Description
Verfahren zur Herstellung von Olefinen
Die vorliegende Erfindung betrifft ein neue Methode zur Synthese von Olefinen mit aromatischen Substituenten unter Verwendung eines sehr einfachen Katalysatorsystems .
Olefine mit aromatischen Substituenten spielen in der industriellen Chemie eine wichtige Rolle, so z. B. als Ausgangsprodukte für Sonnenschutzmittel, Polymere, Feinchemikalien und Wirkstoffvorprodukte von Pharmazeutika, Pflanzenschutzmitteln und Duftstoffen.
Eine Möglichkeit zur Synthese von arylsubstituierten Olefinen ist die sogenannte Heck-Reaktion, bei der Iod-, Brom- oder Chloraromaten ArX (I, Br, Cl) mit Olefinen in Gegenwart von stöchiometrischen Mengen einer Base und katalytischen Mengen einer Palladium- Verbindung wie Pd(PPh3)4 umgesetzt werden (R. Trotz der vielen Publikationen aus Universitätslaboratorien hat die Heck-Reaktion bislang so gut wie keine industrielle Anwendung erfahren. Zu den Gründen gehört die Tatsache, dass die Reaktivität von Halogenaromaten ArX in der Reihenfolge Arl > ArBr > ArCl stark abfällt. So werden meist die Iodide Arl unter relativ milden Bedingungen (80 - 1 10 °C) eingesetzt. Solche Substrate sind jedoch sehr
teuer. Im Falle von Bromaromaten führen die gängigen Katalysatoren bzw. Präkatalysatoren wie Pd(PPh3)4, Pd(OAc)2 in Gegenwart von überschüssigem PPh3 oder P(tolyl)3 zu akzeptablen Ausbeuten bei Reaktionstemperaturen von etwa 140 °C. Nachteilig sind jedoch die hohen Mengen an Palladium (1 - 2 Mol-%), ferner die Tatsche, dass Phosphane erforderlich sind. Deshalb hat es nicht an Versuchen gefehlt, phosphanfreie Katalysatorsysteme für Heck-Reaktionen zu entwickeln. Trotz gewisser Fortschritte (A. S. Carlstroem, T. Frejd, J. Org. A. Herrmann, C. Brossmer, K. Öfele, C.-P. Reisinger, T. Priermeier, M. Beller, H. Fischer, Angew. Chem. 1995, 107, 1989-1992; Angew. Chem., Int. Ed. Engl. 1995, 34, 1844; W. A. Herrmann, C. Brossmer, C.-P. Reisinger, T. H. Riermeier, K. Öfele, M. Beller, Chem.-Eur. J. 1997, 3, 1357-1364; M. Ohff, A. Ohff, M. Ε. van der Boom, D. Milstein, J. Am. Chem. Soc. 1997, 119, 11687-11688; DE 4421730C1; EP 0725049A1) Aufmerksamkeit auf sich gezogen. Tatsächlich lassen sich Bromaromaten glatt umsetzen, sogar mit nur 0.01 Mol-% Palladacyclus. Allerdings sind solche Katalysatoren teuer bzw. erfordern mehrere Synthesestufen unter Verwendung des teuren Tris(o-tolyl)phosphans oder von anderen schwer zugänglichen Phosphanen. Im Falle von bestimmten Chloraromaten wurde kürzlich ein aktives Katalysatorsystem bestehend aus PdCl2(PhCN)2, Ph^Cl" und N,N- Dimethylglycin (DMG) als Additiv beschrieben; in Abwesenheit des Phosphoniumsalzes Ph4P+Cl" erfolgt keine Reaktion (M. T. Reetz, G. Lohmer, R. Schwickardi, Angew. Chem. 1998, HO, 492-495; Angew. Chem., Int. Ed. Engl.
1998, 37, 481-483; M. T. Reetz, G. Lohmer, R. Schwickardi, DE-A 19712388.0, 1997). Allerdings ist dieses Katalysatorsystem für Bromaromaten weniger gut geeignet, ferner sind Phosphoniumsalze erforderlich, die wiederum die Verwendung von Phosphanen nötig machen.
Es wurde nun überraschenderweise gefunden, dass ein verblüffend einfaches, billiges und phosphanfreies Katalysatorsystem glatte Heck-Reaktionen von Bromaromaten bewirkt, und zwar auch dann, wenn geringe Mengen des Katalysators bzw. Präkatalysators eingesetzt werden. Das neue Katalysatorsystem besteht aus billigen Palladium(II)-Salzen in Gegenwart von stickstoffhaltigen Additiven wie N,N-Dimethylglycin (DMG) und einer Base. Somit wird die Verwendung von Phosphanen überflüssig.
Überdies wurde gefunden, daß bei sehr kleinen Mengen von einfachen Palladium- Salzen die Reaktion auch ohne Additiv verläuft, wenn auch bei deutlich erhöhten Reaktionszeiten. Erst bei Konzentrationen von mehr als 0,5 Molprozent Palladium wirkt sich das Additiv stark beschleunigend aus.
Als Katalysatoren dienen gängige Palladium-II-Salze PdXY oder deren CH3CN-, PhCN-Komplexe, wobei typischerweise X = Y = Cl, Br, I, NO2, RCO2 [R = - C22, CF3, CCI3, CH2N(CH3)2, C6H5] oder RSO3 (R = Ci - C22, CF3, C4F9, CCI3, C6H5, /7-CH3C6H4) oder typischerweise X = Cl, Br, I, RCO2 (R = Ci - C22, CF3, CCI3, CH2OCH3, C6H5) und typischerweise Y = C6H5, o-, m-, /7-CH3C6H4, 0-, m-, 7-CI-C6H4, o-, m-, p-CHOCöHt, o-, m-, p-CN-CßΑi, o-, m-, -NO2-C6H4, o-, m-, p-PhCO-CόELi, o-, m-, P-F-C6H4, I-C10H7 oder 2-C10H7. Vorzugsweise wird Pd(OAc)2, PdCl2(PhCN)2, PdCl2, PdCl2(CH3CN)2, CöHsPdCl oder Pd(NO3)2 oder deren dimere oder oligomere Form verwendet.
Als Additive werden stickstoffhaltige Carbonsäuren wie gängige oc- oder ß- Aminosäuren H2N(R)CHCO2H bzw. H2N(R)CHCH2CO2H [R = H, CH3, C6H5, CH2C6H4, CH(CH3)2] oder deren N-alkylierte Formen R'NH(R)CHCO H bzw. R'NH(R)CHCH2CO2H oder R'2N(R)CHCO2H bzw. R'2N(R)CHCH2CO2H [R' = CH3, C2H5, C3H7, C4H9 oder R + R = (CH2)4 oder (CH2)5] bzw. deren Natrium- oder Kaliumsalze, Anthranilsäure oder N,N-Dimethylanthranilsäure oder Pyridincarbonsäuren (oder deren Natrium- oder Kaliumsalze) wie 2-Pyridincarbon-
säure oder aromatische stickstoffhaltige Heterocyclen wie 2,2'-Dipyridyl verwendet. Vorzugsweise wird N,N-Dimethylglycin (DMG) eingesetzt. Das Verhältnis von Additiv zu Palladium bewegt sich zwischen 100 : 1 und 1 : 1, vorzugsweise zwischen 50 : 1 und 1 : 1.
Die Verwendung von diesen Additiven führt dazu, dass die Reaktionszeit und die Reaktionstemperatur deutlich gesenkt werden können. Ferner erhöhen solche Stoffe die Selektivität der Reaktion.
Als Lösungsmittel dienen aprotische dipolare Solventien wie Dimethylformamid (DMF), Dimethylacetamid (DMA), Dimethylsulfoxid, Propylencarbonat, 1,3- Dimethyl-3,4,5,6-tetrahydro-2(lH)-pyrimidinon (DMPU) oder l-Methyl-2- pyrrolidinon (NMP), aber auch protische Lösungsmittel wie Methanol, Ethanol oder Diethylenglycol. Vorzugsweise wird DMF, NMP oder Methanol verwendet.
Als Base dienen Metallsalze wie Natrium-, Kalium-, Cesium-, Calcium- oder Magnesiumsalze von Carbonsäuren oder die entsprechenden Carbonate bzw. Bicarbonate oder Amine wie Triethylamin oder Trioctylamin, vorzugsweise Natriumacetat. Das Verhältnis von Base zu Arylhalogenid bewegt sich zwischen 1 : 1 und 5 : 1, vorzugsweise 1.5 : 1 bis 2 : 1.
Reaktionstemperaturen zwischen 60 °C und 180 °C können gewählt werden, vorzugsweise läßt man die Reaktionen zwischen 100 °C und 140 °C ablaufen.
Was die Arylkomponente ArX angeht, so können recht unterschiedliche Aryl- und Heteroarylhalogenide, vorzugsweise Arylbromide, aber auch -O-tosylate, -O- mesylate oder -O-triflate eingesetzt werden, beispielsweise Benzol-, Naphthalin-, Pyridin- oder Chinolinderivate. Auch Aryldiazoniumsalze können umgesetzt werden.
Bei der Olefinkomponente der vorgenannten Formel II stellen R1, R2 und R3 unabhängig voneinander Wasserstoff, Alkyl-(Cι - Cs), Phenyl, 1- oder 2-Naphthyl, Vinyl, O-Alkyl-(Cι - C8), O-Phenyl, CN, CO2H, CO2-Alkyl-(Cι - C8), CO2- Phenyl, CONH2, CONH-Alkyl-(Cι - C5), CON(Alkyl)2-(Cι - C5), Fluor, Chlor, PO(Phenyl)2, PO(Alkyl) -( - C5), CO-Phenyl, CO-Alkyl-(Cι - C5), NH-Alkyl-
(Ci - C4), SO3H, PO3H, SO3-Alkyl-(Cι - C4) oder SO2-Alkyl-(Cι - C4), ferner kommen cyclische Derivate in Frage, nämlich wenn R1 + R2 = (CH2)n oder R2 + R3 = (CH2)n, wobei n = 2 bis 16, sein kann.

Beispiel 1:
In einem Reaktionsgefäß werden 5.7 mg (0.015 mmol) (C6H5CN)2PdCl2 und 30.9 mg (0.3 mmol) Dimethylglycin eingewogen und anschließend dreimal evakuiert und argoniert. Dazu werden unter Argon 164 mg wasserfreies Natriumacetat, 157 mg (1.00 mmol) Brombenzol und 125 mg (1.2 mmol) Styrol zugewogen. Nach Zugabe von 1 ml NMP wird das Gefäß verschlossen und das Gemisch 10 Stunden bei 130 °C gerührt.
Nach der Reaktion werden die GC-Standards «-Decan und n-Hexadecan zugewogen und das Gemisch mit 2 ml Diethylether versetzt. Man zentrifugiert die Suspension und filtriert anschließend die festen Bestandteile ab. Schließlich wird das Filtrat gaschromatographisch untersucht: 98 % Umsatz bezogen auf Brombenzol; die Ausbeute an Heck-Produkten beträgt ebenfalls 98 % (Selektivität: 96.4 % trans-Stilben, 0.6 % c/s-Stilben und 3.0 % 1,1-Dipheny lernen).
Beispiel 2:
Es wird wie in Beispiel 1 beschrieben vorgegangen, jedoch als Lösungsmittel Methanol anstatt NMP verwendet: 99 % Umsatz bezogen auf Brombenzol; die Ausbeute an Heck-Produkten beträgt ebenfalls 99 % (Selektivität: 97.0 % trans- Stilben, 0.5 % cw-Stilben und 2.5 % 1,1-Dipheny lernen).
Beispiel 3:
Es wird wie in Beispiel 1 beschrieben vorgegangen, jedoch als Lösungsmittel Diethylenglycol anstatt NMP verwendet: 99 % Umsatz bezogen auf Brombenzol; die Ausbeute an Heck-Produkten betrag 98 % (Selektivität: 96.6 % trans-Stilben, 0.4 % c/s-Stilben und 3.0 % 1,1-Dipheny lernen).
Beispiel 4:
Es wird wie in Beispiel 1 beschrieben vorgegangen, jedoch als Lösungsmittel
Ethanol anstatt NMP verwendet: 79 % Umsatz bezogen auf Brombenzol; die
Ausbeute an Heck-Produkten beträgt 69 % (Selektivität: 97.0 % tra/w-Stilben, 0.5 % cw-Stilben und 2.5 % 1,1-Diphenylethen).
Beispiel 5:
Es wird wie in Beispiel 1 beschrieben vorgegangen, jedoch als Lösungsmittel Isopropanol anstatt NMP verwendet: 60 % Umsatz bezogen auf Brombenzol; die Ausbeute an Heck-Produkten beträgt 55 % (Selektivität: 94.4 % trans-Stilben, 2.4 % cw-Stilben und 2.8 % 1,1-Diphenylethen).
Beispiel 6:
Es wird wie in Beispiel 2 beschrieben vorgegangen, jedoch wird die Katalyse an Luft durchgeführt: 91 % Umsatz bezogen auf Brombenzol; die Ausbeute an Heck- Produkten beträgt 90 % (Selektivität: 92.1 % trαns-Stilben, 2.8 % cw-Stilben und 5.1 % 1,1-Diphenylethen).
Beispiel 7:
Es wird wie in Beispiel 1 beschrieben vorgegangen, jedoch als Lösungsmittel DMA anstatt NMP verwendet: 81 % Umsatz bezogen auf Brombenzol; die Ausbeute an Heck-Produkten beträgt 78 % (Selektivität: 96.0 % trans-Stilben, 0.9 % cis- Stilben und 3.1 % 1,1-Diphenylethen).
Beispiel 8:
Es wird wie in Beispiel 1 beschrieben vorgegangen, jedoch als Lösungsmittel DMF anstatt NMP verwendet: 54 % Umsatz bezogen auf Brombenzol; die Ausbeute an Heck-Produkten beträgt 50 % (Selektivität: 96.0 % trans-Stilben, 0.7 % cis- Stilben und 2.6 % 1,1-Diphenylethen).
Beispiel 9:
Es wird wie in Beispiel 1 beschrieben vorgegangen, jedoch wird kein Dimethylglycin eingesetzt: 40 % Umsatz bezogen auf Brombenzol; die Ausbeute an Heck-Produkten beträgt 26 % (Selektivität: 93.5 % trans-Stilben, 0.7 % cis- Stilben und 5.8 % 1,1-Diphenylethen).
Beispiel 10:
Es wird wie in Beispiel 2 beschrieben vorgegangen, jedoch wird kein Dimethylglycin eingesetzt: 8 % Umsatz bezogen auf Brombenzol; die Ausbeute an Heck-Produkten beträgt 8 % (Selektivität: 83.0 % trαn -Stilben, 8 % ris- Stilben und 9 % 1,1-Diphenylethen).
Beispiel 11 :
Es wird wie in Beispiel 3 beschrieben vorgegangen, jedoch wird kein Dimethylglycin eingesetzt: 36 % Umsatz bezogen auf Brombenzol; die Ausbeute an Heck-Produkten beträgt 29 % (Selektivität: 90.0 % trαns-Stilben, 1.3 % cis- Stilben und 8.7 % 1,1-Diphenylethen).
Beispiel 12:
Es wird wie in Beispiel 2 beschrieben vorgegangen, jedoch werden 4.6 mg (0.045 mmol) Dimethylglycin eingesetzt: 99 % Umsatz bezogen auf Brombenzol; die Ausbeute an Heck-Produkten beträgt 99 % (Selektivität: 95.1 % trans- Stilben, 0.3 % cw-Stilben und 3.8 % 1,1-Diphenylethen).
Beispiel 13:

Beispiel 17:

Nach Zugabe von 40 ml NMP wird das Gefäß verschlossen und das Gemisch 10 Stunden bei 130 °C gerührt: 99 % Umsatz bezogen auf Brombenzol; die Ausbeute an Heck-Produkten beträgt 95 % (Selektivität: 93.3 % trans-Stilben, 0.7 % -Stilben und 6.0 % 1,1- Diphenylethen).
Beispiel 18:
Es wird wie im Beispiel 1 beschrieben vorgegangen, jedoch werden 1.0 ml einer PaUadiumstammlösung (3.8 mg [0.01 mmol] PdCl2(C6H5CN)2/ 20.4 mg [0.2 mmol] Dimethylglycin in 100 ml Methanol/Dichlormethan anstatt PdCl2(C6H5CN)2 und Dimethylglycin an der Luft zugewogen. Das Gemisch wird 24 Stunden bei 130°C gerührt: 98 % Umsatz bezogen auf Brombenzol; die Ausbeute an Heck- Produkten beträgt 97 % (Selektivität: 92.7 % tr ns-Stilben, 0.7 % cw-Stilben und 6.6 % 1,1-Diphenylethen).
Beispiel 19:
Es wird wie im Beispiel 1 beschrieben vorgegangen, jedoch werden 0.1 ml einer PaUadiumstammlösung (3.8 mg [0.01 mmol] PdCl2(C6H5CN)2/ 20.4 mg [0.2 mmol] Dimethylglycin in 100 ml Methanol/Dichlormethan anstatt PdCl2(C6H5CN)2 und Dimethylglycin an der Luft zugewogen. Das Gemisch wird 96 Stunden bei 130°C gerührt: 96 % Umsatz bezogen auf Brombenzol; die Ausbeute an Heck- Produkten beträgt 91 % (Selektivität: 92.9 % trαns-Stilben, 0.7 % cw-Stilben und 6.4 % 1,1-Diphenylethen).
Beispiel 20:

Beispiel 21:

Beispiel 22:

Beispiel 24:
Es wird wie in Beispiel 22 beschrieben vorgegangen, jedoch wird das Gemisch 30 Stunden bei 130°C gerührt: 51 % Umsatz bezogen auf Brombenzol; die Ausbeute an Heck-Produkten beträgt 41 % (Selektivität: 93.0 % trans-Stilben, 1.0 % cis- Stilben und 6.0 % 1 ,1-Diphenylethen).
Beispiel 25:
Es wird wie in Beispiel 22 beschrieben vorgegangen, jedoch wird das Gemisch 30 Stunden bei 150°C gerührt: 70 % Umsatz bezogen auf Brombenzol; die Ausbeute an Heck-Produkten beträgt 65 % (Selektivität: 92.0 % trαn -Stilben, 1.0 % cis- Stilben und 7.0 % 1 ,1-Diphenylethen).
Beispiel 26:
Es wird wie in Beispiel 21 beschrieben vorgegangen, jedoch werden 1.0 ml einer Palladiumstammlösung (2.2 mg [0.01 mmol] Pd(OAc)2 in 100 ml Methanol/Dichlormethan) anstatt Pd(OAc)2 an der Luft zugewogen. Das Gemisch wird 24 Stunden bei 130°C gerührt. 77 % Umsatz bezogen auf Brombenzol; die Ausbeute an Heck-Produkten beträgt 89 % (Selektivität: 92.7 % trαns-Stilben, 0.8 % cw-Stilben und 6.5 % 1,1-Diphenylethen).
Beispiel 27:
Es wird wie in Beispiel 21 beschrieben vorgegangen, jedoch werden 0.1 ml einer Palladiumstammlösung (2.2 mg [0.01 mmol] Pd(OAc)2 in 100 ml Methanol/Dichlormethan) anstatt Pd(OAc)2 an der Luft zugewogen. worin Ar die gleiche Bedeutung hat wie bei Formel III, und X Chlor, Brom, OSO2CH3, OSO2-Tolyl, OSO2CF3, OSO2C4F9 oder N2 +Cl" darstellt, mit Olefinen der Formel II,

worin R1, R2 und R3 die gleiche Bedeutung wie bei Formel III haben, in Gegenwart von Palladiumkatalysatoren, dadurch gekennzeichnet, daß man als Katalysator eine Palladium-II- Verbindung der allgemeinen Formel PdXY oder deren CH3CN- oder C6H5CN-Komplexe, wobei X = Y = Cl. Br, I, NO3, RCO2 (R = C, - C22, CF3, CC13, CH2OCH3, C6H5) oder RSO3 ( R = d - C22, CF3, C F9, CC13, C6H5, p-CH3CόH4), einsetzt, und die Umsetzung in Abwesenheit von Phosphoniumsalzen und Phosphanen in Gegenwart eines stickstoffhaltigen Additivs und eines Lösungsmittels und einer Base bei Temperaturen von 60 °C bis 180 °C durchführt.
2. Verfahren nach Anspruch 1, wobei als stickstoffhaltiges Additiv eine α- Aminosäure verwendet wird.
3. Verfahren nach Anspruch 1, wobei als Additiv die N-alkylierte Form einer α- Aminosäure verwendet wird.
4. Verfahren nach Anspruch 3, wobei als Additiv N,N-Dimethylglycin verwendet wird.
5. Verfahren nach Anspruch 1, wobei als Additiv eine heterocyclische Verbindung verwendet wird.
6. Verfahren nach Anspruch 5, wobei als Additiv 2,2'-Dipyridyl verwendet wird.
7. Verfahren nach Ansprüchen 1 - 6, dadurch gekennzeichnet, dass das Verhältnis von Palladium zum stickstoffhaltigen Additiv 1 : 3 bis 1 : 100 beträgt.
8. Verfahren nach Ansprüchen 1 - 7, dadurch gekennzeichnet, dass die Menge des Palladium-Katalysators 0.001 Mol-% bis 3 Mol-% beträgt.
9. Verfahren nach Ansprüchen 1 - 8, wobei als Lösungsmittel ein dipolares aprotisches Lösungsmittel eingesetzt wird.
10. Verfahren nach Anspruch 9, wobei als dipolares aprotisches Lösungsmittel Dimethylsulfoxid, Dimethylformamid, Dimethylacetamid, 1 ,3-Dimethyl- 3,4,5, 6-tetrahydro-2-(lH)-pyrimidon oder N-Methylpyrrolidon verwendet wird.
11. Verfahren nach Ansprüchen 1 - 8, wobei als Lösungsmittel ein protisches Lösungsmittel verwendet wird.
12. Verfahren nach Anspruch 11, wobei als protisches Losungsmittel Methanol, Ethanol oder Diethylenglykol verwendet wird.
13. Verfahren nach Ansprüchen 1 - 12, wobei als Base ein Amin, ein Alkali- oder Erdalkalimetallsalz einer Carbonsäure oder ein Alkali- oder Erdalkali- Carbonat oder -Bicarbonat eingesetzt wird.
14. Verfahren nach Ansprüchen 1 - 13, dadurch gekennzeichnet, dass die Reaktion im Temperaturbereich von 100 - 140 °C durchgeführt wird.
