Abstract
A procedure is provided for the purification of a physiologically active protein substance that has antitumorigenic activity, which is induced by the administration to a rabbit of at least one substance which is able to stimulate the reticuloendothelial system and, afterwards, by injection into the rabbit of endotoxin from a Gram-negative bacterium. The procedure involves bringing a raw solution of said physiologically active protein substance into contact with a basic anion exchanger, in order to adsorb said physiologically active substance on the anion exchanger, elute the physiologically active substance adsorbed, and subject the eluate which contains said physiologically active substance to gel filtration, with a gel that is suitable for the separation of a substance with a molecular weight in the range from 30,000 to 70,000. The purified preparation of said physiologically active substance so obtained can be used as an antitumorigenic agent for the treatment of malignant tumours. Said physiologically active substance is isolated by subjecting the purified preparation successively to affinity chromatography on immobilized Cibacron Blue F3G-A, gel filtration, affinity chromatography on immobilized concanavalin A, preparative polyacrylamide gel electrophoresis, and gel filtration.
