Abstract
Description
The present invention relates to the technical field of polymer surface modification methods, in particular methods adapted to microfluidic devices. In particular, the invention relates to a method of grafting a monolithic polymer on a substrate and microfluidic devices that can be obtained by such a method. It is increasingly sought miniaturized systems to perform liquid chromatography operations in particular, given their portable character, disposable, and the fact that they allow to perform analyzes in volumes of the order of nanoliter. However, the miniaturization of chromatographic techniques (separation technique based on the interaction of a solute with a stationary phase and a mobile phase) involves the synthesis of innovative stationary phases in microfluidic systems. The stationary phase used in such chromatographic systems may consist of a crosslinked polymer in the form of a porous monolith (K. Faure, Electrophoresis, 2010, 31, 2499-2511). Micro-fluidic devices chosen to contain these monolithic phases, which have channels most often 50-100pm in diameter and a few centimeters in length, must be chemically inert to be resistant to solvents. These are generally hydrocarbon plastics (containing only carbon and hydrogen) such as cyclic olefin copolymers (COC). Since the substrates used in the microfluidic devices are most often chemically inert, it is difficult to anchor the stationary phases on these substrates. However, such anchoring, preferably by covalent bonding, is essential to prevent the displacement of the monolith during the analyzes and preserve the rigidity and robustness of the device, but also the chromatographic performance. Already described in the prior art are photo-initiated anchoring methods which make it possible to locate the monolith in certain sections of the microfluidic device. The most frequently used technique employs activation of the surface of the substrate with benzophenone. US Pat. No. 7,431,888 describes, for example, a two-step process, with activation of the surface, then synthesis of the monolith, leading to the surface grafting of a first cross-linked polymer, then to the grafting on this polymer of the desired porous monolith.
However, this method has many disadvantages, such as the formation of homopolymers during irradiations of long duration, so that on the same principle of activation with benzophenone, the same group has developed a three-step process (J. Sep. Science 2007, 30, 1088-1093) in which the benzophenone, then a crosslinking monomer, and finally finally a mixture of monomer and initiator, are successively introduced. At first, a benzophenone solution (photoinitiator) is introduced into the polymeric micro-channels. Its UV irradiation initiates the abstraction of hydrogen on the plastic surface, thus generating a radical on the surface. Then, in a second step, a monomer acting as a crosslinking agent, often a multi-unsaturated monomer (diacrylate for example) is introduced. This reacts with a second UV irradiation with the surface-activated groups and thus covalently anchors to the wall in the form of a cross-linked homopolymer. The multi-unsaturated monomer being relatively short and the degree of crosslinking low, a certain proportion of polymerizable functions remain available for a new polymerization reaction. A reaction mixture for the synthesis of the monolith is then introduced into the microchannels and during a third UV irradiation for the polymerization of this monolith, the crosslinking process will include the polymerizable functions present on the intermediate homopolymer in the final structure of the monolith, resulting in a covalent bond between the stationary phase and the plastic surface. Nevertheless, these two techniques are problematic because of the use of benzophenone which is always ultimately present in a residual manner and makes it impossible to use the substrate obtained for the analysis of amines. In addition, the presence of an intermediate crosslinked polymer layer different in nature from that of the monolith contained in the volume of the channel reduces the chromatographic performance of the device obtained. Another technique (patent WO 2009/048933) consists in activating the surface with an electron beam, grafting an acrylate monomer having a photoreactive function, and reacting this photoreactive function with a reaction mixture based on acrylate monomer. The invention aims to overcome the disadvantages of the prior art, by providing a simpler, more efficient and more economical process.
In this context, the present invention provides a method of modifying the surface of a substrate consisting of a polymer having abstractible hydrogen atoms, in which the substrate is contacted under photoactivation with a mixture containing at least one multi-unsaturated vinyl monomer, a polymerization initiating agent, at least one porogenic agent, so as to carry out, simultaneously by photoactivation, an abstraction of hydrogen on the surface of the polymer by the initiating agent and a polymerization cross-linking of the monomer (s) present leading to the formation of a crosslinked polymer in the form of a porous monolith grafted by covalent bonding to the surface of the substrate.
In the context of the invention, a single reaction mixture is used, and a single irradiation is conducted to trigger both the polymerization and the anchoring. Unlike the previously employed technique which uses two or three reaction mixtures (benzophenone, bifunctional monomer, pre-monolithic mixture), two rinses (between the steps) and three UV irradiations, the process according to the invention is much simpler, more efficient and more economical. In the process according to the invention, the polymerization initiator also plays the role of hydrogen abstractor, this hydrogen abstraction leading to the formation of radicals on the surface of the substrate which react with the radicals formed in the mixture. reaction, which leads to both the grafting and the growth of the monolith formed by polymerization / crosslinking reaction. In addition, the process according to the invention does not use benzophenone.
Another advantage emerges from having only one type of polymer and not an intermediate layer of crosslinked polymer formed of bi-functional monomers located on the surface of the substrate, which subsequently has consequences when the substrate is used in chromatography, on the separative performance of the chromatographic system. The method according to the invention is perfectly suited for fixing stationary phases dedicated to liquid chromatography, but can also be used for any grafting of photoinitiated polymer on a polymeric surface, in particular to modify the chemical properties thereof (charge input, character hydrophilic, ...). The invention also relates to a microfluidic device comprising a substrate made of a polymer comprising abstractible hydrogen atoms in which at least one channel is arranged and on which a crosslinked polymer forming a porous monolith is grafted by covalent bond, directly on the surface of the channel, without intermediate polymeric layer and, in particular, a microfluidic device that can be obtained according to the process of the invention in all its variants of implementation. Such a device is free of benzophenone. Most often, the channel or channels formed in the substrate will have a cross section whose largest dimension (which corresponds to the diameter in the case of channels of circular section or the largest stride of the rectangle in the case of section channels rectangular) is in the range of 10 to 200 microns, preferably in the range of 50 to 100 microns, which dimensions are particularly suitable for miniaturized chromatographic systems. The invention also relates to the use of a device according to the invention in liquid chromatography. The methods and devices according to the invention will be detailed in the description which follows.
The process according to the invention is not limited to particular polymers with respect to the substrate. The only constraint is that the constituent polymer of the substrate must have abstractible hydrogens, that is to say hydrogens capable of being removed under photoactivation, in the presence of a hydrogen abstractor, to form radicals. By way of example, such abstractible hydrogen atoms are, for example, the activated secondary hydrogen atoms, that is to say in the alpha position with respect to a nitrogen atom (amine function) or oxygen (vinyl function, ether function) or tertiary or secondary hydrogen atoms. The chemical functions amines, vinyls, esters, alcohols, alkenes and alkanes may have such hydrogen atoms. The constituent polymer of the substrate is preferably a thermoplastic polymer, which provides molding facilities to obtain a suitable substrate. The constituent thermoplastic polymer of the substrate is, for example, chosen from poly (methyl methacrylates), poly (butyl methacrylates), polyolefins, cyclic olefin copolymers, polyethylenes, polypropylenes and poly (ethylene terephthalates). ), poly (butylene terephthalate), polyimides, polycarbonates and hydrogenated polystyrenes. In particular, a cyclic olefin copolymer, also called COC, may be used. The chemical formula of an exemplary cyclic olefin copolymer is given below, the stars indicating the presence of tertiary hydrogens easily abstractible: In the case of microfluidic devices, the shape of the substrate can be of any type . In particular, miniaturized substrates suitable for liquid chromatography will be preferred. In particular, it is advantageous that the channel or channels formed in the substrate which are most often rectangular in cross section have the largest side of their cross section which belongs to the range of 10 to 200 microns. Often the channels will have a closed cross-section, so that the light will have to pass through the substrate in order to be able to photo-initiate all or part of the channel. Preferably, the constituent polymer of the substrate will then be transparent to light in a wavelength range of from 200 to 400 nm, in particular in a range from 2.40 to 380 nm, the irradiations being generally at a length wavelength of 254 nm, 365 nm or 380 nm. Furthermore, for this type of chromatographic application, the constituent polymer of the substrate is preferably chemically inert with respect to the main solvents used in liquid chromatography. In particular, the constituent polymer of the substrate will not react with acetonitrile, methanol, tetrahydrofuran, 1-propanol, 2-propanol and acetone. Moreover, for this type of organic monolith synthesis, the constituent polymer of the substrate is preferably chemically inert with respect to the main solvents used during this synthesis. In particular, the constituent polymer of the substrate will not react with acetonitrile, alcohols, and in particular methanol, 1-propanol, 2-propanol, cyclohexanol and dodecanol. In the context of the invention, all the reagents necessary to form the desired polymeric monolith, namely the monomer (s), the pore-forming agent (s) and the initiating agent are introduced into the same reaction mixture with which the substrate will be contacted and subjected to photo-irradiation. The initiator agent is, for example, selected from methyl benzoin ether (BME), azobisisobutyronitrile (AIBN), benzoin ether derivatives, benzyl ketals derivatives, alkylphenones and thioxanthones. In the context of the invention, the initiator agent is used in amounts sufficient to allow both the abstraction of hydrogen and the radical polymerization leading to the porous monolith. The proportions of the reaction mixture will be adapted by those skilled in the art, so as to proceed simultaneously to the synthesis of the polymer and its anchoring by abstraction of hydrogen on the wall of the substrate. The amounts of initiator agent will be dependent on the initiator agent used. In most cases, the hydrogen abstraction anchoring and the monolith synthesis are carried out simultaneously if the photoinitiator content is greater than or equal to 0.50 / 0, preferably greater than or equal to 1%, in mass relative to the total mass of the monomer (s) used in the reaction mixture with which the substrate is brought into contact. In general, the amount of initiator agent used will be less than or equal to 10% by weight relative to the total weight of the monomer or monomers used. To improve the pressure resistance and electrochromatographic performance, the skilled person will adapt the photoinitiator content in the reaction mixture depending on the nature of the photoinitiator and the desired monolith. As an illustration, when the initiator agent is BME, it is preferably used in a proportion of 1 to 5%, and preferably about 2.5%, by weight relative to the total mass of the monomers. As an illustration, when the initiator agent is AIBN, it is preferably used in a proportion of 2 to 7%, and preferably about 2.5%, by weight relative to the total mass of the monomers. . For the other reagents necessary for the preparation of the polymeric monolith, reference may be made to the techniques already described in the prior art, in particular by Svec, F .; Frechet, J. M. 3. Anal. Chem. 1992, 64, 820 and J. Chrom. A 2010, 1217, 902-924 and in U.S. Patents 5,334,310; US 5,453,185; 5,728,457 which describe monolithic compositions for chromatographic columns and their method of preparation. The structure of the monoliths obtained is controlled, in particular by varying the composition of the mixture used for the polymerization and / or the polymerization time. Erg particular, monoliths obtained may be macro or microporous substrates or have a double micro / macro porosity.
The radical polymerization by photoactivation is, in particular, carried out with at least one vinyl monomer of valence greater than or equal to 4, called multi-unsaturated. The vinyl monomers of valence equal to 2 are named monounsaturated. "Monomer" means an entity which by a polymerization reaction leads to a covalent assembly repeated a large number of times and forms a macromolecule called polymer. The ethylenic monomers are those whose polymerization results from an addition reaction on an ethylenic double bond. The vinyl monomers are ethylenic monomers bearing a substituent other than an alkyl group. A mono-unsaturated vinyl monomer, also referred to as a valence monomer 2, has only one vinyl group capable of forming covalent bonds in the polymerization process.
A polyunsaturated vinyl monomer (especially valence 4, 6, ...) has several vinyl groups capable of forming covalent bonds in the polymerization process. For example, it is possible to use only a multi-unsaturated vinyl monomer, or a mixture of several multi-unsaturated vinyl monomers, or a mixture of at least one multi-unsaturated vinyl monomer and at least one mono vinyl monomer. -unsaturated. The multi-unsaturated vinyl monomers have at least 2 vinyl groups and act as a crosslinking agent, the polymerization process then leading to a three-dimensional network. Preferably, at least one of the monomers used carries a functional group. The polymer constituting the porous monolith will be, for example, a homopolymer of a multi-unsaturated vinyl monomer, or a copolymer of different multi-unsaturated vinyl monomers or a copolymer of multi-unsaturated vinyl monomers) and monounsaturated (s). The multi-unsaturated vinyl monomer or monomers are, for example, chosen from alkylene diacrylates, alkylene dimethacrylates, hydroxyalkylene diacrylates, hydroxyalkylene dimethacrylates, alkylene bisacrylamides, alkylene bismethacrylamides, in which the alkylene groups preferably comprise from 1 to 18 carbon atoms. carbon, oligoethylene glycol diacrylates, oligoethylene dimethacrylates, diallyl polycarboxylic acid esters, divinyl ethers, pentaerythritol di-, tri-, or tetraacrylates, pentaerythritol di-, tri-, or tetra methacrylates, trimethylopropane triacrylates, trimethylopropane trimethacrylates and phenylene diacrylates. In particular, the monolith can be obtained from a mixture of multi-unsaturated monomers chosen from 2,4 butanediol diacrylate, ethylene dimethacrylate, phenylene diacrylate, trimethylolpropane trimethacrylate, pentaerythritol triacrylate and pentaerythritol diacrylate monostearate. It is possible to use, in addition to the multi-unsaturated monomer (s), a monovinyl monomer bearing a functional group, in particular a hydrophilic, hydrophobic, ionizable or reactive group. By way of example of such monomers bearing a functional group, mention may be made of acrylic acid, methacrylic acid, acrylamide, methacrylamide, alkyl acrylamides, alkyl methacrylamides, alkyl acrylates and methacrylates, and perfluorinated alkyl acrylates. and perfluorinated alkyl methacrylates, hydroxyalkyl acrylates, hydroxyalkyl methacrylates, wherein the alkyl groups include, for example, from 1 to 18 carbon atoms, the vinylazlactones, oligoethyleneoxide acrylates, oligoethyleneoxide methacrylates, the acrylate and methacrylate derivatives which comprise a primary, secondary, tertiary or quaternary amine or zwitterion, and acrylate or methacrylate derivatives which include aromatic groups. More specific examples of monomers carrying a functional group are methyl acrylate, methyl methacrylate, butyl acrylate, butyl methacrylate, tert-butyl acrylate, tert-butyl methacrylate, 2-hydroxyethyl acrylate, hexyl acrylate, hexyl methacrylate, lauryl acrylate, lauryl methacrylate, 2-hydroxyethyl methacrylate, glycidyl acrylate, glycidyl methacrylate, 3-sulphopropyl acrylate, 3-sulphopropyl methacrylate, pentafluorophenyl acrylate, pentafluorophenyl methacrylate, 2,2,3,3,4,4,4-heptafluorobutyl acrylate, 2,2 , 3,3,4,4,4-heptafluorobutyl methacrylate, 1H, 1H-perfluorooctyl acrylate, 1H, 1H-perfluorooctyl methacrylate, acrylamide, methacrylamide, N-ethylacrylamide, N-isopropylacrylamide, N- [3- (dimethylamino) propyl] methacrylamide, 2-acrylamido-2-methyl-1-propanesulfonic acid, 2-acrylamidoglycolic acid, [2- (methacryloyloxy) ethyl] trimethylammonium chloride, [2- (methacryloyloxy) ethyl] dimethyl (3-sulfopropyl) ammonium hydroxide , 2-vinyl-4,4 dimethyl azlactone and naphthyl methacrylate. Preferably, the photoactivation polymerization is carried out between at least one acrylate or methacrylate monomer carrying a functional group and at least one multi-unsaturated acrylate or methacrylate monomer acting as a crosslinking agent.
A - 0% BME ; B - 0,5% BME ; C- 1.5% BME ; D - 2,5% BME. By way of example of acrylate or methacrylate monomer carrying a functional group that may be used, mention may be made of butyl acrylate, butyl methacrylate, hexyl acrylate, hexyl methacrylate, lauryl acrylate, lauryl methacrylate, octadecyl acrylate, octadecyl methacrylate, glycidyl acrylate glycidyl methacrylate, 2-acrylamido-2-methyl-1-propanesulfonic acid, [2- (methacryloyloxy) ethyl] trimethylammonium, and [2- (methacryloyloxy) ethyl] triethylammonium. By way of example of a multi-unsaturated acrylate or methacrylate monomer acting as a crosslinking agent that may be used, mention may be made of 1,4-butanediol diacrylate, ethylene dimethacrylate, trimethylolpropane trimethacrylate, pentaerythritol triacrylate and pentaerythritol diacrylate. monostearate. The polymerization is conducted in the presence of at least one porogenic agent, which confers porosity on the monolith obtained and makes it possible to obtain the desired porosity. The solvent (s) used, when solvents are used, may serve as a blowing agent. The pore-producing agent (s) used will be, for example, chosen from water, acetonitrile, alcohols, esters, ethers, ketones and water-soluble polymer solutions. Preferably, one or more porogenic agents will be chosen from water, acetonitrile, methanol, ethanol, 1-propanol, 1,4-butanediol, dodecanol and cyclohexanol. The pore-generating agent (s), and in particular the solvents used, may, for example, represent from 40 to 80% by weight, preferably from 50 to 70% by weight, relative to the total mass of the mixture used to carry out the polymerization. The% can be extremely variable from one monolith to another because the 3D structure depends on the solubility of the constituents and therefore the skilled person will adjust the% of the various constituents of the mixture to obtain an equilibrium and therefore a homogeneous structure. This adjustment will depend on the monomer (s) used and their relative proportions in the case of several monomers. The mixture used for the polymerization is, in general, a homogeneous mixture, in which the monomer or monomers used are soluble. The order of introduction of the reagents into the mixture used is of little importance, except for the initiating agent which will preferably be introduced last. The reaction mixture will generally comprise at least 10%, preferably at least 15% by weight of monomer (s). The polymerization is, in most cases, carried out under UV irradiation at a wavelength in the range from 200 nm to 400 nm, and more particularly in the range from 250 nm to 380 nm. The irradiation energy is, in general, 0.5 3 cm -2 to 9 3 cm -2, and more particularly 1 to 3 J.cm -2. The irradiation time is, for example, 5 to 60 min, preferably 10 to 30 min. The examples below, with reference to the appended figures, are given purely by way of illustration and make it possible to better understand the invention. FIG. 1 shows the evolution, as a function of the irradiation time, of surface contact angle measurements COC after irradiation of a mixture PEGMA-BME (FIG. 1A) and PEGMA-AIBN (FIG. 1B) containing 0, 03 M PEGMA in acetone at different percentages (by mass) of the photoinitiator BME or AIBN. Figure 2 shows transmission electron microscope (SEM) photographs of a section of the device prepared in Example 2, showing a monolith made in a COC microsystem with different percentages of photoinitiator. A - 0% BME; B - 0.5% BME; C-1.5% BME; D - 2.5% BME. Figure 3 shows the theoretical plateau heights of anthracene as a function of the separation electric field, on a HA-BDDA monolith synthesized with 0.5% BME and 2.5% BME prepared according to Example 3. Mobile phase 70/30 (v / v) ACN / water + 2mM NH4 H2 PO4 + 5mM LiDS. Direct injection ikV, 5s. Example 1 Grafting of Functional Molecules on the COC Substrate - Demonstration of Hydrogen Abstraction This example illustrates the ability of BME and AIBN to simultaneously abstract hydrogen and initiate photo-initiated polymerization. COC plates were grafted by irradiation of a mixture of polyethylene glycol methacrylate (PEGMA) and photoinitiator (BME or AIBN). The choice of Pegma is motivated by the fact that once polymerized, this monomer increases the hydrophilic character of the COC substrate. During this experiment, a solution of PEGMA (Acros Organics, Noisy-Le-Grand, France) with a concentration of 0.03 M in acetone is mixed with a certain percentage by mass of photoinitiator (AIBN or BME). The mass percentages tested with respect to the total mass of monomers are 0%; 0.5%; 2.5% and 5%. The mixture is deposited between two plates of COC (Microfluidic Chipshop,) ena, Germany) with a thickness of each imm, then irradiated at 355 nm under 5 UV tubes of 8W each (Biolink crosslinker, VWR, Strasbourg, France) with a energy corresponding to 0.95] / cm2 for every 10min. The plates are then separated and rinsed extensively with water and ethanol to remove all residual PEGMA. Then, the plates are placed in an ultrasonic bath so as to also remove any unanchored homopolymer, and then dried.
Since the COC is composed solely of carbon and hydrogen, any molecule remaining anchored to the substrate is necessarily bound by the result of a hydrogen abstraction. The PEGMA monomer being divalent, that is to say monounsaturated, it results from this experiment that only the copolymers whose initiation began by abstraction of hydrogen on the COC substrate remain present on the surface after this procedure. rinsing.
The amount of PEGMA copolymer present is therefore directly related to the importance of hydrogen abstraction in the polymer initiation process. Since PEGMA is a hydrophilic monomer and COC is a polymer known for its hydrophobic properties, the grafting rate of PEGMA on the COC can be monitored by contact angle measurement. These measurements were performed by depositing a drop of water and determining the angle using the Digidrop Contact Meter instrument (GBX, Bourg de Peage, France), at room temperature. Figure 1 shows the different results obtained for different irradiation times of the PEGMA-BME (Fig 1A) and PEGMA-AIBN (Fig 1B) mixtures. It is first noted that in the absence of photoinitiator, the presence of PEGMA monomer is not sufficient to induce its anchoring on the COC wall, whatever the duration of irradiation. Regardless of the initiator and its concentration, irradiation at 365 nm does initiate hydrogen abstraction followed by PEGMA anchoring. This anchoring is all the more important as the concentration is important, generating a greater number of anchoring points on the surface, to reach an optimum at 2.5% initiator. A concentration greater than 2.50 / 0 (% by weight based on the weight of monomer) seems to create a screening effect during the irradiation process and therefore a chaotic behavior. For a given concentration, the hydrogen abstraction and the ensuing anchoring increase with the duration of irradiation, until reaching a threshold. Note that when the AIBN, which has a lower photoinitiation efficiency than the BME, is used, it is necessary to irradiate 30 min (2.85 J / cm 2) to obtain maximum anchorage. This example not only demonstrates that BME and AIBN react on the surface of the COC by hydrogen abstraction, but also demonstrates a possible application which consists of chemically modifying the properties of the substrate by photoinitiation, which can be performed in a localized and controlled manner.
EXAMPLE 2 Simultaneous Synthesis and Anchoring of an Acrylate Monolith in a Microchannel of a COC Device This example illustrates the simultaneous synthesis and anchoring of a hexyl acrylate monolith in a cyclic olefin copolymer microsystem for the implementation of electrochromatography. A reaction mixture is prepared from a hydrophobic functional monomer, hexyl acrylate (HA), an anionic functional monomer, 2-acrylamido-2-methyl-1-propanesulfonic acid (AMPS), and a polyvinyl monomer, 1,3 butanediol diacrylate (BDDA). The monomer mixture is composed as follows: 65.4% w / w HA and 33.6% w / w of BDDA + 0.5% AMPS. The porogen consists of 54% acetonitrile, 23 ethanol, 23% water (% w / w). The monomer / porogen ratio is 34.5: 65.5 by weight. The photoinitiator is added at different percentages (by weight relative to the weight of the monomers): 0.5; 1.5; 2.5 and 3%. All reagents and solvents are from Sigma (Isle d'Abeau, France). The microsystem is manufactured by Microfiuidic Chipshop (Jena, Germany). It is cyclic olefin copolymer Topas brand. The structure is molded into a plate of imm thick and closed by a plate 130 pm thick. The channel has a dimension of 58.5mm x 100pm x 100pm. After dissolution of the various reagents and ultrasonic degassing, the reaction mixture is introduced by capillarity into the microsystem without any prior treatment. The device is then irradiated at 365 nm under 5 UV tubes of 8W each (Biolink crosslinker, VWR, Strasbourg, France) for 20 min, ie 1.9] / cm 2. The SEM photographs obtained are presented in FIG. 2 and show that the anchoring of the monolith, which is carried out by abstraction of hydrogen on the COC wall simultaneously with the synthesis, sees its importance depend on the quantity of initiator introduced into the mixture. reaction, the structure of the monolith and in particular the size of the nodules not being related to this amount of initiator. Without initiator, there is no anchoring or monolith synthesis (Fig 2A). As soon as a content of 0.50 / 0 is used, the anchoring appears on the SEM photographs (Fig 2B). The greater the amount of initiator, the greater the number of nodules interconnected between the wall and the monolith (Fig 2B to 2D). This also results in greater pressure resistance, a monolith anchored with 0.50 / 0 BME withstands 4-5 bar, while a monolith synthesized with 2.5% BME withstands a pressure of 30 bar . EXAMPLE 3 Comparison of Electrochromatographic Performances Anchoring by hydrogen abstraction and the synthesis of the monolith are carried out simultaneously when the photoinitiator content is greater than 0.50% (by weight relative to the weight of the monomers). However, to improve the pressure resistance and electrochromatographic performance, the skilled person will have to adapt the photoinitiator content in the reaction mixture depending on the nature of this photoinitiator and the desired monolith. In this example describing the synthesis of an acrylate monolith in a COC microsystem, it has been demonstrated that if 0.5% and 2.5% of photoinitiator lead to simultaneous anchoring and synthesis, the devices obtained with these two grades do not have the same characteristics.
The device is produced according to the protocol described in Example 2. After irradiation, it is rinsed by electroosmotic flow with a mobile phase 70% acetonitrile, 30% water + 2mM ammonium phosphate and 5 mM lithium dodecyl sulfate (LiDS) , under an electric field of 617 V / cm. Electric fields are applied using a voltage generator (Micralyne, Canada). The detection is carried out by fluorescence on a microscope (IX-71, Olympus, France) equipped with a 100W mercury lamp and a 330 nm excitation filter (collection above 400 nm) (XF02-2, Omega, USA). A CCD camera combined with a data analysis software (Alliance Vision, France) allows the collection and conversion of the fluorescence signal, while the chromatographic peaks are analyzed by the Azur software (Datalys, France).
The electrochromatographic performances are recorded in the form of the theoretical plateau height of the anthracene peak after injection of it by a protocol described in a previous publication (Ladner, Y. et al., J. Chrom A, 2010, 1217, 8001-8008) and presented in FIG. 4. The linear velocity of the mobile phase is connected by the electroosmosis phenomenon to the applied electric field. It is observed that the chromatographic performances are very much improved when the anchoring is carried out with a quantity of initiator of 2.5%. The efficiency is multiplied by 3 at the optimum speed of use and by 3.5 at the maximum achievable speed, which is limited by the electric field delivered by the generator. Example 4 - Comparative test and demonstration of the absence or presence of benzophenone The microsystem is manufactured by Microfluidic Chipshop (Jena, Germany). It is cyclic olefin copolymer Topas brand. The structure is molded in a plate 1mm thick and closed by a plate 130 pm thick. The channel has a dimension of 58.5 mm x 100pm x 100pm.
Device manufactured with a first step according to the prior art A first device is prepared with an intermediate layer according to a method similar to Stachowiak et al. (Electrophoresis 2003, 24, 3689-3693). A mixture containing 14000 acetone, 50 mg benzophenone and 600 μl 1,3 butanediol diacrylate is introduced into the microsystem and the whole is irradiated at 365 nm under 5 UV tubes of 8W each (Biolink crosslinker, VWR, Strasbourg, France) for 5min, ie 0.95 J / cm2. The system is then rinsed with ethanol and acetone using a syringe, to remove any residual reagent. Then the system is dried.
Device manufactured according to the invention The second device is prepared without the intermediate layer. It is simply rinsed with acetone and dried.
Then the next step is the same in both cases. The monomer mixture is composed as follows: 66.4% v / v HA and 33.6% v / v of BDDA + 0.50 / o AMPS. The porogen consists of 54% acetonitrile, 23% ethanol, 23% water (% v / v). The monomer / porogen ratio is 34.5: 65.5 by weight. The photoinitiator is added at 2.5% (by weight relative to the weight of the monomers). All reagents and solvents are from Sigma (Isle d'Abeau, France). After dissolution of the various reagents and ultrasonic degassing, the reaction mixture is introduced by capillarity into the two devices. The devices are then irradiated at 365 nm under 5 UV tubes of 8W each (Biolink crosslinker, VWR, Strasbourg, France) for 20 min, ie 1.9 3 / cm 2. The two devices are produced simultaneously and are then tested simultaneously according to protocol below, to highlight the presence or absence of benzophenone.
The general protocol is as follows: The device is first rinsed with a mobile phase consisting of 70% (v / v) acetonitrile, 30% (v / v) water + 2mM ammonium phosphate pH 6 + 5mM of lithium dodecylsulfate for 30min. The fluorescence emanating from the polymer is recorded under an excitation of 405 nm (t 20 nm) and a signal collection> 500 nm. After rinsing, the recorded fluorescence should be negligible. A solution of primary amine derived from naphthalene-2,3-dicarboxaldehyde (NDA) is prepared according to the following protocol: 1m1 of NDA (2.9mM in 50/50 acetonitrile / water) + 1ml of 5mM sodium tetraborate pH 9 + 1mL of primary amine imM. The derivation takes place in a few minutes during which the solution becomes yellow.
The solution is then diluted by 10 in the mobile phase (consisting of 70% (v / v) acetonitrile, 30% (v / v) water + 2mM ammonium phosphate pH 6 + 5mM lithium dodecylsulfate. NDA derivative amine is injected into the device in the form of a crenel, then the device is rinsed for 10 min with the mobile phase. In the case of device containing no benzophenone residues, which is the case of device according to the invention, the fluorescence returns to its initial level after rinsing.
In the case of devices whose monolith was anchored via a treatment with benzophenone, the residues present react with the amine-NDA derivative and the fluorescence after rinsing remains important. In the case where a doubt could remain, repeated injections followed by rinsing lead to a significant fluorescence in the presence of benzophenone, while a negligible fluorescence is observed in the case of a device developed according to the present invention. Test carried out and results: The rinsing with the mobile phase is carried out under an electric field of 683 V / cm for 30 min. Then the amine derivative, here histamine, is injected at 512 V / cm for 10s. Finally, the rinsing is carried out with the mobile phase described above, at 683 V / cm for 10 min. After a first fluorescence measurement, a second injection is performed, followed by rinsing before the second measurement. Table 1 summarizes the evolution of fluorescence relative to the initial fluorescence of the system. Table 1: Evolution of the relative fluorescence relative to the initial fluorescence Injection device with device without benzophenone benzophenone 1 + 173% + 2% 2 + 190% + 6% It is noted that the residual fluorescence after rinsing is very important in the device having been treated with benzophenone, while the device according to the invention has not received benzophenone during its synthesis is non-fluorescent after rinsing. The presence of benzophenone is therefore easily detected by this simple test to be carried out.
