La présente invention concerne un extrait de Curcuma longa ayant moins de 0,1% en poids de curcumine, notamment ayant moins de 0,1% en poids de curcuminoïde, par rapport au poids de l’extrait sec, ainsi qu’une composition dermatologique ou dermo-cosmétique le contenant, pour leur utilisation dans le traitement et/ou la prévention de dermatoses inflammatoires, notamment le psoriasis, la dermatite atopique ou l’acné. Les dermatoses sont des affections de la peau et des muqueuses, qui se caractérisent par des manifestations inesthétiques comme des rougeurs et des plaques de desquamation. Plusieurs pathologies sont regroupées sous la dénomination de dermatoses inflammatoires. On peut citer, à titre d’exemples, le psoriasis, la dermatite atopique, l’eczéma, l’acné, la rosacée, le lichen plan, les prurigos, ou encore les dermites séborrhéiques. Ces dermatoses résultent bien souvent de phénomènes inflammatoires et/ou de désordres immunitaires.Dermatosis are conditions of the skin and mucous membranes, which are characterized by unsightly manifestations such as redness and patches of desquamation. Sur le plan histologique, la dermatite atopique se caractérise comme bien des dermatoses, par une infiltration de lymphocytes, monocytes et de polynucléaires éosinophiles autour de petits vaisseaux et des capillaires. Dans le cas d’affections légères d’une dermatose inflammatoire, des émollients et des kératolytiques sont préconisés. Ces traitements ont pour but de rendre les lésions tolérables pour le malade et ils n’ont souvent qu’un effet suspensif. Pour les affections plus sévères, ce sont des anti-inflammatoires ou des corticoïdes susceptibles de réguler l’inflammation de la peau, qui sont utilisés depuis plusieurs années. Tous ces traitements ont des effets secondaires importants, parfois très lourds pour les patients. Du fait des effets secondaires importants des traitements existants précités pour les troubles cutanés ou du cuir chevelu résultant de l’état d’activation de la réponse immunitaire innée et inflammatoire épidermique de la peau, il existe un réel besoin de disposer de nouveaux actifs cosmétiques ou dermatologiques et de nouvelles compositions cosmétiques ou dermatologiques utilisables en relais ou en accompagnement des traitements desdits troubles cutanés ou du cuir chevelu.In the case of mild conditions of inflammatory dermatosis, emollients and keratolytics are recommended. Les raisons de ces maladies dites auto-immunes ne sont pas encore complètement élucidées mais seraient liées à une dérégulation du système immunitaire notamment au niveau des réponses cellulaires. On peut classer les réponses cellulaires en 3 catégories: Th1, Th2 et récemment Th17. Bien que le psoriasis et la dermatite atopique présentent des différences au niveau des signes cliniques, elles présenteraient des ressemblances notamment au niveau d’IL-17 et IL-22 caractéristiques de la réponse Th17 (Miyagaki and Sugaya, J. Dermatol Sci. 78(2) 89-94, 2015). Il-17 et IL-22 seraient donc des cibles intéressantes pour traiter ces maladies. Ainsi des anticorps monoclonaux dirigés contre IL-17, IL-17R, IL-22, IL-23, TNFa ont été développés pour bloquer la voie Th17 (Lowes et al., Annu Rev Immunol 32, 227-255, 2014). En effet, les inventeurs ont mis en évidence, de façon tout à fait inattendue, qu’un extrait deCurcuma longaprésente des activités pharmacologiques d’intérêt pour le traitement et la prévention des dermatoses inflammatoires et plus particulièrement du psoriasis, de la dermatite atopique ou de l’acné, alors que cet extrait ne contient pas de curcumine et notamment ne contient pas de curcuminoïdes.The object of the present invention is to meet the aforementioned needs. Les cellules sont trypsinisées, c’est-à-dire qu’on enlève le milieu de culture et on met une solution de trypsine pour les décoller, les cellules sont comptées et ensemencées dans des plaques noires à fond clair, de 96 puits à une densité de 50000 cellules par puits dans un volume de 100µl et incubées jusqu’au lendemain. Le milieu est retiré et les cellules sont lavées avec du PBS (Phosphate Buffer Saline – tampon phosphate salin) deux fois puis 70µl d’un composé à tester pour son activité antagoniste est ajouté, solubilisé dans le tampon d’essai (20mM HEPES-HBSS pH 7.4). Les cellules sont ensuite chargées avec le marqueur fluorescent calcique 5. Le colorant a été concentré 10 fois dans l’eau et ensuite dilué au 1/10 dans le tampon. La solution colorante est ajoutée dans les puits et incubée à température ambiante pendant 1 heure. Pour les tests d’antagoniste, le composé à tester est ajouté dans les puits et incubé à température ambiante pendant 10 minutes. Les plaques sont ensuite placées dans le FLIPR (lecteur de plaque d’imagerie par fluorescence) et la fluorescence est enregistrée aux longueurs d’ondes excitation/émission: 488nm/510-570nm. Après 20 secondes, l’agoniste (FPL 64176) (80µl solubilisé dans le tampon d’essai (20mM HEPES-HBSS pH 7,4)) est ajouté et la fluorescence est enregistrée aux mêmes longueurs d’ondes.
