INTRODUCTION
INTRODUCTION
The following report which describes the Patent for Invention refers to an unprecedented drying process for obtaining dry forms (powders) by spraying (nebulization) from aqueous colloidal suspensions of nanocapsules and polymer nanospheres. It is an interesting alternative technology for improving physicochemical stability with the maintenance and / or improvement of the properties of colloidal systems, as well as for the preparation of intermediate products for the production of compartmentalized pharmaceutical forms (capsules and tablets) or not (granules and powders).
Comme il s'agit de formes pharmaceutiques liquides, STATE OF THE ART
In recent decades, various drug support systems, such as liposomes, microesters, microcapsules and nanospheres, have been proposed in order to increase the efficacy and / or reduce the toxicity of certain drugs. Nanocapsules and nanospheres are aqueous polymeric suspensions, which have particles of submicrometric diameter, which differ only in the presence of oil in the formation of the former. As these are liquid dosage forms,

En accord avec la littérature, les nanocapsules d'indométacine sont stables pendant seulement 7 mois (Fessil et al. these aqueous suspensions may exhibit physicochemical instability resulting essentially from the sedimentation of particles or from the chemical degradation of components of the formulation, which constitutes a significant technological problem, since this limitation may compromise the industrial applicability of these pharmaceutical forms. Indeed, the aqueous suspensions of polymer nanocapsules have limited stability, which has already been demonstrated experimentally. In accordance with the literature, indomethacin nanocapsules are stable for only 7 months (Fessil et al.
A study has demonstrated the possibility of using nebulization to obtain a dry product (reconstituted in situ in the respiratory system), from a dispersion of liposomes for pulmonary administration (attached: Goldbach et al. 1993a; and al. 1993b). It should be emphasized that liposomes are drug carriers with a different structure from nanocapsules and nanospheres, the former being liquids prepared from phospholipids and the latter being made of a polymer.
PROBLEMS OF CURRENT METHODS
Methods available for drying liquid forms, leading to potentially more stable solid forms, include lyophilization and nebulization.
However, the high cost for the production of freeze-dried products, associated with the technological difficulties for the selection of cryoprotectors, and the destructuring of these nanocapsules by the low temperatures used are the main drawbacks of the freeze-drying method which justify the impracticability of its industrial application. posterior.
The nebulization is a drying operation of dispersed fluid systems through the division of these into fine goticles inside a drying tower provided with circulating hot air, producing fine dry powders, granules or agglomerates. . This technique has a large number of applications in the pharmaceutical industry and also in the production of excipients, in the formation of complexes, among others. Despite these applications, so far this technique has not yet been described for the drying of colloidal suspensions of nanocapsules or nanospheres.
One of the main advantages of nebulization is the extremely short contact time of the dispersed liquid to be nebulized with the heat source. This allows nebulization to be a technique for drying heat-sensitive products.
In lyophilization, the wide range of particle size distribution presented by the particles requires subsequent processing steps, such as trituration and classification. On the other hand, nebulization makes it possible to obtain powders with particle size characteristics better defined than freeze-dried products, apart from lower hygroscopicity and better flow characteristics. Other advantages of nebulization compared to freeze-drying include its lower cost and the ease of industrial transposition.

Despite the advantages and the various industrial uses of nebulization, so far this technique has not been proposed for the drying of polymer colloidal suspensions.
DESCRIPTION DU PROCÉDÉ
