Definitions
- HPV Human papilloma viruses
- the present invention relates to monoclonal antibodies (MAK) which are directed against the 15 kd E7 protein of human papillomavirus 16 (HPV 16 E7). These antibodies can be used as a diagnostic agent, drug or drug carrier. 6 hybridomas were obtained, the secreted MAK (E71 - E7VI) were characterized. All MAK reacted positively in an "enzyme linked immunosorbent assay (ELISA)", on the solid phase of which an MS-2 / HPV16 E7 fusion protein was adsorbed (see below). No reaction was detected for E7III in the corresponding Western blot.
- MAK monoclonal antibodies
- HPV16 E7 protein does not have the first 8 amino acids. It is fused with an N-terminal "leader peptide", which consists of 100 amino acids of MS-2 polymerase and 9 amino acids from polylinker sequences, and was used in a purity of about 70%.
- ELISA test plates (Nunc, Denmark) were incubated overnight at room temperature with 0.2-0.5 ⁇ g per well of the purified MS-2, MS-2 / HPV16 E7 or MS-2 / HPV18 E7 protein in 100 ⁇ l 7M urea (12 hours, Room temperature). After washing twice with phosphate-buffered saline (PBS), the plates were saturated for one hour at 37 ° C. with 2% bovine serum albumin solution in PBS and were thus ready for use after washing 3 times in PBS.
- PBS phosphate-buffered saline
- H2O2 / chromogen solution 0.01% H2O2, 1 mg / ml orthophenylenediamine in 0.1 M phosphate buffer pH 5.5
- absorbance measurement after which after 10 minutes the Enzyme reaction was stopped by adding 50 ⁇ l 1M H2SO4.
- Hybridomas that secrete antibodies against HPV 16 E7 could be detected by parallel testing in two "enzyme linked immunosorbent assays (ELISA)".
- ELISA enzyme linked immunosorbent assay
- the solid phase of one ELISA MS-2 / HPV16 E7 fusion protein was adsorbed, on the solid phase of the other only MS-2 protein.
- Bound antibodies from hybridoma supernatants were detected by incubation with goat anti-mouse antibodies, that were conjugated to horseradish peroxidase.
- about 150 clones were positive when screened with the "MS-2 ELISA”
- 10 clones were additionally positive with the "MS-2 / HPV16 E7 ELISA”. Of these 10 clones, 6 were stable after two rounds of cloning to generate single clones on a "feeder layer" of peritoneal mouse macrophages (Table 1).

Abstract
Description
Soc. Experiment. Biol. and Med. 135, 543-545) vorher in 10 % SDS Polyacrylamidgelelektrophorese im wesentlichen wie beschrieben (Laemmli, V.K. (1970), Nature 227, 680-682) aufgetrennt waren.
