Description
Nouveaux dérivés de I-(aminophényl)-2-amino-l-éthanol, leur procédé de préparation et leur utilisation en thérapeutique.
La présente invention concerne, en tant que produits industriels, de nouveaux dérivés de I-(aminophényl)-2 amino-l-éthanol. Elle concerne également le procédé de préparation de ces nouveaux dérivés et leur utilisation en therapeutique notamment en tant qu'agents actifs sur le SNC (système nerveux central).
On sait que l'on a déjà proposé en tant que médicaments des composés appartenant à la famille des dérivés de 2-amino-I-pbényl-1-éthanol pour traiter les maladies du SNC et/ou du système cardiovasculaire. On connait, en particulier des résumés des CA 51, 5046i et 51, 5047c (1957), le dichlorhydrate de 1-(4-aminophényl-2-isopropylamino-1-éthanol. Il se trouve que ce produit, qui agit sur le SNC en tant qu'antidépresseur, présente un certain nombre d'inconvénients: il est toxique et a des effets secondaires défavorables : excitation et hyperréactivité.
Selon l'invention on préconise de nouveaux dérivés qui ne présentent pas les inconvénients rappelés ci-dessus, qui agissent en tant qu'antidépresseurs du SNC en manifestant des effets sédatifs et hyporéactifs, d'une part, et en tant que vasodilatateurs périphériques et hypotenseurs, d'autre part.
Les nouveaux dérivés selon l'invention sont caractérisés en ce qu'ils sont choisis parmi l'ensemble constitue par
a) les fumarate et aspartate de 1-(4-aminophenyl)-2-isopro-
pylamino-1-éthanol, et
b) les I-(aminophényl)-2-amino-l-éthanols répondant a la
formule générale
A - CHOH - CH2 - NH - R (I)
(où A représente un groupe 3-aminophényle, 3-amino-4
chlorophényle, 3-amino-4-methoxyphenyle, 4-amino-2
chlorophényle ou 4-amino-2-méthoxyphényle, et R représente
un groupe isopropyle ou tertiobutyle) et leurs sels
d'addition d'acide.
a) les fumarate et aspartate de 1-(4-aminophenyl)-2-isopro-
pylamino-1-éthanol, et
b) les I-(aminophényl)-2-amino-l-éthanols répondant a la
formule générale
A - CHOH - CH2 - NH - R (I)
(où A représente un groupe 3-aminophényle, 3-amino-4
chlorophényle, 3-amino-4-methoxyphenyle, 4-amino-2
chlorophényle ou 4-amino-2-méthoxyphényle, et R représente
un groupe isopropyle ou tertiobutyle) et leurs sels
d'addition d'acide.
Les composés selon l'invention peuvent etre préparés selon une méthode connue en soi par application de mécanismes réactionnels classiques. Le procédé que l'on préconise ici consiste à- soumettre un dérivé I-(nitrophényl) -2-amino-l- éthanol de formule A'-CHOH - CH2 - NH - R (Il) (ou A' est 4-nitrophényle, 3-nitrophényle, 3-nitro-4-chlorophényle, 4-nitro-2-chlorophényle, 3-nitro-4-méthoxyphényle, 4-nitro-2-méthoxy- phényle, et R est isopropyle Q'l tertiobutyle) à une réaction de réduction au moyen d'hydrogène en présence d'un catalyseur, le palladium sur charbon, cette réaction de réduction étant réalisée dans un alcool inférieur notamment le méthanol ou l'éthanol.
Si nécessaire, on peut séparer les iaomères optiques des composes selon l'invention, notamment selon une méthode classique de résolution.
Par sels d'addition d'acide on entend ici les sels d'addition obtenus par réaction de la base libre de formule I avec un acide minéral ou organique. Parmi les acides utilisables pour salifier les bases libres de formule I on peut notamment mentionner les acides chlorhydrique, bromhydrique, sulfurique, nitrique, phosphorique, formique, acétique, benzoïque, oxalique, tartrique, lactique, malique, citrique, fumarique, maléfique, méthanesulfonique, p-toluènesulfonique et aspartique. Les sels préférés des bases de formule I sont les chlorhydrate, fumarate, p-toluènesulfonate et aspartate.
Selon l'invention on préconise une composition thérapeutique qui est caractérisée en ce qu'elle renferme, en association avec un excipient physiologiquement acceptable, une quantité pharmaceutiquement efficace d'un dérivé de 1-(aminophenyl)- 2-amino-éthanol selon l'invention.
On a consigné de façon nullement limitative, dans le tableau I ci-après, les composés préférés selon l'invention, les plus intéressants sur le plan pharmacologique étant les produits des exemples 1 (CRL 40 823), 2 (CRL 40 823 A), 3 (CRL 40 892), 4 (CRL 40 821) et 5 (CRL 40 857).
On a également consigné dans le tableau II ci-après les intermédiaires de synthèse de formule II. Ces intermédiaires sont avantageusement préparés selon le schéma réactionnel suivant

préférence l'éthanol).

NaBH4. On agite encore 2 heures à OOC, acidifie avec 10 ml d'acide acétique, évapore à sec sous vide, reprend à l'eau, essore, sèche et recristallise dans le benzène. On obtient le produit attendu avec un rendement de 83 %. F = 84 - 850C.

Dans 100 ml d'une solution alcoolique à 25 % d'isopropylamine, on ajoute à 20 C, 12,3 g (0,05 mole) de 1-(4-nitrophenyl)-2-bromo-ethanol, apres 48 heures en contact, on évapore à sec sous vide, reprend avec HCl N, filtre sur charbons précipite avec NaOH concentrée, essore, sèche et recristallise dans l'éthanol pour recueillir le 1-(4-nitrophényl)-2-isopropylamino- 1-éthanol. F = 94 - 950C.
Dans 100 ml d'une solution alcoolique à 25 % d'isopropylamine, on ajoute à 20 C, 12,3 g (0,05 mole) de 1-(4-nitrophenyl)-2-bromo-ethanol, apres 48 heures en contact, on évapore à sec sous vide, reprend avec HCl N, filtre sur charbons précipite avec NaOH concentrée, essore, sèche et recristallise dans l'éthanol pour recueillir le 1-(4-nitrophényl)-2-isopropylamino- 1-éthanol. F = 94 - 950C.
On transforme la base libre mise en solution dans l'acétate d'éthyle en chlorhydrate, avec de l'alcool chlorhydrique.
On obtient le chlorhydrate attendu avec un rendement de 73 %.

c) CRL 40 823
On réduit sous atmosphère d'hydrogène 39,1 g (0,15 mole) de chlorhydrate de 1-(4-nitrophényl)-2-isopropylamino1-éthanol dans 400 ml de méthanol et 2 g de Pd/C à 10 %.

L'absorption d'hydrogène terminée, on filtre le Pd/C, évapore à sec sous vide, reprend le résidu avec 100 ml d'éthanol froid et essore le monochlorhydrate de 1-(4-aminophenyl)-2-isopropylamino-
méthanol (F = 156 - 1580C). Celui-ci en solution dans 500 ml d'eau est traité avec 50 ml de soude concentrée. On filtre le précipité ainsi obtenu que l'on lave avec 2 fois 25 ml d'eau froide puis sèche, pour obtenir le 1-(4-aminophenyl)-2-isopropyla- mino-1-éthanol. F = 136 - 1370C.

On maintient le chauffage jusqu'à dissolution complète, refroidit, amorce la cristallisation, essore, lave à l1éthanol, et recristallise dans le mélange méthanol-acétate d'éthyle 1 : 1 v/v) pour obtenir le
CRL 40 823 avec un rendement de 75 %. F = 142 - 143 OC.

1-éthanol

On prépare une solution aqueuse en mélangeant 19,4 g (0,1 mole) de 1-(4-aminophényl)-2-isopropylamino-1-éthanol et 13,3 g (0,1 mole) d'acide aspartique dans 100 ml d'eau. Cette solution est évaporée à sec sous vide, reprise 3 fois à l'éthanol et évaporée pour dessécher le résidu ; celui-ci repris avec 100 ml de méthanol est essoré et séché. On obtient le CRL 40 823 A avec un rendement de 98 %. F = 158 - 1590C.
Obtention de l'aspartate de 1-(4-aminophényl)-2-isopropylamino
1-éthanol

6,9 g (0,34 At-g) de magnésium sont mis à réagir avec 8 ml d'éthanol anhydre et 0,6 ml de tétrachlorure de carbone pour catalyser. En maintenant le mélange réactionnel à 35 - 40 C, on coule goutte à goutte la solution de 54,5 g (0,34 mole) de diéthylmalonate et 30 ml d'éthanol anhydre dans 50 ml d'éther anhydre, puis on ajoute 100 ml d'éther anhydre ; on maintient 4 heures à reflux et ajoute goutte à goutte une solution de 64 g (0,0285 mole) de chlorure de 4-nitro-2-chlorobenzoyle en solution dans 100 ml d'éther anhydre.

Le résidu d'évaporation est ensuite décarboxylé dans un mélange de 240 ml d'acide acétique, 56 ml d'eau et 12,5 ml de S04H2 concentré pendant 5 heures a reflux.
On évapore ensuite à sec, reprend à l'eau, essore, lave au bicarbonate de sodium dilué et à l'eau ; on sèche et recristallise dans le méthanol. On obtient 45,5 g (rendement : 75%) du produit attendu. F = 42 - 430C.
b) 4-nitro-2-chloro- D-bromoacetoEhenone
50 g (0,25 mole) de 4-nitro-2-chloroacétophénone en solution dans 300 ml de chloroforme, sont bromés par addition goutte à goutte de 15,25 ml (44 g) de brome à une température comprise entre 5 et 100C. Après une nuit au repos, on évapore le chloroforme sous vide, reprend le résidu à l'éther isopropylique, essore et recristallise dans l'éthanol. La bromo-cétone attendue est obtenue avec un rendement de 80 %. F = 64 - 650C.
50 g (0,25 mole) de 4-nitro-2-chloroacétophénone en solution dans 300 ml de chloroforme, sont bromés par addition goutte à goutte de 15,25 ml (44 g) de brome à une température comprise entre 5 et 100C. Après une nuit au repos, on évapore le chloroforme sous vide, reprend le résidu à l'éther isopropylique, essore et recristallise dans l'éthanol. La bromo-cétone attendue est obtenue avec un rendement de 80 %. F = 64 - 650C.
c) 1-(4-nitro-2-chlorophényl)-2-bromo-1-éthanol
50 g (0,18 mole) de la bromo-cétone précédente en solution dans 250 ml de méthanol, sont réduit à une température comprise entre - 100 et - 50C, avec 3,42 g (0,09 mole) de NaBH4 ajoutés par petites portions. On agite encore 2 heures à 0 C, acidifie avec 15 ml d'acide acétique, évapore à sec sous vide ; on reprend le résidu d'évaporation à l'eau, essore et lave à l'eau ; on sèche et recristallise dans l'éthanol pour obtenir le produit attendu avec un rendement de 81 %. F = 1020C.
50 g (0,18 mole) de la bromo-cétone précédente en solution dans 250 ml de méthanol, sont réduit à une température comprise entre - 100 et - 50C, avec 3,42 g (0,09 mole) de NaBH4 ajoutés par petites portions. On agite encore 2 heures à 0 C, acidifie avec 15 ml d'acide acétique, évapore à sec sous vide ; on reprend le résidu d'évaporation à l'eau, essore et lave à l'eau ; on sèche et recristallise dans l'éthanol pour obtenir le produit attendu avec un rendement de 81 %. F = 1020C.
d) Chlorhydrate du 1-(4-nitro-2-chlorophényl)-2-isopropylamino-1 méthanol
28 g (0,1 mole) du produit précédent sont ajoutés dans une solution froide de 40 ml d'îsopropylamine et 200 ml d'éthanol. Après 48 heures de contact à 200C et 3 heures à reflux, on évapore à sec sous vide, reprend à l'eau, essore, lave à l'eau et à l'éther isopropylique. On recristallise la base (F = 114 - 1150C) dans l'acétate d'éthyle. On transforme la base libre en chlorhydrate dans l'éthanol chlorhydrique et obtient celui-ci avec un rendement global de 40 %. F = 172 - 174 C avec décomposition.
28 g (0,1 mole) du produit précédent sont ajoutés dans une solution froide de 40 ml d'îsopropylamine et 200 ml d'éthanol. Après 48 heures de contact à 200C et 3 heures à reflux, on évapore à sec sous vide, reprend à l'eau, essore, lave à l'eau et à l'éther isopropylique. On recristallise la base (F = 114 - 1150C) dans l'acétate d'éthyle. On transforme la base libre en chlorhydrate dans l'éthanol chlorhydrique et obtient celui-ci avec un rendement global de 40 %. F = 172 - 174 C avec décomposition.
e) CRL 40 892
On réduit sous atmosphère d'hydrogène 8,85 g (0,03 mole) de chlorhydrate de 1-(4-nitro-2-chlorophényl)-2-isopropyl- amino-1-éthanol en solution dans 120 ml de méthanol et 10 ml d'éthanol chlorhydrique 8N en présence de 0,6 g de Pd/C ; on évapore à sec sous vide, reprend le résidu d'évaporation à l'eau, précipite la base avec NaOH concentrée, essore, lave à l'eau et à l'éther isopropylique. On obtient la base libre qui fond à 91 - 920C.
On réduit sous atmosphère d'hydrogène 8,85 g (0,03 mole) de chlorhydrate de 1-(4-nitro-2-chlorophényl)-2-isopropyl- amino-1-éthanol en solution dans 120 ml de méthanol et 10 ml d'éthanol chlorhydrique 8N en présence de 0,6 g de Pd/C ; on évapore à sec sous vide, reprend le résidu d'évaporation à l'eau, précipite la base avec NaOH concentrée, essore, lave à l'eau et à l'éther isopropylique. On obtient la base libre qui fond à 91 - 920C.
Cette base mise en solution dans l'éthanol est acidifiée avec un équivalent d'acide fumarique ; le fumarate est essoré et recristallisé dans l'éthanol. On obtient le CRL 40 892 avec un rendement de 66 % . F = 145 - 1460C.
HCl (Exemple 4 ; No de code : CRL 40 821) a) Chlorhydrate de 1-(3-nitrophényl)-2-tertiobutylamino-1-éthanol
Dans une solution à 200C de 80 ml de tertiobutylamine et 400 ml méthanol, on ajoute 49,6 g (0,2 mole) de 1-(3-nitrophényl-2-bromo-1-éthanol. Après 48 heures en contact à 20oC, on chauffe 3 heures à reflux, puis évapore à sec sous vide, reprend à l'acétate d'éthyle, lave 3 fois à l'eau, extrait avec
HCl (IN), précipite la solution acide avec NaOH concentrée, essore, sèche et recristallise dans l'acétate d'éthyle. On obtient la base libre (F. = 50 - 51"C) que l'on transforme en chlorhydrate avec de l'éthanol chlorhydrique ; on obtient celui-ci avec un rendement de 60 %.F = 2300C.
Dans une solution à 200C de 80 ml de tertiobutylamine et 400 ml méthanol, on ajoute 49,6 g (0,2 mole) de 1-(3-nitrophényl-2-bromo-1-éthanol. Après 48 heures en contact à 20oC, on chauffe 3 heures à reflux, puis évapore à sec sous vide, reprend à l'acétate d'éthyle, lave 3 fois à l'eau, extrait avec
HCl (IN), précipite la solution acide avec NaOH concentrée, essore, sèche et recristallise dans l'acétate d'éthyle. On obtient la base libre (F. = 50 - 51"C) que l'on transforme en chlorhydrate avec de l'éthanol chlorhydrique ; on obtient celui-ci avec un rendement de 60 %.F = 2300C.
b) Chlorhydrate de 1-(3-aminophényl)-2-tertiobutylamino-1-éthanol
On réduit sous atmosphère d'hydrogène, une solution de 16,5 g (0,08 mole) de chlorhydrate de 1-(3-nitrophényl)-2-tertiobutylamino-1-éthanol dans 175 ml de méthanol avec 1,5 g de Pd/C à 10 %. L'Absorption terminée, on filtre le Pd/C, évapore à sec sous vide et recristallise le résidu dans le mélange ethanol-acétate d'éthyle (1 . 1) v/v. On obtient le
CRL 40 821 avec un rendement de 54 Z. F = 178 - 1800C.
On réduit sous atmosphère d'hydrogène, une solution de 16,5 g (0,08 mole) de chlorhydrate de 1-(3-nitrophényl)-2-tertiobutylamino-1-éthanol dans 175 ml de méthanol avec 1,5 g de Pd/C à 10 %. L'Absorption terminée, on filtre le Pd/C, évapore à sec sous vide et recristallise le résidu dans le mélange ethanol-acétate d'éthyle (1 . 1) v/v. On obtient le
CRL 40 821 avec un rendement de 54 Z. F = 178 - 1800C.

28 g (0,1 mole) de C3 -bromo-4-chloro-3-nitroacétophénone (F = 90 - 910C) sont mis en suspension dans 160 ml de méthanol, puis soumis à une réduction à - 10 C et sous agitation au moyen de 1,9 g (0,05 mole) de NaBH4.Après une heure à OOC, on acidifie avec 10 ml de CH3COOH, évapore à sec sous vide, reprend le résidu d'évaporation à l'éther, lave à l'eau, HCl dilué, HC03Na dilué et à l'eau, sèche et évapore ; on obtient le produit attendu avec un rendement quantitatif. F = 35 - 360C.

F = 118- 1190C.

On obtient ainsi le dichlorhydrate monohydraté de 1-(3-amino-4-chlorophényl)-2-isopropylamino-1- éthanol (CRL 40 857) avec un rendement global (9tapes "a" à "c") de 58 %. F = 130 - 1310C.

L'antagonisme très net des hypothermies induites par l'apomorphine, la réserpine ou l'oxotrémorine permet sans conteste d'attribuer au CRL 40 823 des propriétés anti2- dépressives . Ces effets qui se produisent en l'absence de stimulation 3(-adrénergique et d'activité anticholinergique, rapporchent le CRL 40 823 des antidépresseurs du type stimulant /1-adré- nergique.
A ces effets antidépresseurs s > ajoute une composante sédative nette (sédation, hyporéactivité, hypomotilité, diminution de l'agressivité).
Par ailleurs, les essais cliniques chez l'homme ont permis de confirmer que le CRL 40 823 est un bon agent antidépresseur.

Après 18 heures de séjour dans les actimêtres, les souris (6 par dose, 12 témoins) reçoivent le CRL 40 892 . Elles sont aussitôt replacées dans leurs enceintes respectives et une demi-heure plus tard. on enregistre leur motilité pendant 30 minutes.

Le CRL 40 892 n'entraîne pas de modification du délai d'apparition des convulsions et de la mort consécutives à une anoxie asphyxique provoquée par un curarisant tel que le triiodoéthylate de gallamine.

CONCLUSION
Le CRL 40 892 présente les effets d'une activité antidépressive (antagonisme des hypothermies induites par l'apomor- phine, la réserpine ou l'oxotrémorine). Par ailleurs, le CRL 40 892 présente aux fortes doses chez la souris et le rat, une composante sédative (sédation, hyporéactivité, hypothermie, hypomotilité, diminution de l'agressivité) et un effet anticonvulsivant vis-à-vis de l'électrochoc.

on observe chez la souris une sédation et une hyporéactivité, et à la dose de 128 mg/kg I.P. une hypomotilité.
En ce qui concerne l'action sur le SNC, on constate que le CRL 40 821 antagonise I'hypothermie induite par l'apomorphine, la réserpine et l'oxotrémorine. Ceci dénote un effet antidépresseur.
L'absence de signes de stimulation sympathique du type Gz (salivation, piloérection, antagonisme du ptosis réserpinique) permet de différencier le CRL 40 821 des substances amphétaminiques et des agents antidépresseurs tricycliques. Enfin, le CRL 40 821 présente un effet antiagressif.
Par ailleurs, sur le système cardiovasculaire on observe chez le chien anesthésié (à partir de la dose de 10 mg/kg I.D.) et chez le rat éveillé spontanément hypertendu (à partir de la dose de 0,5 mg/kg P.O.) une action hypotensive nette.
Par ailleurs on note chez le chien un effet vasodilatateur périphérique notamment au niveau de l'artère fémorale et de l'artère vertébrale.
En clinique, le CRL 40 821 s'est avéré être un excellent agent hypotenseur chez l'homme

Aux doses de 128 mg/kg et 64 mg/kg I.P., chez la souris on observe une sédation et une hyporéactivité au toucher.
Le CRL 40 857 agit en tant qu'agent antidépresseur sur le SNC, car il s'oppose aux trois modèles d'hypothermie (apomorphine, réserpine et oxotrémorine). On observe egalement qu'il potentialise modérément les stéréotypies induites par l'amphétamine. Aux doses élevées, le CRL 40 857 provoque une hypomotilité, un effet antiagressif, et s'oppose aux effets convulsivants de l'électrochoc.
Par ailleurs, sur le système cardiovasculaire, on constate que le CRL 40 857, administré par voie I.V. chez le chien anesthésié, provoque (i) une vasodilatation fémorale importante à partir de la dose de 0,5 mg/kg (cet effet vasodilatateur est accompagné de tachycardie) et (ii) une hypotension modérée apparaissant à la dose de 10 mg/kg I.V.

CRL 40 827 a un effet hypotenseur net et durable (supérieur à 7 heures) à la dose de 20 mg/kg P.O. Cet effet se manifeste de façon moins intense a la dose de 10 mg/kg P.O.

Procédé de préparation d'un composé selon la revendication 1, caractérisé en ce que l'on soumet un dérivéI-(nitrophényl)-2-amino-1 -éthanol de formuleA'- CHOH - CH2 - NH - R (II) (où A' est 4-nitrophényle, 3-nitrophényle, 3-nitro-4-chlorophenyle, 4-nitro-2-chlorophényle, 3-nitro-4-méthoxyphényle, 4-nitro-2méthoxyphényle, et R est isopropyle ou tertiobutyle) à une réaction de réduction au moyen d'hydrogène en présence d'un catalyseur, le palladium sur charbon, cette réaction de réduction étant réalisée dans un alcool inférieur notamment le méthanol ou I 'éthanol.

Produit intermédiaire intervenant dans la synthèse d'un composé selon la revendication 1 lors de la mise en oeuvre du procédé de la revendication 10, ledit produit intermédiaire étant caractérisé en ce qu'il répond à la formule généraleA' - CHOH - CH2 - NH - R où A' est 4-nitro-2-chlorophenyle, 3-nitro-4-chlorophényle ou 3-nitro-4-méthoxyphényle, et R est CH(CH3)2 ou C(CH3)3.
