Abstract
Description
Die Erfindung betrifft enzymatisch unterstützte Äscher- und Beizverfahren wobei alkalische Lipasen, vorzugsweise in Kombination mit proteolytischen Enzymen zur Anwendung kommen.The invention relates to enzymatically supported liming and pickling processes in which alkaline lipases, preferably in combination with proteolytic enzymes, are used.
Der gezielte Einsatz von Enzymen bei der Lederherstellung nahm seinen Anfang mit der Einführung der enzymatischen Beize durch Dr. Otto Röhm im Jahre 1907 (DE-PS 200 519). Seit dieser Zeit wurde - auf dem Hintergrund eines zunehmenden oekologischen Bewußtseins - die Anwendung von Proteasen bei verschiedenen Teiloperationen der Wasserwerkstatt vorgeschlagen und auch praktisch verwirklicht (vgl. E. Pfleiderer und R. Reiner in Biotechnology Ed. H.-J. Rehm S, 729 - 743, VCH 1988). Auch Amylasen, insbesondere in Kombination mit Proteasen haben ebenfalls Eingang in das Beiz-Verfahren der Wasserwerkstatt gefunden (US-A 4 273 876). Die gleichzeitige Anwendung von Lipase und Amylase (in Form des Pankreatins) in Anwesenheit von Desoxycholsäure ist aus dem ungar. Patent 3325 (Chem. Abstr. 77, 7341 k) bekannt. Naturgemäß bietet sich die Verwendung von Lipasen zur Entfettung von Häuten und Fellen, insbesondere der stark fetthaltigen Schweinshäute und Schaffelle und von Abfällen an. Allerdings stehen Anwendungsempfehlungen bei der Entfettung (z.B. L.H. Posorske J. Am. Oil Chem. Soc. 61 (11) 1758 - 1760 (1984); K. Yeshodha et al. Leather sci (Madras) 25 (2) 77 - 86 (1978), Chem. Abstr. 89, 199097; T. Nielsen Fette, Seifen, Anstrichm. 87 (1) 15 - 19 (1985) auch negative Erfahrungen z.B. bei gepickelten und entkälkten Schafsblößen gegenüber (vgl. A. Vulliermet et al. Technicuir 16 (4) 64 - 76 (1982), Chem. Abstr. 97, 57467 q; Chem. Abstr. 82, 113205g). In der letzteren Literaturstelle wird ein enzymatischer Fettabbau mittels Lipasen bzw. lipasehaltiger Enzympraeparationen in einem pH-Bereich unterhalb 8, vorzugsweise im mäßig sauren pH-Bereich in Erwägung gezogen.
In "Biotechnology" Ed. H.-J. Rehm Bd. 7a loc.cit S. 644 wird angemerkt, daß mikrobielle und pankreatische Lipasen (E.C.3.1.1.3) nicht als Waschmittel-Enzyme angewendet werden können, wegen der notorischen Instabilität unter alkalischen Bedingungen, ganz abgesehen vom Preis derselben. Gegen eine gemeinsame Anwendung von Lipasen und Proteasen spricht von vorneherein die Abbauwirkung von Proteasen gegenüber Proteinen, wie sie die Lipasen darstellen.
In jüngster Zeit wird ein enzymatisch unterstütztes Weichverfahren für Häute und Felle empfohlen bei dem die Weichflotten
- A) Lipasen mit einem Wirkungsoptimum im pH-Bereich 9 bis 11
- B) Proteasen mit Wirksamkeit im pH-Bereich 9 - 11 und
- C) Grenzflächenaktive Agentien enthalten, wobei der pH-Wert der Weichflotte im Bereich 9 - 11 liegt (vgl. deutsche Patentanmeldung P 39 22 748.0).
In "Biotechnology" Ed. H.-J. Rehm Vol. 7a loc.cit p. 644 notes that microbial and pancreatic lipases (EC3.1.1.3) cannot be used as detergent enzymes because of the notorious instability under alkaline conditions, not to mention the price thereof. From the outset, the degradation effect of proteases against proteins, as represented by the lipases, speaks against the joint use of lipases and proteases.
Recently, an enzymatically supported softening process for hides and skins has been recommended in which the soft liquors are used
- A) Lipases with an optimum effect in the pH range 9 to 11
- B) Proteases with activity in the pH range 9-11 and
- C) Containing surface-active agents, the pH of the soft liquor being in the range 9-11 (cf. German patent application P 39 22 748.0).
Als besonders geeignet werden dabei aus Aspergillus-Arten gewonnene und speziell gewisse, genetisch veränderte Stämme befunden, beispielsweise eine alkalische Lipase aus einem durch Rekombination gewonnenen Aspergillus-oryzae-Stamm mit ausgeprägtem Aktivitäts-Optimum zwischen pH 9 und 11 sowie eine unter der Bezeichnung "LIPOLASE 100 T" im Handel befindliche Lipase (NOVO INDUSTRI A/S, DK 2880 Bagsvaerd).Especially suitable are certain genetically modified strains obtained from Aspergillus species, for example an alkaline lipase from an Aspergillus oryzae strain obtained by recombination with a pronounced optimum activity between pH 9 and 11 and one under the name "LIPOLASE 100 T "commercially available lipase (NOVO INDUSTRI A / S, DK 2880 Bagsvaerd).
Nach wie vor stellt die Verarbeitung sehr fettreicher Rohware (wie Schweine, Schaffelle, Abfälle etc.) die Lederhersteller vor schwierige Probleme. Diese Probleme lassen sich unter den Stichworten: unzureichende Durchäscherung und Grundreinheit der Blößen nach dem Hautaufschluß sowie Bildung störender Kalkseifen, die zu unangenehmen Verschmierungen auf der Haut führen können, einordnen.
Auch die Beize fettreicher Blößen bereitet Schwierigkeiten, da ein oberflächlich anhaftender Fettfilm die Penetration der Beizenzyme hindern und der optimalen Grundlockerung entgegenwirken kann.The processing of very fat-rich raw materials (such as pigs, sheepskins, waste, etc.) continues to pose difficult problems for leather manufacturers. Die Lehre der oben genannten deutschen Patentanmeldung P 39 22 748.0 geht nicht über die Anwendung bestimmter Lipasen in der Weiche, d.h. im pH-Bereich 9 - 11 hinaus. Da diese Enzyme laut Herstellerangabe ihr pH-Optimum im Bereich 10 - 11 besitzen, schien die Anwendungsempfehlung gemäß der oben genannten deutschen Patentanmeldung wenigstens hinsichtlich dieses Parameters innerhalb eines vernünftigerweise in Betracht zu ziehenden Bereichs zu liegen. Ein Überschreiten dieses pH-Bereichs schien von vorneherein kaum erfolgversprechend, mußte der Fachmann doch, je weiter man sich vom genannten Bereich entfernt, mit erheblich reduzierter Wirksamkeit und verminderter Stabilität rechnen.
Es wurde nun gefunden, daß überraschenderweise alkalische Lipasen AL für die ein pH-Optimum bei ca. 9 - 11, insbesondere 10 - 11, charakteristisch ist, in der Wasserwerkstatt bei den Teilschritten
- a) des Äschers im pH-Bereich 11,5 - 14, insbesondere 12 - 13,5, speziell 12 - 13 und
- b) der Beize im pH-Bereich 5 - 11,5, insbesondere 7 - 9,5, speziell 8 - 9 in den diesen Teilschritten entsprechenden wäßrigen Flotten vorteilhaft angewendet werden können.

Dabei ist die Wirkung besonders ausgeprägt, wenn die genannten Lipasen in einer Enzymkombination EK zusammen mit neutralen bzw. alkalischen Proteasen P zur Anwendung kommen, die dem fraglichen Teilschritt entsprechend ausgewählt sind. Dabei handelt es sich vorzugsweise um die in der Technik einschlägig angewendeten Proteasen. Unter dem Äscher sei der bekannte Prozeß der Oberhautschwellung und Lockerung bis zur Entfernung der Haare und Grannen unter Einwirkung von alkalischen Äscher-Chemikalien verstanden (vgl. F. Stather, Gerbereichemie und Gerbereitechnologie, S. 166 - 199, Akademie-Verlag 1967; Ullmann's Encyclopedia of Industrial Chemistry 5th Ed. Vol., A15, 259 - 282, VCH 1990). Je nach der Verfahrensführung kann der Äscher haarerhaltend oder haarzerstörend gestaltet werden. Der Äscher wird im allgemeinen im pH-Bereich 12 - 13 durchgeführt, entweder in Form des sogenannten "Hydroxyläschers", wo insbesondere Calciumhydroxid neben Alkalihydroxid, Ammoniak und anderen Erdalkalihydroxiden eingesetzt wird oder in Form des sogenannten Sulfidäschers, dessen wirksame Bestandteile Alkali- oder Erdalkalisulfide gegebenenfalls in Mischung mit anderen basischen Alkalien bzw. Erdalkalien sind. Das erfindungsgemäße Äscherverfahren schließt sich sehr weitgehend an die Verfahren des Standes der Technik an (Ullmann's Encyclopedia of Industrial Chemistry 5th Ed. Vol. 15A, 259 - 282, VCH (1990); Ullmanns Enzyklopädie der Techn. Chemie, 4. Aufl. Bd. 16, S. 119 - 120 Verlag Chemie (1978), 3. Aufl. Bd. 11, S. 609, Urban & Schwarzenberg 19..).
Der Arbeitsgang des Äschers kann erfindungsgemäß mit einer Flottenlänge von 50 - 250, vorzugsweise 80 bis 150 % Wasser bezogen auf das Gewicht der Häute durchgeführt werden.

In den Schritten der Entkälkung und Beize, welche in der Wasserwerkstatt dem Äscher nachgeschaltet sind, werden die Häute und Felle neutralisiert und enzymatisch geäschert. Die Häute bzw. Felle werden dabei zunächst gewaschen und mittels vorzugsweise schwacher Säuren, beispielsweise organischen Säuren wie Milchsäure, Ameisensäure, Essigsäure, Buttersäure, Propionsäure, oder Dicarbonsäuren u.ä. oder schwach saurer anorganischer Verbindungen wie Natriumbisulfit, Sulfophthalsäure, Ammonsulfat oder auch Kohlensäure entkälkt. Im allgemeinen achtet man bei der Entkälkung darauf, daß ein für den nachfolgenden Enzymeinsatz in der Beize günstiger pH-Bereich resultiert. Für pankreatische Enzyme liegt dieser Bereich bei pH 7,5 - 8,2. Die nachfolgende Beize dient zur Entfernung von Epidermis- und Haarresten und zum zusätzlichen Hautaufschluß. In der Regel wird nach einer gewissen Zeit die enzymatische Beizkomponente insbesondere Enzyme des pankreatischen Komplexes zugegeben. Zu den Enzymen des pankreatischen Komplexes können auch Lipasen gehören (DE-A 37 04 465). Für die Beiztemperatur hat sich der Bereich zwischen 32 und 37 Grad C als zweckmäßig erwiesen. Die Beizdauer hält sich im allgemeinen im Bereich 1 Stunde bis 3 Stunden.
Vorzugsweise enthalten die enzymatischen Ansätze, speziell die mit der Enzymkombination EK durchgeführten noch an sich bekannte Sequestriermittel SM, vorab zwecks Vermeidung von Kalkseifen.
Ferner hat sich der Zusatz von emulgierwirksamen Substanzen ES bewährt, der zu besonders guter Fettemulgierung führt. Im Gegensatz zu den einschlägig verwendeten Lipasen des Standes der Technik mit einem Einsatzbereich von pH 6 - 9 haben die erfindungsgemäß eingesetzten Lipasen ein ausgeprägtes Wirkungsoptimum (z.B. gegenüber Olivenöl oder Tributyrin) zwischen pH 9 und 11. Derartige alkalische Lipasen wurden speziell für die Waschmittelindustrie entwickelt. Sie sind mikrobiologischen Ursprungs. Potentielle Quellen für derartige, gegebenenfalls genetisch veränderte Mikroorganismen-Stämme sind insbesondere Pilze und Bakterien. Gewisse alkalische Lipasen kommen z.B. in Pseudomonas-Stämmen vor. Weiter kommen Rhizopus sp., Candida sp., Chromobacterium sp. als Lipase-Lieferanten infrage. Weitere wichtige Lipase-Produzenten sind Geotrichium sp., Aspergillus sp., Mucor sp., Penicillium sp., Corynebacterium sp., Propionibacterium sp. und Achromobacter sp.. Genannt seien speziell Rhizopus arrhizus und Rh. oryzae, Candida cyclindracea, Chromobacterium viscosum, Geotrichium Candidum, Mucor miehi, Mucor pusillus, Penicillium roqueferti und P. cyclopium, Corynebacterium acne, Propionibacterium shermanii, Achromobacter lipolyticum, Aspergillus niger, insbesondere Aspergillus oryzae. Als besonders geeignet wurden auch gewisse genetisch veränderte Stämme befunden, z.B. eine alkalische Lipase aus einem durch Rekombination gewonnenen Aspergillus oryzae-Stamm mit ausgeprägtem Aktivitätsoptimum zwischen pH 9 und 11 oder eine unter der Bezeichnung ® Lipolase TM 30 T im Handel befindlichen Lipase (NOVO INDUSTRI A/S, DK 2880 Bagsvaerd, Dänemark).
In herkömmlicher Weise wird die Aktivitätsbestimmung von Lipasen mit Olivenöl als Substrat durchgeführt, ferner mit Triacetin und Tributyrin. [Vgl. M. Sémériva et al. Biochemistry 10, 2143 (1971); Pharmaceutical Enzymes, edited by R. Ruyssen and A. Lauwers 1978, (FIP)]. Soweit die fettspaltende Aktivität in Kilo-Lipase-Units (Einheit = KLCA) ausgedrückt wird, wird unter den Standardbedingungen 40 Grad C, pH = 5,5 mit Tributyrin als Substrat gearbeitet. (Vgl. M. Sémériva, oben zitierte Literaturstelle).
Für die Zwecke der vorliegenden Erfindung wird die Lipaseaktivität in LCA-Einheiten angegeben, wobei jedoch bei pH 9,5 gemessen wird. Man bewegt ca. 1 1/2 Stunden unter diesen Bedingungen bevor man die Enzyme, insbesondere die Enzymkombination EK-II- als Richtwert in Anteilen von ca. 0,3 Gew.-%, vorzugsweise mit etwa der gleichen Menge Kalkhydrat wie bereits angewendet und läßt zunächst für eine kürzere Zeit unter ständigem Bewegen, so dann über einen größeren Zeitraum, beispielsweise ca. 16 Stunden unter gelegentlichem Rühren reagieren.
Nach Ablassen der Flotte und Waschen, vorzugsweise mit ca. 150 % Wasser bei 28 Grad C erhält man Blößen von sehr guter Qualität. Es wurde gefunden, daß Häute und Felle, welche mit einer alkalischen Protease bei pH 8 - 11 4 - 20 Stunden vorbehandelt bzw. geweicht werden und anschließend bei demselben pH Wert 2 - 6 Stunden mit der alkalischen Lipase im gleichen oder einem neuen Bad versetzt werden, für eine anschließende proteolytische Enthaarung hervorragend zubereitet sind. Vorteilhafterweise schließt sich an die Weiche ein Haarimmunisierungsschritt an, bei dem - in Anlehnung an DE-A 38 02 640 - Kalkhydrat und organische Thioverbindungen zusammen mit Aminen in ca. 80 % Wasser bei ca. pH 12 zur Anwendung kommen können. Dann folgt gewöhnlich eine Haarlockerungsstufe. Bei Verwendung der erfindungsgemäß einzusetzenden Enzymkombinationen EK-II kommt man mit einer stark reduzierten Menge an Sulfid aus, beispielsweise 0,4 Gew.-% Natriumsulfhydrat (72 %ig) bezogen auf die Häute. Nach relativ kurzer Zeit, beispielsweise ca. 2 Stunden sind die Häute haarfrei. Man gibt zweckmäßig noch ca. 70 Gew.-% Wasser zusammen mit ca. 2 Gew.-% Kalkhydrat und ca. 0,3 Gew.-% Natronlauge (50 %) zu und läßt über einen gewissen Zeitraum, zweckmäßig ca. 14 Stunden bei 28 Grad C unter intervallmäßigem, kurzzeitigem Bewegen weiterlaufen. Anschließend wird die Flotte abgelassen und die weitere Bearbeitung in betriebsüblicher Weise fortgesetzt. Besonders ausgeprägt ist die Wirkung in Kombination mit entsprechenden neutralen und alkalischen Proteasen; die unter den Stichworten:
- Verbesserte Lockerung des Pigmentgrundes
- Verbesserte Entfettung der Blößen
- weniger Mastfalten und Narbenzug
- Durchführung einer sulfidfreien wie auch einer sulfidarmen haarerhaltenden Enthaarung
