Abstract
Description
The subject of the present invention is medicaments constituted by new 2-aminoethanols of general formula I
non toxiques de ces derniers.where X is O, NH or pY-Ph-CH2-N, where Y means hydrogen,
methyl methoxy or chloro, as well as the addition salts
non-toxic of these.
The invention also extends to the manufacturing process
cation of the above products.
Schéma I:
The new 2-aminoethanols are obtained from
of the Piperonal in three diapres synthesis operations on
Diagram I below, when X is O or pY-Ph-CH2-N
Diagram I:
According to this scheme, the Piperonal undergoes the reaction
Grignard with methylmagnesium iodide
providing compound II in a single synthesis operation,
without the need to isolate the intermediate carbinol.
Obtaining halohydrin III (Z = Br, C1)
is carried out with the corresponding N-halosuccinimide
(NZS; Z = Br, C1) in dimethyl sulfoxide and water.
The reaction of III with the amines, which leads
to the products I forming the subject of the invention, takes place
in different reaction media. In the dissol
non polar, like toluene, the reaction is
slow although it flows at a speed sufficient to
make it practical; in polar solvents like
ethanol (at reflux) or dimethyl sulfoxide (at 500C) the reaction is favored.
Quand X est NH, l'obtention de I steffectue convenablement par hydrogénation catalytique de l'homologue benzylé, I (X = C6H5-CH2-N), d'après le Schéma 2
Schéma 2:
When X is NH, the production of I takes place suitably by catalytic hydrogenation of the benzylated homolog, I (X = C6H5-CH2-N), according to Scheme 2
Diagram 2:
This hydrogenation takes place under the ordinary conditions of this reaction, that is to say in dissolution of an alcohol, such as ethanol, and in the presence of a catalyst, such as palladium carbon.

EXEMPLE I : 3,4-méthylènedioxystyrène (TI)
Spectre IR (BrK), cm-1 ; 3470, 3330, 2980, 2920, 2830, 2700, 2660, 1490, 1475, 1435, 1240, 1030 920, 805. Compound I (X = C6H5-CH2-N, base) dissolved in ethanol is hydrogenated with Pd / C 10% until the theoretical amount of hydrogen has been absorbed. The catalyst is filtered, the ethanol is concentrated, it is taken up in water and extracted with methylene chloride. From the yellowish solid obtained, the dihydrochloride is formed by adding ethanolic solution of ClH (g) 11N to this solid dissolved in methylene chloride. After a few hours of cooling, it is filtered and dried. I1 present m.p. 162 C (d). IR spectrum (BrK), cm-1; 3470, 3330, 2980, 2920, 2830, 2700, 2660, 1490, 1475, 1435, 1240, 1030 920, 805.
