Description
- Diese Anmeldung ist eine Teilfortführung der U.S.-Seriennummer 07/771.760, angemeldet am 4. Oktober 1991, deren Inhalte hiermit durch Bezugnahme in dieser Offenbarung eingeschlossen werden. Die hierin beschriebene Erfindung wurde im Verlauf der Arbeit unter Grant Nummer CA-57227-01 von den Nationalen Gesundheitsinstituten durchgeführt. Die Regierung der Vereinigten Staaten hat an dieser Erfindung bestimmte Rechte.
- Hintergrund der Erfindung
- In dieser Anmeldung wird durch arabische Nummern in Klammern auf verschiedene Veröffentlichungen Bezug genommen. Vollständige Zitate für diese Veröffentlichungen können am Ende der Beschreibung direkt vor den Patentansprüchen gefunden werden. Die Offenbarungen dieser Veröffentlichungen in ihrer Vollständigkeit sind hiermit durch Bezugnahme in diese Anmeldung aufgenommen, um den Stand der Technik, zu dem diese Erfindung gehört, vollständiger zu beschreiben.
- Krebs ist eine Erkrankung, bei der eine Zellpopulation auf die Kontrollmechanismen, welche normalerweise die Proliferation und Differenzierung steuern, in verschiedenem Ausmaß unreaktiv geworden ist. Über viele Jahre gab es zwei Hauptstrategien zur chemotherapeutischen Behandlung von Krebs: a) Blockierung von hormonabhängiger Tumorzell-Proliferation durch Eingriff in die Produktion oder die periphere Wirkung von Sexualhormonen; und b) direktes Abtöten von Krebszellen durch Aussetzen derer von cytotoxischen Stoffen, die sowohl neoplastische als auch normale Zellpopulationen verletzen.
- Erst seit kurzem wird die Krebstherapie auch durch die Induktion von terminaler Differenzierung der neoplastischen Zellen angestrebt (1). Es wurde berichtet, dass in Zellkultur-Modellen die Differenzierung durch Exposition der Zellen von verschiedenen Stimulantien, einschließlich zyklischer AMP und Vitamin-A Säure (2, 3), Aclarubicin und anderen Anthracyclinen (4) ausgelöst wurde.
- Es gibt reichlich Belege, dass die neoplastische Transformierung nicht notwendigerweise das Potential von Krebszellen zur Differenzierung zerstört (1, 5, 6). Es gibt viele Beispiele von Tumorzellen, die auf die normalen Regulatoren der Poliferation nicht reagieren und die in der Expression ihres Differenzierungsprogramms blockiert zu sein scheinen, und die dennoch zur Differenzierung und zur Einstellung der Replikation induziert werden können. Eine Vielfalt von Agenzien, einschließlich einiger relativ einfacher polarer Verbindungen (5, 7–9), Derivative von Vitamin D und Vitamin-A Säure (10–12), Steroidhormone (13), Wachstumsfaktoren (6, 14), Proteasen (15, 16), Tumorpromotoren (17, 18) und Inhibitoren der DNA- oder RNA-Synthese (4, 19–24) können verschiedene transformierte Zelllinien und primäre menschliche Tumor-Explantate induzieren, so dass sie differenziertere Eigenschaften exprimieren.
- Frühe Untersuchungen durch die Erfinder der vorliegenden Anmeldung identifizierten eine Reihe an polaren Verbindungen, die bei einer Anzahl von transformierten Zelllinien (8, 9) effiziente Induktoren der Differenzierung waren. Von diesen war der effizienteste Induktor die hybride polare/apolare Verbindung N,N'-Hexamethylen-bisacetamid (HMBA)(9).
- Die Verwendung dieser polaren/apolaren Verbindung zur Induktion von erythroleukämischen Mäusezellen (MELC) zur Erythroid-Differenzierung mit einer Suppression der Onkogenität, bewies sich als ein nützliches Modell zur Untersuchung von Induktoren-vermittelter Differenzierung von transformierten Zellen (5, 7–9). Eine durch HMBA-induzierte MELC-terminate Erythroid-Differenzierung ist ein mehrstufiger Prozeß. Nach Zusetzen von HMBA zu MELC (745A-DS19) in Kultur gibt es eine latente Periode von 10 bis 12 Stunden, bevor eine Vorbestimmung zu einer terminalen Differenzierung nachgewiesen wird. Das Vorbestimmen wird als die Fähigkeit der Zellen, eine terminale Differenzierung trotz des Entfernens des Induktors (25) zu exprimieren, definiert. Bei einer kontinuierlichen Exposition von HMBA gibt es eine fortschreitende Zunahme von Zellen, welche differenzieren. Die Erfinder der vorliegenden Anmeldung berichteten, dass MELC-Zelllinien, die gegen relativ geringe Mengen an Vincristin resistent gemacht wurden, auf die induzierende Wirkung von HMBA deutlich empfindlicher werden und sie können mit einer kleinen oder keiner latenten Periode zur Differenzierung induziert werden (26).
- HMBA ist in der Lage, phänotypische Veränderungen, die bei vielen unterschiedlichen Zelllinien (5) einer Differenzierung entsprechen, zu induzieren. Die Eigenschaften der durch den Arzneistoff induzierten Wirkung wurden bei dem erythroleukämischen Zellsystem bei Mäusen (MELC) am umfassendsten untersucht (5, 25, 27, 28). Die MELC-Induktion der Differenzierung ist sowohl Zeit- als auch konzentrationsabhängig. Die minimale Konzentration, die erforderlich ist um bei den meisten Stämmen in vitro eine Wirkung zu zeigen, liegt bei 2 bis 3 mM; die minimale Dauer einer kontinuierlichen Exposition, die allgemein erforderlich ist, um in einem wesentlichen Anteil (> 20%) der Population eine Differenzierung ohne Weiterführung der Wirkstoffsexposition zu induzieren, liegt bei etwa 36 Stunden.
- Das primäre Ziel der Wirkung von HMBA ist nicht bekannt. Es gibt Anzeichen dafür, dass die Proteinkinase C auf dem Weg der Inducer-vermittelten Differenzierung beteiligt ist (29). Die Untersuchungen in vitro stellten eine Basis für die Evaluation des Potenzials von HMBA als zelldifferenzierendem Mittel bei der Behandlung von Krebs beim Menschen bereit (30). Verschiedene klinische Versuche der Phase I mit HMBA wurden abgeschlossen (31–36). Klinische Versuche haben gezeigt, dass diese Verbindung bei Patienten mit Krebs eine therapeutische Reaktion induzieren kann (35, 36). Diese klinischen Versuche der Phase I haben jedoch auch gezeigt, dass die potenzielle Wirksamkeit von HMBA teilweise durch dosisabhängige Toxizität limitiert wird, die es verhindert, dass optimale Blutspiegel erreicht werden und dadurch, dass über einen verlängerten Zeitraum die intravenöse Verabreichung von großen Mengen des Mittels benötigt werden.
- Kürzlich berichteten die Erfinder der vorliegenden Anmeldung von mehreren Verbindungen, die mit HMBA verwandt sind, mit polaren Gruppen, die durch apolare Verbindungen getrennt sind, die auf einer molaren Basis ebenso aktiv sind (37) oder 100 Mal aktiver sind als HMBA (38). Als Klasse jedoch wurde herausgefunden, dass die symmetrischen Dimere wie HMBA und verwandte Verbindungen nicht die besten zelldifferenzierenden Mittel sind.
- EP-A-0 576 941 offenbart die Verbindung 5-{N-[2,5-Di(benzyloxy)benzoyl]amino}-pentylcarbohydroxam-Säure und ihre Verwendung als Medikament.
- WO 93/07/48 beschreibt Verbindungen, die eine terminale Differenzierung induziert mit der Struktur
- In Proc. Natl. Acad. Sci. (1991) Band 58, S. 5542–5546 werden zelldifferenzierende Mittel offenbart, die Derivate von Bishydroxamsäure sind.
- Acta Cient. Venez (1981) 232(5) S. 411–416 offenbart die Verbindung 6-Benzamidohexanoyldihydroxamsäure.
- Unerwarteterweise wurde herausgefunden, dass die besten Verbindungen zwei Reste mit polaren Enden umfassen, die durch eine flexible Kette aus Methylengruppen getrennt sind, wobei eine oder beide der polaren Endgruppen ein großer hydrophober Rest ist. Vorzugsweise unterscheiden sich die polaren Endgruppen voneinander und nur eine ist ein großer hydrophober Rest. Diese Verbindungen sind unerwarteterweise tausendmal aktiver als HMBA und zehnmal mehr aktiv als Verbindungen, die mit HMBA in verwandt sind.
- Diese neue Klasse von Verbindungen der vorliegenden Erfindung kann bei der selektiven Induktion der terminalen Differenzierung von neoplastischen Zellen von Nutzen sein und somit bei der Behandlung von Tumoren bei Patienten helfen.
- Zusammenfassung der Erfindung
- Die vorliegende Endung stellt eine Verbindung mit der Struktur bereit: worin R eine Piperidin-, Thiazol- oder Phenylgruppe ist, die mit einer Methyl-, Cyano-, Nitro-, Thio-, Trifluormethyl-, Amino-, Aminocarbonyl-, Methylcyano-, Chlor-, Fluor-, Brom-, Iod-, 2,3-Difluor-, 2,4-Difluor-, 2,5-Difluor-, 3,4-Difluor-, 3,5-Difluor-, 2,6-Difluor-, 1,2,3-Trifluor-, 2,3,6-Trifluor-, 2,4,6-Trifluor-, 3,4,5-Trifluor-, 2,3,5,6-Tetrafluor-, 2,3,4,5,6-Pentafluor-, Azido-, Hexyl-, t-Butyl-, Phenyl-, Carboxyl-, Hydroxyl-, Methyloxy-, Phenyloxy-, Phenylaminooxy-, Phenylaminocarbonyl-, Methyloxycarbonyl-, Methylaminocarbonyl-, Dimethylamino-, Dimethylaminocarbonyl-, oder Hydroxylaminocarbonylgruppe substituiert ist und n eine gerade Zahl von 4 bis 8 oder ein pharmazeutisch verträgliches Salz davon ist.

- Zusätzlich stellt die vorliegende Erfindung ein in vitro-Verfahren zur selektiven Induktion einer terminalen Differenzierung von neoplastischen Zellen zur Verfügung, wobei die Proliferation solcher Zellen gehemmt wird, was ein Inkontaktbringen der Zellen mit einer wirksamen Menge einer beliebigen der vorstehenden Verbindungen, die für die selektive Induktion einer terminalen Differenzierung wirksam ist, unter geeigneten Bedingungen umfasst.
- Die vorliegende Erfindung stellt ebenfalls die Verwendung der vorstehenden Verbindungen zur Herstellung eines Arzneimittels, welches für die selektive Induktion einer terminalen Differenzierung solcher neoplastischen Zellen wirksam ist und dadurch ihre Proliferation hemmt, für die Behandlung von Patienten, die einen Tumor haben, der durch die Proliferation von neoplastischen Zellen charakterisiert wird, zur Verfügung.
- Die vorliegende Erfindung stellt ebenfalls ein Arzneimittel umfassend eine pharmazeutisch verträgliche Menge einer beliebigen der vorstehenden Verbindungen oder pharmazeutisch verträgliche Salze davon und einen pharmazeutisch verträglichen Träger zur Verfügung.
- Schließlich stellt die vorliegende Erfindung das vorstehend definierte Arzneimittel, allein oder in Kombination mit einem Antitumormittel in einer Form mit verlängerter Freisetzung zur Verfügung.
- Detaillierte Beschreibung der Erfindung
- Die vorliegende Erfindung stellt eine Verbindung mit der Struktur bereit: worin R eine Piperidin-, Thiazol- oder Phenylgruppe ist, die mit einer Methyl-, Cyano-, Nitro-, Thio-, Trifluormethyl-, Amino-, Aminocarbonyl-, Methylcyano-, Chlor-, Fluor-, Brom-, Iod-, 2,3-Difluor-, 2,4-Difluor-, 2,5-Difluor-, 3,4-Difluor-, 3,5-Difluor-, 2,6-Difluor-, 1,2,3-Trifluor-, 2,3,6-Trifluor-, 2,4,6-Trifluor-, 3,4,5-Trifluor-, 2,3,5,6-Tetrafluor-, 2,3,4,5,6-Pentafluor-, Azido-, Hexyl-, t-Butyl-, Phenyl-, Carboxyl-, Hydroxyl-, Methoxy-, Phenyloxy-, Phenylaminooxy-, Phenylaminocarbonyl-, Methoxycarbonyl-, Methylaminocarbonyl-, Dimethylamino-, Dimethylaminocarbonyl-, oder Hydroxylaminocarbonylgruppe substituiert ist und n eine gerade Zahl von 4 bis 8 oder ein pharmazeutisch verträgliches Salz davon ist.

- In einer bevorzugten Ausführungsform der vorstehend definierten Verbindung ist R eine substituierte Phenylgruppe. In einer stärker bevorzugten Ausführungsform ist die Phenylgruppe mit einer Methyl-, Cyano-, Nitro-, Thio-, Trifluormethyl-, Amino-, Aminocarbonyl-, Methylcyano-, Chlor-, Fluor-, Brom-, Iod-, 2,3-Difluor-, 2,4-Difluor-, 2,5-Difluor-, 3,4-Difluor-, 3,5-Difluor-, 2,6-Difluor-, 1,2,3-Trifluor-, 2,3,6-Trifluor-, 2,4,6-Trifluor-, 3,4,5-Trifluor-, 2,3,5,6-Tetrafluor-, 2,3,4,5,6-Pentafluor-, Azido-, Hexyl-, t-Butyl-, Phenyl-, Carboxyl-, Hydroxyl-, Methyloxy-, Phenyloxy-, Benzyloxy-, Phenylaminooxy-, Phenylaminocarbonyl-, Methyloxycarbonyl-, Methylaminocarbonyl-, Dimethylamino-, Dimethylaminocarbonyl-, oder Hydroxylaminocarbonylgruppe substituiert.

- Die vorliegende Erfindung stellt des Weiteren ein in vitro-Verfahren zur selektiven Induktion einer terminalen Differenzierung von neoplastischen Zellen zur Verfügung, wobei die Vermehrung solcher Zellen gehemmt wird, was ein Inkontaktbringen der Zellen mit einer wirksamen Menge einer beliebigen der vorstehenden Verbindungen, die für die selektive Induktion einer terminalen Differenzierung wirksam ist, unter geeigneten Bedingungen umfasst.
- Das Inkontaktbringen muss kontinuierlich über einen längeren Zeitraum, d. h. über mindestens 48 Stunden, bevorzugt über etwa 4–5 Tage oder länger durchgeführt werden.
- Das Verfahren soll in vitro ausgeführt werden, das Inkontaktbringen kann durch Inkubation der Zellen mit der Verbindung erreicht werden. Die Konzentration der Verbindung, die mit den Zellen in Kontakt gebracht wird, sollte etwa zwischen 1 μM bis etwa 25 mM, bevorzugt zwischen 4 μM und etwa 5 mM liegen. Die Konzentration hängt von der einzelnen Verbindung und dem Stadium der neoplastischen Zellen ab.
- Das Verfahren kann ebenfalls eine anfängliche Behandlung der Zellen mit einem Antitumormittel umfassen, um sie gegen ein Antitumormittel resistent zu machen und die entstandenen resistenten Zellen danach unter geeigneten Bedingungen mit einer wirksamen Menge eines der vorstehend erwähnten Verbindungen in Kontakt zu bringen, welche ausreicht, um die terminale Differenzierung derartiger Zellen selektiv zu induzieren.
- Das Antitumormittel kann eines von zahlreichen chemotherapeutischen Mitteln sein, wie ein alkylierendes Mittel, ein Antimetabolit, ein hormonelles Mittel, ein Antibiotikum, Colchicin, ein VincaAlkaloid, L-Asparaginase, Procarbazin, HydroxylHarnstoff, Mitotan, NitrosoHarnstoffe oder ein ImidazolCarboxamid. Geeignete Mittel sind jene Mittel, die die Depolarisierung von Tubulin fördern. Vorzugsweise ist das Antitumormittel Colchicin oder ein VincaAlkaloid; besonders bevorzugt sind Vinblastin und Vincristin. In Ausführungsformen, in welchen das Antitumormittel Vincristin ist, werden die Zellen vorzugsweise so behandelt, dass sie gegen Vincristin bei einer Konzentration von etwa 5 mg/ml resistent sind. Die Behandlung der Zellen dahingehend, dass sie gegen ein Antitumormittel resistent werden, kann durch Inkontaktbringen der Zellen mit dem Mittel über einen Zeitraum von mindestens 3–5 Tagen erreicht werden. Das Inkontaktbringen der entstandenen Zellen mit einem der vorstehenden Verbindungen wird wie zuvor beschrieben durchgeführt.
- Die vorliegende Erfindung stellt ebenfalls die Verwendung von jeder beliebigen der vorstehenden Verbindungen oder von pharmazeutisch verträglichen Salzen davon für die Herstellung eines Arzneimittels, welches für die selektive Induktion einer terminalen Differenzierung solcher neoplastischen Zellen wirksam ist und dadurch ihre Proliferation hemmt, für die Behandlung von Patienten, die einen Tumor haben, der durch die Proliferation von neoplastischen Zellen charakterisiert wird, zur Verfügung. Außerdem stellt die Erfindung die Verwendung einer Verbindung mit der Struktur: worin R eine substituierte oder unsubstituierte Phenyl-, Piperidin- oder Thiazolgruppe ist und n eine gerade Zahl von 4 bis 8 oder ein pharmazeutisch verträgliches Salz davon ist, für die Verwendung eines Arzneimittels zur selektiven Induktion einer terminalen Differenzierung von neoplastischen Zellen bereit. Und vorzugsweise, wobei die selektive Induktion die Proliferation der neoplastischen Zellen bei der Behandlung von Patienten hemmt, die einen Tumor haben, der durch die Proliferation von neoplastischen Zellen charakterisiert wird. Die Verwendung der vorliegenden Erfindung zielt auf die Behandlung von menschlichen Patienten mit Tumoren ab. Es ist jedoch ebenfalls wahrscheinlich, dass die Verwendung bei der Behandlung von Tumoren bei anderen Säugern wirksam wäre. Der Begriff Tumor soll jeden Krebs, der durch die Proliferation von neoplastischen Zellen verursacht wird, wie Lungenkrebs, akutes lymphoides Myelom, Blasenmelanom, Nierenkarzinom, Brustkarzinom, oder Kolorektalkarzinom umfassen. Die Verabreichung der Verbindung an den Patienten kann oral oder parenteral erfolgen. Bis jetzt hat sich die intravenöse Verabreichung als wirksam erwiesen. Die Verabreichung der Verbindung muss kontinuierlich über einen längeren Zeitraum durchgeführt werden, etwa mindestens über 3 Tage und bevorzugt mehr als 5 Tage. In den am stärksten bevorzugten Ausführungsformen wird die Verabreichung kontinuierlich über mindestens 10 Tage ausgeführt und wird in Intervallen wiederholt, wobei die Verabreichung in jedem Intervall kontinuierlich über mindestens 10 Tage ausgeführt wird. Die Verabreichung kann zum Beispiel in so kurzen Intervallen wie 5–10 Tagen bis zu etwa 25–35 Tagen und fortlaufend über mindestens 10 Tage während eines jeden solchen Intervalls ausgeführt werden. Der optimale Zeitraum eines Intervalls wird abhängig von dem Typ des Patienten und des Tumors variieren. Im Falle einer akuten Leukämie, dem sogenannten myelodysplastischen Syndrom, scheint zum Beispiel eine fortlaufende Infusion indiziert, so lange der Patient den Wirkstoff ohne Toxizität vertägt und es eine positive Reaktion gibt.
- Die Menge der an den Patienten verabreichten Verbindung ist weniger als eine Menge, die beim Patienten eine Toxizität verursachen würde. In bestimmten Ausführungsformen ist die Menge, die an den Patienten verabreicht wird, weniger als die Menge, die eine Konzentration im Plasma des Patienten hervorruft, die den toxischen Wert der Verbindung erreicht oder übersteigt. Die Konzentration der Verbindung im Plasma des Patienten wird bevorzugt bei etwa 1,0 mM aufrechterhalten. Es wurde mit HMBA herausgefunden, dass eine Verabreichung der Verbindung in einer Menge von etwa 5 g/m2/Tag bis etwa 30 g/m2/Tag, insbesondere etwa 20 g/m2/Tag wirksam ist, ohne dass eine Toxizität beim Patienten hervorgerufen wird. Die optimale Menge der Verbindung, die bei der Umsetzung der vorliegenden Erfindung an den Patienten verabreicht werden sollte, wird von der bestimmten Verbindung, welche verwendet wird und dem Krebstyp, der behandelt wird, abhängen.
- Es ist beabsichtigt, dass diese Erfindung, zusätzlich zu den vorstehend aufgelisteten Verbindungen, die Verwendung von Homologa und Analoga solcher Verbindungen umfasst. In diesem Kontext sind Homologa Moleküle, die wesentliche strukturelle Ähnlichkeiten mit den vorstehend beschriebenen Verbindungen haben, und Analoga sind Moleküle, die wesentliche biologische Ähnlichkeiten haben, wobei strukturelle Ähnlichkeiten nicht berücksichtigt werden.
- Die Verwendung kann auch die anfängliche Verabreichung einer Menge eines Antitumormittels an einen Patienten umfassen, um die Zellen gegen ein Antitumormittel resistent zu machen und eine nachfolgende Verabreichung einer wirksamen Menge einer beliebigen der vorstehenden Verbindungen oder pharmazeutisch verträglicher Salze davon an den Patienten umfassen, welche wirksam sind, um eine terminale Differenzierung derartiger neoplastischer Zellen zu induzieren und dadurch ihre Proliferation zu hemmen.
- Das Antitumormittel kann eines von zahlreichen chemotherapeutischen Mitteln sein, wie ein alkylierendes Mittel, ein Antimetabolit, ein hormonelles Mittel, ein Antibiotikum, Colchicin, ein VincaAlkaloid, L-Asparaginase, Procarbazin, HydroxylHarnstoff Mitotan, NitrosoHarnstoffe oder ein Imidazol-Carboxamid. Geeignete Mittel sind jene Mittel, die die Depolarisation von Tubulin fördern. Das Antitumormittel ist vorzugsweise Colchicin oder ein VincaAlkaloid; besonders bevorzugt sind Vinblastin und Vincristin. In Ausführungsformen, in welchen das Antitumormittel Vincristin ist, wird eine Menge verabreicht, um die Zellen bei einer Konzentration von etwa 5 mg/ml gegen Vincristin resistent zu machen. Die Verabreichung des Mittels wird im Wesentlichen wie vorstehend beschrieben für die Verabreichung jede beliebige der Verbindungen durchgeführt. Vorzugsweise geht die Verabreichung des Mittels über einen Zeitraum von mindestens 3–5 Tagen. Die Verabreichung einer beliebigen der vorstehenden Verbindungen wird wie zuvor beschrieben durchgeführt.
- Die vorliegende Erfindung stellt ebenfalls ein Arzneimittel umfassend eine therapeutisch wirksame Menge eines der vorstehenden Verbindungen oder pharmazeutisch verträglicher Salze davon und einen pharmazeutisch verträglichen Träger wie steriles Pyrogen-freies Wasser zur Verfügung. Vorzugsweise ist die therapeutisch verträgliche Menge eine Menge, die für die selektive Induktion einer terminalen Differenzierung von geeigneten neoplastischen Zellen wirksam ist und weniger als eine Menge, die bei einem Patienten Toxizität hervorruft. Des Weiteren stellt die vorliegende Erfindung ebenfalls ein Arzneimittel umfassend einen pharmazeutisch verträglichen Träger und eine therapeutisch wirksame Menge einer Verbindung zur Verfügung mit der Struktur: worin R ein unsubstituiertes Phenyl und n eine gerade Zahl von 4 bis 8 oder ein pharmazeutisch verträgliches Salz davon ist bereit.
- Die vorliegende Erfindung stellt das vorstehende Arzneimittel in Kombination mit einem Antitumormittel zur Verfügung. Das Antitumormittel kann jedes der zuvor beschriebenen Mittel sein.
- Schließlich stellt die vorliegende Erfindung das vorstehende Arzneimittel, allein oder in Kombination mit einem Antitumormittel in einer Form mit verlängerter Freisetzung zur Verfügung. Mit einer Form mit verlängerter Freisetzung meinen die Anmelder den Einschluss der Arzneimittel in eine pharmazeutisch verträgliche Formulierung, die die verlängerte Freisetzung einer therapeutisch wirksamen Menge der Verbindungen dieser Erfindung über einen Zeitraum bereitstellt, welche notwendig ist, um die beabsichtigte therapeutische Wirkung zu erzielen. Formulierungen von Arzneimitteln mit verlängerter Freisetzung erlauben eine weniger häufige Verabreichung der Verbindung und stellen eine Verabreichung des Arzneimittels an oder nahe dem Zielbereich im System eines Lebewesens zur Verfügung. Formulierungen mit verlängerter Freisetzung und Verfahren zum Einschluss von Arzneimitteln darin sind dem Durchschnittsfachmann bekannt. Beispiele umfassen solche Formulierungen wie Einschluss in Ionenaustauschharzen (U.S.-Patent Nr. 5,296,228 an Chang et al.), Xanthangummis (U.S.-Patent Nr. 5,292,534 an Valentine et al.), Mikrosphären (U.S.-Patent Nr. 5,288,502 an McGinity et al.), Hydrogele (U.S.-Patent Nr. 5,266,325 an Kuzma et al.) und feste Formen wie wachsähnliche oder fettähnliche hydrophobe Stoffe, die wasserunlösliche Polymere (U.S.-Patent Nr. 5,270,055 an Moest), sind jedoch nicht auf sie beschränkt. Verfahren zur Verabreichung von Verbindungen zur verlängerten Freisetzung sind ebenfalls auf dem Fachgebiet bekannt und umfassen die chirurgische Implantation von in mikroverkapselten pharmazeutischen Verbindungen nahe der beabsichtigten Zielstelle (U.S.-Patent Nr. 5,290,271 an Jernberg) sowie den Einschluss der Verbindung in transdermale Pflaster (U.S.-Patent Nr. 5,298,256 an Flockhart et al und U.S.-Patent Nr. 5,290,561 an Farhadieh et al.), sind jedoch nicht auf sie beschränkt. Der Text der vorstehend zitierten Patente und die darin offenbarten Bezugnahmen sind hierdurch in ihrer Gesamtheit in diese Offenbarung durch Bezugnahme eingeschlossen.
- Die Erfindung wird im Abschnitt der folgenden Versuchsdetails veranschaulicht. Dieser Abschnitt wird gezeigt, um das Verständnis der Erfindung zu unterstützen, und sollte nicht so ausgelegt werden, dass es die Erfindung auf irgendeine Weise beschränkt, wie in den Patentansprüchen, die danach folgen, gezeigt wird.
- Versuchsdetails
- Zellen und Materialien
- MELC 745A-DS19-Zellen und die Varianten von MELC, die von dieser Zelllinie abstammen, nämlich die Vincristin-resistenten MELC-V3.17- und VCR.C(2)15-Zelllinien (26) und die gegen Dimethylsulfoxid resistente Zelllinie DR10 (39), wurden in minimalem essentiellem α-Medium, welches 10% fötales Kälberserum (16) enthielt, gehalten. Die Zellkulturen für alle Versuche wurden mit Zellen in der logarithmischen Wachstumsphase (Zellen vom Tag 2 der Kultur) in einer Dichte von 105 Zellen/ml gestartet. Induktorverbindungen wurden in die nachstehend angezeigten Endkonzentrationen zugesetzt, in Kulturmedium ohne fötales Kälberserum gelöst, wenn nicht anders angezeigt. Zelldichte und Benzidin-Reaktivität wurden wie beschrieben bestimmt (16).
- Die Vorbestimmung zur terminalen Differenzierung, charakterisiert durch eine begrenzte Zellteilung (Koloniezahl < 32 Zellen) und die Anhäufung von Hämoglobin (auf Benzidin reaktive Kolonien), wurde durch einen Kolonie-Klonierungstest unter Verwendung von 2% Methylcellulose wie beschrieben (25) (siehe Tabelle 1 hinsichtlich der Ergebnisse) getestet.
- In einer 25 ml-Flasche wurde eine Lösung aus 0,371 g 7-Aminoheptansäure mit 0,3145 g NaOH in 12 ml Wasser auf 0°C abgekühlt und dann wurden 0,5 ml Benzoylchlorid in 8 ml trockenem THF tropfenweise 30 Minuten lang zugesetzt. Nach 3,5 Stunden Rühren wurde das THF abgedampft und die Lösung wurde auf einen pH-Wert von 1 angesäuert. Der entstandene Niederschlag von 7-Benzoylamidoheptanoylhydroxamsäure wurde aufgefangen und mit Ether gewaschen. Er wurde durch NMR und Massenspektroskopie (M + 1 = 250) charakterisiert. Dann wurden 0,20 g dieser Amidsäure drei Stunden lang mit 0,1750 Carbonyldiimidazol in 10 ml trockenem THF behandelt. Zu dieser rührenden Lösung wurden 0,1114 g Hydroxylamid-Hydrochlorid zugesetzt und die Lösung wurde über Nacht bei Raumtemperatur gerührt. Dann wurden 3 ml 0,1 N HCl zugesetzt, das THF wurde abgedampft, und der Rückstände wurde in 5 ml Ethylacetat und 3 ml Salzlösung aufgenommen. Die hergestellte HydroxamSäure war als elfenbeinfarbener Feststoff in der organischen Schicht vorhanden; sie wurde durch Filtration in einer Ausbeute von 60% aufgefangen. Sie wurde durch NMR und Massensprektrum (M + 1 = 265) charakterisiert und hatte einen Schmelzpunkt von 105°C.
- Auf eine ähnliche Weise wurden Analoga mit n = 5 oder 6, und mit R = p-Cyanophenyl, m-Cyanophenyl und Thiophenyl, durch die Verwendung des geeigneten Carbon-säurechlorids und 7-Aminoheptansäure oder 6-Aminohexansäure im ersten Schritt hergestellt.
- Zu einer eiskalten Lösung aus 5 ml Suberoylchlorid in 20 ml THF wurden tropfenweise unter Rühren 1,37 ml Methanol und 4,7 ml Triethylamin in 40 ml THF zugesetzt. Nach 19 Stunden wurde eine Lösung aus 3,2032 g 4-Aminopyridin und 4,7 ml Triethylamin in 250 ml THF tropfenweise unter Rühren und Eiskühlung zugesetzt. Nach 24 Stunden wurde eine kleine Menge weißen Feststoffs durch Filtration entfernt, das THF wurde abgedampft und das Rohprodukt wurde chromatographiert, um 2,8879 g des Methylesters dieses Amidesters zu erhalten, und wurde zu einer Lösung von 0,9866 g Hydroxylamin-Hydrochlorid in 17 ml Methanol mit 0,8887 g NaOH zugesetzt, und die gefilterte Lösung wurde zwei Tage bei Raumtemperatur stehen gelassen. Das präzipitierte Salz der Hydroxamsäure wurde mit wenig Ethanol gewaschen und in 0,1242 g Essigsäure in 10 ml Wasser gerührt. Nach 48 Stunden waren 0,2291 g der Hydroxamsäure kristallisiert, und sie wurde aufgefangen und aus Methanol rekristallisiert, um das reine Produkt, mit einem Schmelzpunkt von 202–203°C zu erhalten. Es wurde durch NMR und Massenspektrum (M + 1 = 266) charakterisiert.
- Auf eine ähnliche Weise wurden die 2-Pyridyl- und 3-Pyridyl-Analoga, unter Verwendung der geeigneten Amine, hergestellt.
- Zu 3,0 g 6-Aminocapronsäure in 150 ml THF wurden 3,5 ml Triethylamin, dann 3 ml m-Chlorphenylisocyanat zugesetzt. Nach Stehen lassen über Nacht wurde die Lösung filtriert und durch Abdampf konzentriert. Dann lieferte eine Verteilung zwischen Wasser und Ether, gefolgt von einer Ansäuerung der wässrigen Schicht auf einen pH-Wert von 3,0, ein Präzipitat der Ureidocarboxylsäure in einer Ausbeute von 35%, was durch NMR und Massenspektrum (M – 1 = 285) charakterisiert wurde. Dieses wurde dann durch Behandlung von 0,0418 g der Säure mit 0,321 g Carbonyldiimidazol in 25 mg THF in das Hydroxamsäure-Produkt umgewandelt. Nach zwei Stunden bei Raumtemperatur wurde die Lösung mit 0,1948 g Hydroxylamin-Hydrochlorid behandelt und 20 Stunden gerührt. Dann wurden 15 ml 0,1 N HCl und 25 ml Ethylacetat zugesetzt, und das THF wurde abgedampft. Das Produkt erschien als Kristalle in der organischen Schicht und wurde in einer Ausbeute von 38% aufgefangen. Es hatte einen Schmelzpunkt von 162–163°C und wurde durch NMR und eine Elementaranalyse charakterisiert: C, 51,62; H, 5,82; N, 13,47. berechnet C, 52,0; H 6,05; N, 14,00.
- Auf eine ähnliche Weise wurden die unsubstituierten Phenyl-Analoga aus Phenyl-Isocyanat hergestellt.

Claims (18)
- Verbindung mit der Struktur: wobei R eine Piperidin- oder Thiazolgruppe oder eine Phenylgruppe ist, die mit einer Methyl-, Cyano-, Nitro-, Thio-,Trifluormethyl-, Amino-, Aminocarbonyl-, Methylcyano-, Chlor-, Fluor-, Brom-, Iod-, 2,3-Difluor-, 2,4-Difluor-, 2,5-Difluor-, 3,4-Difluor-, 3,5-Difluor-, 2,6-Difluor-; 1,2,3-Trifluor-, 2,3,6-Trifluor-, 2,4,6-Trifluor-, 3,4,5-Trifluor-, 2,3,5,6-Tetrafluor-, 2,3,4,5,6-Pentafluor-, Azid-, Hexyl-, t-Butyl-, Phenyl-, Carboxyl-, Hydroxyl-, Methyloxy-, Phenyloxy-, Phenylaminooxy-, Phenylaminocarbonyl-, Methyloxycarbonyl-, Methylaminocarbonyl-, Dimethylamino-, Dimethylaminocarbonyl- oder Hydroxylaminocarbonylgruppe substituiert ist, und n eine ganze Zahl von 4 bis 8 ist, oder ein pharmazeutisch verträgliches Salz davon.
- 7-Benzoylamidoheptanoyl-hydroxamsäure oder ein pharmazeutisch verträgliches Salz davon.
- Verbindung nach Anspruch 4, wobei R eine substituierte Phenylgruppe ist.
- Verbindung nach Anspruch 5, wobei die Phenylgruppe mit einer Methyl-, Cyano-, Nitro-, Thio-; Trifluormethyl-, Amino-, Aminocarbonyl-, Methylcyano-, Chlor-, Fluor-, Brom-, Iod-, 2,3-Difluor-, 2,4-Difluor-, 2,5-Difluor-, 3,4-Difluor-, 3,5-Difluor-, 2,6-Difluor-, 1,2,3-Trifluor-, 2,3,6-Trifluor-, 2,4,6-Trifluor-, 3,4,5-Trifluor-, 2,3,5,6-Tetrafluor-, 2,3,4,5,6-Pentafluor-, Azid-, Hexyl-, t-Butyl-, Phenyl-, Carboxyl-, Hydroxyl-, Methyloxy-, Phenyloxy-, Benzyloxy-, Phenylaminooxy-, Phenylaminocarbonyl-, Methyloxycarbonyl-, Methylaminocarbonyl-, Dimethylamino-, Dimethylaminocarbonyl- oder Hydroxylaminocarbonylgruppe substituiert ist.
- In-vitro-Verfahren zur selektiven Induktion der terminalen Differenzierung von neoplastischen Zellen und damit zur Hemmung der Proliferation derartiger Zellen, umfassend das Inkontaktbringen der Zellen unter geeigneten Bedingungen mit einer wirksamen Menge der Verbindungen nach einem der Ansprüche 1 bis 8 oder eines pharmazeutisch verträglichen Salzes davon, wirksam zur selektiven Induktion der terminalen Differenzierung.
- Verwendung der Verbindungen nach einem der Ansprüche 1 bis 8 oder eines pharmazeutisch verträglichen Salzes davon für die Herstellung eines Arzneimittels, das zur Behandlung von Patienten mit einem Tumor wirksam ist, wobei der Tumor durch die Proliferation neoplastischer Zellen gekennzeichnet ist.
- Arzneimittel, umfassend einen pharmazeutisch verträglichen Träger und eine therapeutisch wirksame Menge der Verbindungen nach einem der Ansprüche 1 bis 8 oder eines pharmazeutisch verträglichen Salzes davon.
- Arzneimittel nach Anspruch 11 oder 12, wobei die wirksame Menge eine Menge ist, die zur selektiven Induktion der terminalen Differenzierung von geeigneten neoplastischen Zellen wirksam ist und geringer ist als eine Menge, die Toxizität in einem Patienten verursacht.
- Arzneimittel nach Anspruch 11 oder 12 in Kombination mit einem Antifumormittel.
- Arzneimittel nach Anspruch 12 in langzeitwirkender Form.
- Arzneimittel nach Anspruch 14 in langzeitwirkender Form.
- Verwendung der Verbindung mit der Struktur: wobei R eine substituierte oder unsubstituierte Phenyl-, Piperidin- oder Thiazolgruppe und n eine ganze Zahl von 4 bis 8 ist, oder eines pharmazeutisch verträglichen Salzes davon für die Herstellung eines Arzneimittels, das zur Behandlung von Patienten mit einem Tumor wirksam ist, wobei der Tumor durch die Proliferation neoplastischer Zellen gekennzeichnet ist.
- Verwendung nach Anspruch 17, wobei die neoplastischen Zellen für eine terminale Differenzierung selektiv induziert sind.
