Lateral Flow Assav
Die vorliegende Erfindung betrifft einen Lateral Flow Assay, ein Verfahren zur Quantifizierung eines Analyten sowie eine Vorrichtung zum Auslesen eines La teral Flow Assays.
Lateral Flow Assay (LFA) finden weit verbreitet Anwendung zum Nachweis von verschiedensten Analyten in verschiedenen Proben. Dabei werden die LFA so wohl in gut ausgestatten Laboren oder aber außerhalb der Labore von untrai niertem Personal als Point-of-Care-Test (POCT) eingesetzt. In einem typischen LFA wird eine flüssige Probe (z.B. eines Patienten) auf das Probenaufgabefeld des LFA aufgeben. Die Probe interagiert mit den einzelnen Komponenten des LFA und verursacht optische Veränderungen des LFA. Diese optischen Indikato ren können durch den Anwender oder aber auch durch optische Auslesegeräte beobachtet und ausgewertet werden. Ein Auslesegerät besitzt dabei typischer weise einen optischen Sensor, der die Variation des optischen Signals sehr ak kurat erfasst.
Die LFAs basieren dabei auf den Prinzipien der Immunoassays, bei denen die Anwesenheit einer Substanz, bezeichnet als Analyt, in flüssigen Proben wie Blut, Urin, Speichel oder Umweltproben nachgewiesen wird. Eine flüssige Probe, wel che den Analyten enthält, wird dabei auf das Probenaufgabenfeld des LFA auf gegeben.
Typischerweise besteht ein LFA aus einer Probenzone, einer Reaktionszone und einer Nachweiszone.
Die Probenzone wird auch als Probenaufgabefeld oder Sample Pad bezeichnet.
Die Reaktionszone umfasst Affinitätsmoleküle, die auch als Rezeptorelemente bezeichnet werden.
Auch die Nachweiszone umfasst Affinitätsmoleküle.
Dabei ist ein Rezeptorelement direkt nach dem Probenaufgabefeld eingebracht und ein zweites Rezeptorelement auf der sogenannten Testzone des LFA immo bilisiert. Dabei sind die beiden Rezeptorelemente so positioniert, dass das erste Rezeptormolekül nach Kontakt mit der Probe zusammen mit dem Analyten bis zu dem zweiten Rezeptorelement durch kapillare Flusskräfte transportiert wird. Das zweite Rezeptorelement, welches auf der festen Phase fixiert ist, bindet den Analyten oder das erste Rezeptorelement aus der Probe.
Wird die Probe auf das Aufgabefeld gegeben, interagiert der Analyt aus der Probe mit dem Rezeptorelement direkt nach dem Probenaufgabefeld. Dieses Rezeptorelement ist in der Regel an optisch signalgebende Partikel oder Mole küle (Markierung) konjugiert. Die Probe, mit dem Analyten und dem markierten Rezeptormolekül ("Markierungskomplex") fließt anschließend weiter zu der Testzone, auf der ein zweites Rezeptorelement immobilisiert ist. An der Test zone interagieren Analyt und die beiden Rezeptorelemente, wodurch sich ein optisches Signal, je nach verwendeter Markierung, auf der Testzone ausbildet. Dieses Signal kann durch ein Auslesegerät quantifiziert werden.
Bindet das erste Rezeptorelement den Analyten, welcher wiederum an das zweite Rezeptorelement bindet, spricht man von einem Sandwich Assay. Dabei steigt mit zunehmender Analytkonzentration die Signalintensität auf der Testli nie.
Ist nur ein Rezeptorelement vorhanden und zusätzlich ein Molekül das struktur gleiche oder strukturähnliche Abschnitte des Analyten besitzt, spricht man von einem kompetitiven Assay. Dabei kann zum Beispiel die Markierung an den Ana lyten (oder ein strukturähnliches Molekül) gebunden werden. Dieses Reagenz (Markierungskomplex) befindet sich dann in definierter Menge direkt nach dem Probenaufgabefeld und wandert nach Probenaufgabe mit dem Analyten aus der Probe bis zur Testzone, an welcher das Rezeptorelement fixiert ist.
In einem anderen Assayformat ist das Rezeptorelement an die Markierung ge bunden und direkt nach dem Probenaufgabefeld eingebracht. Der Analyt oder ein strukturähnliches Molekül ist in der Testzone immobilisiert. Dabei wandert der Analyt aus der Probe mit dem Konjugat ebenfalls bis zur Testzone. An der der Testzone interagieren dann Analyt aus der Probe, Rezeptormolekül und der in den Assay eingebrachte Analyt miteinander und verursachen ein Signal in der Testzone. Bei beiden kompetitiven Assayformaten sinkt die Signalintensität mit zunehmender Analytkonzentration.
Die feste Phase des LFA, welche die Testzone enthält, stellt dabei eine Zone bereit, in der ein Rezeptorelement oder der Analyt (oder strukturähnliches Mo lekül), bezeichnet als Ligand, immobilisiert ist. Dabei werden poröse Materialien wie Nitrocellulose, Nylon, Cellulose Acetat und andere poröse Materialien als feste Phase eingesetzt. Das Probenaufgabenfeld besteht zumeist aus Glasfaser oder Cellulosemembranen. In das an das Probenaufgabenfeld anschließende Pad werden die Markierungskomplexe eingebracht, wobei diese Membran eben falls zumeist aus Glasfaser und Cellulosemembranen bestehen.
Einige LFA Formate werden auch als Dipstick Format eingesetzt, bei denen ein Rezeptormolekül ebenfalls an die feste Phase gebunden ist. Dieser Teststreifen wird dann in die Probe getaucht, welche den Analyten enthält. Diese Probe kann dann entweder den Markierungskomplex freisetzen oder aber der Markierungs komplex kann der Probe vorher zugesetzt werden. Die Markierungskomplexe interagieren dann mit den Analyten aus der Probe und/oder dem Rezeptormo lekül auf der festen Phase und bilden entweder ein farbiges Feld oder Linie in der Testzone aus, oder nicht.
Ein LFA kann auch zur Detektion mehrere Analyten verwendet werden. Zum Beispiel werden beim Nachweis von Drogen aus biologischen Proben mehrere Drogen nachgewiesen. Solche LFA werden zumeist mit mehreren Testzonen an- geboten, wobei auf einer Testzone genau eine Droge nachgewiesen werden kann.
Bei dem Nachweis von kleinen Analyten (100-1000 Da) werden zumeist kom petitive Assayformate eingesetzt, bei denen mit zunehmender Analytkonzent- ration das Signal in der Testzone sinkt, wie in folgenden Patenten gezeigt: U.S. 4,703,017; US 5,451,504; und US 5,798,273
Beim Nachweis von größeren Analyten (1.000-1.000.000 Da) werden zumeist Sandwich Assayformate eingesetzt, bei denen das Signal mit zunehmender Ana- lytkonzentration in der Testzone ansteigt. Unter anderem sind Sandwich Assay formate auf LFA Basis in folgenden Patenten beschrieben: US 20150094227.
Wie bereits erwähnt, werden die Rezeptormoleküle mit farbigen Molekülen oder Partikeln markiert, um den Komplex aus Analyt und Antikörper oder Antikörpern sichtbar zu machen. Typische Markierungen für LFA sind farbige Nanopartikel oder Farbstoffe. Es gibt eine Vielzahl von Protokollen, die die Anbindung der Markierung an Liganden beschreiben. Traditionell werden LFA durch das Auge des Anwenders ausgewertet. Allerdings gibt es eine Vielzahl von Anwendungen, bei der eine Quantifizierung der Analytkonzentration notwendig ist. Daher wer den zur Quantifizierung verschiedenste Auslesegeräte eingesetzt, die durch die Intensität der Färbung der Testzone auf die Analytkonzentration in der Probe schließen können.
Solche Auslesegerät besitzen zumeist eine Leuchtquelle, mit der die Testzone bestrahlt wird und eine Detektionseinheit, die die Lichtintensität des durch die Testzone modifizieren Lichtes quantifiziert. Dabei wird zumeist das Licht der Leuchtquelle (zum Beispiel eine LED), welches von der Testzone entweder re flektiert oder absorbiert wird, durch eine oder mehrere Photodioden oder eine Kamera detektiert. Typischerweise besitzen Ausleseeinheiten mehrere LEDs, um die verschieden Test- und Kontrollzonen zu quantifizieren. Das gemessene Sig nal wird durch das Gerät anschließend in eine Analytkonzentration umgewandelt und dem Anwender auf einem Display angezeigt. Zum Beispiel werden solche Ausleseeinheiten in EP 1484601, KR102005597B1, US 20150094227, US 7,297,529 beschrieben.
Ein typisches Problem von den beschrieben Lateral Flow Assays und der Quan tifizierung des Signals ist der beschränkte dynamische Bereich. LFA besitzen dabei nur einen Messbereich von ca. 1,5-2 log Stufen. Dies reicht für die Quan tifizierung einiger Analyten nicht aus. Beispielsweise ist es notwendig, das CRP im Blut im Konzentrationsbereich von 0,5 - 150 pg/ml und damit über einen Messbereich von 2,5 Log-Stufen genau zu quantifizieren. Die dafür beschrieben LFA Verfahren sind zumeist unzureichend.
Einige Maßnahmen wurden bereits beschrieben, um bessere Messbereiche zu erzielen, zum Beispiel durch fluoreszenzbasierte Markierungen und das Auswer ten der Fluoreszenzintensität der Testbanden (US20100135857A1; US10048259B2). Andere Erfindungen beschreiben Signalverstärkungsreaktio nen (US20160169887A1) oder benutzen Photothermische Sensoren als Detek tionssystem und Metallnanopartikel als Markierungsmoleküle, um die Sensitivi- tät und den dynamischen Bereich zu erhöhen (US10094823B2; US9625385B2; US10094823B2). Dabei können mit den genannten Methoden verbesserte Mess bereiche erzielt werden. Allerdings erhöht sich dabei der messtechnische Auf wand und damit die Kosten des Auslesegerätes.
Aufgabe der vorliegenden Erfindung war es, einen Lateral Flow Assay bereit zu stellen, der eine Quantifizierung eines Analyten erlaubt, insbesondere über ei nen größeren Messbereich als im Stand der Technik.
Gelöst wird die Aufgabe durch einen Lateral Flow Assay mit
- eine Probenzone zum Aufträgen einer Probe
- einer Reaktionszone mit ersten Affinitätsmolekülen
- einer Nachweiszone mit zweiten Affinitätsmolekülen
dadurch gekennzeichnet, dass die ersten Affinitätsmoleküle eine optisch mess bare Markierung aufweisen, die mindestens 2 verschiedene Absorptionsmaxima unterschiedlicher Höhe aufweist, die zur Quantifizierung verwendet werden.
Der erfindungsgemäße Lateral Flow Assay weist, wie im Stand der Technik üb lich, eine Probenzone, eine Reaktionszone und mindestens eine Nachweiszone auf.
In der Probenzone wird die Probe aufgetragen, um in der sich daran anschlie ßenden Reaktionszone erste Affinitätsmoleküle zu binden. In der Nachweiszone sind zweite Affinitätsmoleküle immobilisiert, so dass sich dort im Sinne eines Sandwich-Assays Komplexe aus ersten Affinitätsmolekülen, Analyt und zweiten Affinitätsmolekülen anreichern.
Entscheidend ist, dass die ersten Affinitätsmoleküle eine optisch messbare Mar kierung aufwerfen, die zwei verschiedene Absorptionsmaxima unterschiedlicher Höhe aufweist, die so zur Quantifizierung verwendet werden können.
Wichtig ist, dass die beiden Absorptionsmaxima eine unterschiedliche Höhe auf weisen, so dass sie, je nach Konzentration des Analyten, unterschiedlich hohe Absorptionswerte geben.
Erfindungsgemäß wird bei beiden Absorptionsmaxima gemessen. Je nach Kon zentration des Analyten sind entweder die ersten Messungen im Bereich des ersten Absorptionsmaximum oder die im zweiten Absorptionsmaximum in einem Bereich, der eine gute und sichere Quantifizierung erlaubt.
Die ersten und zweiten Affinitätsmoleküle können beispielsweise Antikörper o- der Antikörperfragmente sein. Auch Protein A, Protein G, Streptavidin, Neutra- vidin oder Derivate davon sind geeignet. Dem Fachmann sind weitere geeignete Affinitätsmoleküle bekannt.
Die optisch messbare Markierung wird bevorzugt durch Markierungsverbindun gen erreicht, die aus Farbstoffen, Nanomaterialen oder Kombinationen hiervon ausgewählt sind.
Als Nanomaterialien eignen sich insbesondere Metallnanomaterialien, Latexna- nomaterialien, cellulosebasierte Nanomaterialen, Silica-Nanomaterialien oder Kombinationen hiervon.
Die optisch messbare Markierung mit zwei verschiedenen Absorptionsmaxima kann zum einen durch zwei verschiedene Markierungsverbindungen erreicht werden. Beispielsweise könnte die Markierung ein blauer und ein roter Farbstoff sein, die Absorptionsmaxima unterschiedlicher Höhe aufweisen.
Bevorzugt wird jedoch eine Markierungsverbindung verwendet, die mindestens zwei verschiedene Absorptionsmaxima unterschiedlicher Höhe aufweist.
Die erfindungsgemäß eingesetzten zwei verschiedenen Absorptionsmaxima lie gen bevorzugt mindestens 50 nm auseinander, bevorzugt mindestens 70 nm auseinander und noch mehr bevorzugt mindestens 100 nm auseinander.
Bevorzugt liegt das eine Absorptionsmaximum mindestens 1,5 mal höher als das andere Absorptionsmaximum. Bevorzugt ist das Verhältnis mindestens 2 oder mindestens 3. Es ist dabei egal, ob das Absorptionsmaximum, das höher ist als das andere Absorptionsmaximum, bei höherer oder niedriger Wellenlänge liegt.
Gemessen wird die Absorbanz oder Transmission der Markierungsmoleküle bei beiden Absorptionsmaxima. Durch die unterschiedliche Höhe der beiden Absorp tionsmaxima werden unterschiedliche Messbereiche erhalten.
Der Lateral Flow Assay der vorliegenden Erfindung kann auch weitere übliche Merkmale enthalten, beispielsweise eine Kontrollzone, in der die korrekte
Funktionsweise des Tests überprüft wird und eine Saugzone, um das Wandern der Proben zu fördern.
Gegenstand der Erfindung ist auch ein Verfahren zur Quantifizierung eines Ana- lyten umfassend die Schritte
- Aufträgen einer Probe auf den erfindungsgemäßen Lateral Flow Assay
- Messen des Signals bei mindestens zwei Wellenlängen
- Berechnen der Konzentration des Analyten.
Gegenstand der Erfindung ist auch eine Vorrichtung zum Auslesen des erfin dungsgemäßen Lateral Flow Assay umfassend
- mindestens eine Beleuchtungseinrichtung mit mindestens zwei Wellenlängen
- eine Haltevorrichtung für einen Lateral Flow Assay
- mindestens eine Detektionseinrichtung zur Messung der mindestens zwei Wellenlängen der Beleuchtungseinrichtung
- eine Auswerteinheit zur Berechnung der Konzentration des Analyten.
Üblicherweise werden bei LFAs zur Markierung Detektionskonjugate eingesetzt, welche über ein definiertes Absorptionsmaximum verfügen, wie zum Beispiel Goldnanopartikel mit einem Absorptionsmaxima zwischen 520-540 nm. Mit ei nem Auslesegerät kann die Färbung der Testzone, verursacht durch zum Bei spiel Goldnanopartikel quantifiziert werden. Dabei ist der dynamische Bereich des Verfahrens dadurch determiniert, das die Färbung in der Testzone mit we nigen Nanopartikeln gesättigt ist und von den Auslesegeräten nicht mehr auf gelöst werden kann. Dieses Problem tritt auch bei Verwendung von anderen gefärbten Nanomaterialen wie beispielsweise Latexnanopartikeln, Cellulose-ba- sierte Nanopartikeln oder anderen Metall-basierten Metallnanopartikeln auf. Bei Verwendung von weniger gefärbten Nanomaterialien kann der dynamische Be reich zu höheren Konzentrationen verschoben werden, wobei dann die
Sensitivität des Verfahrens geringer wird und sich die Gesamtbreite des dyna mischen Bereichs nicht ändert.
In dem hier beschrieben Verfahren werden Markierungskomplexe eingesetzt, welche über mindestens zwei verschiedene Absorptionsmaxima bei deutlich un terschiedlichen Wellenlängen verfügen. Außerdem unterscheiden sich die Ab sorptionsmaxima des Markierungskomplexes in der Höhe. Damit können die Markierungskomplexe durch optische Messverfahren unterschiedlich sensitiv detektiert werden. Bei geringen Mengen des Markierungskomplexes, zum Bei spiel wenn die Menge an Analyt der Nachweisgrenze des Messverfahrens ent spricht, wird nur das stärkere Absorptionsmaximum des Markierungskomplexes erkannt, wobei das zweite, schwächer Absorptionsmaximum nicht detektiert werden kann. Bei höheren Konzentrationen des Markierungskomplexes ist die quantifizierbare Menge des Markierungskomplexes entsprechend vom Messsys tem limitiert. Das Messsystem kann ab einer entsprechenden Menge an Markie rungskomplex das optische Signal unterschiedlicher Mengen des Markierungs komplex nicht mehr auflösen. Stehen zwei Absorptionsmaxima desselben Mar kierungskomplexes zur Verfügung, kann bei Verwendung des schwächeren Ab sorptionsmaxima die Menge des Markierungskomplexes noch quantifiziert wer den, wenn das Messverfahren bei Messung des stärkeren Absorptionsmaxima bereits die Sättigung erlangt hat.
Die verwendeten Markierungen verfügen dabei über zwei oder mehrere Absorp tionsmaxima im Bereich zwischen 200-2000 nm. Typischerweise werden Gold- Nanostäbchen, Silber-Nanoplättchen oder Silica-Nanoshells eingesetzt. Es kön nen aber auch Latex oder Silica-Nanopartikel verwendet werden, die verschie dene Farbstoffe enthalten. Die verschiedenen Absorptionsmaxima liegen dabei typischerweise um mindestens 100 nm auseinander. In Figur 2 sind beispielhaft Spektren von Gold-Nanostäbchen mit unterschiedliche Absorptionsspektren dargestellt. Diese Nanostäbchen verfügen über ein Absorptionsmaxima im Be reich zwischen 520-540 nm und ein zweites Absorptionsmaxima im Bereich zwi schen 600-800 nm. Die Form und die Größe der Markierung können sich für die
Anwendung unterscheiden, bevorzugt ist aber, dass die Materialien homogen in Größe und Form innerhalb eines Verfahrens sind. Die Gold-Nanostäbchen der beispielhaft gezeigten Spektren haben dabei eine Länge zwischen 50-70 nm und ein Durchmesser von 10-20 nm.
Diese Nanomaterialien sind außerdem an ein oder mehrere Affinitätsmoleküle auf der Oberfläche gebunden. Dabei wurden die Liganden entweder durch pas sive Adsorption oder kovalent durch chemische Reaktionen an die Nanomateri alien gebunden. Als Ligand werden dabei zum Beispiele Antikörper eingesetzt.
Ein weiterer Aspekt des Verfahrens stellt ein Auslesegerät dar, welches die Menge des Markierungskomplexen in der Testzone quantifizieren kann. Dabei wird die Menge des in der Testzone angereicherte Markierungskomplexes bei zwei verschieden Wellenlängen quantifiziert. Eine bevorzugte Vorrichtung be sitzt dabei eine oder mehrere Lichtquellen, welches Licht entsprechend der ver schiedenen Wellenlänge der Absorptionsmaxima emittiert. Außerdem besitzt das Gerät einen oder mehrere Detektoren der das Licht, welches von der Test zone reflektiert oder absorbiert, detektiert. Der Detektor kann dabei eine Ka mera sein, welche die Testzone abbildet und anschließend die Verfärbung der Testbande durch das Gerät anschließend quantifiziert wird. Eine andere Mög lichkeit für einen Detektor sind Photodioden, welche das Licht, das auf der Membran reflektiert oder absorbiert wird, detektieren. Um Licht verschiedenen Wellenlängen zu emittieren können entweder verschiedene LED eingesetzt wer den oder es wird eine Lichtquelle eingesetzt und im Anschluss das emittierte Licht durch optische Bauteile wie optische Filter oder Gitter so modifiziert, das nur noch Licht definierter Wellenlängen die Testzone erreicht. Das Auslesegerät quantifiziert dabei die Menge des Markierungskomplexes unter Anwendung von Licht der beiden verschiedenen Wellenlängen. Die Wellenlängen des Lichtes ent sprechen dabei den Absorptionsmaxima des Markierungskomplexes. Durch An wendung geeigneter Algorithmen kann anschließend das Auslesegerät die Kon zentration des Analyten berechnen. Eine schematische Darstellung ist Figur 1 gezeigt.

Da Gold-Nanostäbchen über Plasmonenresonanz verfügen, welche durch die Größe der Stäbchen beeinflusst werden können, können die Absorptionseigen schaften durch die Synthesebedingungen gesteuert werden. Das Absorptions spektrum eines 10 nm im Durchmesser und 40 nm langen Goldnanostäbchens besitzt zwei Absorptionsmaxima. Ein Absorptionsmaximum, verursacht durch die quer gerichtete Oberflächen Plasmonenresonanz des Gold-Nanostäbchens, liegt bei 525 nm. Das zweite Absorptionsmaximum, verursacht durch die längs gerichtete Oberflächenplasmonenresonanz liegt bei 800 nm. Dabei besitzt das Absorptionsmaxima bei 800 nm eine 4,1 mal so hohe Absorption wie das Ma- xima bei 525 nm.
Markierunaskomplex
Diese Goldnanostäbchen werden zur Herstellung eines Detektionsmoleküls ver wendet. Dazu wurden anti-CRP Antikörper passiv an die Citrat-stabilisierten Gold-Nanostäbchen gebunden, indem der Antikörper direkt zu einer Lösung der Nanostäbchen gegeben wurde. Nach einer 30-minütigen Inkubation wurden restliche Bindungsstellen auf den Nanostäbchen mit BSA geblockt und der Mar kierungskomplex durch Zentrifugation bei 8000 g und erneutem re-suspendie- ren in Wasser gewaschen. Die Lösung wurde auf eine OD 10 bei 525 nm einge stellt.
Nanostäbchen-basierter LFA
Die Einbringung des Markierungskomplexes in eine Glasfasermembran erfolgte mit einem Airjet Quanti Modul der Biodot XYZ Series Dosierplattform. Mit der Sprühfunktion wurde der Markierungskomplex mit einem Druck von ca. 4 psi und einer Dosierrate von 10 pl cm-1 auf das Pad gesprüht. Anschließend wurde die Glasfasermembran mit dem eingebrachten Markierungskomplex (Reaktions zone) für mindestens zwei Stunden bei 37 °C im Trockenschrank getrocknet. Zur Beschichtung einer Nitrocellulosemembran wurden mit der Cavro Einheit der Biodot Dosierplatform 1 pg anti-CRP Ak/mI in PB Puffer mit einer Dosierrate von 1 mI/cm aufgegeben. Nach der Beschichtung wurde die Membran für 1 h bei 37°C getrocknet. Für die Herstellung von gebrauchsfertigen Teststreifen wurden ein Probenzone, eine Reaktionszone, eine Nachweiszone und eine Saugzone auf eine Backing Card geklebt und anschließend in 4 mm breite Streifen geschnit ten.
Ausleseaerät
Das Auslesegerät besteht aus eine Beleuchtungsquelle, welche über zwei LED mit den Wellenlängen 525 nm und 800 nm verfügt. Diese LED wurden so plat ziert, dass sie das Licht gleichmäßig auf die Nachweiszone des Lateral Flow As- says emittieren. Über der Nachweiszone befindet sich eine Kamera, die ein Bild von der Nachweiszone aufnimmt. Das Gerät nimmt sowohl ein Bild mit der Ka mera bei einer Beleuchtung mit 525 nm, als auch ein Bild mit einer Beleuchtung
bei 800 nm auf. Anschließend kann durch einen Algorithmus aus den Bildern eine Konzentration des CRP in der Probe berechnet werden.
Verfahren
Es werden 100 mI eine Probe auf die Probenzone des LFA aufgegeben und 10 Minuten inkubiert. Nach 10 Minuten wird die Verfärbung der Nachweiszone mit dem Auslesegerät vermessen und die CRP Konzentration ausgegeben. Dabei konnten über einen Messbereich von 0,5-200 pg CRP mit einem Variationskoef fizienten von <15% CRP in den CRP-haltigen Proben nachgewiesen werden. Fi gur 3 zeigt dabei den Messbereich des Verfahrens bei Anwendung von 525 nm als Detektionswellenlänge und bei Anwendung von 800 nm als Wellenlänge so wie den resultierenden gesamten Messbereich.
Beispiel 2
Es wurden Silbernanomaterialien, welche über zwei verschiedene Absorptions- maxima aufgrund der Plasmonenresonanz verfügen hergestellt. Ein Absorpti onsmaximum lag bei 400 nm und das zweite bei 600 nm. Dabei besitzt das Absorptionsmaximum bei 600 nm eine 5 mal so hohe Absorption wie das Maxi mum bei 400 nm.
An diese Silbernanomaterialien wurden anti-TSH Antikörper konjugiert und die ses Konjugat anschließend als Markierungskomplex verwendet. Für einen Late ral Flow Assay wurde eine Nitrocellulosemembran ebenfalls mit anti-TSH Anti körpern beschichtet.
Der Markierungskomplex wurde zu der Probe gegeben und die beschichtete Nit rocellulose wurde in diese Probe eingetaucht, 5 Minuten inkubiert und anschlie ßend mit einem Auslesegerät, welches den in der Nachweiszone angereicherten Markierungskomplex bei zwei verschiedenen Wellenlängen quantifizieren kann, ausgewertet. Dabei konnten über einen Messbereich von 0,05-100 mIII/ml TSH mit einem Variationskoeffizienten von <15% TSH in den TSH-haltigen Proben nachgewiesen werden.
Beispiel 3
Es wurden zwei verschiedene Farbstoffe mit unterschiedenen Absorptionsma- xima und Extinktionskoeffizienten in Silica-Nanopartikel eingeschlossen. Ein Ab sorptionsmaximum lag bei 450 nm und das zweite bei 620 nm. Dabei besitzt das Absorptionsmaximum des Silicananopartikels bei 620 nm eine 2,5 mal so hohe Absorption wie das Maxima bei 450 nm.
An diese Silcananopartikel wurden kovalent anti-IgE Antikörper immobilisiert. Dieser Markierungskomplex wurde in eine Glasfasermembran eingebracht und im Vakuum getrocknet. Zur Beschichtung einer Nitrocellulosemembran wurde 1 pg/cm einer Mischung von vier bekannten Eiallergene (Ovomucoid, Ovalbumin, Ovotransferrin und Lysozyme) in PBS Puffer auf eine Nitrocellulosemembran do siert. Für die Herstellung von gebrauchsfertigen Teststreifen wurden eine Pro benzone, eine Nachweiszone, eine Reaktionszone und eine Saugzone auf eine Backing Card geklebt und anschließend in 4 mm breite Streifen geschnitten. Diese Teststreifen wurden in ein Gehäuse platziert.
Nach Aufgabe von 100 pl einer Probe auf die Probenzone wurde der Test in die Ausleseeinheit platziert. Die Ausleseeinheit verfügt über zwei LED mit den Wel lenlängen 450 nm und 620 nm und kann das reflektierte Licht bei 450 nm als auch bei 620 nm der Testzone durch eine Photodiodenzeile detektieren. Dabei konnten über einen Messbereich von 2-5000 kU/l IgE mit einem Variationsko effizienten von <15% in den Eiallergen-spezifischen IgE-haltigen Proben nach gewiesen werden.
Claims
1. Lateral Flow Assay mit einer Probenzone zum Aufträgen einer Probe einer Reaktionszone mit ersten Affinitätsmolekülen einer Nachweiszone mit zweiten Affinitätsmolekülen dadurch gekennzeichnet, dass die ersten Affinitätsmoleküle eine optisch messbare Markierung aufweisen, die mindestens 2 verschiedene Absorptionsmaxima unterschiedlicher Höhe aufweist, die zur Quantifizierung verwendet werden.
2. Lateral Flow Assay nach Anspruch 1 wobei die ersten und die zweiten Affi nitätsmoleküle unabhängig voneinander Antikörper, Antikörperfragmente, Antigene, Antigenfragmente, Antigen-ähnliche Strukturen, Protein A, Pro tein G, Streptavidin oder Neutravidin sind.
3. Lateral Flow Assay nach einem der Ansprüche 1 oder 2, wobei die optische messbare Markierung durch Markierungsverbindungen ausgewählt aus Farbstoffen, Nanomaterialien oder Kombinationen hiervon erfolgt.
4. Lateral Flow Assay nach einem der vorhergehenden Ansprüche, wobei die Nanomaterialen Metallnanomaterialien, Latexnanomaterialien, cellulose basierte Nanomaterialen, Silica-Nanomaterialien oder Kombinationen hier von sind.
5. Lateral Flow Assay nach einem der vorhergehenden Ansprüche, wobei die optische messbare Markierung zwei verschiedene Markierungsverbindun gen umfasst.
6. Lateral Flow Assay nach einem der vorhergehenden Ansprüche, wobei die optische messbare Markierung eine Markierungsverbindungen umfasst, die
mindestens 2 verschiedene Absorptionsmaxima unterschiedlicher Höhe aufweist.
7. Lateral Flow Assay nach einem der vorhergehenden Ansprüche, wobei zu sätzlich eine Kontrollzone vorhanden ist.
8. Lateral Flow Assay nach einem der vorhergehenden Ansprüche, wobei zu sätzlich eine Saugzone vorhanden ist.
9. Lateral Flow Assay nach einem der vorhergehenden Ansprüche, wobei zwei der mindestens zwei Absorptionsmaxima einen Abstand von mindestens 100 nm aufweisen.
10. Lateral Flow Assay nach einem der vorhergehenden Ansprüche, wobei ei nes der mindestens zwei Absorptionsmaxima mindestens 1,5 mal höher ist als das Absorptionsmaximum eines anderen Absorptionsmaximums.
11. Verfahren zur Quantifizierung eines Analyten umfassend die Schritte
Aufträgen einer Probe auf einen Lateral Flow Assay nach einem der Ansprüche 1 bis 10
Messen des Signals bei mindestens zwei Wellenlängen, wobei die min destens zwei Wellenlängen Absorptionsmaxima der optischen Markie rung entsprechen
Berechnen der Konzentration des Analyten.
12. Vorrichtung zum Auslesen des Lateral Flow Assay nach einem der Ansprü che 1 bis 10 umfassend mindestens eine Beleuchtungseinrichtung mit mindestens zwei Wel lenlängen eine Haltevorrichtung für einen Lateral Flow Assay
mindestens eine Detektionseinrichtung zur Messung der mindestens zwei Wellenlängen der Beleuchtungseinrichtung eine Auswerteinheit zur Berechnung der Konzentration des Analyten.
13. Verwendung eines Lateral Flow Assay nach einem der Ansprüche 1 bis 10 zur Bestimmung der Konzentration eines Analyten durch Mes sen der optischen Absorption bei zwei verschiedenen Wellenlängen die Absorptionsmaxima der optischen Markierung entsprechen.
