Description
- Diese Erfindung betrifft ein Verfahren zum Bereitstellen eines Substrats, insbesondere einer medizinischen Vorrichtung oder einem Teil einer derartigen Vorrichtung, die bzw. der zum Einführen in den menschlichen Körper vorgesehen ist, mit einer Beschichtung, welche bei in Kontakt Bringen mit einem wässrigen Fluid gleitfähig wird, und Substrate, insbesondere medizinische Vorrichtungen, welche mit derartigen hydrophilen Beschichtungen ausgestattet sind.
- Es ist allgemein bekannt, Substrate wie medizinische Vorrichtungen oder Teile derartiger Vorrichtungen mit einer hydrophilen Beschichtung zu versehen, um die Reibung zu verringern, wenn die Vorrichtung in eine nasse Umgebung wie den menschlichen Körper eingeführt wird, zu verringern. Derartige hydrophile Beschichtungen wurden auch als gleitfähige oder "schlüpfrige" Beschichtungen bezeichnet.
- Katheter und andere medizinische Vorrichtungen, die zur Einführung in Blutgefäße, die Harnröhre, Körpergänge und dergleichen verwendet werden, und Führungsdrähte, die mit derartigen Vorrichtungen verwendet werden, sind Beispiele von medizinischen Vorrichtungen, die mit hydrophilen Beschichtungen versehen werden können. Katheter für Ballonangioplastie und Biopsie sind spezifische Beispiele derartiger Katheter.
- Substrate und medizinische Gegenstände oder Vorrichtungen, bei denen es erwünscht sein kann, sie mit einer hydrophilen Beschichtung zu versehen, und Verfahren zum Versehen von derartigen Substraten und Gegenständen oder Vorrichtungen mit hydrophilen Beschichtungen wurden in einer großen Anzahl von Literaturstellen beschrieben, wovon nachstehend Beispiele erwähnt werden.
- US-Patent Nr. 4,119,094 offenbart eine Methode zum Beschichten eines Substrats mit einem Polyvinylpyrrolidon-Polyurethan Interpolymer. In dem Verfahren werden ein Polyisocyanat und ein Polyurethan in einem Lösungsmittel wie etwa Methylethylketon auf ein Substrat aufgebracht und das Lösungsmittel abgedampft, wonach Polyvinylpyrrolidon in einem Lösungsmittel auf das behandelte Substrat aufgebracht und das Lösungsmittel abgedampft wird.
- US-Patent Nr. 5,091,205 offenbart eine Methode zum Bereitstellen eines Substrats mit einer hydrophilen gleitfähigen Beschichtung, wobei das Substrat zunächst mit einer Polyisocyanatlösung in Kontakt gebracht wird, um Kopplung bereitzustellen, danach mit einer Poly(carbonsäure)lösung in Kontakt gebracht wird, wobei eine Beschichtung bereitgestellt wird, und schließlich ofengetrocknet wird.
- Methylethylketon ist das bevorzugte Lösungsmittel für die Polyisocyanate und Dimethylformamid für die Poly(carbonsäure). Es ist erwähnt, daß die Polyisocyanate emulgiert werden können um eine Öl-in-Wasser-Emulsion zu bilden, in welchem Fall die reaktiven Isocyanatgruppen jedoch durch geeignete chemische Gruppen geschützt werden müssen.
- EP-Patent Nr. 0 106 004 B1 offenbart eine Methode zum Bilden einer hydrophilen Beschichtung auf einem Substrat durch Aufbringen einer Beschichtung aus einer Lösungsmittellösung eines Polyisocyanats, wobei eine Kopplungsbeschichtung gebildet wird, gefolgt von Aufbringen einer Lösungsmittellösung eines hydrophilen Copolymers aus Monomeren, ausgewählt aus Vinylpyrrolidon, Vinylmethylether oder Vinylpyridin, und einem Monomer, das aktiven Wasserstoff enthält, der mit Isocyanat reagiert, wobei eine kovalente Bindung zwischen der Kopplungsbeschichtung und dem hydrophilen Copolymer gebildet wird.
- EP-Patent Nr. 0 166 998 B1 offenbart eine Methode zur Behandlung der Oberfläche eines medizinischen Instruments. Die Oberfläche wird mit einer Lösung eines Polymers mit einer reaktiven funktionellen Gruppe in einem organischen Lösungsmittel behandelt, gefolgt von Behandlung mit einem wasserlöslichen Polymer, ausgewählt aus Maleinsäureanhydridpolymeren, Cellulosepolymeren, Polyethylenoxidpolymeren und wasserlöslichen Nylons oder Derivaten davon, wobei die reaktive funktionelle Gruppe kovalent an das wasserlösliche Polymer gebunden wird, wonach das behandelte Substrat gegebenenfalls mit Wasser in Kontakt gebracht wird.
- US-Patent Nr. 5,077,352 offenbart eine Methode, wobei eine flexible, gleitfähige organische Polymerbeschichtung gebildet wird, durch Aufbringen eines Gemisches aus einem Isocyanat, einem Polyol und einem Poly(ethylenoxid) in einer Trägerflüssigkeit auf eine zu beschichtende Oberfläche. Die Trägerflüssigkeit wird entfernt und das Gemisch reagiert, wobei eine Polyurethanbeschichtung mit assoziiertem Poly(ethylenoxid) gebildet wird. Methylenchlorid, Chloroform, Dichlorethan, Acetonitril, Dichlorethylen und Methylenbromid werden als geeignete Trägerflüssigkeiten erwähnt.
- Die internationalen Patentanmeldungen WO92/9289, WO92I9290 und WO9219288 offenbaren Verfahren, um verschiedene medizinischen Vorrichtungen mit einer Polyurethanoberfläche mit einer Beschichtung eines hydrophilen Poly(meth)acrylamids bereitzustellen. Vor dem Auftrag der hydrophilen Beschichtung wird das Substrat mit einer Verbindung behandelt, welche funktionelle Gruppen aufweist, die mit dem Polyurethan bzw. dem Poly(meth)acrylamid reagieren können, typischerweise einem Diisocyanat oder einem Isocyanat mit höherer Funktionalität in einem organischen Lösungsmittel.
- Ein Nachteil der Methoden gemäß den vorstehend genannten Dokumenten besteht darin, daß die Bereitstellung der hydrophilen Beschichtung üblicherweise die Verwendung organischer Lösungsmittel oder toxischer Chemikalien, beispielsweise von Polyisocyanaten, umfaßt, welche Umweltprobleme und/oder Gesundheitsrisiken darstellen können. Um die Verwendung von Lösungsmitteln zu vermeiden, wurden einige Lösungsmittel-freie Methoden entwickelt.
- Die europäische Patentanmeldung Nr. 92 100 787.8, Veröffentlichungsnummer EP 0 496 305 A2, offenbart eine Methode zur Herstellung eines medizinischen Formgegenstands, der mit einer gleitfähigen Beschichtung versehen ist. Eine Beschichtungszusammensetzung, umfassend eine Mischung aus Polyurethan und Polyvinylpyrrolidon, wird mit einem Substratpolymer coextrudiert, wobei ein Formgegenstand mit einer Schicht aus der Beschichtungszusammensetzung darauf, welche bei in Kontakt Bringen mit Wasser gleitfähig wird, erhalten wird.
- US-Patent Nr. 5,041,100 offenbart eine Methode zum Beschichten eines Substrats mit einem Gemisch aus Poly(ethylenoxid) upd einer wässrigen Dispersion eines Strukturkunststoffmaterials, z. B. Polyurethan. Wie in Spalte 2, Zeilen 15-21 angegeben, ist das Poly(ethylenoxid) ohne Vernetzung in innig dispergierter Beziehung mit dem Strukturkunststoffmaterial vermischt, wobei eine hydrophile Komponente des Systems bereitgestellt wird, die an die Oberfläche austreten kann oder nahe der Oberfläche eingeschlossen sein kann, wobei dieser eine hydrophile Eigenschaft vermittelt wird und Reibung, insbesondere nach Hydratisierung, verringert wird.
- Die in den vorstehend erwähnten Dokumenten beschriebenen Methoden haben den Nachteil, daß das Interpolymer-Netzwerk, welches das hydrophile Polymer physikalisch an das Substrat bindet, nach längerer Turbulenzströmung oder Durchnässung oftmals zusammenbricht, und daß die hydrophile Spezies ausgewaschen werden kann, wodurch der Gegenstand unzureichend gleitfähig wird.
- Schließlich offenbart die internationale Patentanmeldung Nr. WO91/9756 eine Methode, um ein medizinisches Instrument mit einer hydrophilen reibungsarmen Beschichtung zu versehen, wobei das Verfahren die Schritte des Bildens einer Innenlage aus einer wässrigen Polymeremulsion und einer Außenlage aus einer wässrigen Lösung eines wasserlöslichen hydrophilen Polymers, und des Trocknens der zwei Lagen gleichzeitig nach Aufbringen der Außenlage, durch Erwärmen auf eine Temperatur oberhalb 100ºC, umfaßt.
- Die vorstehende Methode beseitigt die Verwendung organischer Lösungsmittel und resultiert in einer fest an das Substrat gebundenen Beschichtung. Die Verwendung von Vernetzungstemperaturen oberhalb 100ºC beschränkt jedoch die Verwendung der Methode, da viele Vorrichtungen, beispielsweise Ballonkatheter aus Poly(ethylenterephthalat) (PET) gegenüber derartigen Temperaturen nicht beständig sind.
- Die vorliegende Erfindung ist auf ein Verfahren zum Bereitstellen eines Substrats, insbesondere einer medizinischen Vorrichtung oder einem Teil einer derartigen Vorrichtung, die bzw. der zum Einführen in den menschlichen Körper vorgesehen ist, mit einer hydrophilen Beschichtung, welche bei in Kontakt Bringen mit einem wässrigen Fluid gleitfähig wird, gerichtet, wobei das Verfahren es unter anderem ermöglicht, Vorrichtungen zu beschichten, welche gegenüber hohen Bearbeitungstemperaturen empfindlich sind, wie etwa (PET) Ballonkatheter. Das hydrophile Polymer wird kovalent an das Polymer der ersten Beschichtung gebunden.
- Wie die Methode gemäß WO91/9756 verwendet das erfindungsgemäße Verfahren wässrige Beschichtungszusammensetzungen, das Verfahren der vorliegenden Erfindung kann jedoch bei erheblich niedrigeren Temperaturen, beispielsweise bei Raumtemperatur durchgeführt werden.
- Als ein weiterer Vorteil führt das erfindungsgemäße Verfahren zu einer sehr abriebbeständigen Beschichtung.
- Das erfindungsgemäße Verfahren umfaßt:
- a) Beschichten eines Substrats mit einer ersten wässrigen Beschichtungszusammensetzung, umfassend eine wässrige Dispersion oder Emulsion eines Polymers mit organischen säurefunktionellen Gruppen und ein polyfunktionelles Vernetzungsmittel mit funktionellen Gruppen, die mit organischen sauren Gruppen reagieren können, und Trocknen der Beschichtung, wobei eine im wesentlichen wasserunlösliche Beschichtungslage erhalten wird, welche noch funktionelle Gruppen umfaßt, die mit organischen sauren Gruppen reaktiv sind, und
- b) in Kontakt Bringen der getrockneten, in a) erhaltenen Beschichtungslage mit einer zweiten wässrigen Beschichtungszusammensetzung, umfassend eine wässrige Lösung oder Dispersion eines hydrophilen Polymers mit organischen säurefunktionellen Gruppen, und Trocknen der kombinierten Beschichtung, wodurch das hydrophile Polymer durch das Vernetzungsmittel an das Polymer der ersten Beschichtungszusammensetzung gebunden wird.
- Ein Aspekt der vorliegenden Erfindung betrifft eine zum Einführen in den menschlichen Körper vorgesehene medizinische Vorrichtung, umfassend ein zum Einführen in den menschlichen Körper geeignetes Substrat, wobei die Oberfläche des Substrats mit einer vernetzten Polymerzusammensetzung beschichtet ist, die Zusammensetzung eine erste Polymerschicht, gebildet aus einer mindestens partiellen Reaktion einer wässrigen Dispersion oder Emulsion eines Polymers mit vorhandenen reaktiven organischen säurefunktionellen Gruppen mit einem polyfunktionellen Vernetzungsmittel, welches mit den organischen säurefunktionellen Gruppen reagieren kann, und eine zweite hydrophile Polymerschicht mit vorhandenen organischen säurefunktionellen Gruppen, die mit dem Vernetzungsmittel reagieren können, umfaßt. Wie hierin weiter beschrieben, wird die erste Polymerschicht vor dem Aufbringen der zweiten Polymerschicht im wesentlichen vernetzt. Es bleiben ausreichend funktionelle Gruppen des Vernetzungsmittels übrig, um an der kovalenten Bindung mit der zweiten Polymerschicht teilzunehmen. Diese kovalente Bindung ermöglicht eine hervorragende Haftung der gleitfähigen hydrophilen Lage an der ersten Polymerbeschichtung. Die Beschichtung hat eine hervorragende Verschleißbeständigkeit, Gleitfähigkeit, und kann in extrem dünnen Schichten aufgetragen werden, so daß die mechanischen Eigenschaften des Substrats, auf das sie aufgetragen wird, nicht beeinträchtigt werden. Dies ist insbesondere von Bedeutung, wenn die Beschichtung auf einen dünnwandigen aufblasbaren Ballon auf einem für Angioplastie verwendeten Ballonkatheter aufgebracht werden soll.
- Als ein Teil der vorliegenden Erfindung wird auch ein reaktiver Beschichtungsfilm erachtet, welcher zum Binden von hydrophilen Polymeren mit vorhandenen organischen säurefunktionellen Gruppen geeignet ist, wobei der Beschichtungsfim das Reaktionsprodukt einer wässrigen Dispersion oder Emulsion eines Polymers mit organischen säurefunktionellen Gruppen mit einem polyfunktionellen Vernetzungsmittel, welches mit den organischen säurefunktionellen Gruppen reagieren kann, ist, wobei das Reaktionsprodukt noch reaktive funktionelle Gruppen des polyfunktionellen Vernetzungsmittels umfaßt. Dieser Aspekt der Erfindung soll denjenigen Teil der Beschichtung vor weiterer Reaktion mit der hydrophilen zweiten Beschichtung, welche aufgebracht wird um Gleitfähigkeit zu erhalten, abdecken. Somit werden in dieser Ausführungsform Gegenstände abgedeckt, welche nur mit der ersten Beschichtung beschichtet worden sind und zum Aufbringen einer hydrophilen Beschichtung, die bei Kontakt mit Wasser Gleitfähigkeit vermitteln kann, vorbereitet sind.
- Ein weiterer Aspekt der vorliegenden Erfindung umfaßt eine hydrophile Polymerbeschichtung, welche bei Kontakt mit einem wässrigen Medium gleitfähig werden kann, wobei die Beschichtung mindestens zwei kovalent aneinander gebundene Polymerlagen umfaßt, wobei ein vernetztes Netzwerk gebildet wird, wobei das vernetzte Netzwerk das Reaktionsprodukt ist aus:
- a) einer ersten Polymerlage, umfassend eine wässrige Dispersion oder Emulsion eines Polymers mit organischen säurefunktionellen Gruppen und ein polyfunktionelles Vernetzungsmittel, welches mit den organischen säurefunktionellen Gruppen reagieren kann, und
- b) einer zweiten wässrigen Polymerlage, umfassend eine Lösung oder Dispersion eines hydrophilen Polymers mit organischen säurefunktionellen Gruppen.
- Die hydrophile Polymerbeschichtung der ersten Polymerschicht kann aus einer Reihe hierin angegebener Polymere ausgewählt werden. Besonders bevorzugt sind jedoch die wasserbasierten Polyurethanpolymere und Polyacrylsäurepolymere. Die zweite Polymerschicht kann ebenfalls aus einer großen Vielzahl von Polymeren, welche aufgrund der Gegenwart ihrer organischen Säurefunktionalität an die erste Beschichtungslage gebunden werden können, ausgewählt werden. Besonders bevorzugt sind die Polyacrylsäure-Polymere und die Acrylamid-Acrylsäure- Copolymere.
- Wie vorstehend erwähnt, haben medizinische Vorrichtungen, die mindestens teilweise mit den Beschichtungen der vorliegenden Erfindung beschichtet sind, gegenüber dem bekannten Stand der Technik besondere Vorteile, indem sie aufgrund der Gleiteigenschaften der äußeren Beschichtung mit einem niedrigeren Reibungswiderstand leicht in den Körper eingeführt werden können. Zusätzlich wird die Haftung der äußeren Beschichtung gegenüber dem bekannten Stand der Technik aufgrund der kovalenten, zwischen den zwei Beschichtungslagen auftretenden Bindung verbessert. Wie hierin erwähnt, ist die Beschichtung von aufblasbaren Angioplastieballons, die ein integraler Bestandteil von Angioplastieballonkathetern sind, eine spezifische, für die erfindungsgemäßen Beschichtungen vorgesehene Anwendung. Zusätzlich sind andere medizinische Vorrichtungen wie etwa Führungsdrähte und dergleichen beabsichtigt. Die Vorrichtungen müssen nicht notwendigerweise zur Verwendung innerhalb des Körpers vorgesehen sein, und äußere Verwendungen sind ebenfalls beabsichtigt.
- Die vorliegende Erfindung sieht auch ein Kit vor, das als eine erste Komponente eine wässrige Dispersion eines Polymers mit organischer Säurefunktionalität umfaßt, als eine zweite Komponente ein polyfunktionelles Vernetzungsmittel, das mit der organischen Säurefunktionalität der ersten Komponente reaktiv ist, und als eine dritte Komponente eine wässrige Lösung oder Dispersion eines hydrophilen Polymers mit organischer Säurefunktionalität, und welches nach Vernetzen und in Kontakt Bringen mit Wasser Gleitfähigkeit vermittelt.
- In vorliegenden Kontext soll der Begriff "organische Säuregruppe" jede Gruppierung umfassen, welche einen organischen, sauren, ionisierbaren Wasserstoff enthält. Beispiele von funktionellen Gruppierungen, welche organischen, sauren, ionisierbaren Wasserstoff enthalten, sind die Carbon- und Sulfonsäuregruppen. Der Begriff "organische säurefunktionelle Gruppen" soll alle Gruppen umfassen, welche unter den Reaktionsbedingungen ähnlich wie die organischen Säuregruppen funktionieren, beispielsweise Metallsalze derartiger Säuregruppen, insbesondere Alkalimetallsalze wie Lithium-, Natrium- und Kaliumsalze, und Erdalkalisalze wie Calcium- oder Magnesiumsalze, und quaternäre Aminsalze derartiger Säuregruppen, insbesondere quaternäre Ammoniumsalze.
- Das Polymer mit organischen säurefunktionellen Gruppen, welches in der ersten wässrigen Beschichtungszusammensetzung vorhanden ist, wird unter entsprechender Berücksichtigung der Art des zu beschichtenden Substrats ausgewählt. Typischerweise wird das Polymer in der ersten Beschichtungszusammensetzung aus Vinylmonomereinheiten-enthaltenden Homo- und Copolymeren, Polyurethanen, Epoxyharzen und Kombinationen davon ausgewählt. Das Polymer in der ersten Beschichtungszusammensetzung wird bevorzugt aus Polyurethanen, Polyacrylaten, Polymethacrylaten, Polyisocrotonaten, Epoxyharzen, Acrylat-Urethan-Copolymeren und Kombinationen davon, mit organischen säurefunktionellen Gruppen, ausgewählt. In einer besonders bevorzugten Ausführungsform des erfindungsgemäßen Verfahrens wird das Polymer in der ersten Beschichtungszusammensetzung ausgewählt aus Homo- und Copolymeren mit einer erheblichen Menge an organischen säurefunktionellen Gruppen in ihrer Struktur, die als ein interner Emulgator wirken können. Eine spezifische Klasse von Polyurethanen, die in der ersten Beschichtungszusammensetzung verwendet werden können, sind die sogenannten wasserbasierten Polyurethane, zu denen die sogenannten intern emulgierten wasserbasierten Polyurethane gehören, welche Carbonsäuregruppen und/oder Sulfonsäuregruppen enthalten, gegebenenfalls als Salze derartiger Gruppen, da interne Emulgatoren besonders bevorzugt sind.
- Beispiele von wasserbasierten Polyurethanen sind diejenigen, welche unter der Handelsbezeichnung NeoRez von Zeneca Resins, beispielsweise NeoRez-940, NeoRez-972, NeoRez-976 und NeoRez-981; unter der Handelsbezeichnung Sancure von Sanncor, beispielsweise Sancure 2026, Sancure 2710, Sancure 1601 und Sancure 899; unter den Handelsbezeichnungen U21 und U21X von B. F. Goodrich; und unter den Handelsbezeichnungen Bayhydrol LS-2033, Bayhydrol LS-2100, Bayhydrol LS-2952 und Bayhydrol LS-2990 von der Bayer AG erhältlich sind.
- Eine weitere spezifische Klasse von Polymeren, die sich in der ersten Beschichtungszusammensetzung als besonders geeignet herausgestellt haben, sind Acrylat-Urethan-Copolymere, beispielsweise die Acrylurethan- Copolymerdispersionen, welche von Zeneca Resins unter den Handelsbezeichnungen NeoPac E-106, NeoPac E-121, NeoPac E-130 und NeoRez R-973 erhältlich sind.
- Die Konzentration des Polymers in der ersten Beschichtungszusammensetzung beträgt üblicherweise von etwa 2 bis etwa 60 Gew.-% und bevorzugt von etwa 5 bis etwa 40 Gew.-%, berechnet als Polymerfeststoffe, bezogen auf das Gesamtgewicht der ersten Beschichtungszusammensetzung.
- Zusätzlich zu einem oder mehreren Polymeren mit organischen säurefunktionellen Gruppen umfaßt die erste wässrige Beschichtungszusammensetzung eine oder mehrere polyfunktionelle Vernetzungsmittel mit funktionellen Gruppen, die mit organischen Säuregruppen reagieren können. Polyfunktionelle Vernetzungsmittel mit funktionellen Gruppen, die mit organischen Säuregruppen reagieren können, sind in der Technik bekannt. Beispielsweise wurden derartige polyfunktionelle Vernetzungsmittel für eine externe Vernetzung von Polyurethanen verwendet.
- Besonders bevorzugte polyfunktionelle Vernetzungsmittel zur Verwendung im erfindungsgemäßen Verfahren sind polyfunktionelle Aziridine und polyfunktionelle Carbodiimide.
- Polyfunktionelle Aziridine und polyfunktionelle Carbodiimide und deren Verwendung als Vernetzungsmittel sind in der Technik bekannt.
- Das von Zeneca Resins unter der Handelsbezeichnung NeoCryl CX 100 erhältliche Vernetzungsmittel und das von EIT Industries unter der Handelsbezeichnung XAMA- 7 erhältliche Vernetzungsmittel sind spezifische Beispiele von polyfunktionellen Aziridin-Vernetzungsmitteln, welche im erfindungsgemäßen Verfahren verwendet werden können, und das von Union Carbide unter der Handelsbezeichnung Ucarlink XL-29SE erhältliche Vernetzungsmittel ist ein spezifisches Beispiel eines polyfunktionellen Carbodiimid-Vernetzungsmittels, welches im erfindungsgemäßen Verfahren verwendet werden kann.
- Die geeigneten polyfunktionellen Aziridine umfassen das trifunktionelle Aziridin der nachfolgenden Formel:
- Das polyfunktionelle Vernetzungsmittel ist bevorzugt ein Vernetzungsmittel mit mehr als zwei funktionellen Gruppen pro Molekül. Weiterhin sollte bemerkt werden, daß eine Kombination von polyfunktionellen Vernetzungsmitteln im erfindungsgemäßen Verfahren verwendet werden kann.
- Die funktionellen Gruppen auf dem Vernetzungsmittel dienen mindestens zwei Zwecken. Der erste Zweck besteht darin, die erste Polymerbeschichtung zu vernetzen. Der zweite Zweck besteht darin, an der kovalenten Bindung mit den in der zweiten (hydrophilen) Polymerbeschichtung vorhandenen organischen Säuregruppen teilzunehmen. Somit muß ausreichend Funktionalität im Vernetzungsmittel vorhanden sein, um beide Zwecke zu erfüllen. Das bedeutet, die verwendete Menge an Vernetzungsmittel muß ausreichend sein, so daß ausreichend funktionelle Gruppen vorhanden sind um die erste Polymerbeschichtung im wesentlichen zu vernetzen, und daß genügend nicht-abreagierte funktionelle Gruppen übrig bleiben um die zweite hydrophile Schicht kovalent zu binden.
- Ein Hinweis darauf, daß nicht genügend funktionelle Gruppen des Vernetzungsmittels vorhanden sind, ist eine unzureichende Bindung der zweiten Schicht. Dies wird offensichtlich durch fehlende Verschleißbeständigkeit, und derartige Beschichtungen können von dem Substrat, auf das sie aufgebracht worden sind, leicht abgewischt werden.
- Die Konzentration des Vernetzungsmittels in der ersten Beschichtungszusammensetzung liegt üblicherweise im Bereich von etwa 0,2 bis etwa 30 Gew.-%, und bevorzugt im Bereich von etwa 0,5 bis etwa 20 Gew.-%.
- Wie in der Technik bekannt, kann die erste wässrige Beschichtungszusammensetzung andere herkömmliche Additive umfassen, wie Egalisierungsmittel, verschiedene Stabilisatoren, pH-Einstellmittel, Antischaummittel, Colösungsmittel, etc., soweit mit der vorgesehenen Verwendung des beschichteten Substrats kompatibel.
- Die Beschichtung aus der ersten wässrigen Beschichtungszusammensetzung wird getrocknet, wobei eine im wesentlichen wasserunlösliche Beschichtungslage erhalten wird, welche noch funktionelle, mit organischen Säuregruppen reaktive funktionelle Gruppen umfaßt. Danach wird die erhaltene getrocknete Beschichtung mit einer zweiten wässrigen Beschichtungszusammensetzung, umfassend eine wässrige Lösung oder Dispersion eines hydrophilen Polymers mit organischen säurefunktionellen Gruppen in Kontakt gebracht, wonach die zweite Beschichtung getrocknet wird, wodurch des hydrophile Polymer durch das Vernetzungsmittel an das Polymer der ersten Beschichtungszusammensetzung gebunden wird.
- Hydrophile Polymere zur Verwendung in hydrophilen gleitfähigen Beschichtungen sind in der Technik bekannt. Im erfindungsgemäßen Verfahren kann jedes hydrophile Polymer (Homo- oder Copolymer oder Gemisch aus einem oder mehreren derartiger Polymere) verwendet werden, vorausgesetzt daß es organische säurefunktionelle Gruppen in seiner Struktur enthält, welche mit dem polyfunktionellen Vernetzungsmittel mit funktionellen Gruppen, die mit organischen Gruppen reagieren können, reagieren können, um eine hydrophile Beschichtung zu bilden, welche bei in Kontakt Bringen mit einem wässrigen Fluid gleitfähig wird.
- Das hydrophile Polymer kann Monomereinheiten eines oder mehrerer Monomere mit organischen säurefunktionellen Gruppen umfassen. Bevorzugte Beispiele derartiger Monomere umfassen Acrylsäure, Methacrylsäure und Isocrotonsäure.
- Zusätzlich dazu, daß es Monomereinheiten mindestens eines Monomers mit organischen säurefunktionellen Gruppen umfaßt, kann das hydrophile Polymer Monomereinheiten mindestens eines hydrophilen Monomers ohne jegliche organische säurefunktionelle Gruppen, wie etwa Vinylpyrrolidon und Acrylamid, enthalten. Ein bevorzugtes Beispiel eines Copolymers zur Verwendung in oder als das hydrophile Polymer im erfindungsgemäßen Verfahren ist ein Acrylsäure- Acrylamid-Copolymer. Das von Allied Colloids unter der Handelsbezeichnung Versicol WN 33 erhältliche Acrylamid-Acrylsäure-Copolymer ist ein spezifisches Beispiel für ein derartiges Copolymer.
- Die Fähigkeit, bei Hydratisierung gleitfähig zu werden, ist ein kritischer Aspekt der vorliegenden Erfindung. Der Grad an Gleitfähigkeit, welcher bei Kontakt mit wässrigem Medium erzeugt wird, hängt von einer Reihe von Faktoren ab, umfassend den Typ des hydrophilen Polymers, sein Molekulargewicht, den Grad, zu dem es an das wässrige Medium ausgesetzt wird, sowie die Gegenwart von Mitteln, welche eine Benetzung erleichtern. Davon ist das Molekulargewicht am wichtigsten. Der in der vorliegenden Erfindung geeignete Molekulargewichtsbereich hängt von dem besonderen Typ an ausgewähltem Polymer ab. Das Molekulargewicht des hydrophilen Polymers in der zweiten Beschichtungszusammensetzung wird typischerweise im Bereich von etwa 100.000 bis etwa 15 Millionen, insbesondere von etwa 150.000 bis etwa 10 Millionen liegen. Hydrophile Polymere mit einem Molekulargewicht im Bereich von etwa 400.000 bis etwa 10 Millionen und insbesondere von etwa 7,5 Millionen haben sich besonders geeignet zur Verwendung im erfindungsgemäßen Verfahren herausgestellt. Die vorstehend erwähnten Acrylamid-Acrylsäure-Copolymere fallen unter dieses bevorzugte Molekulargewicht.
- Die Konzentration des hydrophilen Polymers in der zweiten Beschichtungszusammensetzung wird typischerweise von etwa 0,1 bis 5 Gew.-%, bevorzugt von etwa 0,5 bis etwa 3 Gew.-%, berechnet als Feststoffe hydrophiles Polymer, bezogen auf das Gesamtgewicht der zweiten Beschichtungszusammensetzung, betragen.
- In einer bevorzugten Ausführungsform des erfindungsgemäßen Verfahrens können die funktionellen Gruppen des Vernetzungsmittels bei einer Temperatur unterhalb 120ºC und bevorzugt bei einer Temperatur unterhalb 100ºC mit den organischen säurefunktionellen Gruppen des Polymers in der ersten Beschichtungszusammensetzung und den organischen säurefunktionellen Gruppen des hydrophilen Polymers reagieren. Der Trocknungsschritt für die zweite Beschichtung kann bei einer Temperatur unterhalb 120ºC und bevorzugt bei einer Temperatur unterhalb 100ºC durchgeführt werden, obwohl natürlich höhere Trocknungstemperaturen verwendet werden können, falls dies gewünscht ist und sie mit der Art des zu beschichteten Substrats kompatibel sind. Ein Metallsubstrat könnte beispielsweise bei einer höheren Temperatur getrocknet werden.
- Die vorliegende Erfindung ist jedoch auf die spezifische Absicht ausgerichtet, daß sie bei relativ niedrigen Temperaturen und insbesondere bei Umgebungs- oder Raumtemperatur wirksam ist, um eine Verwendung mit wärmeempfindlichen Substraten zu ermöglichen. In einer weiteren bevorzugten Ausführungsform des erfindungsgemäßen Verfahrens können die funktionellen Gruppen des Vernetzungsmittels bei einer Temperatur im Bereich von 10-70ºC, bevorzugt bei einer Temperatur im Bereich von 15-35ºC mit den organischen säurefunktionellen Gruppen des Polymers in der ersten Beschichtungszusammensetzung und den organischen säurefunktionellen Gruppen der zweiten Beschichtung (des hydrophilen Polymers) reagieren. Eine derartige Reaktivität des Vernetzungsmittels ermöglicht es, das Substrat bei einer Temperatur im Bereich von 10-70ºC und bevorzugt bei einer Temperatur im Bereich von 15-35ºC, wie etwa bei Raumtemperatur, zu beschichten, obwohl natürlich höhere Trocknungstemperaturen verwendet werden können, falls gewünscht.
- Die Trocknungsdauer wird von der Trocknungstemperatur abhängen, wobei höhere Trocknungstemperaturen kürzere Trocknungsdauern erfordern, und umgekehrt. Es liegt jedoch im gewöhnlichen Können eines Fachmanns, eine geeignete Kombination von Trocknungstemperaturen und Trocknungsdauer für eine spezifische Beschichtung zu ermitteln.
- Oftmals wird Trocknen bei Raumtemperatur während etwa 12 Stunden angemessen sein.
- Weiterhin sollte bemerkt werden, daß die funktionellen Gruppen des Vernetzungsmittels nicht notwendigerweise die gleiche Reaktivität gegenüber den organischen säurefunktionellen Gruppen des hydrophilen Polymers und den organischen säurefunktionellen Gruppen der ersten Beschichtungszusammensetzung haben, und daß die Trocknungsbedingungen in a) bzw. b) unter entsprechender Berücksichtigung dieser Reaktivitäten ausgewählt werden.
- Das erfindungsgemäße Verfahren kann zum Beschichten vieler unterschiedlicher Arten von Substraten verwendet werden. Ein Verwendungsgebiet von besonderem Interesse ist das Beschichten medizinischer Gegenstände zur Verwendung in oder an dem Körper, insbesondere von Kathetern, Führungsdrähten oder Teilen derartiger Gegenstände.
- Ballonkatheter, und insbesondere Ballonkatheter für perkutane Angioplastie sind empfindliche Gegenstände, die sich als mit herkömmlichen Verfahren schwierig zu beschichten herausgestellt haben. Ein wichtiger Teil eines Ballonkatheters ist der aufblasbare Ballon, welcher in einem Ballonkatheter für perkutane Angioplastie eine sehr geringe Wandstärke, d. h. in der Größenordnung von etwa 20 um aufweisen kann. Im Zustand, in dem der Ballonkatheter in ein Blutgefäß eingeführt wird, ist der Ballon in eine mehrlagige Konstruktion zusammengefaltet. Es ist daher von großer Bedeutung, daß eine auf die Wand eines derartigen Ballons aufgetragene hydrophile Beschichtung die Zunahme der Wandstärke des Ballons auf einem Minimum hält. Weiterhin ist es wichtig, daß der Ballon aus einem Material hergestellt ist, welches zu einem Ballon mit geringer Wandstärke verarbeitet werden kann, dabei adäquate Festigkeit beibehält und weiterhin die notwendige Biokompatibilität besitzt. Polyethylenterephthalat (PET) weist diese Kombination von Eigenschaften auf, war aber schwierig mit einer hydrophilen Beschichtung zu beschichten. Gemäß der vorliegenden Erfindung wurde jedoch offenbart, daß ein PET-Ballon mit einer Wandstärke von nur etwa 20 um wirksam mit einer hydrophilen Beschichtung mit einer Schichtdicke von etwa 2-3 um beschichtet werden kann, ohne den Ballon zu beschädigen, und die erforderliche Gleitfähigkeit bereitstellt. Die vorliegende Erfindung erreicht dies, da das Verfahren unter Verwendung wässriger Beschichtungszusammensetzungen durchgeführt werden kann, im Gegensatz zu Systemen auf Basis von organischen Lösungsmitteln, und das Trocknen unter milden Bedingungen stattfindet. z. B. einfaches Lufttrocknen der Beschichtung bei Raumtemperatur. Wie vorstehend erwähnt, kann das Trocknen der kombinierten Beschichtungen beispielsweise während etwa 12 bis etwa 24 Stunden bei Raumtemperatur durchgeführt werden.
- Wie vorstehend erwähnt, kann das erfindungsgemäße Verfahren jedoch zum Beschichten vieler unterschiedlicher Substrate verwendet werden, umfassend Substrate, ausgewählt aus polymeren Substraten, nicht-polymeren Substraten und Kombinationen davon.
- Beispielsweise umfassen die geeigneten polymeren Substrate diejenigen, ausgewählt aus der Gruppe, bestehend aus Olefinpolymeren, insbesondere Polyethylen, Polypropylen, Polyvinylchlorid, Polytetrafluorethylen (PTFE), Polyvinylacetat und Polystyrol; Polyestern, insbesondere Poly(ethylenterephthalat); Polyurethanen; Polyharnstoffen; Silikonkautschuken; Polyamiden, insbesondere Nylons; Polycarbonaten; Polyaldehyden; Naturkautschuken; Polyether-Ester- Copolymeren; und Styrol-Butadien-Copolymeren. Diese Liste ist natürlich nicht einschränkend.
- Insbesondere kann das polymere Substrat ausgewählt werden aus der Gruppe, bestehend aus Poly(ethylenterephthalat), Polyurethanen, Polyethylen, Nylon 6, Nylon 11 und Polyether-Ester-Copolymeren.
- Beispiele geeigneter nicht-polymerer Substrate umfassen diejenigen, ausgewählt aus der Gruppe, bestehend aus Keramiken, Metallen, Gläsern und dergleichen.
- Kombinationen von polymeren Substraten und nicht-polymeren Substraten sowie Kombinationen eines oder mehrerer polymerer Substrate und/oder eines oder mehrerer nicht-polymerer Substrate können mit dem erfindungsgemäßen Verfahren ebenfalls beschichtet werden.
- Die Erfindung betrifft auch ein beschichtetes Substrat, wie durch das erfindungsgemäße Verfahren erhältlich, und eine medizinische Vorrichtung, insbesondere einen Katheter oder einen Führungsdraht, welche mit einer Beschichtung, wie mit dem erfindungsgemäßen Verfahren erhältlich, versehen ist.
- Eine besonders bevorzugte erfindungsgemäße medizinische Vorrichtung ist ein Ballonkatheter für perkutane Angioplastie, wovon mindestens der Ballonteil mit einer derartigen Beschichtung versehen ist.
- Die Erfindung wird in den nachfolgenden nicht-einschränkenden Beispielen, welche lediglich bevorzugte Ausführungsformen der Erfindung darstellen, weiter erläutert.
- Eine erste Beschichtungszusammensetzung wurde hergestellt durch Zugeben der nachfolgenden Bestandteile nacheinander in ein Becherglas, unter geeigneter Bewegung, bis sie gründlich gemischt waren.

- 0,5% Fluorad FC-129 Vorratslösung: (hergestellt durch Verdünnung von 1 ml Fluorad FC-129 in 100 ml Wasser) 10 ml

- NeoRez R981 (von Zeneca Resins) ist ein wasserbasiertes aliphatisches Polyurethan auf Polyester-Basis, das Carbonsäuregruppen als internen Emulgator enthält, durch Triethylamin (TEA) stabilisiert ist, und einen Feststoffgehalt von 32% und einen pH-Wert bei 25ºC von 7,5-9,0 hat. Es enthält 5,3% N-Methylpyrrolidon als Colösungsmittel. NeoCryl CX 100 (von Zeneca Resins) ist ein polyfunktionelles Aziridin-Vernetzungsmittel. Fluorad FC-129 (von 3M) wird als ein Egalisierungsmittel zugegeben. Ammoniumhydroxid wird zur Einstellung des pH-Werts der Lösung verwendet.
- Eine zweite Beschichtungszusammensetzung, wie folgt, wurde hergestellt:
- 1,2% wässrige Lösung von Versicol WN23 : 400 ml
- Die obige Lösung wurde hergestellt durch Zugabe einer entsprechenden Menge von Versicol WN-Pulver zu Wasser unter Bewegung während mehrerer Stunden, wobei eine klare homogene Lösung erhalten wurde. Versicol WN23 (von Allied Colloids) ist ein Acrylsäure-Acrylamid-Copolymer mit einem Molekulargewicht von 7,5 · 10&sup6;.
- Ein Substrat wurde hergestellt durch Extrudieren eines Gemisches von zwei Sorten Polyether-Ester-Blockcopolymer ARNITEL EM 740 und EM630 (von Akzo) mit BaSO&sub4; zu einer Röhre. Die Röhre wurde in die erste, vorstehend hergestellte Beschichtungszusammensetzung getaucht und 40 Minuten bei Umgebungstemperatur (Raumtemperatur) getrocknet. Danach wurde die Röhre in die zweite Beschichtungszusammensetzung getaucht und über Nacht bei Umgebungstemperatur getrocknet. Die beschichtete Oberfläche zeigte bei in Kontakt Bringen mit Wasser sehr gute Gleitwirkung. Weiterhin hatte die Beschichtung eine sehr gute Verschleißbeständigkeit und Abriebbeständigkeit, wobei die Beschichtung sogar unter hoher Kraft fest an der Oberfläche gehalten wurde.
- Eine erste Beschichtungszusammensetzung wurde auf die gleiche Weise wie in Beispiel 1 unter Verwendung der nachfolgenden Bestandteile hergestellt:
- U21 X: 250 ml
- Wasser: 100 ml
- NeoCryl CX 100: 10 ml
- U21X (von B.F. Goodrich) ist eine aliphatische Polyurethandispersion auf Polyester- Basis, die Carbonsäuregruppen als internen Emulgator enthält und durch TEA stabilisiert ist. Die Dispersion hat einen Feststoffgehalt von etwa 30%, einen pH-Wert von 8,5 und eine Viskosität von 75 cps. Die Dispersion enthält 8,3% N- Methylpyrrolidon als Colösungsmittel.
- Eine zweite Beschichtungszusammensetzung, wie folgt, wurde auf die gleiche Weise wie in Beispiel 1 hergestellt:
- 1,2% wässrige Lösung von Versicol WN23:
- Ein Ballonkatheter mit einem Poly(ethylenterephthalat) (PET) -Ballon wurde mit den obigen Beschichtungszusammensetzungen in der nachfolgenden Weise beschichtet. Der PET-Ballon wurde aufgeblasen und durch Eintauchen mit der ersten Beschichtungszusammensetzung beschichtet und 30 Minuten bei Umgebungstemperatur getrocknet. Danach wurde der Ballon in die zweite Beschichtungszusammensetzung getaucht und über Nacht bei Umgebungstemperatur getrocknet. Die resultierende getrocknete Beschichtung wurde mittels Elektronenstrahlen in einer Dosis von 2 · 25 KGray sterilisiert.
- Die erhaltene Beschichtung zeigte bei in Kontakt Bringen mit Salzlösung hervorragende Schlüpfrigkeit und Gleitfähigkeit. Die Verschleißbeständigkeit und Abriebbeständigkeit der Beschichtung waren ebenfalls hervorragend.
- Eine erste Beschichtungszusammensetzung wurde hergestellt, wie in Beispiel 1 beschrieben, unter Verwendung der nachfolgenden Bestandteile:
- Bayhydrol LS-2033: 250 ml
- Wasser: 250 ml
- 0,5 Fluorad FC-129 Vorratslösung: 10 ml
- 34% NH&sub4;OH: 4 ml
- NeoCryl CX 100: 20 ml
- Bayhydrol LS-2033 (von Bayer A.G.) ist ein wasserbasiertes Polyurethan, das durch Sulfonatgruppen stabilisiert ist. Das wasserbasierte Polyurethan wie geliefert hat einen pH-Wert von 6,5-7,5, und die Sulfonatgruppen liegen in Form des Natriumsalzes vor. Das Polyurethan hat einen Feststoffgehalt von 40%. Die Dispersion enthält kein Colösungsmittel.
- Eine zweite Beschichtungszusammensetzung wurde hergestellt, wie in Beispiel 1 beschrieben, unter Verwendung der nachfolgenden Bestandteile:
- Versicol WN23 Lösung: 400 ml 1% (w/w)
- Versicol WN23
- NeoRez R960 : 1,0 ml
- Eine Polyurethanröhre (Tecoflex EG-93A, von Thermedics, Inc.) wurde in die erste Beschichtungszusammensetzung getaucht und 10 Minuten bei 60ºC in einem Ofen getrocknet. Danach wurde die Röhre in die zweite Beschichtungszusammensetzung getaucht, 10 Minuten bei 60ºC in einem Ofen getrocknet und ein weiteres Mal in die zweite Beschichtungszusammensetzung getaucht, wonach sie über Nacht bei Umgebungstemperatur getrocknet wurde. Die Beschichtung zeigte bei in Kontakt Bringen mit Wasser hervorragende Schlüpfrigkeit und Gleitfähigkeit.
- Ein Glasträger wurde unter Verwendung der nachfolgenden Beschichtungszusammensetzungen und der gleichen Beschichtungsverfahren und Trocknungsbedingungen wie in Beispiel 1 beschichtet.
- Erste Beschichtungszusammensetzung:
- NeoRez R-940: 100 ml
- NeoCryl CX 100: 4 ml
- NeoRez R-940 (von Zeneca Resins) ist ein wasserbasiertes aromatisches Polyurethan auf Polyetherbäsis.
- Zweite Beschichtungszusammensetzung:
- 1,2% wässrige Lösung von Versicol WN23: 400 ml
- Die Beschichtung zeigte bei in Kontakt Bringen mit Wasser hervorragende Schlüpfrigkeit und Gleitfähigkeit.
- Unter Verwendung der gleichen Beschichtungsverfahren wie in Beispiel 1 beschrieben, wurde ein Edelstahlsubstrat mit den nachfolgenden Beschichtungszusammensetzungen beschichtet.
- Erste Beschichtungszusammensetzung:
- NeoPac E121: 250 ml
- Wasser: 100 ml
- 34% NH&sub4;OH: 2 ml
- NeoCryl CX 100: 16 ml
- Zweite Beschichtungszusammensetzung:
- 1% wässrige Lösung von Versicol WN23 : 400 ml
- erste Beschichtungszusammensetzung 1,5 ml
- Die resultierende Beschichtung zeigte bei in Kontakt Bringen mit Wasser hervorragende Schlüpfrigkeit und Gleitfähigkeit.
- Unter Verwendung der gleichen Beschichtungsverfahren wie in Beispiel 1 beschrieben, wurde ein PET-Substrat mit den nachfolgenden Beschichtungszusammensetzungen beschichtet.
- Erste Beschichtungszusammensetzung:
- Bayhydrol LS 2033: 200 ml
- NeoRez R-940: 100 ml
- Triethylamin: 2 ml
- Wasser: 200 ml
- NeoCryl CX 100: 10 ml
- Zweite Beschichtungszusammensetzung:
- 0,8% wässrige Lösung von Versicol WN23: 400 ml
- Die resultierende Beschichtung zeigte bei in Kontakt Bringen mit Wasser hervorragende Schlüpfrigkeit und Gleitfähigkeit.
- Eine Glasplatte wurde wie nachstehend beschrieben mit den nachfolgenden Beschichtungszusammensetzungen beschichtet.
- Erste Beschichtungszusammensetzung:
- Sancure 899: 200 ml
- NeoPac E121: 100 ml
- Acrysol TT-615: (vorverdünnt mit dem gleichen Gewicht Wasser) 1 ml
- SAG 710: /ml
- 34% NH&sub4;OH: 4 ml
- Zweite Beschichtungszusammensetzung:
- 1% wässrige Lösung von Versicol WN23: 400 ml
- Die erste Beschichtungszusammensetzung wurde auf die Glasplatte aufgebürstet und 1 Stunde bei Raumtemperatur getrocknet. Danach wurde die zweite Beschichtungszusammensetzung auf die vorbeschichtete Glasoberfläche aufgesprüht und über Nacht bei Umgebungstemperatur getrocknet. Die erhaltene Beschichtung zeigte bei in Kontakt Bringen mit Wasser hervorragende Schlüpfrigkeit und Gleitfähigkeit.
- Acrysol TT-615 ist ein von Rohm und Haas Company erhältliches Verdickungsmittel und SAG 710 ist ein von OSI Specialties, Inc. erhältliches Antischaummittel.
- Erste Beschichtungszusammensetzung:
- Sancure 899: 250 ml
- 0,5% Fluorad FC-129 Vorratslösung: 10 ml
- 34% NH&sub4;OH: 4 ml
- Wasser: 200 ml
- Ucarlink XL-29SE: 40 ml
- Zweite Beschichtungszusammensetzung:
- 1% wässrige Lösung von Versicol WN23: 400 ml
- Ein Ballon aus Polyurethan (Impranil ELN, von der Bayer A.G.) wurde in die erste Beschichtungszusammensetzung getaucht und 40 Minuten bei Umgebungstemperatur getrocknet. Danach wurde der Ballon in die zweite Beschichtungszusammensetzung getaucht, 30 Minuten bei Umgebungstemperatur getrocknet und danach ein weiteres Mal in die zweite Beschichtungszusammensetzung getaucht. Die Beschichtung wurde mittels EtO (Ethylenoxid) Sterilisierung sterilisiert. Die Beschichtung zeigte bei in Kontakt Bringen mit Wasser hervorragende Schlüpfrigkeit und Gleitfähigkeit.
- Ucarlink XL-29SE ist ein von Union Carbide erhältliches polyfunktionelles Carbodiimid.
- Eine PET-Röhre wurde wie nachstehend beschrieben mit den nachfolgenden Beschichtungszusammensetzungen beschichtet.
- Erste Beschichtungszusammensetzung:
- NeoPac E121: 250 ml
- Wasser: 250 ml
- Ucarlink XL-29SE: 40 ml
- Zweite Beschichtungszusammensetzung:
- 1% wässrige Lösung von Versicol WN23: 400 ml
- erste Beschichtungszusammensetzung: 1 ml
- Die PET-Röhre wurde in die erste Beschichtungszusammensetzung getaucht und 30 Minuten luftgetrocknet. Danach wurde die vorbeschichtete Röhre in die zweite Beschichtungszusammensetzung getaucht und 30 Minuten luftgetrocknet, gefolgt von 24-stündigem Trocknen bei 60ºC. Die Beschichtung zeigte bei in Kontakt Bringen mit Wasser hervorragende Schlüpfrigkeit und Abriebbeständigkeit.
- NeoRez R-981, NeoRez R-940, NeoRez R-961, NeoRez R-972, NeoRez R-976, NeoRez R-973, NeoPac E-106, NeoPac E-130, NeoPac E-121, NeoCryl CX 100, Fluorad FC-129, U21, U21X, Versicol WN23, Bayhydrol LS-2033, Bayhydrol LS- 2100, Bayhydrol LS-2952, Bayhydrol LS-2990, Sancure 899, Sancure 2710, Sancure 1601, Sancure 2026, Ucarlink XL-29SE, Acrysol TT-615 und SAG 710 sind Marken, welche in einem oder mehreren der benannten Länder registriert sein können.
Claims (28)
1. Verfahren zum Bereitstellen eines Substrats mit einer hydrophilen
Beschichtung, welche bei in Kontakt Bringen mit einem wässrigen Fluid
gleitfähig wird, wobei das Verfahren umfaßt:
a) Beschichten des Substrats mit einer ersten wässrigen
Beschichtungszusammensetzung, umfassend eine wässrige Dispersion
oder Emulsion eines Polymers mit organischen säurefunktionellen
Gruppen und ein polyfunktionelles Vernetzungsmittel mit funktionellen
Gruppen, die mit organischen sauren Gruppen reagieren können, und
Trocknen der ersten Beschichtungszusammensetzung, wobei eine im
wesentlichen wasserunlösliche Beschichtungslage erhalten wird, welche
noch Gruppen umfaßt, die mit organischen sauren Gruppen reaktiv sind,
und
b) in Kontakt Bringen der ersten getrockneten Beschichtungslage mit einer
zweiten wässrigen Beschichtungszusammensetzung, umfassend eine
wässrige Lösung oder Dispersion eines hydrophilen Polymers mit
organischen säurefunktionellen Gruppen, und Trocknen der kombinierten
Beschichtungen, wobei eine Bindung des hydrophilen Polymers an das
Polymer der ersten Beschichtungszusammensetzung durch das
Vernetzungsmittel bewirkt wird.
2. Verfahren nach Anspruch 1, wobei das hydrophile Polymer durch kovalente
Bindung an das Polymer der ersten Beschichtungszusammensetzungen
gebunden wird.
3. Verfahren nach Anspruch 1, wobei die organischen säurefunktionellen Gruppen
aus freien Carbon- oder Sulfonsäuregruppen, Metallsalzen, insbesondere
Alkalimetallsalzen derartiger Säuregruppen und quaternären Aminsalzen
derartiger Säuregruppen ausgewählt werden.
4. Verfahren nach Anspruch 1, wobei das polyfunktionelle Vernetzungsmittel aus
polyfunktionellen Aziridinen, polyfunktionellen Carboimiden und Kombinationen
davon ausgewählt wird.
5. Verfahren nach Anspruch 1, wobei das polyfunktionelle Vernetzungsmittel ein
Vernetzungsmittel mit mehr als zwei funktionellen Gruppen pro Molekül ist.
6. Verfahren nach Anspruch 1, wobei das Polymer in der ersten
Beschichtungszusammensetzung aus Homo- und Copolymeren mit einer
erheblichen Menge an organischen säurefunktionellen Gruppen in ihrer Struktur
ausgewählt wird.
7. Verfahren nach Anspruch 1, wobei das Polymer in der ersten
Beschichtungszusammensetzung aus der Gruppe, bestehend aus
Polyurethanen, Polyacrylaten, Polymethacrylaten, Polyisocrotonaten,
Epoxyharzen, Acrylat-Urethan-Copolymeren und Kombinationen davon
ausgewählt wird.
8. Verfahren nach Anspruch 7, wobei das Polymer in der ersten
Beschichtungszusammensetzung ein Acrylat-Urethan-Copolymer und/oder ein
Polyurethan umfaßt.
9. Verfahren nach Anspruch 1, wobei das hydrophile Polymer Monomereinheiten
mindestens eines Monomers mit organischen säurefunktionellen Gruppen
umfaßt.
10. Verfahren nach Anspruch 9, wobei das hydrophile Polymer aus der Gruppe,
bestehend aus Acrylsäure, Methacrylsäure, Isocrotonsäure und Kombinationen
davon ausgewählt wird.
11. Verfahren nach Anspruch 9, wobei das hydrophile Polymer weiter
Monomereinheiten ohne organische säurefunktionelle Gruppen umfaßt.
12. Verfahren nach Anspruch 9, wobei das hydrophile Polymer ein Acrylamid-
Acrylsäure-Copolymer umfaßt.
13. Verfahren nach Anspruch 1, wobei das hydrophile Polymer in der zweiten
Beschichtungszusammensetzung ein Molekulargewicht im Bereich von etwa
100.000 bis etwa 15 Millionen aufweist.
14. Verfahren nach Anspruch 13, wobei das hydrophile Polymer in der zweiten
Beschichtungszusammensetzung ein Molekulargewicht im Bereich von etwa
150.000 bis etwa 10 Millionen aufweist.
15. Verfahren nach Anspruch 14, wobei das hydrophile Polymer in der zweiten
Beschichtungszusammensetzung ein Molekulargewicht im Bereich von etwa
400.000 bis etwa 10 Millionen aufweist.
16. Verfahren nach Anspruch 1, wobei die Konzentration des hydrophilen Polymers
in der zweiten Beschichtungszusammensetzung von etwa 0,1 bis etwa 5%,
bezogen auf das Gewicht des Feststoffgehalts, beträgt.
17. Verfahren nach Anspruch 16, wobei die Konzentration des hydrophilen
Polymers in der zweiten Beschichtungszusammensetzung von etwa 0,5 bis
etwa 3%, bezogen auf das Gewicht des Feststoffgehalts, beträgt.
18. Verfahren nach Anspruch 1, wobei die Konzentration des Polymers in der
ersten Beschichtungszusammensetzung von etwa 2 bis etwa 60%, bezogen auf
das Gewicht des Feststoffgehalts, beträgt.
19. Verfahren nach Anspruch 1, wobei die Konzentration des Polymers in der
ersten Beschichtungszusammensetzung von etwa 5 bis etwa 40%, bezogen auf
das Gewicht des Feststoffgehalts, beträgt.
20. Verfahren nach Anspruch 1, wobei die Konzentration des Vernetzungsmittels in
der ersten Beschichtungszusammensetzung im Bereich von etwa 0,2 bis etwa
30 Gew.-% liegt.
21. Verfahren nach Anspruch 1, wobei die funktionellen Gruppen des
Vernetzungsmittels mit den organischen säurefunktionellen Gruppen des
Polymers in der ersten Beschichtungszusammensetzung und des hydrophilen
Polymers in der zweiten Beschichtungszusammensetzung bei einer Temperatur
unterhalb 120ºC reagieren können.
22. Verfahren nach Anspruch 21, wobei die funktionellen Gruppen des
Vernetzungsmittels mit den organischen säurefunktionellen Gruppen des
Polymers in der ersten Beschichtungszusammensetzung und des hydrophilen
Polymers in der zweiten Beschichtungszusammensetzung bei einer Temperatur
in einem Bereich von 10-70ºC reagieren können.
23. Medizinische Vorrichtung, vorgesehen zum Einführen in den Körper,
umfassend:
ein zum Einführen in den Körper geeignetes Substrat, wobei die Oberfläche des
Substrats mit einer vernetzten Polymerzusammensetzung beschichtet ist, die
Zusammensetzung eine erste Polymerschicht, gebildet aus einer mindestens
partiellen Reaktion einer wässrigen Dispersion oder Emulsion eines Polymers
mit vorhandenen reaktiven organischen säurefunktionellen Gruppen mit einem
polyfunktionellen Vernetzungsmittel, welches mit den organischen sauren
Gruppen reagieren kann, und eine zweite hydrophile Polymerschicht mit
vorhandenen organischen säurefunktionellen Gruppen, die mit dem
Vernetzungsmittel reagieren können, umfaßt.
24. Reaktiver Beschichtungsfilm, geeignet zum Binden von hydrophilen Polymeren
mit organischen säurefunktionellen Gruppen, wobei der Beschichtungsfilm das
Reaktionsprodukt einer wässrigen Dispersion oder Emulsion eines Polymers
mit organischen säurefunktionellen Gruppen mit einem polyfunktionellen
Vernetzungsmittel, welches mit den organischen säurefunktionellen Gruppen
reagieren kann, ist, wobei das Reaktionsprodukt noch reaktive funktionelle
Gruppen des polyfunktionellen Vernetzungsmittels umfaßt.
25. Hydrophile Polymerbeschichtung, welche bei in Kontakt Bringen mit einem
wässrigen Medium gleitfähig werden kann, wobei die Beschichtung mindestens
zwei kovalent aneinander gebundene Polymerlagen umfaßt, wobei ein
vernetztes Netzwerk gebildet wird, wobei das vernetzte Netzwerk das
Reaktionsprodukt ist aus:
a) einer ersten Polymerlage, umfassend eine wässrige Dispersion oder Emulsion
eines Polymers mit organischen säurefunktionellen Gruppen und ein
polyfunktionelles Vernetzungsmittel, welches mit den organischen
säurefunktionellen Gruppen reagieren kann, und
b) einer zweiten wässrigen Polymerlage, umfassend eine wässrige Lösung oder
Dispersion eines hydrophilen Polymers mit organischen säurefunktionellen
Gruppen.
26. Hydrophile Polymerbeschichtung nach Anspruch 25, wobei die erste
Polymerlage aus der Gruppe, bestehend aus wasserbasierten Polyurethan- und
Acryl-Urethan-Copolymeren ausgewählt ist und die zweite Polymerlage aus der
Gruppe, bestehend aus Polyacrylsäure-Polymer und Acrylamid-Acrylsäure-
Copolymeren ausgewählt ist.
27. Medizinische Vorrichtung, welche mindestens teilweise mit der
Polymerbeschichtung nach Anspruch 25 beschichtet ist.
28. Gegenstand nach Anspruch 27, wobei die medizinische Vorrichtung ein
Angioplastieballon ist.
