the process of preparation thereof, including the case where R is hydrogen, which comprises the reduction by sodium borohydride of a quaternary salt of formula <FORM:1003645/C2/2> wherein R is hydrogen or C1 to C5 alkyl, and X represents the anion of a monobasic acid. The quaternary salt of the above formula is prepared by Bischler and Napieralski method of cyclising an indolylethylpyrrolidone of general formula <FORM:1003645/C2/3> wherein R is H or C1-C5 alkyl. The indolylethylpyrrolidones of the above formula are prepared by condensation of tryptamine with a gammahalobutyric acid alkyl ester, which may carry an alkyl substituent containing 1 to 5 carbon atoms on the methelene chain, in the presence of a basic condensing agent e.g. potassium carbonate in an inert solvent.ALSO:The invention comprises pharmaceutical compositions containing an inert pharmacologically acceptable carrier and at least one indolo-indolizine derivative of general formula <FORM:1003645/A5-A6/1> wherein R is hydrogen or alkyl containing 1 to 5 carbon atoms, for oral and parenteral administration the said compositions having a sedative action.
