Abstract
Description
Mehrkanal-Tropfengeneratoren oder auch Dispensiereinrichtungen, wie sie häufig
im Stand der Technik genannt werden, dienen insbesondere dem Befüllen von
Kavitäten von Mikrotitrationsplatten mit Flüssigkeit.
Sie bestehen in der Regel aus einer Pumpe und einem mit dieser über eine Leitung
verbundenen Verteiler. Die aus dem Stand der Technik bekannten Lösungen
verfolgen das Ziel, mit möglichst geringen Volumenunterschieden Kavitäten zu
befüllen, das heißt, daß an allen Ausgängen des Verteilers zu jeder Zeit das gleiche
Volumen abgegeben wird.
Um diese Aufgabe zu lösen, wurden insbesondere unterschiedliche konstruktive
Lösungen für den Verteiler entwickelt. Häufig ist der Verteiler als Röhrenverteiler
aufgebaut. Dieser verbietet jedoch aufgrund seines Funktionsprinzips eine exakt
gleichmäßige Verteilung.
Ausgänge, deren Entfernung zum Eingang des Verteilers kürzer ist als weiter
entfernte Ausgänge werden bevorzugt durchströmt. Im Patent US 5,334,352 ist
eine Variante eines solchen Verteilers beschrieben. Dort wird mit Hilfe von
Einbauten und gezielten Querschnittsverengungen versucht, dieses Verhalten zu
verbessern. Anordnungen dieser Art sind jedoch auf die Eigenschaften der zu
verteilenden Flüssigkeiten abgestimmt und sind für andersartige nur bedingt
einsetzbar. Die Abgabe kleiner Tropfen V<10µl ist mit dieser Anordnung nur
möglich, indem die abzugebende Flüssigkeit in Kontakt mit einer anderen
Oberfläche kommt, andernfalls löst sich der Tropfen aufgrund der
Oberflächenspannkraft nicht vom Ausgang ab.
Eine Dispensiereinrichtung mit einer weiteren Variante eines Verteilers ist aus
EP 0 180 591 B1 bekannt. Dieser besteht grundsätzlich aus einem horizontalen
Verteilerrohr, von dem aus mindestens zwei Füllrohre zuerst nach oben und dann
nach einem Umlenkbereich in einen Abflußbereich nach unten führen. Im
Umlenkbereich sind die Füllrohre untereinander über ein Ausgleichsrohr verbunden.
Vorteilhafterweise sollte die Leitung zwischen der Pumpe und dem Verteilerrohr aus
elastischem Material sein, um die relativ harten Pumpenstöße etwas ausgleichen
und die Flüssigkeit in den Füllrohren nach Beendigung eines jeden Pumpvorganges
um etwa 1 - 2 mm zurückzuziehen, wodurch ein Nachtropfen sicher verhindert
werden soll. Um Volumina gleicher Menge von ca. 0,5 - 10 µl mehrfach simultan
abgeben zu können, ist diese Dispensiereinrichtung nicht geeignet, da schon eine
geringfügig unterschiedliche Flüssigkeitssäulenhöhe in den Abflußbereichen der
Füllrohre nach dem Zurückziehen der Flüssigkeit zu unterschiedlichen
Abgabevolumina an den einzelnen Auslässen beim nächsten Dosiervorgang führen
kann.
Andere Verteilerstrukturen sind aus der Drucktechnik bekannt, um z.B. Druckfarbe
mit einer homogenen Dicke auf eine Druckwalze aufzubringen. Eine Anordnung
aus dem Patent US 5,441,204 benutzt eine mehrfache Aufspaltung des
Flüssigkeitsstroms, um eine gleichmäßige Verteilung an einem linienförmigen
Ausgang zu erreichen. Bei diesem Prinzip ist eine Aufteilung des Eingangsstromes
erreichbar, bei der jeder Ausgang den gleichen Volumenstrom aufweist,
unabhängig von den fluidischen Eigenschaften der verteilten Flüssigkeit. Die
Abgabe von Tropfen ist durch ein elektrostatisches Prinzip realisiert, indem die
kontinuierlich geförderte Flüssigkeit elektrostatisch aufgeladen und durch ein
elektrisches Feld von der Oberfläche abgelöst wird. Einsetzbar ist ein derartiger
Verteiler nur innerhalb von Anordnungen mit definierten elektrischen Feldern, was

Der erfindungsgemäße Mehrkanal-Tropfengenerator besteht aus den in Fig. 1
schematisch dargestellten Komponenten:
Die Dosiereinrichtung 1 besteht aus einem Vorratsgefäß 5 und einem Dosierer 6,
der die Flüssigkeit vom Vorratsgefäß 5 über die Verbindung 2 in einen Verteiler 3 an
dessen Ausgängen Auslässe 4 angeordnet sind, die eine Abgabe der Tröpfchen
ermöglichen, befördert. Die Flüssigkeitsmenge muß genau einstellbar und mit
hoher Wiederholgenauigkeit gefördert werden. Magnetventile in Verbindung mit
einem Druckgefäß oder Taumelkolbenpumpen sind vorteilhaft einsetzbar. Die starke
Pulsation der Dosiereinrichtungen 1 erzeugt einen Druckimpuls, der über die
Verbindung 2 zum Verteiler 3 und auf die Auslässe 4 übertragen wird und die dort
die Abgabe kleiner Tropfen ermöglicht.
Die Verbindung ist ein vorzugsweise schlauchartiges Bauteil definierter Elastizität
um die Flankensteilheit der Druckimpulse zu verringern.
Der Verteiler 3 ist ein planares Kanalsystem aus kaskadierten T-Stücken mit einem
Eingang 12 und mehreren Ausgängen 13, wie in Fig. 1 und 2 dargestellt, das
beispielsweise in Plattenmaterial eingearbeitet sein kann. Eine derartige Struktur
gewährleistet bei minimalen Totvolumen eine exakt gleichmäßige Verteilung der
durchströmenden Flüssigkeit auf die Ausgänge, unabhängig von deren fluidischen
Eigenschaften. Weiterhin bewirkt der Verteiler 3 eine gleichmäßige Aufteilung der
Druckimpulse vom Eingang auf die Ausgänge. Aufgrund des im Vergleich zu den
Auslässen 4 großen Querschnitts der Kanäle des Kanalsystems hat der Verteiler 3
nur einen vernachlässigbaren Anteil an den dynamischen Eigenschaften des

An den Ausgängen sind Auslässe 4 angeordnet, die eine Querschnittsverengung
darstellen und somit den größten fluidischen Widerstand im Mehrkanal-Tropfengenerator
bilden. Damit die Auslässe 4 einen gleich großen fluidischen
Widerstand bilden, müssen sie eine hohe Fertigungsgenauigkeit aufweisen, die zum
Beispiel bei Präzisionsrohren erfüllt sind. Fertigungstoleranzen innerhalb des
Kanalsystems werden dadurch ausgeglichen. Darüber hinaus besitzt die
Flüssigkeitssäule innerhalb der Auslässe 4 die größte Trägheit im Gesamtsystem und
hat daher wesentlichen Einfluß auf dessen dynamisches Verhalten. Für das
Verständnis des Mehrkanal-Tropfengenerators ist insbesondere für die Abgabe
kleinster Tropfen über die Betrachtung der statischen Parameter eine dynamische
Betrachtung notwendig, die anhand eines elektrischen Ersatzschaltbildes erfolgen
soll. Aus Fig. 1 leitet sich das detaillierte elektrische Ersatzschaltbild ab (Fig. 2), das
sich auf das vereinfachte elektrische Ersatzschaltbild (Fig. 3) reduzieren läßt. Dieses
stellt im wesentlichen einen Reihenschwingkreis mit Stromanregung dar, der mit
den Differentialgleichungen aus der Elektrotechnik beschrieben werden kann. Die
Dosiereinrichtung 1 wird durch die Stromquelle 7 mit Schalter 8 wiedergegeben,
die elastische Verbindung durch die Kapazität 9. Der Verteiler wird vernachlässigt.
Die Auslässe sind als Widerstände 10 und Trägheiten 11 dargestellt.
Bei quasistatischer Betrachtung des Mehrkanal-Tropfengenerators würde sich bei
langsamer Zugabe von Flüssigkeit durch die Dosiereinrichtung 1 an den Auslässen 4
Tropfen bilden, die abreißen, wenn das Gewicht des Tropfens dessen
Oberflächenspannkraft überschreitet. Danach bleibt an den Auslässen 4 ein
sogenannter Meniskus zurück, der zu Ungenauigkeiten führt. Derart erzeugte
Tropfen haben bei Wasser ein Volumen von ca. 10-20µl und entsprechen somit
nicht der Aufgabenstellung.
Es gehört daher zu den erfindungswesentlichen Merkmalen, daß die
Dosiereinrichtung 1 das abzugebende Volumen mit einer hohen
Beförderungsgeschwindigkeit über die Verbindung 2 und den Verteiler 3 zu den
Auslässen 4 befördert. Dort bilden sich Flüssigkeitsstrahlen mit dem Durchmesser
der Auslässe 4. Die Trägheit verhindert eine Tropfenbildung. Wird der
Flüssigkeitsstrom abrupt unterbrochen, reißt der Strahl aufgrund der hohen
kinetischen Energie ab, bevor ein Tropfen entsteht. Es bleibt jedoch auch hier eine
unbestimmte Menge Flüssigkeit an den Auslässen 4 zurück und bildet einen
Meniskus. Um das Ergebnis des folgenden Abgabezyklus nicht zu verfälschen, muß
dieser Flüssigkeitsrest entfernt werden. Dies könnte durch einen inversen Betrieb
der Dosiereinrichtung 1 erfolgen, was aufgrund der hohen Geschwindigkeiten aber
zu einem stark erhöhtem technischen Aufwand führen würde. Diese Aufgabe
erfüllt im erfindungsgemäßen Mehrkanal-Tropfengenerator die als elastisches
Element ausgeführte Verbindung 2, die in der Lage ist, diese Funktion zu
übernehmen. Zu Beginn eines Abgabezyklusses nimmt die Verbindung 2, indem
sich ihr Querschnitt ausdehnt, einen Teil des geförderten Volumens auf und gibt es
im weiteren Verlauf teilweise wieder ab. Wird der Volumenstrom unterbrochen,
bewirkt die Trägheit der Flüssigkeitssäulen in den Auslässen 4 für ein kurzzeitigen
Fortbestand des Volumenstromes. Das strömende Volumen wird von der Elastizität
der Verbindung 2 geliefert, wobei ein Unterdruck entsteht. Kommen die
Flüssigkeitssäulen zum Stillstand, bewirkt dieser Unterdruck ein Zurücksaugen der
Flüssigkeitsreste aus den Auslässen 4.
Der chronologische Ablauf eines Dosierzyklus (dargestellt in Fig. 5, wobei VD den
von der Dosiereinrichtung 1 erzeugten Volumenstrom und VA den an den Auslässen
bewirkten Volumenstrom darstellt) stellt sich, unter der Voraussetzung der
ausgeglichenen Druckverhältnisse vorher, wie folgt dar:
Zum Zeitpunkt t=0 beginnt die Dosiereinrichtung 1 einen Volumenstrom zu
fördern.
Dieser kann aufgrund der Trägheiten in den Auslässen 4 zunächst nicht abfließen
und bewirkt somit eine Druckerhöhung, die zu einer Volumenexpansion der
elastischen Verbindung 2 führt. Die Flüssigkeitssäulen in den Auslässen 4 werden
beschleunigt, und es bilden sich darin Volumenströme aus. Der Druck in der
Verbindung 2 sinkt bis auf ein Niveau ab, bestimmt vom fluidischen Widerstand und
dem Durchfluß, wobei ein Teil des gespeicherten Volumens abgegeben wird. Dies
entspricht dem quasistatischen Zustand.
Bei Unterbrechung des zugeführten Volumenstroms zum Zeitpunkt t=1 bewirken
die Trägheiten zunächst ein Fortbestehen der Strömung, gespeist aus dem Vorrat
des in der Verbindung 2 gespeicherten Volumens, bis dort eine
Volumenkontraktion verbunden mit einem Unterdruck entsteht, der die Strömung
bremst. Dieser Unterdruck bleibt in der Verbindung 2 bestehen, wenn die Strömung
zum Stillstand kommt. Dadurch kehrt sich die Richtung der Strömung um und die
Flüssigkeit wird
von den Auslässen 4 in den Verteiler zurückgesaugt. Der Flüssigkeitsrest an den
Auslässen wird somit entfernt.
Dieses Verhalten des Volumenstromes an den Auslässen VA ist von der Abstimmung
des fluidischen Schwingkreises abhängig.
