Abstract
L'invention concerne une nouvelle utilisation des dérivés du niobium à l'état d'oxydation 4 ou 5 comme principe actif pour la fabrication de médicaments utiles dans le traitement et/ou la prévention des désordres du métabolisme des glucides et/ou des lipides. Les dérivés du niobium utiles selon l'invention sont sous forme de sels minéraux ou de sels ou complexes organométalliques. Les médicaments sont utiles pour le traitement du diabète, de l'hypercholestérolémie, de l'hyperlipidémie, de l'hypertriglycéridémie, et des complications associées à ces pathologies.

05 La présente invention concerne l'utilisation de dérivés du niobium comme principe actif de médicaments utiles dans le trai¬ tement et/ou la prévention des désordres du métabolisme des glu¬ cides et/ou des lipides.
La recherche de nouveaux dérivés hypoglycémiants pour le
10 traitement du diabète est très importante, car, dans l'état actuel, les substances disponibles (insuline, biguanides, chlofibrates, sulfamides, etc.) aident au contrôle de celui-ci via une régula- ' tion de la glycémie, traitent les "effets" du diabète et les troubles du métabolisme qui y sont associés, mais n'en traitent pas
15 les causes inconnues pour la plupart et ne le soignent pas. De plus, ces traitements sont de longue durée, généralement à vie, et contraignants pour l'individu (injections quotidiennes). Ils s'accompagnent donc forcément d'effets secondaires et de nouvelles pathologies apparaissent au cours des années (insulino-
20 résistance, cécité, etc.).
Avec la découverte et l'étude depuis quelques années des oligo-éléments essentiels, certains ions métalliques ont été décrits comme ayant des propriétés insulinomimétiques. C'est le cas tout particulièrement des sels minéraux du vanadium comme cela res-
25 sort de la mise au point bibliographique intitulée "The bio- inorganic Chemistry of Vanadium" effectuée récemment par Dieter REHDER (Angew. Chem. Int. Ed. Engl. 1991, 30., 148-167) de certains dérivés du molybdène et d'autres du chrome (en particulier un pep- tide naturel non encore totalement identifié (nommé GTF, et un
30 dérivé baptisé dithiochrome, cf. W0 91/01738, publié le 21.02.1991). De plus, il a été découvert de nombreuses métallo- . enzymes faisant intervenir le V, Cr ou Mo naturellement, ou en rem¬ placement d'un autre métal manquant ou déficitaire. l'hypothèse actuelle est que les ions métalliques comme
35 le vanadate ont une action biologique importante par analogie avec
les ions phosphate, en particulier car ils ont sensiblement la même taille et Les mêmes Longueurs de liaisons métal-oxygène.
De nombreux travaux ont été effectués concernant l'acti¬ vité du vanadium dans Le traitement de L'hyperglycémie. Il s'agit pour l'essentiel de documents de littérature scientifique où il n'est question que de dérivés minéraux du vanadium.
Dans la Littérature depuis 1985, de nombreuses études décrivent Les effets hypoglycémiants de dérivés minéraux d'ions vanadate (+5) ou vanadyle (+4). Une excellente mise au point bibliographique se trouve dans la Thèse de Doctorat en Pharmacie de Jean-Jacques M0NG0LD, septembre 1991, Université de Montpellier I, France.
Le brevet européen EP-0 264 278 décrit des compositions à activité insulino-mimétique contenant des vanadates et des peroxo- vanadates minéraux.
Par ailleurs, le demanderesse a décrit dans ses demandes internationales WO 91/07406 et WO 91/13892 ainsi que dans sa demande française non encore publiée et déposée Le 21 mai 1991 sous le numéro 91 06 174 des complexes organo- étalliques de métaux de transition à structure porphyrinique utiles notamment dans le traitement de L'hyperglycémie.
Le brevet européen EP-0 305 264 décrit des composés organo inéraux de vanadyle obtenus à partir d'esters en C,-C1n de La cystéine. Le brevet japonais JP 2 292 217 décrit une activité antidiabétique pour 7 complexes vanadyLe-dioxo (tartrate, gluco- nate, alonate, oxalate, lactate, salycilate et acétyl-acétonate), ainsi que pour 2 complexes vanadyle-monoamino-monothio : Le com¬ plexe de L'ester methylique de La cystéine avec Le vanadyLe et celui de la 2-amino-éthane-thiol.
Des sels minéraux du niobium ainsi que quelques dérivés organiques du niobium (cyclopentadiényles et isothiocyanates) ont d'autres utilisations médicales (dentifrice, agent de contraste pour radiographie ou imagerie RMN, ou utilisation en cancérologie). Ils sont donc administrés à des humains et Leur toxicité est g connue. Par exemple. Le Merck Index, 11 Edition, signale que le
chlorure d'oxovanadate (VOCl, produit n° 9834) a une dose létale orale DL-0 = 140 g/kg pour des rats (ce qui fait 41,2 mg de métal/ kg), alors que le chlorure de niobium (NbC produit n 6473) a une dose Létale orale DL-. = 830 mg/kg pour des souris (ce qui fait 285 mg de métal/kg). Il semblerait donc que le niobium ait une toxicité moindre que le' vanadium, qui lui-même est beaucoup moins toxique que le chrome VI.
La synthèse de dérivés ou de complexes du' niobium est bien connue : par exemple elle peut se faire suivant les méthodes répertoriées par Leopold Gmelins, et décrites dans : Handbuch der anorgani schen Chemie, 8 Auflage, Niobium (System Nummer 49), Teit B4, Verbindungen, VerlagCheππe-GMBH, Weinheim/Bergstrasse, 1973, en particulier aux pages 334-472 pour ce qui est des com¬ plexes organiques. Les sels minéraux du niobium peuvent être pré- parés suivant des procédures connues et décrites par exemple dans l'ouvrage ci-dessus, des sulfates de niobium à la page 303.
Des alcoxydes de niobium, des dialkylamides de niobium et des aminoalcoxydes de niobium peuvent également être préparés en suivant par exemple les procédures décrites dans les brevets EP 0 450 717 et EP 0 442 563.
Des complexes de niobium avec des hydroxy acides (comme les acides tartrique, lactique, glycolique et malique), peuvent être préparés en suivant par exemple les procédures décrites par E. RUZDIC et N. BRNICEVIC dans Inorganica Chimica Acta, 1984, ^88, 99-103.
Des complexes peroxo de niobium avec des dérivés orga¬ niques comme les acides tartrique, lactique, glycolique, dipicoli- nique et malique peuvent être préparés en suivant par exemple les procédures décrites par A.C. Dengel et W.P. Griffith, Polyhedron, 1989, , 1371-1377.
A l'heure actuelle, le niobium n'est nullemnet considéré comme un oligo-élément essentiel et n'est pas utilisé pour la pré¬ vention ou le traitement des désordres du métabolisme des glucides et/ou des lipides.
Par ailleurs, l'analogie avec les ions phosphates qui peut être faite, comme on l'a vu ci-dessus dans le cas du vanadium, n'est nullement justifiée dans le cas du niobium.
Donc, rien ne rendait évident que le niobium présente- rait une activité thérapeutique similaire à celle du vanadium dans le traitement du diabète et des maladies associées.
Or, La demanderesse a maintenant découvert que des dérivés du niobium au degré d'oxydation 4 ou 5 avaient une activité comparable à celle des vanadyle et vanadate dans le traitement du diabète et de façon plus large s'avéraient actifs pour le traite¬ ment et la prévention des désordres du métabolisme des glucides et/ ou des lipides et des complications associées à ces pathologies.
La demanderesse a découvert que cette activité pouvait être encore notablement accrue par compLexation desdits dérivés par différents ligands organiques.
D'une façon générale, on entendra par complexe selon l'invention le produit de réaction d'un dérivé du niobium au degré d'oxydation 4 ou 5 avec un ligand organique. Il s'agit, en général, de mélanges de complexes correspondant à La fixation par des Liaisons à 1 et/ou 2 électrons d'un nombre variable d'hétéroatomes suivant le ligand utilisé dont la disposition spatiale autour du métal et le degré de coordination du métal peuvent varier
Les complexes ci-dessus ayant la particularité de se former très facilement par contact, soit en solution, soit en sus- pension, d'un dérivé du niobium au degré d'oxydation 4 ou 5 avec un Ligand organique, l'invention couvre non seulement L'ut lisation desdits complexes déjà formés comme principe actif pour la fabrica¬ tion d'un médicament utile dans le traitement et la prévention des désordres du métabolisme des glucides et/ou des lipides, mais également l'utilisation du mélange des deux types de constituants (dérivé du niobium au degré d'oxydation 4 ou 5 et Ligand organique) dont la compLexation sera réalisée in vivo lors de l'administration de La composition pharmaceutique.
On désignera par "association" aussi bien le mélange des deux types de constituants que le produit de la réaction de com¬ pLexation.
Ainsi, selon une caractéristique essentielle, la. présente invention concerne une nouvelle utilisation des dérivés du niobium à l'état d'oxydation 4 ou 5 comme principe actif pour obtenir un médicament utile dans le traitement des désordres du métabolisme des glucides et/ou des lipides.
Parmi les maladies dont te traitement ou la prévention est particulièrement visé dans le cadre de la présente invention, on citera en tout premier lieu le diabète mais également t'hyper- cholestérolémie, l ' hyperglycéridémie, l ' hyperlipidémie ainsi que les maladies associées à ces pathologies.
A titre d'exemples de pathologies associées, on citera l'hypertension artérielle, l 'arthériosc lérose, l'insuffisance cai— diaque, l'ischémie périphérique ou des pathologies oculaires évo¬ luant vers La cécité. Selon une première variante de l'invention, le niobium est sous la forme d'un dérivé minéral.
Avantageusement, ce dérivé minéral est soluble dans l'eau.
On choisira, de préférence parmi ces sels, des sels donnant des solutions de pH compris entre 3 et 9, de préférence proche de la normalité.
A titre d'exemples de dérivés minéraux utilisables, on citera les oxydes, sulfates, halogénures, chlorhydrates, métallates de sels alcalins ou d'ammonium, ainsi que leurs hydrates ou leurs complexes avec un solvant.
A titre d'oxyde, on citera tout particulièrement Nb_0,-. A titre de chlorure, on citera NbCl, et NbCl,-. Parmi les sulfates préférés, on citera tout particulière¬ ment celui répondant à La formule (NH, ),Nb(S0, ) , . A titre de complexe de ces dérivés minéraux avec un sol¬ vant, on citera par exemple le complexe entre NbCl, et le THF.
Les dérivés organiques utilisables selon l'invention sont tout particulièrement sous la forme de sels ou de complexes dans lesquels le niobium se trouve au degré d'oxydation 4 ou 5. Parmi les sets ou complexes organiques avec le niobium, on citera tout particulièrement ceux avec des dérivés organiques
contenant au moins un hétéro-atome ou une fonction à caractère électro-donneur susceptible de donner un ou deux électrons.
A titre d'hétéro-atome à caractère électro-donneur, on citera L'oxygène, l'azote, Le soufre. A titre de fonctions à caractère électro-donneur, on citera des fonctions acide carboxylique, acide sulfurique, acide phosphorique, aminé, alcool, thiol, éther, •thioéther.
A titre de dérivés organiques préférés, on citera Les complexes du niobium avec des Ligands comportant au moins un doublet électronique libre, de préférence plusieurs, c'est-à-dire avec des dérivés organiques dits polydentates, et en particulier bidentates, tridentates et tétradentates.
Comme famille de complexes préférés, on peut citer en exemple, sans être limitatif, les complexes du niobium avec les aminés, Les polya ines, les alcools, les polyols, les sucres, les polymères de sucres, Les nucléotides, les thioLs, Les amino-acides, les amino-alcools, Les guanines, Les oxymes, Les dicétones-1 ,3, Les acides carboxyliques, phosphoriques, sulfuriques, carboxydi- thioîques, carbamadithioîques, les phénols, Les catéchols, les bases de Schiff, les protéines, les peptides, Les dérivés polyfonc- tionnels contenant des hétéroatomes comme l'oxygène, le soufre, L'azote, des cycles contenant ces hétéroatomes.
Ces dérivés organiques et complexes ont L'avantage de se former aisément par contact dudit ligand avec un dérivé minéral ou un sel organique de niobium à l'état d'oxydation 4 ou 5 soit en solution, soit en suspension, dans un solvant. On se reportera aux exemples de synthèse donnés à titre d'exemples ilLustratifs mettant en évidence La simplicité de formation des complexes et sels orga¬ niques du niobium où le niobium est un degré d'oxydation 4 ou 5, utiles selon l'invention.
Comme dérivé du niobium utile pour la préparation des complexes ou association utiles selon la présente invention, on citera les oxydes, halogénures, en particulier chlorure, oxy- halogénures, sulfate, étallate de métal alcalin ou d'ammonium, acétate, acétylacétonate ainsi que Leurs dérivés sous forme d'hydrate ou de complexe avec un solvant.
A titre d'exemples de sels et complexes utilisables selon l'invention, on citera les oxalate, tartrate, alcoolate, .catécho- late.
Parmi tes familles de complexes ou association préférés selon l'invention, on citera : a) les complexes ou associations du niobium à l'état d'oxydation 4 ou 5 avec l'inositol, un phosphate d'inositot ou un de leurs dérivés :
Plus précisément, ces complexes ou associations sont obtenus par mélange ou réaction de 0,1 à 6 moles, de préférence 1 à 3 moles d'un dérivé du niobium au degré d'oxydation 4 ou 5 tel que défini ci-dessus pour -1 mole d'un dérivé d'inositot correspondant à la formule :

Le dérivé du niobium sera au degré d'oxydation 4 ou 5 et avantageusement sous la forme d'un oxyde, d'un halogénure, d'un oxyhalogénure, d'un sulfate, d'un métallate de métal alcalin ou d'ammonium, d'un acétate, d'un acétylacétonate, ledit dérivé pouvant être sous forme d'un hydrate ou d'un complexe avec un solvant . L'acide aminé est avantageusement choisi parmi les acides aminés naturels.
Selon une variante avantageuse, on choisit l'acide aminé parmi les acides aminés naturels présentant une chaîne latérale contenant des hétéroatomes. Ainsi, on choisira avantageusement l'acide aminé dans le groupe constitué du tryptophane, de la lysine, de la tyrosine, de la serine, de la thréonine, de l'arginine, de l'acide aspartique, de la glutamine, de l 'asparagine, du glutamate, de l'aspartate. Ces acides aminés peuvent donc coordiner ou comptexer soit un deuxième ion métallique, soit le même.
Parmi les acides aminés avantageusement choisis, on trouve également la proline.
Les dérivés d'acides aminés utiles selon l'invention sont les dérivés d'acides aminés sous la forme d'esters, d'amides ou d'hydroxamates, c'est-à-dire des dérivés d'acides aminés dans lesquels au moins une fonction acide carboxylique ou a iné libre a été transformée par des réactions bien connues de l'homme de l'art. Les dérivés d'acide aminé utilisés pour former les com¬ plexes du nobium utiles selon l'invention se trouvent avanta- geusement sous l'une des formes suivantes :

A titre d'exe pLes de tels groupements, on citera des groupements méthyle, éthyle, tinoléyle, gamma-tinolényle, oléyle, phényle, benzyle, ricinoléyte.
Selon une autre variante, te dérivé d'acide aminé est sous la forme d'un monoamide et l'un des groupements R' ou R" ou

Selon une autre variante, te dérivé -d'acide aminé est un ester de sucre, on citera par exempte les esters obtenus par condensation d'un glucose ou d'un fructose sur la fonction acide d'un a inoacide.
Selon une autre variante de l'invention, le dérivé d'acide aminé est sous la forme d'un hydroxamate. ' Selon une autre variante, le dérivé d'acide aminé est une aminé secondaire ou tertiaire. Parmi ces aminés secondaires ou tertaires, on citera tout particulièrement celles pour Lesquelles

Cette réaction se fait Le plus souvent à température ambiante ou à une température modérée, généralement inférieure à 50°C.
Cette réaction peut être réalisée soit en solution dans l'eau, soit dans un solvant.
A titre de solvant,- on citera tes mélanges hydro- alcooliques, par exemple les mélanges contenant 100 à 70 % d'eau pour 0 à 30 % d'alcool.
L'alcool entrant dans la constitution du milieu hydro- alcoolique est avantageusement choisi parmi les alcools en C.-C, ι 4 linéaires ou ramifiés, avantageusement l'alcool éthylique.
On peut également réaliser la réaction en suspension dans un agent organique. A titre d'exemple, on citera le THF, le DMF,' le dichloro éthane. Les complexes organo- étal tiques décrits précédemment sont utilisés pour ta préparation de médicaments utiles dans le traitement ou la prévention de maladies telles que le diabète, l' hypercholestérolémie, l ' ypertriglycéridémie," l 'hyperlipidemie ainsi que des complications associées à ces pathologies. Ces complexes sont également utiles pour te traitement et/ou la prévention de l ' insutino-rési stance associée ou non à une forme quelconque de diabète.
Comme on l'a dit précédemment, le niobium se trouve au degré d'oxydation 4 ou 5 dans Les dérivés minéraux ou organo- métalliques utiles selon l'invention.
Selon une variante particulièrement avantageuse, il s'y trouve au degré d'oxydation 5.
Par ailleurs, la demanderesse a constaté de façon tout à fait surprenante que l'utilisation d'un mélange d'un dérivé minéral ou d'un complexe de niobium utile selon l'invention avec un dérivé minéral ou un complexe d'un dérivé du vanadium au degré d'oxyda¬ tion 4 ou 5 permettait d'obtenir des effets antidiabétiques à des doses plus faibles que ce que L'on pourrait attendre compte tenu de l'activité de chacun des deux dérivés ou complexes. L'invention concerne donc également l'utilisation d'un mélange d'un sel ou com¬ plexe du niobium au degré d'oxydation 4 ou 5 et d'un sel ou corn-
plexe du vanadium au degré d'oxydation 4 ou 5, comme matière active pour la fabrication d'un médicament utile pour le traitement ou la prévention des désordres du métabolisme des glucides et/ou des lipides, notamment du diabète. A titre de mélange synergique, on citera par exemple, le mélange des complexes du niobium et du vanadium avec la proline, plus particulièrement celui d'un mélange d'un complexe de la pro¬ line avec te vanadium 4 et d'un complexe de la proline avec le nio¬ bium 5. La présente invention concerne donc l'utilisation des complexes décrits précédemment ou du mélange de leurs deux constituants non encore complexés mais susceptibles de se comptexer dans l'organisme après administration de la composition pharma¬ ceutique pour préparer un médicament utile dans le traitement et/ou la prévention du diabète, de L' hypercholestérolémie, de l'hyper- lipidé ie et les complications associées à ces pathologies ainsi que de L' însulino-résistance associée ou non à une forme de diabète.
Les compositions pharmaceutiques renfermant Les complexes ou associations de L'invention pourront se trouver sous différentes formes. Les dérivés du niobium, sous forme de sels, complexes ou mélanges de L'invention seront incorporés dans un excipient, véhi¬ cule, ou support pharmaceutiquement acceptable.
Comme excipients, véhicules ou supports pharmaceutique- ment acceptables, on peut utiliser tout excipient, véhicule ou sup¬ port bien connu de l'homme de l'art. On peut citer par exemple, et de façon non limitative Le Lactose, L'amidon de mais, le glu¬ cose, la gomme arabique. L'acide stéarique ou le stéarate de magnésium, La dextrine, Le mannitol. Le taLc ou une huile d'origine naturelle riche en acides gras insaturés essentiels, etc. En parti¬ culier, si cela s'avère éventuellement nécessaire, on peut utiliser d'autres additifs biens connus de L'homme de l'art tels que des stabilisants, des desséchants, des Liants, des tampons de pH, etc. Un avantage des complexes décrits précédemment est qu'ils pourront être utilisés directement sans purification préalable
autre que l'élimination du solvant, en particulier lorsqu'il s'.agit d'un solvant organique.
Les compositions pharmaceutiques peuvent être admi- - nistrées de différentes manières, en particulier par voie orale, permuqueuse (linguale, nasale, oculaire). Elles peuvent être égale¬ ment sous forme injectable et destinées à une injection sous- cutanée, intramusculaire ou intraveineuse.
Elles peuvent également être utilisées en application locale, par exemple sous forme de patchs. Les associations organo-métal tiques représentent avan¬ tageusement de 5 à 80 % en poids par rapport au poids total de la préparation pharmaceutique, et se trouvent incorporées dans un excipient, véhicule ou support pharmaceutique acceptable et compa¬ tible avec te mode d'administration envisagée. Les compositions pharmaceutiques selon l'invention lors¬ qu'elles sont destinées à une administration orale contiennent avantageusement de 10 à 5 000 nanomoles de dérivé métallique par dose.
Les compositions sous forme de formulations injectables contiennent avantageusement de 20 à 10 000 nanomoles de dérivé métallique par dose injectable de 1 à 10 ml.
Les compositions destinées à une application locale, en particulier sous forme de patch, contiennent avantageusement de 0,1 à 100 micromoles de dérivé métallique par dose applicable. Les exemples suivants sont donnés à titre purement illus- tratif et nullement limitatif de L'invention. Ils mettent en évi¬ dence La facilité de préparation des sels et complexes utiles selon l'invention, l'utilisation des différents dérivés du niobium pour préparer des compositions pharmaceutiques et l'efficacité de la matière active.
Exemple 1
Préparation d'un oxalate de niobium :
On prépare une solution aqueuse concentrée de Nb.,0,- à Laquelle on ajoute une solution saturée et chaude de monooxalate de sodium NaHC_0,. En refroidissant il précipite du monooxalate de
sodium qu'on filtre. On abandonne ensuite cette solution sous la hotte aspirante jusqu'à ce qu'environ les deux tiers de L'eau se soient évaporés. Les cristaux incolores apparus sont filtrés, lavés à L'alcool et séchés sous vide.
Exemple 2
Préparation d'un tartrate de niobium :
On prépare une solution aqueuse de Nb^Ot- à laquelle on ajoute une solution de monotartrate d'ammonium C,Hg0,N. On chauffe ensuite cette solution à 85 C durant 2 h en Laissant l'eau s'éva¬ porer. En refroidissant par un bain eau-glace rien ne précipite. La solution est alors concentrée sous vide jusqu'à 10 ml environ, et précipitée par addition d'alcool. On filtre et lave Le précipité à L'alcool, puis on le sèche sous vide.
Exemple 3
Préparation d'un alcoolate de niobium :.
On coule une solution de 5 mmol de NbCL,:THF dans du THF anhydre sous azote, sur une suspension dans de l'hexane, de 20 mmol de l 'alcoolate de lithium du phytol, alcoolate préparé in situ pré¬ céderont par addition à 0 C de 20 mmol de nBuLi sur 20 mmol de phytol. On chauffe ensuite cette solution à 45 C durant 2 h. Les solvants sont ensuite évaporés sous vide et te produit séché à 40 C à la pompe à palettes.
Exemple 4
Préparation d'un complexe mixte d'éthanolate de niobium et de pro- linol :
On prépare une solution de 2 mmol de Nb(OEt),- dans du toluène à laquelle on ajoute une solution de 4 mmol de 2- pyrroLidine méthanoL dans du toluène. On chauffe ensuite cette solution à 85 C durant 5 h. En refroidissant par un bain eau-glace rien ne précipite. La solution est alors évaporée sous vide à cette température.
Exemp le 5
Préparation d'un catéchotate de niobium :
Une solution de 2,5 mmol de NbCl,- dans du toluène anhydre est chauffée au reflux sous azote, puis on y ajoute de 20 mmol de catéchot recristallisé. On chauffe ensuite cette solution à 85 C durant 16 h. En refroidissant il précipite des aiguilles rouges. On place le tout au froid (+6 C) durant 48 h, puis on filtre Le préci¬ pité, qu'on lave à l'hexane, puis sèche sous vide.
Exemple 6
Préparation d'un complexe de niobium et de pyridine :
On additionne un excès de pyridine à une solution de 5 mmol de NbCl,:THF dans de l'éthanol absolu. On Laisse sous agita¬ tion toute La nuit, puis on refroidit à 0 C et on- filtre le préci- pité rouge qui se forme. Ce précipité est séché sous vide à 30 C.
Exemple 7
Préparation d'un dérivé de niobium avec l'acide diéthylcarbamadi- thioîque : On fait réagir 1,25 mmol NbCl,:THF avec 5 mmol de
Et?NCS-,Na dans 35 ml d'acétonitri le durant 2 h, puis on évapore le solvant sous vide. Le composé pourpre foncé obtenu est séché à 50 C sous vide de la pompe à palettes.
Exemple 8
Préparation d'un complexe de niobium et de diéthylamine :
On additionne 4 équivalents de diéthylamine à une solu¬ tion aqueuse de Nb_0ς (fraîchement préparé). On laisse sous agita¬ tion 4 h, puis on évapore l'eau à 70 C au rotavapor.
Exemple 9
Préparation d'un complexe de niobium et d'otéylamine :
On additionne 2 équivalents d'une solution alcoolique d'otéylamine à une solution aqueuse de Nb_0,-. On laisse sous agita- tion une nuit, puis on évapore l'eau à 45 C sous vide.
Exemple 10
Préparation d'un dérivé de niobium avec un aminodiol :
On fait réagir 2 équivalents d'une solution aqueuse d'hydroxy-2,3 propylamine avec une solution aqueuse de Nb_0--. On porte au reflux durant une nuit, puis on évapore L'eau à 60 C sous vide de la trompe à eau. Le composé obtenu est ensuite séché à 55 C sous vide de la pompe à palettes.
Exemple 11 Préparation d'un complexe de proline avec du niobium 5 :
Dans une solution de THF anhydre et sous azote, on sus¬ pend 25 millimoles de L-proline (2,875 g), puis on ajoute en une seule fois 5 millimoles de NbC (1,35 g). La solution jaune qui en résulte devient de plus en plus foncée très rapidement (orange, puis rouge, puis brune, puis noire avec des reflets bleus) au fur et à mesure que la proline de dissout. Puis elle se décolore très rapidement jusqu'à devenir très légèrement jaune. On ajoute un peu de méthanol, puis on évapore les solvants sous vide (trompe à eau) en chauffant à 40 C. On parfait Le séchage sous vide de La pompe à palettes, à 75 C. Il en résulte une très fine pellicule jaune-clair sur Le ballon, pratiquement indécollable. Pour récupérer le pro¬ duit, on le dissout dans 50 ml d'eau et on le transvase dans un flacon, ou il se solidife en une sorte de gélatine blanche.
Exemple 12
Préparation d'un sel de proline avec du niobium 4 :
Dans une solution de THF anhydre et sous azote, on sus¬ pend 20 millimoles de L-proline (2,3 g), puis on ajoute en une seule fois 5 millimoles du complexe du chlorure de niobium avec Le THF soit NbCL.rTHF (1,895 g). La solution rouge-ocre qui en résulte devient rapidement de plus en plus foncée (brune-noire avec des reflets jaune-vert). On ajoute un peu de méthanol, puis on évapore les solvants sous vide (trompe à eau) en chauffant à 40 C. Il en résulte un produit gommeux foncé très hygroscopîque.
Exemp t e 13
Préparation de complexes du pyrazole avec le niobium 5 :
On ajoute 2,5 mmol (0,675 g) de NbCl-. dissous dans 15 ml de THF à une solution de THF anhydre sous azote contenant 0,851 g (12,5 mmol) de pyrazole. Le mélange obtenu se colore en jaune foncé très rapidement. Après 2 h d'agitation, on évapore te solvant, puis sèche à la pompe à palettes à 45 C durant 3 h.
Exempte 14 Préparation de complexes de la cytosine avec te niobium 5 :
5 mmol de cytosine (0,556 g) sont mises en suspension dans 20 ml de THF anhydre sous azote, puis on ajoute 15 ml d'une - solution de THF contenant 2,5 mmol de NbCl,- (0,675 g). Tout te pré¬ cipité blanc en suspension se colore petit à petit en jaune. Après 3 h, on ajoute du méthanol, te mélange se trans¬ forme alors en un gel blanc.
Exemple 15
Préparation de complexes 1/1 d'inositot hexaphosphate (acide, phytique) et de niobium :
Dans 10 ml de THF anhydre où l'on a suspendu 2,5 mmol du sel de sodium de l'acide phytique, on additionne 2,5 mmol de penta- chlorure de niobium Nb(Cl)-. (0,675 g) dissous dans 15 ml de THF. La solution jaune devient blanche et il demeure une poudre blanche en suspension. Après 2 h d'agitation, on ajoute 15 ml de MeOH. Puis on évapore tes solvants au rotavapor.
Exemple 16
Formulation dans n'importe quel diluant ou excipient pharmaceuti- quement acceptable : a) Exempte de composition pour administration par voie orale :
Un complexe de métal préparé suivant l'un quelconque des exemples 1 à 15 est mélangé dans un broyeur, à raison de 10 à
5 000 nanomoles de métal (de préférence de 20 à 250 nanomoles), à un polymère de sucre, par exemple du dextran ou de La cellulose, avec du stéréate de zinc, en quantités suffisantes pour réaliser un
comprimé de 100 milligrammes, qui est ensuite enrobé de gomme ara¬ bique et de sorbîtan monostérate. Avantageusement, on peut préparer des gélules gastrorésistantes à bases de gélatine, de dioxyde de titane et d'ester de cellulose.
b) Exemple de composition pour administration par injection :
Un complexe de métal préparé suivant l'un quelconque des exemples 1 à 15 et qui est soluble dans l'eau, est dissous à raison de 20 à 10 000 nanomoles de métal (de préférence de 50 à 500 nano- moles), dans 1 à 10 millilitres d'eau désîonisée stérile (de préfé¬ rence de 2 à 4 mi LLi litres) , puis il est ajusté en pH et en sels minéraux de manière à être le plus proche possible du sérum physio¬ logique (pH 7,4 et 9 g/L), par des acides des bases, ou des sels minéraux constitués d'éléments qu'on trouve dans le sang, de préfé- rence H+, H0~, Na+, Cl".
c) Exemple de composition pour application Locale :
Un complexe de métal préparé suivant l'un quelconque des exemples 1 à 15 précédents est émuLsifié, micronisé ou suspendu à raison de 0,1 à 100 micromoles de métal (de préférence de 1 à
20 micromoles), dans une émulsion d'eau, de glycérol, de mono-, di- et triglycérides d'acides gras, et d'alcools gras.
Exemple 17 Détermination d'un effet hypoglycémiant :
L'injection de 60 mg/kg de streptozotocine par voie intraveineuse chez le rat provoque, dans les 24 h suivant l'injec¬ tion. L'induction d'un diabète sucré stable, irréversible et sen¬ sible à l 'insuline. L'hyperglycémie consécutive peut être réduite par L'administration de substances à propriétés hypoglycémiantes.
a) Protocole
Les tests ont été réalisés sur des rats mâles de souche Wistar (6 par lot), provenant du centre d'élevage de la Faculté de Pharmacie de Montpellier.
Les animaux sont maintenus en observation pendant 4 j avant le début des essais. En début d'essai les animaux pèsent en moyenne 180 g. Pendant ta période d'observation tes animaux, répartis par cage de 3, reçoivent nourriture et eau de boisson a_d_ libitum et sont soumis à une température comprise entre 21 et 23 C et à un cycle jour/obscurité de 12 h.
Dans tous les essais, le diabète a été induit par injec¬ tion de streptozotocine SIGMA en tampon citrate à pH 4,5.
Les animaux d'un poids moyen de 180 g sont anesthésiés à t'éther. Une injection intraveineuse de streptozotocine est pra¬ tiquée au niveau de la veine du pénis ; une glycémie de contrôle est effectuée 72 h après l'administration de la streptozotocine ; seuls les animaux présentant une glycémie supérieure ou égale à 3 g/l (en moyenne 3,8 g/l) sont sélectionnés et soumis au traite- ment par la substance testée.
Les dérivés et complexes du niobium ont été administrés dans les conditions suivantes d'injection : on a administré aux rats des doses correspondant à 120 micromoles de niobium par kg les deux premiers jours et 60 micromoles par kg les 13 jours suivants.
Le contrôle de la glycémie est effectué te matin à 9 h, moment de ta journée où ta glycémie des rats diabétiques témoins ou traités, soumise à de grandes variations durant la journée, est la plus élevée. Des rats diabétiques ne reçoivent que l'excipient (sérum physiologique) et servent de témoins diabétiques ; enfin des rats "témoins blancs" de même poids, n'ayant pas reçu d'injection de streptozotocine sont conservés afin de comparaison des consom¬ mations d'eau et de nourriture.
b) Résultats
Le contrôle quotidien de la glycémie ne nous a pas permis de mettre en évidence L'effet hypoglycémiant durant les deux pre¬ miers jours du traitement. Une chute importante et significative de la glycémie par rapport à celle des rats témoins apparaît dès Les jours suivants et se maintient dans La plupart des cas même après arrêt du traitement. Celle-ci est accompagnée d'une réduction éga-
lement significative de La polyphagie et de La polydipsie, qui elles se manifestent dès les premiers jours. Les consommations de nourriture et d'eau recouvrent des valeurs tendant à se rapprocher de celles des rats témoins non diabétiques. On donne dans les figures jointes en annexe les consomma¬ tions d'eau, de nourriture et L'évolution de la glycémie des rats traités par différents dérivés du niobium.
Les résultats sont représentés, pour chaque dérivé, sous forme de diagramme-barre ; La barre noire correspondant aux résultats obtenus avec les témoins diabétiques et La barre hachurée correspondant aux résultats obtenus avec Les rats traités par des dérivés utiles selon l'invention.
Les figures 1a, 1b et 1c représentent respectivement les variations de consommation d'eau, de nourriture et les variations du taux de glycémie au cours du temps pour un lot de rats diabé¬ tiques traités par Le sulfate de niobium répondant à la formule (NH,),Nb(S0,), et référencé "sulfate" sur Les figures, en compa¬ raison avec les résultats obtenus pour un lot de rats diabétiques non traités. Les figures 2a, 2b et 2c représentent respectivement les variations de consommation d'eau, de nourriture et les variations du taux de glycémie au cours du temps pour un Lot de rats diabé¬ tiques traités par Le produit de L'exemple 4 en comparaison avec les résultats obtenus pour un lot de rats diabétiques non traités. Les figures 3a, 3b et 3c représentent respectivement les variations de consommation d'eau, de nourriture et les variations du taux de qlycémie au cours du temps pour un Lot de rats diabé¬ tiques traités par le produit de l'exemple 7 en comparaison avec les résultats obtenus pour un Lot de rats diabétiques non traités. Les figures 4a, 4b et 4c représentent respectivement Les variations de consommation d'eau, de nourriture et les variations du taux de glycémie au cours du temps pour un Lot de rats diabé¬ tiques traités par le produit de L'exemple 11 en comparaison avec les résultats obtenus pour un Lot de rats diabétiques non traités. Les figures 5a, 5b et 5c représentent respectivement les variations de consommation d'eau, de nourriture et les variations
du taux de glycémie au cours du temps pour un lot de rats diabé¬ tiques traités par le produit de l'exemple 13 en comparaison avec les résultats obtenus pour un lot de rats diabétiques non traités.
Les figures 6a, 6b et 6c représentent respectivement les variations de consommation d'eau, de nourriture et les variations, du taux de glycémie au cours du temps pour un lot de rats diabé¬ tiques traités par le produit de l'exemple 14 en comparaison avec les résultats obtenus pour un lot de rats diabétiques non traités.
Les figures 7a, 7b et 7c représentent respectivement Les variations de consommation d'eau, de nourriture et les variations du taux de glycémie au cours du temps pour un lot de rats diabé¬ tiques traités par te produit de t'exempte 15 en comparaison avec Les résultats obtenus pour un lot de rats diabétiques non traités.
On notera que Les valeurs données dans ces figures sont des moyennes obtenues pour l'ensemble d'un lot.
Comme dans certains cas, un ou deux animaux du Lot ne sont pas corrigés, ces moyennes sont légèrement supérieures aux paramètres correspondants des sujets sains.
On a constaté une très nette diminution de la consomma- tion d'eau et de nourriture accompagnée d'une chute importante de la glycémie.
Par ailleurs, L'évolution pondérale ainsi que celle du ratio consommation de nourriture/prise de poids mettent clairement en évidence un effet hypoglycemiant qu'aucun traitement actuel du diabète ne procure puisqu'en particulier il se prolonge dans la majorité des cas après arrêt du traitement.
Par ailleurs, il est à noter que l'observation des para¬ mètres subjectifs, tels que l'aspect des animaux et leur comporte¬ ment, montre éqalement l'effet positif du traitement. La mesure du taux de cholestérol et de triglycérides met également en évidence une amélioration.
Tout comme L'homme, les rats diabétiques développement des désordres métaboliques (hypercholestérolémie, hyperlipidé ie, hypertriglycéridémie...) ainsi que des pathologies telles qu'hyper- tension artérielle, arthériosc lérose, insuffisance cardiaque,
ischémie périphérique, ou encore des pathologies oculaires évoluant vers la cécité.
On a montré que sur des rats traités, devenus normogtycé- îques, n'apparaissaient pas Les désordres et complications ci- dessus décrits.
Claims
1. Utilisation des dérivés du niobium à l'état d'oxyda¬ tion 4 ou 5 comme principe actif pour la fabrication de médicaments utiles dans le traitement et/ou la prévention des désordres du métabolisme des glucides et/ou des lipides.
2. Utilisation selon La revendication 1, caractérisée en ce que Le dérivé du niobium à L'état d'oxydation 4 ou 5 est un dérivé minéral.
3. Utilisation selon la revendication 2, caractérisée en ce que ledit dérivé minéral est soluble dans l'eau.
4. Utilisation selon la revendication 3, caractérisée en ce que te dérivé minéral soluble dans l'eau conduit à des solutions de pH compris entre 3 et 9, de préférence voisin de 7.
5. Utilisation selon la revendication 1, caractérisée en ce que te niobium est sous forme d'un dérivé organique où te niobium est au degré d'oxydation 4 ou 5,' ledit dérivé étant un set organique ou un complexe organique.
6. Utilisation selon la revendication 5, caractérisée en ce que ledit dérivé organique contient au moins un hétéro-atome ou une fonction à caractère électro-donneur.
7. Utilisation selon La revendication 5 ou 6, caracté¬ risée en ce que Le dérivé organique est un complexe avec un ligand portant au moins un doublet électronique libre.
8. Utilisation selon l'une des revendications 5 à 7, caractérisée en ce que le dérivé du niobium est un sel ou un com¬ plexe avec un produit organique appartenant à La famille des aminés, polya ines, alcools, polyols, sucres, polymères de sucres, nucléotides, thiols, amino-acides, amino-alcools, guanines, oxymes, dicétones-1,3, acides carboxyliques, phosphoriques, sulfuriques, carboxydithioïques, carbamadithioîques, phénols, catéchols, bases de Schiff, protéines, peptides, dérivés polyfonctionnels contenant des hétéro-atomes comme L'oxygène, Le soufre, l'azote, des cycles contenant ces hétéro-atomes.
9. Utilisation selon la revendication 1, caractérisée en ce que le dérivé du niobium est complexé par un ligand organique ou mélangé à un tel Ligand, ledit mélange étant susceptible de se comptexer dans L'organisme après administration du médicament.
10. Utilisation selon la revendication 9, caractérisée en ce que le ligand est l'inositoL, un phosphate d'înositoL ou un de Leurs dérivés.
11. Utilisation selon la revendication 9, caractérsée en ce que Ligand est un acide aminé ou un dérivé d'acide aminé.
12. Utilisation selon la revendication 9, caractérisée en ce que le ligand est un composé organique contenant un cycle renfermant au moins deux atomes d'azote.
13. Utilisation selon La revendication 9, caractérisée en ce que le Ligand est Le pyrocatéchol ou un de ses dérivés.
14. Utilisation selon la revendication 9, caractérisée en ce que le ligand est un dérivé de l'acide dithiocarbamique ou de ses sels.
15. Utilisation selon l'une des revendications 1 à 14, caractérisée en ce que le dérivé du niobium est mélangé à un sel ou complexe du vanadium est au degré d'oxydation 4 ou 5.
16. Utilisation d'un mélange de complexes du niobium au degré d'oxydation 4 ou 5 avec La proline et de complexes du vana¬ dium au degré d'oxydation 4 ou 5 avec la proline comme principe actif pour La fabrication d'un médicament utile pour Le traitement du diabète.
17. Utilisation selon l'une des revendications 1 à 16, caractérisée en ce que Ledit médicament est utile pour Le traite¬ ment du diabète.
18. Utilisation selon l'une des revendications 1 à 16, caractérisée en ce que Ledit médicament est utile pour le traite¬ ment ou La prévention du diabète, de L'hyperchoLesteroLemie, de l'hyperlipidémie ou de l1 hypertrîglycéridéππ'e des complications associées à ces pathologies.
19. Utilisation selon l'une des revendications 1 à 16, caractérisée en ce que ledit médicament est utile pour le traitement et/ou la prévention de L' insulino-résistance.
20. Utilisation selon l'une des revendications 1 à 19, caractérisée en ce que le niobium est au degré d'oxydation 5.
