Abstract

Procedimiento para el estudio de la activación funcional de leucocitos, plaquetas y otras células, producida in vivo o inducida in vitro, basada en la estabilización de proteínas de membrana citoplasmática y su detección mediante técnicas de citometría cuantitativa sin manipulación adicional de la muestra que comprende las etapas de: a) incubar la muestra de forma secuencial con un inhibidor o una mezcla de inhibidores específicos de proteasas y con una mezcla de anticuerpos conjugados directamente con fluorocromos; b) medir mediante citometría cuantitativa las emisiones fluorescentes de los fluorocromos unidos a las células a través de anticuerpos, c) analizar los resultados obtenidos mediante análisis multiparamétrico para identificar las subpoblaciones celulares de interés y dentro de ellas las que expresan las proteínas de membrana estabilizadas cuantificando su expresión de forma objetiva en base a la intensidad de fluorescencia detectada.
