Abstract

PROCEDE DE PREPARATION D'UNE GLOBULINE ANTI-LYMPHOCYTES T A PARTIR DU SERUM D'ANIMAUX IMMUNISES. ON UTILISE POUR L'IMMUNISATION DES CELLULES DE LA LIGNE CELLULAIRE JM.

Description

La présente invention se rapporte à un procédé de prépa-
ration d'une globuline anti-lymphocytes T. On utilise des immunosuppresseurs à haute activité dans les transplantations d'organes pour empêcher le rejet par 1' organisme de l'organe transplanté et dans les transplanta- tions de moelle osseuse pour le traitement des maladies donneur-contre-hôte. Jusqu'à maintenant, on les préparait en injectant des cellules lymphatiques dans le corps d'un animal,
par exemple un cheval ou un lapin, et en recueillant la globu-
line anti-lymphocytes dans le sérum des animaux immunisés. L' inconvénient de ce procédé de préparation réside en ce que l'on utilise des cellules lymphatiques dont le prélèvement
pose des problèmes d'éthique et dont on ne dispose qu'en quan-
tités limitées.
L'invention vise à utiliser pourla préparation d'immuno-
suppresseurs des cellules cultivées in vitro.
La demanderesse a trouvé avec surprise que l'on pouvait
préparer un immunosuppresseur possédant une excellente activi-
té en utilisant pour l'immunisation des cellules de la ligne cellulaire JM. La préparation et les propriétés de la ligne cellulaire JM sont décrites dans la publication de U. Schneider, H.U. Schwenk et G. Bornkamm dans Int. J. Cancer, 19, 621-626 (1977). La ligne cellulaire JM est déposée sous le n0 I-147 à l'Institut Pasteur, Collection Nationale de Culture des Microorganismes (C.N.C.M.). Les cellules sont multipliables à
volonté in vitro sans perdre leurs propriétés.
Un autre but de l'invention réside dans la préparation in vitro d'une globuline anti-lymphocytes T. On y parvient
conformément à l'invention en hybridant des cellules de myélo-
me capables de synthétiser des anticorps, avec des lymphocytes B provenant d'êtres humains ou d'animaux et possédant une
spécificité contre l'antigène introduit sous la forme de cel-
lules de la ligne cellulaire JM ou de leurs produits. Une par-
tie des cellules hybrides formée dans ces conditions forme en continu des anticorps dirigés contre l'antigène. Ainsi donc, après séparation de ces cellules hybrides, il est possible de préparer in vitro de la globuline anti-lymphocytes T. La méthode pour l'hybridation cellulaire est décrite par exemple par D. Gdtze dans "Immunforschurg in Klinik und Labor",
wissenschaftliche Information, Fresenius-Stiftung, numéro spé-
cial Aout 1977, pages 5 et suivantes.
Les lymphocytes B immunisants sont de préférence recueil-
lis à partir du tissu lymphatique des souris.
L'exemple qui suit illustre l'invention sans toutefois
en limiter sa portée. Dans cet exemple, on décrit la prépara-
tion de globuline anti-thymocytes par ulisation des lymphoblas-
tes T de la ligne cellulaire JM.
EXEMPLE
La préparation consiste en les stades opératoires sui-
vants: -1. Centrifugation des cellules de lymphoblastes T 2. Réglage de la suspension cellulaire à une concentration en cellules de 30 x 106 cellules par ml à l'aide d'un milieu
cellulaire physiologique (CL 15).
3. Immunisation des lapins par administration intraveineuse
de 3 ml de suspension cellulaire dans la veine de l'oreil-
le. 4. Après une durée d'immunisation de 4 semaines, saignée des
animaux.
5. Récolte du sérum par centrifugation du sang.
6. Formation d'un lot de sérum des lapins après détermina-
tion du titre en anticorps et après les examens de stéri-
lité.
7. Absorption du sérum par des cellules humaines variées.
8. Purification de l'antisérum pour la fraction de globuline
par précipitation au sulfate d'ammonium suivie d'une chro-
matographie sur colonne.
9. Réglage à une teneur déterminée en protéines (20 mg/ml) de
la fraction d'immunoglobuline.
10. Ultra-centrifugation et filtration stérile de la globuline antithymocytes. 11. Introduction de l'immunoglobuline en ampoules et exécution
des contrôles finals (contrôles de la qualité, de la sté-
rilité et de la pyrogénécité).

Claims (4)

REVENDICATIONS
1. Procédé de préparation d'une globuline anti-lynphocytes T à partir du sérum d'animaux immunisés, caractérisé en ce que l'on utilise pour l'immunisation des cellules de la ligne cellulaire JM.
2. Procédé selon la revendication 1,caractérisé en ce que l'on recueille la globuline antilymphocytes T dans le
sérum de lapins immunisés.
3. Procédé selon la revendication 1, pour préparer une globuline antilymphocytes T à l'aide de cellules hybrides obtenues par hybridation de cellules de myélome aptes à la
synthèse d'anticorps, avec des lymphocytes B provenant d'ani-
maux ou d'êtres humains et possédant une spécificité contre un antigène introduit, caractérisé en ce que l'on utilise des cellules d'antigènes de la ligne cellulaire JM ou leurs produits.
4. Procédé selon la revendication 3, caractérisé en ce
que l'on utilise des lymphocytes B de souris.
