Abstract
Die Erfindung betrifft eine neuartige Öl-in-Wasser-Nanoemulsion auf Pflanzenölbasis, die sich insbesondere für die Bereitstellung von fettlöslichen Substanzen eignet. Die Nanoemulsion besteht ausschließlich aus natürlichen Substanzen und eignet sich daher in besonderem Maße für die orale Applikation. Darüber hinaus zeichnet sich die Nanoemulsion durch eine hohe Langzeitstabilität und Lagerungs fähigkeit aus. Die Nanoemulsion lässt sich auf dem Gebiet der Pharmakologie oder Kosmetik sowie auch als Zusatz in Lebensmitteln verwenden. Ein Verfahren zur Herstellung der neuartigen Öl-in-Wasser-Nanoemulsion wird ebenfalls bereitgestellt.
Description
NEUE OL-IN-WASSER-NANOEMULSION
Die Erfindung betrifft eine neuartige Öl-in-Wasser-Nanoemul- sion auf Pflanzenölbasis, die sich insbesondere für die Be reitstellung von fettlöslichen Substanzen eignet. Die Nano- emulsion besteht ausschließlich aus natürlichen Substanzen und eignet sich daher in besonderem Maße für die orale Applikati on. Darüber hinaus zeichnet sich die Nanoemulsion durch eine hohe Langzeitstabilität und Lagerungsfähigkeit aus. Die Nano emulsion lässt sich auf dem Gebiet der Pharmakologie oder Kos metik sowie auch als Zusatz in Lebensmitteln verwenden. Ein Verfahren zur Herstellung der neuartigen Öl-in-Wasser-Nano- emulsion wird ebenfalls bereitgestellt.
HINTERGRUND DER ERFINDUNG
Als Nanoemulsionen werden in der Regel kinetisch stabile kol loidale Systeme bezeichnet, die eine Tröpfchengröße in der Größenordnung von 100 nm oder weniger aufweisen. Nanoemulsio nen weisen im Vergleich zu herkömmlichen Emulsionen verbesser te funktionelle Eigenschaften auf, die ihre Verwendung in vie len industriellen Bereichen attraktiv macht. So werden Nano emulsionen beispielsweise zur Verabreichung von lipophilen Substanzen in der Pharmakologie, in der Kosmetik und in der Lebensmittelindustrie eingesetzt.
Ein häufig beobachtetes Problem bei Nanoemulsionen ist ihre geringe Stabilität sowie ihre unzureichende Fähigkeit, lipophile Substanzen in ausreichender Menge bereitzustellen. Emulsionen, die in der Lage sind, hohe Mengen an lipophilen Substanzen zu verabreichen, erweisen sich in der Praxis häufig als verhältnismäßig instabil, sodass sie nur einige Wochen stabil gelagert werden können. Umgekehrt weisen stabile Emul sionen mit geringer Tröpfchengröße regelmäßig eine geringe Ka pazität bei der Beladung der Ölphase auf, sodass die Mengen
der zu verabreichenden Substanzen gering ist. Die Herstellung von Nanoemulsionen, die über ausgewogene Eigenschaften verfü gen, d.h. einerseits eine hohe Lagerstabilität zeigen und an dererseits über eine hohe Beladungskapazität verfügen, ist in der Praxis äußerst schwierig. Aufgabe der vorliegenden Erfin dung ist es daher, eine Nanoemulsion mit ausgewogenen Eigen schaften bereitzustellen, die sich über einen Zeitraum von mehreren Monaten oder sogar Jahren lagern lässt und sich gleichzeitig durch eine hohe Beladungskapazität der Ölphase auszeichnet. Die Nanoemulsion sollte gut verträglich und wenig toxisch sein, damit sie Anwendung in pharmazeutischen sowie auch in Lebensmitteln finden kann.
BESCHREIBUNG DER ERFINDUNG
Gemäß der vorliegenden Erfindung wird diese Aufgabe durch eine Auswahl von Bestandteilen gelöst, die zu Nanoemulsionen füh ren, welche auch bei Lagerung äußerst stabil sind, und darüber hinaus mit großen Mengen an lipophilen Substanzen beladen wer den können, die nach Verabreichung an einen Probanden mit ei ner hohen Bioverfügbarkeit an diesen bereitgestellt werden.
In einem ersten Aspekt betrifft die vorliegende Erfindung so mit eine Öl-in-Wasser-Nanoemulsion, wobei die Öl-in-Wasser- Nanoemulsion folgende Bestandteile umfasst:
(a) eine Ölphase, die ein Pflanzenöl umfasst;
(b) eine kontinuierliche Phase, die Wasser und Glycerin um fasst;
(c) einen Emulgator, der ausgewählt ist aus der Gruppe beste hend aus Lecithin, Mono- und Diglyceriden von Fettsäuren, Milchsäure und/oder Citronensäure und Mischungen davon;
(d) Ethanol; wobei die Nanoemulsion einen mittleren Teilchendurchmesser (Dm) von 70 nm oder weniger aufweist.
Als mittlerer Teilchendurchmesser (Dm) wird vorliegend der mittlere Durchmesser der in der kontinuierlichen Phase disper gierten Öltröpfchen verstanden. In einer Ausführungsform be trägt der mittlere Teilchendurchmesser der Nanoemulsion weni ger als 70 nm, vorzugsweise weniger als 65 nm, weniger als 60 nm, weniger als 55 nm, weniger als 50 nm, weniger als 45 nm, oder sogar weniger als 40 nm. So kann der mittlere Teilchen durchmesser der erfindungsgemäßen Nanoemulsion im Bereich von 40-70 nm liegen, wie z.B. im Bereich von 45-65 nm oder im Be reich von 50-60 nm.
Die Nanoemulsion der vorliegenden Erfindung umfasst eine Ölphase, die in einer kontinuierlichen wässrigen Phase disper giert ist. Die Ölphase umfasst erfindungsgemäß ein Pflanzenöl. Bei dem Pflanzenöl kann es sich um ein LCT-Pflanzenöl oder um ein MCT-Pflanzenöl handeln.
In einer besonders bevorzugten Ausführungsform ist das Pflan zenöl ein LCT-Pflanzenöl. Im Rahmen der vorliegenden Erfindung werden als LCT-Pflanzenöle solche Pflanzenöle angesehen, die zu 70% (w/w) oder mehr aus langkettigen Triglyceriden (long- chain triglycerides, LCT) bestehen, bezogen auf das Gesamtge wicht des Pflanzenöls. Als langkettige Triglyceride werden vorliegend Triglyceride verstanden, die mit Fettsäuren veres- tert sind, welche eine Kettenlänge von C14-C24 aufweisen. Zu diesen Fettsäuren zählen u.a. die gesättigten Fettsäuren Myristinsäure (C14), Palmitinsäure (C16), Stearinsäure (C18), Arachinsäure (C20), Behensäure (C22) und Lignocerinsäure (C24) sowie die ungesättigten Fettsäuren Palmitoleinsäure (C16), Öl säure (C18), Gadoleinsäure (C20) und Cetoleinsäure (C22).
Pflanzenöle, die zu 70% oder mehr aus langkettigen Triglyc eriden bestehen, umfassen unter anderem Sonnenblumenöl, Raps öl, Olivenöl, Leinöl, Maiskeimöl, Weizenkeimöl, Palmöl, Hanföl und Sojaöl.
In einer weiteren Ausführungsform ist das Pflanzenöl ein MCT- Pflanzenöl. Im Rahmen der vorliegenden Erfindung werden als MCT-Pflanzenöle solche Pflanzenöle angesehen, die zu weniger als 70% (w/w) aus langkettigen Triglyceriden bestehen, bezogen auf das Gesamtgewicht des Pflanzenöls. Diese Pflanzenöle ent halten im Gegensatz zu den LCT-Pflanzenölen einen hohen Anteil von mittelkettigen Triglyceriden. Als mittelkettige Triglyceride werden vorliegend Triglyceride verstanden, die mit Fettsäuren verestert sind, welche eine Kettenlänge von C6- C12 aufweisen. Zu diesen Fettsäuren zählen die gesättigten Fettsäuren Capronsäure (C6), Caprylsäure (C8), Caprinsäure (CIO) und Laurinsäure (C12). MCT-Pflanzenöle, die für die Durchführung der Erfindung verwendet werden können, weisen üb licherweise einen Anteil von mittelkettigen Triglyceriden von 20% (w/w) oder mehr, vorzugsweise 30% (w/w) oder mehr auf, be zogen auf das Gesamtgewicht des Pflanzenöls. Bevorzugte MCT- Pflanzenöle sind im Rahmen der Erfindung Kokosöl und Palmkernöl .
In einer Ausführungsform umfasst die Ölphase der erfindungsge mäßen Nanoemulsion Palmöl. In einer Ausführungsform umfasst die Ölphase der erfindungsgemäßen Nanoemulsion Sonnenblumenöl. In einer weiteren Ausführungsform umfasst die Ölphase der er findungsgemäßen Nanoemulsion Rapsöl. In einer noch weiteren Ausführungsform umfasst die Ölphase der erfindungsgemäßen Na noemulsion Olivenöl. In einer noch weiteren Ausführungsform umfasst die Ölphase der erfindungsgemäßen Nanoemulsion Leinöl. In einer noch weiteren Ausführungsform umfasst die Ölphase der erfindungsgemäßen Nanoemulsion Maiskeimöl. In einer noch wei teren Ausführungsform umfasst die Ölphase der erfindungsgemä-
ßen Nanoemulsion Weizenkeimöl. In einer noch weiteren Ausfüh- rungsform umfasst die Ölphase der erfindungsgemäßen Nanoemul sion Sojaöl. In einer noch weiteren Ausführungsform umfasst die Ölphase der erfindungsgemäßen Nanoemulsion Kokosöl. In ei ner noch weiteren Ausführungsform umfasst die Ölphase der er findungsgemäßen Nanoemulsion Palmkernöl.
Der Anteil des Pflanzenöls beträgt erfindungsgemäß zwischen 5% und 15% (w/w), bezogen auf das Gesamtgewicht der Emulsion. Das Pflanzenöl kann beispielsweise in einer Menge von mehr als 5% (w/w), mehr als 6% (w/w), mehr als 7% (w/w), mehr als 8% (w/w), mehr als 9% (w/w), mehr als 10% (w/w), on mehr als 11% (w/w), mehr als 12% (w/w), mehr als 13% (w/w) oder mehr als 14% (w/w) vorhanden sein. Besonders bevorzugt ist es, dass das Pflanzenöl in einer Menge von 5-10% (w/w) in der Emulsion vor handen ist, z.B. in einer Menge von 6-10% (w/w), 7-10% (w/w) oder 8-10% (w/w).
Die erfindungsgemäße Nanoemulsion umfasst neben der Ölphase eine kontinuierliche Phase, die neben anderen möglichen Be standteilen Wasser und Glycerin umfasst. Der Anteil an Glyce rin in der kontinuierlichen Phase kann je nach Einsatzgebiet der Emulsion variieren. Der Anteil an Glycerin beträgt jedoch erfindungsgemäß zwischen 30% und 70% (w/w), bezogen auf das Gesamtgewicht der Emulsion. In einer Ausführungsform der Er findung beträgt der Anteil an Glycerin in der erfindungsgemä ßen Nanoemulsion mindestens 30% (w/w), mindestens 35% (w/w), mindestens 40% (w/w), mindestens 45% (w/w), mindestens 50% (w/w), mindestens 55% (w/w), mindestens 60% (w/w) oder mindes tens 65% (w/w), bezogen auf das Gesamtgewicht der Emulsion. So kann der Anteil an Glycerin in der Nanoemulsion 35-70% (w/w), 40-70% (w/w), 45-70% (w/w), 50-70% (w/w), 55-70% (w/w) oder zwischen 60-70% (w/w) betragen, bezogen auf das Gesamtgewicht der Emulsion.
Die erfindungsgemäße Nanoemulsion umfasst ferner einen oder mehrere Emulgatoren, die die Herstellung der Nanoemulsion er leichtern und die Emulsion nach ihrer Herstellung stabilisie ren. Der oder die Emulgatoren wirken als grenzflächenaktive Substanzen, welche die Grenzflächenspannung an der Öl-Wasser- Phasengrenzfläche verringern. Der oder die verwendeten Emula toren sind erfindungsgemäß ausgewählt aus der Gruppe bestehend aus Lecithin sowie Mono- und Diglyceriden von Fettsäuren, Milchsäure und/oder Citronensäure und Mischungen davon. In ei ner bevorzugten Ausführungsform umfasst die Nanoemulsion Leci thin als Emulgator. In einer anderen bevorzugten Ausführungs form umfasst die Nanoemulsion Monoglyceride und/oder Diglyceride von Fettsäuren, Milchsäure und/oder Citronensäure oder Mischungen davon als Emulgator. Dies bedeutet, dass die Nanoemulsion Glycerin umfassen kann, welches einfach oder zweifach mit einer Säure verestert ist. Bei der Säure kann es sich um eine gesättigte oder ungesättigte Fettsäure, Milchsäu re und/oder Citronensäure handeln. Die Fettsäuren Linolsäure, Ölsäure, Stearinsäure sind besonders bevorzugt.
Die Nanoemulsion kann somit eine oder mehrere der folgenden Verbindungen als Emulgator umfassen: Glyceryl Citrat, Glyceryl Lactat, Glyceryl Linoleat, Glyceryl Oleat (Imwitor 948), Glyceryl Stearat (Imwitor 491), Glyceryl Lactat Citrat, Glyceryl Linoleat Citrat, Glyceryl Stearat Citrat (Imwitor 372P), Glyceryl Oleat Citrat, Glyceryl Linoleat Lactat, Glyceryl Stearat Lactat, Glyceryl Oleat Lactat, Glyceryl Oleat Linoleat, Glyceryl Stearat Linoleat, Glyceryl Oleat Stearat und Mischungen der genannten Verbindungen.
In einer bevorzugten Ausführungsform umfasst die Nanoemulsion Lecithin als einzigen Emulgator. In einer anderen bevorzugten Ausführungsform umfasst die Nanoemulsion Glyceryl Citrat als einzigen Emulgator. In einer noch anderen bevorzugten Ausfüh rungsform umfasst die Nanoemulsion Glyceryl Lactat als einzi-
gen Emulgator. In einer noch anderen bevorzugten Ausführungs form umfasst die Nanoemulsion Glyceryl Linoleat als einzigen Emulgator. In einer noch anderen bevorzugten Ausführungsform umfasst die Nanoemulsion Glyceryl Oleat als einzigen Emulga tor.
Es können jedoch in vorteilhafter Weise auch Mischungen dieser Emulgatoren eingesetzt werden. In einer besonders bevorzugten Ausführungsform handelt es sich bei dem Emulgator, der im Rah men der Herstellung der erfindungsgemäßen Nanoemulsion verwen det wird, um eine Mischung aus Mono- und Diglyceriden von Milchsäure, Citronensäure, Linolsäure und Ölsäure (Glyceryl Citrat/Lactat/Linoleat/Oleat ). Eine solche Mischung aus ver schiedenen Emulgatoren wird von mehreren Anbietern unter der Bezeichnung Imwitor 375 vertrieben. In einer besonders bevor zugten Ausführungsform handelt es sich somit bei dem in der Nanoemulsion vorhandenen Emulgator um Imwitor 375.
In einer weiteren bevorzugten Ausführungsform handelt es sich somit bei dem in der Nanoemulsion vorhandenen Emulgator um Glyceryl Stearat, das von mehreren Anbietern unter der Be zeichnung Imwitor 491 vertrieben wird. In einer noch weiteren bevorzugten Ausführungsform handelt es sich somit bei dem in der Nanoemulsion vorhandenen Emulgator um Glyceryl Stearat Citrat, das von mehreren Anbietern unter der Bezeichnung Imwitor 372 P vertrieben wird. In einer besonders bevorzugten Ausführungsform handelt es sich somit bei dem in der Nanoemul sion vorhandenen Emulgator um Glyceryl Oleat, das von mehreren Anbietern unter der Bezeichnung Imwitor 948 vertrieben wird.
Es ist erfindungsgemäß bevorzugt, dass die Gesamtmenge an Emulgator in der Nanoemulsion zwischen 2-10% (w/w), stärker bevorzugt zwischen 3-10% (w/w), 4-10% (w/w), 5-10% (w/w), 6- 10% (w/w), 7-10% (w/w) oder 8-10% (w/w) liegt, bezogen auf das Gesamtgewicht der Emulsion. So beträgt die Gesamtmenge an
Emulgator in der erfindungsgemäßen Nanoemulsion vorzugsweise mehr als 3% (w/w), mehr als 4% (w/w), mehr als 5% (w/w), mehr als 6% (w/w), mehr als 7% (w/w) oder mehr als 8% (w/w). Eine Gesamtmenge an Emulgator von 5-6% (w/w) ist vorliegend beson ders bevorzugt.
In einer Ausführungsform handelt es sich bei dem Emulgator in der Nanoemulsion um Lecithin, das in einer Gesamtmenge zwi schen 3-10% (w/w), 4-10% (w/w), 5-10% (w/w), 6-10% (w/w), 7- 10% (w/w) oder 8-10% (w/w) vorliegt, bezogen auf das Gesamtge wicht der Emulsion. Die Gesamtmenge an Lecithin in der Nano emulsion beträgt vorzugsweise mehr als 3% (w/w), mehr als 4% (w/w), mehr als 5% (w/w), mehr als 6% (w/w), mehr als 7% (w/w) oder mehr als 8% (w/w). Eine Gesamtmenge an Lecithin von 5-6% (w/w) ist vorliegend besonders bevorzugt.
In einer anderen Ausführungsform handelt es sich bei dem Emul gator in der Nanoemulsion um eine Mischung aus zwei oder meh reren der Verbindungen Glyceryl Citrat, Glyceryl Lactat, Glyceryl Linoleat, Glyceryl Oleat (Imwitor 948), Glyceryl Stearat (Imwitor 491), Glyceryl Lactat Citrat, Glyceryl Linoleat Citrat, Glyceryl Stearat Citrat (Imwitor 372P), Glyceryl Oleat Citrat, Glyceryl Linoleat Lactat, Glyceryl Stearat Lactat, Glyceryl Oleat Lactat, Glyceryl Oleat Linoleat, Glyceryl Stearat Linoleat und Glyceryl Oleat Stearat, wobei die Mischung in einer Gesamtmenge zwischen 3- 10% (w/w), 4-10% (w/w), 5-10% (w/w), 6-10% (w/w), 7-10% (w/w) oder 8-10% (w/w) in der Nanoemulsion vorliegt, bezogen auf das Gesamtgewicht der Emulsion. Die Mischung kann in einer Gesamt menge von mehr als 3% (w/w), mehr als 4% (w/w), mehr als 5% (w/w), mehr als 6% (w/w), mehr als 7% (w/w) oder mehr als 8% (w/w) vorliegen. Eine Gesamtmenge von 5-6% (w/w) der Mischung in der Emulsion ist vorliegend besonders bevorzugt, bezogen auf das Gesamtgewicht der Emulsion. Bei der Mischung handelt
es sich vorzugsweise um Imwitor 375 (Glyceryl Cit- rat/Lactat/Linoleat/Oleat ).
In einer anderen Ausführungsform handelt es sich bei dem Emul gator in der Nanoemulsion um eine Mischung aus Lecithin mit einer oder mehreren der Verbindungen Glyceryl Citrat, Glyceryl Lactat, Glyceryl Linoleat, Glyceryl Oleat (Imwitor 948), Glyceryl Stearat (Imwitor 491), Glyceryl Lactat Citrat, Glyceryl Linoleat Citrat, Glyceryl Stearat Citrat (Imwitor 372P), Glyceryl Oleat Citrat, Glyceryl Linoleat Lactat, Glyceryl Stearat Lactat, Glyceryl Oleat Lactat, Glyceryl Oleat Linoleat, Glyceryl Stearat Linoleat und Glyceryl Oleat Stearat, wobei die Mischung in einer Gesamtmenge zwischen 3- 10% (w/w), 4-10% (w/w), 5-10% (w/w), 6-10% (w/w), 7-10% (w/w) oder 8-10% (w/w) in der Nanoemulsion vorliegt, bezogen auf das Gesamtgewicht der Emulsion. Die Mischung aus Lecithin mit ei ner oder mehreren der oben genannten Glycerinester kann in ei ner Gesamtmenge von mehr als 3% (w/w), mehr als 4% (w/w), mehr als 5% (w/w), mehr als 6% (w/w), mehr als 7% (w/w) oder mehr als 8% (w/w) vorliegen. Eine Gesamtmenge von 5-6% (w/w) der Mischung in der Emulsion ist vorliegend besonders bevorzugt, bezogen auf das Gesamtgewicht der Emulsion. Bei der Mischung handelt es sich vorzugsweise um eine Mischung aus Lecithin und Imwitor 375 (Glyceryl Citrat/Lactat/Linoleat/Oleat).
Als weiteren Bestandteil umfasst die erfindungsgemäße Nano emulsion Ethanol. Das Ethanol ist vorzugsweise in der Nano emulsion in einer Menge von 2-20% (w/w) vorhanden. Das Ethanol kann beispielsweise in einer Menge von mehr als 2% (w/w), mehr als 3% (w/w), mehr als 4% (w/w), mehr als 5% (w/w), mehr als 6% (w/w), mehr als 7% (w/w), mehr als 8% (w/w), mehr als 9% (w/w), mehr als 10% (w/w), mehr als 11% (w/w), mehr als 12% (w/w), mehr als 13% (w/w), mehr als 14% (w/w), mehr als 15% (w/w), mehr als 16% (w/w), mehr als 17% (w/w), mehr als 18% (w/w) oder mehr als 19% (w/w) vorhanden sein. Besonders bevor-
zugt ist es, dass das Ethanol in einer Menge von 5-10% (w/w) in der Emulsion vorhanden ist, z.B. in einer Menge von 6-10% (w/w), in einer Menge von 7-10% (w/w) oder in einer Menge von 8-10% (w/w).
In einer Ausführungsform umfasst die Ölphase der erfindungs gemäßen Nanoemulsion eine schwer wasserlösliche oder wasserun lösliche lipophile Verbindung, die nach Aufnahme der Nanoemul sion freigesetzt werden soll. Die schwer wasserlösliche oder wasserunlösliche lipophile Verbindung ist vorzugsweise ausge wählt aus der Gruppe bestehend aus Cannabidiol (CBD), Cannabigerol (CBG), Cannabichromen (CBC), Cannabinol (CBN), Tetrahydrocannabinol (THC), Melatonin, Resveratrol, Asta- xanthin, Coenzym Q10, Vitamin A, Vitamin C, Vitamin E, Vitamin D3, Vitamin K2, Flavonoide, Glutathion, ß-Caryophyllen, 2- Arachidonylglycerol (2-AG), Palmitoylethanolamid (PEA), Anand- amid, Oleoylethanolamid (OEA) und oligomeren Proanthocyan- idinen (OPC). In einer besonders bevorzugten Ausführungsform umfasst die Ölphase der erfindungsgemäßen Nanoemulsion Cannabidiol (CBD), das nach Aufnahme oder Verabreichung der Nanoemulsion an einen Probanden oder Patienten freigesetzt wird.
Neben den oben ausführlich beschriebenen Bestandteilen kann die erfindungsgemäße Nanoemulsion weitere optionale Bestand teile umfassen, je nach Einsatzgebiet der jeweiligen Emulsion. So ist es in bestimmten Ausführungsformen vorteilhaft, wenn die Ölphase ein ätherisches Öl umfasst. Dieser Bestandteil kann einerseits als Konservierungsmittel oder Antioxidations mittel dienen. Andererseits kann das ätherische Öl den Ge schmack einer für die orale Verabreichung vorgesehenen Nano emulsion beeinflussen sowie auch das Lösungsverhalten der üb rigen Bestandteile. Geeignete ätherische Öle umfassen bei spielsweise Orangenschalenöl, Zitronenöl und Pfefferminzöl.
Das ätherische Öl wird erfindungsgemäß in einer Menge von 0,5- 5% (w/w) eingesetzt,
Darüber hinaus kann die erfindungsgemäße Nanoemulsion Substan zen umfassen, welche die Bioverfügbarkeit der zu verabreichen den lipophilen Verbindung verstärken. Geeignete Substanzen zur Erhöhung der Bioverfügbarkeit sind gemäß einer besonders be vorzugten Ausführungsform ausgewählt aus der Gruppe bestehend aus Piperin, Curcumin, Resveratrol, Quercetin, Menthol, Narin gin, Bergamottin, Kaempferol und Rutin.
Die Ölphase der erfindungsgemäßen Nanoemulsion kann ferner Öl säure und/oder Ethyloleat umfassen. Ölsäure und Ethyloleat können in der Nanoemulsion als Co-Emulgatoren oder Lösungsmit tel agieren. Ferner können Sie die Reifung (Ripening) der Na noemulsion verhindern und/oder verzögern. Ölsäure und Ethyloleat können in einer Menge von insgesamt 0,5-10% (w/w) in der Nanoemulsion vorhanden sein.
Darüber hinaus kann die erfindungsgemäße Nanoemulsion einen oder mehrere Zuckerester umfassen. Bei dem Zuckerester kann es sich um einen Saccharoseester handeln. Geeignete Saccharoseester, die sich als Zusatz für die erfindungsgemäßen Nanoemulsionen eignen, umfassen unter anderem Saccharose- stearat, Saccharosepalmitate, Saccharosemyristate und/oder Saccharoselaurate. Der oder die Zuckerester können in einer Menge von insgesamt 0,5-3% (w/w) in der Nanoemulsion vorhanden sein. In einer besonders bevorzugten Ausführungsform handelt es sich bei dem Saccharoseester um Saccharosestearat der fol genden Struktur:
Die erfindungsgemäße Nanoemulsion weist den besonderen Vorteil auf, dass bei ihrer Herstellung lediglich auf rein natürliche Verbindungen zurückgegriffen werden kann. So lassen sich er- findungsgemäß auch Nanoemulsionen mit den besonderen, vorteil haften Eigenschaften hersteilen, ohne dabei auf ethoxylierte Verbindungen zurückgreifen zu müssen. In einer bevorzugten Ausführungsform enthält die erfindungsgemäße Nanoemulsion nur eine minimale Menge an ethoxylierten Verbindungen, wie z.B. ethoxylierte Glycerylfettsäureester oder Sorbitanfettsäure- ester, wie z.B. Polysorbat 80. In einer besonders bevorzugten Ausführungsform ist die Gesamtmenge an ethoxylierten Verbin dungen in der erfindungsgemäßen Nanoemulsion kleiner als 0,5% (w/w), stärker bevorzugt kleiner als 0,4% (w/w), kleiner als 0,3% (w/w), kleiner als 0,2% (w/w), kleiner als 0,1% (w/w), kleiner als 0,09% (w/w), kleiner als 0,08% (w/w), kleiner als 0,07% (w/w), kleiner als 0,06% (w/w), kleiner als 0,05% (w/w), kleiner als 0,04% (w/w), kleiner als 0,03% (w/w), kleiner als 0,02% (w/w), kleiner als 0,01% (w/w), kleiner als 0,005% (w/w) oder kleiner als 0,001% (w/w). Besonders bevorzugt ist es er findungsgemäß, dass die vorliegend beschriebene Nanoemulsion keine ethoxylierten Verbindungen, wie z.B. ethoxylierte Glycerylfettsäureester oder Sorbitanfettsäureester, enthält.
Die Nanoemulsion der vorliegenden Erfindung zeichnet sich durch eine besonders hohe Lagertähigkeit aus. Dies bedeutet, dass sich die mittlere Teilchengröße der Nanoemulsion auch nach längerer Lagerung nicht signifikant verändert. Erfin dungsgemäß ist bevorzugt, dass sich die mittlere Teilchengröße der Nanoemulsion nach einer Lagerung für 2 Monate, stärker be-
vorzugt 4 Monate, noch stärker bevorzugt 6 Monate, eine Größe von 100 nm nicht überschreitet. Die Lagerung findet vorzugs weise in einem Temperaturbereich von 20-24°C statt.
In einer Ausführungsform weist die erfindungsgemäße Nanoemul- sion nach 2 Monaten Lagerung bei einer Temperatur von 20-24°C eine mittlere Teilchengröße von 100 nm oder weniger auf, und vorzugsweise 80 nm oder weniger. In einer weiteren Ausfüh rungsform weist die erfindungsgemäße Nanoemulsion nach 4 Mona ten Lagerung bei einer Temperatur von 20-24°C eine mittlere Teilchengröße von 100 nm oder weniger auf, und vorzugsweise 80 nm oder weniger. In einer noch weiteren Ausführungsform weist die erfindungsgemäße Nanoemulsion nach 6 Monaten Lagerung bei einer Temperatur von 20-24°C eine mittlere Teilchengröße von 100 nm oder weniger auf, und vorzugsweise 80 nm oder weniger.
Besonders bevorzugte Nanoemulsionen der vorliegenden Erfindung werden nachfolgend beispielhaft aufgeführt. So betrifft die vorliegende Erfindung in einer Ausführungsform eine Öl-in- Wasser-Nanoemulsion, die folgende Bestandteile umfasst:
(a) eine Ölphase, die ein Pflanzenöl umfasst, welches mittelkettige und/oder langkettige Triglyceride umfasst, wobei das Pflanzenöl in einer Menge von 5-15% (w/w) ent halten ist, bezogen auf das Gesamtgewicht der Emulsion;
(b) eine kontinuierliche Phase, die Wasser und Glycerin um fasst, wobei Glycerin in einer Menge von 30-70% (w/w) ent halten ist, bezogen auf das Gesamtgewicht der Emulsion;
(c) einen Emulgator, der ausgewählt ist aus der Gruppe beste hend aus Lecithin, Mono- und Diglyceriden von Fettsäuren, Milchsäure und/oder Citronensäure und Mischungen davon, wobei der Emulgator in einer Menge von 2-10% (w/w) enthal ten ist, bezogen auf das Gesamtgewicht der Emulsion;
(d) Ethanol, das in einer Menge von 2-20% (w/w) enthalten ist, bezogen auf das Gesamtgewicht der Emulsion; wobei die Nanoemulsion einen mittleren Teilchendurchmesser (Dm) von 70 nm oder weniger aufweist.
In einer anderen Ausführungsform betrifft die vorliegende Er findung eine Öl-in-Wasser-Nanoemulsion, die folgende Bestand teile umfasst:
(a) eine Ölphase, die ein Pflanzenöl umfasst, welches mittelkettige und/oder langkettige Triglyceride umfasst, wobei das Pflanzenöl in einer Menge von 5-8% (w/w) enthal ten ist, bezogen auf das Gesamtgewicht der Emulsion;
(b) eine kontinuierliche Phase, die Wasser und Glycerin um fasst, wobei Glycerin in einer Menge von 50-70% (w/w) ent halten ist, bezogen auf das Gesamtgewicht der Emulsion;
(c) einen Emulgator, der ausgewählt ist aus der Gruppe beste hend aus Lecithin, Mono- und Diglyceriden von Fettsäuren, Milchsäure und/oder Citronensäure und Mischungen davon, wobei der Emulgator in einer Menge von 5-8% (w/w) enthal ten ist, bezogen auf das Gesamtgewicht der Emulsion;
(d) Ethanol, das in einer Menge von 5-10% (w/w) enthalten ist, bezogen auf das Gesamtgewicht der Emulsion; wobei die Nanoemulsion einen mittleren Teilchendurchmesser (Dm) von 70 nm oder weniger aufweist.
In einer anderen Ausführungsform betrifft die vorliegende Er findung eine Öl-in-Wasser-Nanoemulsion, die folgende Bestand teile umfasst:
(a) eine Ölphase, die ein oder mehrere Pflanzenöle ausgewählt aus der Gruppe bestehend aus Palmöl, Sonnenblumenöl, Oli venöl und Sojaöl umfasst;
(b) eine kontinuierliche Phase, die Wasser und Glycerin um fasst, wobei Glycerin in einer Menge von 50-70% (w/w) ent halten ist, bezogen auf das Gesamtgewicht der Emulsion;
(c) einen Emulgator, wobei es sich bei dem Emulgator um Leci thin handelt, welches in einer Menge von 5-8% (w/w) ent halten ist, bezogen auf das Gesamtgewicht der Emulsion;
(d) Ethanol, das in einer Menge von 5-10% (w/w) enthalten ist, bezogen auf das Gesamtgewicht der Emulsion; wobei die Nanoemulsion einen mittleren Teilchendurchmesser (Dm) von 70 nm oder weniger aufweist.
In einem weiteren Aspekt betrifft die vorliegende Erfindung ein Verfahren zur Herstellung einer wie oben beschriebenen Na noemulsion, wobei das Verfahren Schritte umfasst, bei denen man
(a) Glycerin, Wasser, Ethanol und einen oder mehrere Emulgato ren ausgewählt aus der Gruppe bestehend aus Lecithin, Mo no- und Diglyceriden von Fettsäuren, Milchsäure und/oder Citronensäure und Mischungen davon vormischt;
(b) ein Pflanzenöl zufügt, das überwiegend mittelkettige oder langkettige Triglyceride umfasst; und
(c) die Mischung einer Hochdruckhomogenisierung bei einem Druck von 800-1500 bar unterzieht.
Im Schritt (a) des erfindungsgemäßen Verfahrens werden Glyce rin, Wasser, Ethanol und ein oder mehrere Emulgatoren ausge wählt aus der Gruppe bestehend aus Lecithin, Mono- und Diglyceriden von Fettsäuren, Milchsäure und/oder Citronensäure und Mischungen derselben miteinander vermischt. Die einzelnen Bestandteile können in den jeweiligen Mengen unter Rühren in ein geeignetes Gefäß gegeben werden, wie z.B. in einem Glasbe hälter oder Glaskolben.
In Schritt (b) des erfindungsgemäßen Verfahrens wird ein Pflanzenöl zufügt, das mittelkettige oder langkettige Triglyceride umfasst. Pflanzenöle, die sich für die Herstel lung der erfindungsgemäßen Nanoemulsionen eignen, sind oben beschrieben worden. In einer Ausführungsform handelt es sich bei dem Pflanzenöl, das der zuvor hergestellten Mischung zuge setzt wird, um ein Pflanzenöl, das zu über 50% aus mittelkettigen Triglyceriden besteht, wie z.B. Kokosöl oder Palmkernöl. In einer anderen Ausführungsform handelt es sich bei dem Pflanzenöl, das der zuvor hergestellten Mischung zuge setzt wird, um ein Pflanzenöl, das zu über 50% aus langkettigen Triglyceriden besteht, wie z.B. Sonnenblumenöl, Rapsöl, Olivenöl, Leinöl, Maiskeimöl, Weizenkeimöl oder Soja öl.
In einer Ausführungsform umfasst die Ölphase der erfindungsge mäßen Nanoemulsion Palmöl. In einer anderen Ausführungsform umfasst die Ölphase der erfindungsgemäßen Nanoemulsion Sonnen blumenöl. In einer weiteren Ausführungsform umfasst die Ölphase der erfindungsgemäßen Nanoemulsion Hanföl. In einer weiteren Ausführungsform umfasst die Ölphase der erfindungsge mäßen Nanoemulsion Rapsöl. In einer noch weiteren Ausführungs form umfasst die Ölphase der erfindungsgemäßen Nanoemulsion Olivenöl. In einer noch weiteren Ausführungsform umfasst die Ölphase der erfindungsgemäßen Nanoemulsion Leinöl. In einer noch weiteren Ausführungsform umfasst die Ölphase der erfin-
dungsgemäßen Nanoemulsion Maiskeimöl. In einer noch weiteren Ausführungsform umfasst die Ölphase der erfindungsgemäßen Na noemulsion Weizenkeimöl. In einer noch weiteren Ausführungs form umfasst die Ölphase der erfindungsgemäßen Nanoemulsion Sojaöl. In einer noch weiteren Ausführungsform umfasst die Ölphase der erfindungsgemäßen Nanoemulsion Kokosöl. In einer noch weiteren Ausführungsform umfasst die Ölphase der erfin dungsgemäßen Nanoemulsion Palmkernöl.
Im Schritt (c) des erfindungsgemäßen Verfahrens wird die aus dem vorhergehenden Schritt (b) erhaltende Mischung einer Hoch druckhomogenisierung bei einem Druck von 800-1500 bar unterzo gen, um die Größe der Öltröpfchen in der kontinuierlichen Pha se zu verringern.
Dieser Schritt wird vorzugsweise in einer für die Hochdruck homogenisierung geeigneten Vorrichtung durchgeführt. Vorrich tungen für die Hochdruckhomogenisierung sind im Stand der Technik hinreichend bekannt und können von mehreren kommerzi ellen Anbietern bezogen werden. Geeignete Vorrichtungen umfas sen beispielsweise den LM10 Microfluidizer oder den M-110L Microfluidizer (Microfluidics, Westwood, USA). Vorrichtungen zur Mikrofluidisierung verringern die durchschnittliche Größe der Öltröpfchen in der kontinuierlichen Phase, indem man die Ausgangsemulsion bei hohem Druck und hoher Geschwindigkeit durch eine Kammer mit geometrisch definierten Kanälen leitet. Die Kammer der Mikrofluidisierungsvorrichtung kann eine Viel zahl von geometrisch definierten Kanälen umfassen, wie z.B. 2, 3, 4, 5, 6, 7, 8, 9, 10 oder mehr Kanäle. Die Kammer der Mikrofluidisierungsvorrichtung besteht vorzugsweise aus Kera mik oder aus rostfreiem Stahl.
Im Rahmen der vorliegenden Erfindung wird die Hochdruckhomoge nisierung, z.B. die Mikrofluidisierung, bei einem Druck zwi schen 800-1500 bar durchgeführt. Besonders bevorzugt ist es
erfindungsgemäß, dass die Hochdruckhomogenisierung, z.B. die Mikrofluidisierung, bei einem Druck von mindestens 800 bar, stärker bevorzugt mindestens 850 bar, mindestens 900 bar, min destens 950 bar, mindestens 1000 bar, mindestens 1050 bar, mindestens 1100 bar, mindestens 1150 bar, mindestens 1200 bar, mindestens 1250 bar, mindestens 1300 bar, mindestens 1350 bar, mindestens 1400 bar, oder mindestens 1450 bar durchgeführt wird.
Zur Erreichung der geringen Teilchengröße kann es vorteilhaft sein, die aus Schritt (b) erhaltene Mischung mehrfach durch die Vorrichtung zur Hochdruckhomogenisierung zu leiten, da die Teilchengröße in der kontinuierlichen Phase sich mit jedem Durchlauf weiter verringert. Im Rahmen des Verfahrens ist es bevorzugt, die aus Schritt (b) erhaltene Mischung 2-15 Mal, vorzugsweise 2-10 Mal durch die Vorrichtung zur Hochdruckhomo genisierung zu leiten. So kann die aus Schritt (b) erhaltene Mischung vorzugsweise mindestens 2 Mal, mindestens 3 Mal, min destens 4 Mal, mindestens 5 Mal, mindestens 6 Mal, mindestens 7 Mal, mindestens 8 Mal, mindestens 9 Mal oder mindestens 10 Mal durch die Vorrichtung zur Hochdruckhomogenisierung gelei tet werden.
Die erfindungsgemäßen Nanoemulsion können in vielen Gebieten Anwendung finden. So können sie beispielsweise auf dem Gebiet der Medizin oder Pharmakologie verwendet werden, um lipophilen pharmazeutische Wirkstoffe zu verabreichen. In einem weiteren Aspekt betrifft die vorliegende Erfindung somit eine wie oben beschriebene Nanoemulsion zur Verwendung in der Medizin oder Pharmakologie. Die Nanoemulsion kann für diese Zwecke für ver schiedene Formen der Verabreichungen formuliert sein. Es ist jedoch bevorzugt, dass die Nanoemulsion für die orale Verab reichung formuliert ist.
In einem noch weiteren Aspekt betrifft die vorliegende Erfin dung somit die Verwendung einer wie oben beschriebenen Nano- emulsion zur Herstellung einer pharmazeutischen oder kosmeti schen Zusammensetzung. Vorzugsweise umfasst die pharmazeuti sche oder kosmetische Zusammensetzung eine lipophile Verbin dung, die pharmazeutisch oder kosmetisch aktiv ist, wie z.B. Cannabidiol (CBD). Die Erfindung betrifft auch die einer wie oben beschriebenen Nanoemulsion zur Herstellung eines Lebens mittels oder Futtermittels, wie z.B. eines Getränks.
Die Erfindung betrifft daher auch die Verwendung einer wie oben beschriebenen Nanoemulsion als Zusatzstoff in Lebensmit teln oder in Kosmetika. Da die Nanoemulsion bei topischer Ap plikation die oberen Hautschichten durchdringen und tiefere Hautschichten erreichen kann, eignet sich diese insbesondere für die Bereitstellung von lipophilen kosmetischen oder phar mazeutischen Wirkstoffen an tiefere Hautschichten.
In einem weiteren Aspekt betrifft die vorliegende Erfindung eine wie oben beschriebene Nanoemulsion zur Verwendung in ei nem therapeutischen Verfahren, bei dem ein lipophiler pharma zeutisch aktiver Wirkstoff verabreicht wird. In einem noch weiteren Aspekt betrifft die vorliegende Erfindung eine wie oben beschriebene Nanoemulsion zur Bereitstellung einer lipophilen Verbindung an ein Subjekt, das diese Verbindung be nötigt. Bei der lipophilen Verbindung handelt es sich vorzugs weise um Cannabidiol (CBD).
BESCHREIBUNG DER FIGUREN
Figur 1 zeigt die Ergebnisse der Partikelgrößenbestimmung der in Beispiel 1 hergestellten Nanoemulsion 1.
Figur 2 zeigt die Ergebnisse der Partikelgrößenbestimmung der in Beispiel 1 hergestellten Nanoemulsion 2.
Figur 3 zeigt die Ergebnisse der Partikelgrößenbestimmung der in Beispiel 1 hergestellten Nanoemulsion 3.
Figur 4 zeigt die Ergebnisse der Partikelgrößenbestimmung der in Beispiel 1 hergestellten Nanoemulsion 4.
Figur 5 zeigt die Ergebnisse der Bestimmung der Langzeitstabi- lität der in Beispiel 1 hergestellten Nanoemulsion 1.
Figur 6 zeigt die Werte für den Polydispersitätsindex (PDI) der in Beispiel 1 hergestellten Nanoemulsion 1.
BEISPIELE
Die nachfolgenden Beispiele sollen die vorliegende Erfindung veranschaulichen. Sie sind jedoch nicht als Einschränkung des Bereichs der Erfindung zu verstehen.
Beispiel 1: Herstellung der Nanoemulsionen
Nanoemulsion 1: Zur Herstellung einer ersten Nanoemulsion wur de zunächst eine Präemulsion aus Lecithin P75 (Lipoid GmbH), Glycerin (99%ig, Gustav Heess GmbH), Ethanol (96%ig, Sigma- Aldrich) und unraffiniertem rotem Palmöl (Gustav Heess GmbH) hergestellt. Dazu wurden 6% (w/w) Lecithin bei 40°C unter kon stantem Rühren in ein Gemisch aus 60% (w/w) Glycerin, 16%
(w/w) demineralisiertem Wasser und 10% (w/w) Ethanol gelöst.
Nach 3 Stunden wurde 8% (w/w) unraffiniertes rotes Palmöl als
Ölphase zugegeben. Die Präemulsion wurde durch Rühren mit ei nem VISCO-JET bei 800 U/min und 40°C für zwei Stunden erhal-
ten. Anschließend wurde die Präemulsion 12-mal bei 1400 bar durch einen Microfluidizer (LM10 Microfluidizer, Microfluidics International Corporation, Canada) geleitet. Nach jedem Durch lauf wurde das Gemisch auf unter 20°C gekühlt.
Nanoemulsion 2: Zur Herstellung einer zweiten Nanoemulsion wurde zunächst eine Präemulsion aus Imwitor 375 (IOI Oleo GmbH), Glycerin (99%ig, Gustav Heess GmbH), Ethanol (96%ig, Sigma-Aldrich) und unraffiniertem rotem Palmöl (Gustav Heess GmbH) hergestellt. Dazu wurden 6% (w/w) Imwitor 375 bei 40°C unter konstantem Rühren in ein Gemisch aus 60% (w/w) Glycerin, 16% (w/w) demineralisiertem Wasser und 10% (w/w) Ethanol ge löst. Nach 3 Stunden wurde 8% (w/w) unraffiniertes rotes Palm öl als Ölphase zugegeben. Die Präemulsion wurde durch Rühren mit einem VISCO-JET bei 800 U/min und 40°C für zwei Stunden erhalten. Anschließend wurde die Präemulsion 12-mal bei 1400 bar durch einen Microfluidizer (LM10 Microfluidizer, Micro- fluidics International Corporation, Canada) geleitet. Nach je dem Durchlauf wurde das Gemisch auf unter 20°C gekühlt.
Nanoemulsion 3: Zur Herstellung einer dritten Nanoemulsion wurde zunächst eine Präemulsion aus Lecithin P75 (Lipoid GmbH), Glycerin (99%ig, Gustav Heess GmbH), Ethanol (96%ig, Sigma-Aldrich), Zuckerester (Sisterna SP70-C, Sisterna) und unraffiniertem rotem Palmöl (Gustav Heess GmbH) hergestellt. Dazu wurden 5,5% (w/w) Lecithin und 0,5% (w/w) Zuckerester bei 40°C unter konstantem Rühren in ein Gemisch aus 60% (w/w) Gly cerin, 16% (w/w) demineralisiertem Wasser und 10% (w/w) Etha nol gelöst. Nach 3 Stunden wurde 8% (w/w) unraffiniertes rotes Palmöl als Ölphase zugegeben. Die Präemulsion wurde durch Rüh ren mit einem VISCO-JET bei 800 U/min und 40°C für zwei Stun den erhalten. Anschließend wurde die Präemulsion 12-mal bei 1400 bar durch einen Microf luidizer (LM10 Microfluidizer, Microfluidics International Corporation, Canada) geleitet. Nach jedem Durchlauf wurde das Gemisch auf unter 20°C gekühlt.
Nanoemulsion 4: Zur Herstellung einer vierten Nanoemulsion wurde zunächst eine Präemulsion aus Imwitor 375 (IOI Oleo GmbH), Glycerin (99%ig, Gustav Heess GmbH), Ethanol (96%ig, Sigma-Aldrich), Zuckerester (Sisterna SP70-C, Sisterna) und unraffiniertem rotem Palmöl (Gustav Heess GmbH) hergestellt. Dazu wurden 5,5% (w/w) Imwitor 375 und 0,5% (w/w) Zuckerester bei 40°C unter konstantem Rühren in ein Gemisch aus 60% (w/w)
Glycerin, 16% (w/w) demineralisiertem Wasser und 10% (w/w) Ethanol gelöst. Nach 3 Stunden wurde 8% (w/w) unraffiniertes rotes Palmöl als Ölphase zugegeben. Die Präemulsion wurde durch Rühren mit einem VISCO-JET bei 800 U/min und 40°C für zwei Stunden erhalten. Anschließend wurde die Präemulsion 12- mal bei 1400 bar durch einen Microf luidizer (LM10 Microfluidizer, Microfluidics International Corporation, Cana- da) geleitet. Nach jedem Durchlauf wurde das Gemisch auf unter 20°C gekühlt.
Beispiel 2: Bestimmung der Partikelgröße
Die Partikelgrößenbestimmung der in Beispiel 1 hergestellten Nanoemulsionen erfolgte durch dynamische Lichtstreuung (DLS, Malvern Nano ZS90, Malvern, UK). Die Proben (1 ml) wurden in 300 ml demineralisiertem Wasser dispergiert. DLS-Messungen wurden bei 25°C und 173° Streuwinkel durchgeführt.
Das Ergebnis ist in den Figuren 1-4 dargestellt. Wie aus den Figuren ersichtlich ist, betrug der mittlere Teilchendurchmes ser der Nanoemulsion 1 etwa 47,8 nm. Der mittlere Teilchen durchmesser der Nanoemulsion 2 betrug etwa 65,7 nm. Der mitt lere Teilchendurchmesser der Nanoemulsion 3 betrug etwa 53,2 nm. Der mittlere Teilchendurchmesser der Nanoemulsion 4 betrug etwa 61,5 nm.
Beispiel 3: Bestimmung der Langzeitstabilität
Proben der in Beispiel 1 hergestellten Nanoemulsion 1 wurden unter Lichtausschluss bei Raumtemperatur und 4°C gelagert. Es wurde im Abstand von einem Monat Proben der Nanoemulsionen entnommen und mittels DLS-Messungen charakterisiert.
Das Ergebnis ist in Figur 5 dargestellt. Es wurde festge stellt, dass auch eine Lagerung von 8 Monaten den mittleren Teilchendurchmesser nicht wesentlich veränderte, was auf die Lagerstabilität der Emulsionen verweist.
Die aus den Ergebnissen der DLS-Messungen errechneten Werte für den Polydispersitätsindex (PDI) sind in Figur 6 gezeigt.
Claims
1. Öl-in-Wasser-Nanoemulsion, umfassend
(a) eine Ölphase, die ein Pflanzenöl umfasst, wobei es sich vorzugsweise um ein LCT-Pflanzenöl handelt;
(b) eine kontinuierliche Phase, die Wasser und Glycerin umfasst;
(c) einen Emulgator, der ausgewählt ist aus der Gruppe bestehend aus Lecithin, Mono- und Diglyceriden von Fettsäuren, Milchsäure und/oder Citronensäure und Mi schungen davon;
(d) Ethanol; wobei die Nanoemulsion einen mittleren Teilchendurchmesser (Dm) von 70 nm oder weniger aufweist.
2. Nanoemulsion nach Anspruch 1, wobei der Emulgator eine Mi schung aus Mono- und Diglyceriden von Milchsäure, Citronensäure, Linolsäure und Ölsäure ist und vorzugsweise in einer Menge von 2-10% (w/w), mehr bevorzugt 5-6% (w/w) vorliegt.
3. Nanoemulsion nach Anspruch 1, wobei der Emulgator Lecithin ist und vorzugsweise in einer Menge von 2-10% (w/w), mehr bevorzugt 5-6% (w/w) vorliegt.
4. Nanoemulsion nach einem der Ansprüche 1 bis 3, wobei die Ölphase ferner Ölsäure und/oder Ethyloleat umfasst.
5. Nanoemulsion nach einem der Ansprüche 1-4, wobei die Ölphase ferner ein ätherisches Öl umfasst.
6. Nanoemulsion nach einem der Ansprüche 1-5, wobei die kon tinuierliche Phase 30-70% (w/w) Glycerin umfasst.
7. Nanoemulsion nach einem der Ansprüche 1-6, wobei das Etha nol in der Nanoemulsion in einer Menge von 2-20% (w/w), und vorzugsweise 5-10% (w/w), vorhanden ist.
8. Nanoemulsion nach einem der Ansprüche 1-7, die ferner ei nen Bioverfügbarkeitsverstärker ausgewählt aus der Gruppe bestehend aus Piperin, Curcumin, Resveratrol, Quercetin, Menthol, Naringin, Bergamottin, Kaempferol und Rutin ent hält.
9. Nanoemulsion nach einem der Ansprüche 1-8, wobei die Nano emulsion keine ethoxylierten Verbindungen enthält.
10. Nanoemulsion nach einem der Ansprüche 1-9, wobei die mitt lere Teilchengröße der Nanoemulsion nach einer Lagerung von 6 Monaten eine Größe von 100 nm nicht überschreitet.
11. Nanoemulsion nach einem der Ansprüche 1-9, die ferner ei nen Zuckerester, vorzugsweise einen Saccharoseester, um fasst.
12. Nanoemulsion nach einem der Ansprüche 1-11 zur Verwendung in der Medizin.
13. Nanoemulsion nach einem der Ansprüche 1-11 zur Verwendung in einem therapeutischen Verfahren, bei dem ein lipophiler pharmazeutisch aktiver Wirkstoff verabreicht wird.
14. Verwendung einer Nanoemulsion nach einem der Ansprüche 1- 11 zur Herstellung einer pharmazeutischen oder kosmeti schen Zusammensetzung.
15. Verwendung einer Nanoemulsion nach einem der Ansprüche 1- 11 als Zusatzstoff in Lebensmitteln.
16. Verwendung einer Nanoemulsion nach einem der Ansprüche 1- 11 als Zusatzstoff in Kosmetika.
17. Verwendung nach einem der Ansprüche 15 oder 16, wobei die Nanoemulsion Cannabidiol (CBD) umfasst.
18. Verfahren zur Herstellung einer Nanoemulsion nach einem der Ansprüche 1-11, bei dem man
(a) Glycerin, Wasser, Ethanol und einen Emulgator, der ausgewählt ist aus der Gruppe bestehend aus Lecithin, Mono- und Diglyceriden von Fettsäuren, Milchsäure und/oder Citronensäure und Mischungen davon vor mischt;
(b) ein Pflanzenöl zufügt, wobei es sich vorzugsweise um ein LCT-Pflanzenöl handelt; und
(c) die Mischung einer Hochdruckhomogenisierung bei einem Druck von 800-1500 bar unterzieht.
