Abstract
Description
Derivatives of 2-halophenyl morpholine, process for their preparation and their use in therapy
The present invention relates, as novel industrial products, to 2-phenyl-morpholine derivatives, namely more specifically 2-halophenyl-morpholine derivatives of type 2 (halophenyl) -4-alkyl-morpholine. It also relates to the use of these novel therapeutics, especially as CNS antidepressant agents. It also relates to their preparation process.

Chem. Chimica Therapeutica 11 (No. 3), pages 201-207 (1976), wherein said 2-phenyl-morpholine derivatives are proposed or contemplated as exciting agents, stimulants, tranquilizers, sedatives, anti-inflammatories, analgesics and / or or hypotensives.
L'article de N. BUSCH et al., précité, qui signale que (i) la nature du substituant sur l'atome d'azote en position 4 du cycle morpholinyle est un des éléments essentiels dans l'interaction substance- récepteur tryptaminergique, et Cii) la nature du substituant sur le cycle phényle "module" l'activité des dérivés de 2-phényl-morpholine, enseigne que, sur le plan des propriétés antidépressives, les composés ayant un groupe isopropyle en position 4 du cycle morpholinyle sont les plus efficaces, et que sur le cycle phényle le groupe CF3 donne une meilleure activité que les groupes Cl et F. The article by N. BUSCH et al., Supra, which states that (i) the nature of the substituent on the nitrogen atom at the 4-position of the morpholinyl ring is one of the essential elements in the tryptaminergic substance-receptor interaction, and Cii) the nature of the substituent on the phenyl ring "modulates" the activity of the 2-phenyl-morpholine derivatives, teaches that, in terms of antidepressant properties, the compounds having an isopropyl group at the 4-position of the morpholinyl ring are the more efficient, and that on the phenyl ring the CF3 group gives a better activity than Cl and F groups.
On vient de trouver de façon surprenante que les composés selon l'invention, qui sont structurellement différents des dérivés halogénés de 2phényl-morpholine antérieurement connus, sont thérapeutiquement plus intéressants contrairement à l'enseignement dudit article de N. BUSCH et al. It has surprisingly been found that the compounds according to the invention, which are structurally different from the previously known halogenated derivatives of 2-phenyl-morpholine, are therapeutically more advantageous, contrary to the teaching of the article by N. BUSCH et al.
d'addition,
c) la 2-(3-chlorophényl)-4- méthyl
morpholine et ses sels d'addition,
d) la 2-(3,4-dichlorophényl)-4- éthyl
morpholine et ses sels d'addition, et
e) la 2-(2-fluorophenyl)-4-ethyl-
uiorpholine et ses sels d'addition.The new 2-halophenyl-4-alkyl-morpholine derivatives according to the invention, which correspond to the general formula
where Ar is a halophenyl group and R is a C1-C2 alkyl group, are characterized in that they are selected from the group consisting of
a) 2- (4-chlorophenyl) -4-methyl
morpholine and its addition salts,
b) 2- (2-chlorophenyl) -4
methyl morpholine and its salts
addition,
c) 2- (3-chlorophenyl) -4-methyl
morpholine and its addition salts,
d) 2- (3,4-dichlorophenyl) -4-ethyl
morpholine and its addition salts, and
e) 2- (2-fluorophenyl) -4-ethyl-
uioropholine and its addition salts.
The term "addition salts" here means, on the one hand, the acid addition salts obtained by reaction of the free bases according to the invention with inorganic and organic acids and, on the other hand, 'ammonium. Among the acids that can be used for salifying said free bases, mention may in particular be made of hydrochloric, hydrobromic, acetic, formic, propionic, oxalic, fumaric, maleic, succinic, benzoic, cinnamic, mandelic, citric, malic, tartaric, aspartic and glutamic acids, methanesulfonic, p-toluenesulfonic. Among the compounds making it possible to obtain ammonium salts, mention may in particular be made of IC 3 and ClCH 3.
Acid addition salts are preferred to ammonium salts. Among the acid addition salts, the hydrochlorides, given in a nonlimiting manner in Table I below, are the most interesting. The preferred products according to the invention are those of Examples 2,4 and especially 5, as well as the corresponding free bases.
The compounds according to the invention can be prepared according to a method known per se by application of conventional reaction mechanisms.
The process of preparation which is recommended here is to cyclize a 2- (N-alkyl-Np-hydroxyethyl) amino-1- (halophenyl) -1-ethanol of formula
(where Ar and R are defined as above) using concentrated H 2 SO 4 at a temperature between
O and OC, for at least 1 hour.
Advantageously this cyclization will be carried out with concentrated sulfuric acid having a density of 1.84.
A. The total synthesis of the compounds according to the invention from a halophenacyl bromide of formula III and an N-p-hydroxyethyl-alkylamine INt, is shown schematically in diagram A.

Of course, in such a composition the active ingredient, which is selected from the set of free bases according to the invention, their non-toxic addition salts and mixtures thereof, occurs in a pharmaceutically effective amount.

III. INTERACTION AVEC L'APOMORPHINE.III. INTERACTION WITH APOMORPHINE.
Batches of 6 mice receive the
CRL 41 015 half an hour before the subcutaneous injection of a dose of 1 or 16 mg / kg of apomorphine. It is found that, at a high dose (32 mg / kg), the CRL 41 015 opposes the hypothermic action of apomorphine, without modifying the behavior of verticalization and stereotypies. It is also noted that at doses of 2 and 8 mg / kg, CRL 41 015 causes hypothermia (maximum 8 mg / kg) which then disappears with the increase in doses administered.
CRL 41 015 is administered to batches of 6 rats half an hour before the subcutaneous injection of 0.5 mg / kg of apomorphine. It is observed that CRL 41 015 leaves the stereotypies induced by apomorphine unchanged.
IV.
INTERACTION WITH AMPHETAMINE,
Amphetamine (2 mg / kg) is injected intraperitoneally to batches of 6 rats half an hour after the administration of CRL 41 015. It is observed that CRL 41 015 leaves unchanged amphetamine stereotypies.
V. INTERACTION AVEC LA RESERPINE.V. INTERACTION WITH THE RESERPINE.
Four hours after the intraperitoneal injection of 2.5 mg / kg reserpine, batches of 6 mice receive CRL 41 015 to observe any changes in temperature and ptosis. It is found that at the dose of 32 mg / kg, CRL 41 015 opposes hypothermia and ptosis reserpine.
VI.
INTERACTION WITH OXOTRENORINE

XII I. CONCLUSIONS. XII I. CONCLUSIONS.
The results of the neuropsychopharmacological tests make it possible to draw the following conclusions.
CRL 41 015 has, depending on the doses, biphasic effects on temperature (hypothermia observed before administration of the reagents on the tests of interactions with apomorphine or with oxotremorine) and on motility (hypomotility and, consequently, effects on motility after hypoxia and aggression).
The fugitive excitation observed at high doses (mice and rats) may be a reflection of reversal of effects; it may be possible to relate to it the antagonism of barbiturate sleep.
CRL 41 015 causes antidepressant effects in mice (antagonism of hypothermia caused by apomorphine, reserpine and oxotremorine, and decreased "despair" immobility).
These effects are accompanied by signs of peripheral postsynaptic o-adrenergic stimulation (antagonism of the reserpinic ptosis, salivation).
Briefly, the CRL 41 015 has, according to the doses used, sedative effects and antidepressant effects.

ESSAIS RELATIFS AUX AUTRES PRODUITS.TESTS RELATING TO OTHER PRODUCTS.
In brief, according to the operating procedures given above for CRL 41 015, it is found that CRL 40 919 (Example 1), CRL 40 923 (Example 2),
CRL 40,925 (Example 3) and CRL 41051 (Example 4) show sedative and antidepressant effects.
In particular, CRL 40 919, which has a DL-O greater than 128 mg / kg and less than 256 mg / kg ip, exacerbates the lethal effects of electroshock at a dose of 64 mg / kg. , decreases intergroup aggression, aggravates reserpine hypothermia, and causes in mice and rats, according to
The study of global behavior, hypothermia and hypomotility.
The neuropsychopharmacological study of CRL 40 923 reveals for this product
- a sedative component (sedation, hyporeactivity, hypothermia, hypomotility, decreased vitreous aggressiveness3,
an antidepressant component (antagonism of hypothermia induced by apomorphine, loxotremorine, or reserpine, and decreased duration of immobility in the behavioral despair test),
a postsynaptic Qc-adrenergic stimulation component (antagonism of the reserpinic ptosis, piloerection, salivation), and
- an effect on asphyxial anoxia which is a reflection of hyporeactivity, hypothermia and low anticonvulsant activity.
- des effets de type sédatif apparaissant plutôt chez le rat, ou aux faibles doses chez la souris sédation, hyporéactivité, hypothermie, hypomotilite, diminution de l'agressivité, potentialisation de l'effet hypnogène du barbital ou du pentobarbital, retard d'apparition des convulsions et de la mort apyres anoxie asphy xique (dû à la diminution de déperdition énergétique en rapport avec 1'hypothermie et l'hypomotilité);
être la conséquence d'une vasoconstriction terminale produite par une stimulation OC des vaisseaux de la queue;
- une composante sans doute anticholinergique, présente à toutes les doses chez le rat et à forte dose chez la souris, objectivée par une mydriase, un antagonisme des effets péripheriques, des tremble- ments et de l'hypothermie produits par l'oxotremorine;
The neuropsychopharmacological profile of CRL 40 925 has a complex entanglement of several types of effects, namely
stimulant effects appearing at a high dose, especially in the mouse, and is all the more obvious when the animal is under conditions of basal activity (excitation with presence of stereotyped movements, increase in reactivity, increase in spontaneous motor activity and recovery of motor activity in the mouse accustomed to its enclosure, improvement of motor recovery after hypobaric hypoxia, decrease in the immobility of "despair")
- sedative-type effects appearing rather in rats, or at low doses in mice sedation, hyporeactivity, hypothermia, hypomotility, reduced aggressiveness, potentiation of the hypnogenic effect of barbital or pentobarbital, delayed onset of apophysic convulsions and death anoxia asphyxia (due to the decrease in energy loss in relation to hypothermia and hypomotility);
- postsynaptic stimulant-like postsynaptic stimulant effects at high dose in mice: exophthalmia, tremor, salivation, mydriasis, antagonism of ptosis reserpine; ischemia of the tip of the tail, visible in high doses, may
be the consequence of a terminal vasoconstriction produced by an OC stimulation of the vessels of the tail;
- a probably anti-cholinergic component, present at all doses in the rat and at high dose in the mouse, objectified by a mydriasis, an antagonism of the peripheral effects, tremors and hypothermia produced by oxotremorine;
- an anti-convulsant effect vis-à-vis tonic seizures caused by electroshock.
(i) aux doses faibles, le CRL 41 051 provoque une diminution de l'activité motrice et une hypothermie (responsables de l'aggravation de la récupération motrice après hypoxie et du retard d'apparition des convulsions et de la mort aprds anoxie) ; (ii) aux doses plus élevées, cette hypothermie et cette hypomotilité deviennent moins importantes et peuvent laisser place à une hyperthermie ou a une hypermotilité (responsables de l'augmentation du nombre de passages punis au test des quatre plaques, de l'amélioration de la récupération motrice après hypoxie, et du raccourcissement du délai d'apparition des convulsions et de la mort après anoxie); ;
- un effet anti-convulsivant à dose élevé.CRL 41 051 also presents a complex neuropsychopharmacological profile,
antidepressant effects objectified by the antagonism of hypothermia induced by apomorphine, oxotremorine or reserpine; however, whereas at the level of interaction with apomorphine and oxotremorine, the intensity of the antagonism crott with the doses of CRL 41 051, one observes at 16 mg / kg a maximum of antagonism vis- against reserpine, then this antagonism decreases with increasing doses of CRL 41 051, to transform itself at a high dose into a potentiation of reserpine hypothermia,
- Bisphasic effects as a function of the doses in terms of motor activity and temperature:
(i) at low doses, CRL 41 051 causes a decrease in motor activity and hypothermia (responsible for worsening motor recovery after hypoxia and delayed onset of convulsions and death after anoxia); (ii) at higher doses, this hypothermia and hypomotility become less important and may give way to hyperthermia or hypermotility (responsible for increasing the number of passages punished by the four-plate test, improving motor recovery after hypoxia, and shortening of the onset of seizures and death after anoxia); ;
peripheral stimulatory -adrenergic effects revealed by the antagonism of the reserpinic ptosis and the presence of salivation
- an effect on the behavioral despair "(decrease of the duration of immobility) revealing in particular of an anti-depressive activity;
a high dose anti-convulsant effect.
Clinically, good results have been obtained in the treatment of depressive states in humans by administering a daily oral dose of 3-6 mg CRL 41015, this daily dose being divided into 2 to 3 doses per day, CRL 41015 being presented in the form of capsules or tablets.
