Description
The present invention aims primarily to provide a new active to prevent the occurrence of dysaesthesic sensations.
The present invention relates to the use, in particular cosmetic use, of an effective amount of at least one long UV-A (or UVA1) filter and preferably at least one Merocyanine of formula (1) or (2) for preventing appearance of dysaesthetic sensations. In general, sensitized skin is defined by a particular reactivity of the skin. However, this reactivity is not an immunological process. Its mechanism is said to be nonspecific.
This cutaneous reactivity is characterized by the manifestation of signs of discomfort in response to the contacting of the subject with a triggering element that can have various origins. This may include exposure to air pollution.
It is clearly established that fine particles are one of the most harmful components of pollution. Those that result from combustion are particularly dangerous because they can have very small sizes and thus creep through the tissues. It has been shown that passage of ultrafine particles from the pulmonary alveoli to the bloodstream is possible (Elder et al., 2006, Clin Occup Environ Environ, 5, 797, Nemmar et al., 2014, Circulation, 105, 411). Moreover, it is known that the fine particles carry toxic molecules which are adsorbed on their surface and thus capable of being sold in a medium favorable to their desorption. Among these molecules, Polycyclic Aromatic Hydrocarbons or PAHs are the majority and can be represented by Benzo (a) pyrene (BaP).
Abramovits W et al. Department of Medicine, Baylor University Hospital and Texas A&M Medical School, 3500 Gaston Avenue, Dallas, TX 75246, USA).These signs of discomfort, which appear after contact with the subject triggering skin sensitization, such as BaP, are defined by dysaesthetic sensations. By dysaesthetic sensations, sensations felt in a cutaneous area such as tingling, tingling, itching or tightness. These subjective signs are characteristic of sensitized skin. They exist most often in the absence of visible signs such as redness and flaking. These dysaesthetic sensations are notably related to a low cellular survival of keratinocytes associated with an interleukin release. In particular proinflammatory mediator such as interleukin-la (IL-la). IL-1α secretion is known to correlate with the onset of dysaesthesic sensations such as tingling, tingling, itching, or tightness (Role of interleukin 1 in atopy dermatitis, Abramovits W et al Department of Medicine, Baylor University Hospital and Texas A & M Medical School, 3500 Gaston Avenue, Dallas, TX 75246, USA).
As previously stated, skin sensitization is not an immunological process triggered from an allergen and is reflected only by these sensations dysaesthetic.
For obvious reasons, the absence of visible signs makes it difficult to diagnose sensitive or sensitized skin. Most often this diagnosis is based on the interrogation of the patient. This symptomatology also has the advantage of making it possible to differentiate sensitive or sensitized skin from contact irritation or allergy for which, on the other hand, there are visible signs.
Consequently, the development of products intended to prevent skin sensitization necessitated the availability of tools for assessing the sensory response of the skin. The first tools were inspired from their conception of the essential characteristic of sensitive skin or sensitized namely the presence of signs of discomfort induced by a topical application.
Thus, the "stinging test" with lactic acid was the first test proposed. It is performed by recording tingling sensation reported by a volunteer after application of a lactic acid solution at 10% on the wings of the nose. Subjects reporting moderate or severe tingling sensations are referred to as "stingers" and are considered to have sensitized skin.
For the purposes of the present invention, sensitized skin is characterized by dysaesthesic sensations, tightness, tingling, itching and / or tingling that appear following contact with the skin to various factors such as exposure to air pollution. There is still a need for new active ingredients to reduce skin sensitization. There remains, therefore, a need for new assets that can prevent the appearance of dysesthetic sensations.
The object of the present invention is to satisfy these needs.
Surprisingly, the applicant has shown that long UV-A filters can meet these expectations.
Indeed the inventors have observed, unexpectedly that the long UV-A filters applied to the skin, allowed to avoid the appearance of dysesthetic sensation when it is in contact with pollution particles. Indeed, under the conditions of daily UV irradiation, under realistic conditions these pollution particles induce a significant death of keratinocytes associated with a secretion of interleukin pro-inflammatory, interleukin-la and these signs disappear in the presence long UV-A filter.
Thus, according to a first subject, the invention relates to the topical use of at least one long UV-A filter to prevent the occurrence of dysaesthetic sensations.
According to another of its aspects, the present invention relates to a long UV-A filter for its topical use as an active ingredient in a composition intended to prevent the appearance of dysaesthetic sensations.
According to another of its aspects, the present invention relates to a method, in particular a cosmetic method, for preventing the occurrence of dysaesthesic sensations, in particular as defined above, in an individual, comprising at least one step of topical administration to said individual. at least one long UV-A filter.
As specified below, the compositions containing it are formulated to be compatible with the topical mode of administration.
By "prevention" is meant according to the invention the fact of reducing the risk of occurrence or slowing down the occurrence of a given phenomenon, such as, for example, the appearance of dysaesthesic sensations, the death of keratinocytes and / or the secretion of It has, according to one aspect of the present invention.

EAU QSF 3. Photostabilité des formules solairesWATER QSF 3. Photostability of solar formulas
4. TraitementBefore doing the experiments on cells it is necessary to check the photostability of all the solar formulas used in the study. For this, each formula is spread as described above and then irradiated for 2 hours with an Oriel solar simulator equipped with a Xenon100OW bulb, a dichroic mirror and the UVJ filter. The UVJ filter is a WG320 (Schott) whose thickness is calculated so that the emitted light has a UVA / UVB ratio between 23 and 32 (Christiaens F et al., Photochem Photobiol 2005 Jul-Aug; 81 (4): 874-8). The transmission spectrum is measured with a dual monochromator spectroradiometer before and after irradiation (Macam). From Figure 1, the compositions used are photostable. 4. Treatment
After 48h, the culture medium is replaced by phosphate buffer (PBS) with or without benzapyrene (BaP) at concentrations of OnM, 6nM or 12nM. The plates are put back in the oven for 30min. After this time the 24-well plates are placed under the lens of the simulator, on top of which are placed the PMMA plates on which the solar formulas have been spread beforehand and the whole is irradiated with daily UV during 30, 60, 90 or 120min. In parallel, identically treated cells are kept in the dark. After irradiation, the PBS is replaced with fresh medium and the plates are put in an oven. 24h after, an MTT test is performed to evaluate cell survival.

En présence des compositions filtrantes la survie cellulaire est nettement améliorée avec un avantage marqué net pour la Composition 1 renfermant un filtre UVA long comparativement à la composition 2 ne renfermant pas de filtre UVA long.FIG. 5 shows the percentage of cell survival after increasing irradiation with daily UV in the presence of 6nM BaP and according to the compositions used. In the absence of filtration (Placebo Composition 3) after 30 minutes of irradiation, a significant decrease in survival appears (of the order of 50%) and increases with increasing irradiation time. In the presence of the filtering compositions the cell survival is markedly improved with a clear advantage for the Composition 1 containing a long UVA filter compared to the composition 2 does not contain a long UVA filter.
• BaP 12nM• BaP 12nM
En présence des compositions filtrantes la survie cellulaire est nettement améliorée avec un avantage très important avec la Composition 1 renfermant un filtre UVA long comparativement à la composition 2 ne renfermant pas de filtre UVA long.FIG. 6 shows the percentage of cell survival after increasing irradiation with daily UV in the presence of 12nM BaP and as a function of the compositions used. In the absence of filtration (Placebo Composition 3) after 30 minutes of irradiation more than 80% of the cells died. In the presence of the filtering compositions the cell survival is markedly improved with a very important advantage with the Composition 1 containing a long UVA filter compared to the composition 2 containing no long UVA filter.
CONCLUSIONCONCLUSION
The experimental results were carried out under realistic treatment conditions: UVA illuminations between 6 and 27J / cm 2 and UVJ 10 and 42 J / cm 2, and very small amounts of pollutant (BaP at 6 or 12 nm) . Ds demonstrate that under these conditions, a strong phototoxicity is manifested on keratinocytes in culture in the absence of long UVA filter. On the other hand, if we look at the difference in protection provided by the two solar formulas (composition 1 and composition 2), we can see that cell survival is more important under the formula of Composition 1. Analysis of transmission spectra both formulas highlight the UV zone between 370 and 400nm. Thus a better coverage of this part of the spectrum provides a very significantly higher protection.
