Description
- Die vorliegende Erfindung betrifft wasserlösliche organische Salze von Kreatin der allgemeinen Formel I:
- wobei A das Anion einer Mono-, Bi- oder Tricarbonsäure darstellt. Bevorzugte Anionen sind Citrat, Maleat, Fumarat, Tartrat oder Malat.
- Kreatin oder N-(Aminoiminomethyl)-N-methylglycin ist ein Sarcosin-Derivat, das in Muskelgewebe vieler Vertebraten, einschließlich Menschen, hauptsächlich in Kombination mit Phosphorsäure in Form von Phosphorylkreatin vorliegt, und ist bei der Energieübertragung von Mitochondrien wie ATP-Verwendungsstellen beteiligt.
- Zahlreiche Studien deuten darauf hin, daß eine Beziehung zwischen der Kreatin- (Phosphorylkreatin)-Konzentration in den Muskeln mit der Funktion der Aufrechterhaltung eines hohen intracellulären ATP/ADP-Verhältnisses und der maximalen erträglichen physischen Anstrengung besteht (Annu. Rev. Biochem. 54: 831-862, 1985; Science 24: 448 -452, 1981; BESSMAN S. P., und F. SAVABI. The role of the phospocreatine energy shuttle in exercise and muscle hypertrophy. - Der Kreatin-Anstieg in der Nahrung kann daher nützlich sein, um die Plasma-Kreatin- Konzentrationen auf Spiegel zu bringen, die signifikante Werte von Kreatin selbst im Muskel liefern. Die kurze Kreatin-Halbwertszeit im Plasma (1 bis 1,5 Stunden) erfordert jedoch, daß diese Spiegel schnell erreicht werden, und dies wird angesichts des Bioverfügbarkeitsgrads von Kreatin nur durch Gabe von hohen Dosen von 5 bis 10 g (bei mittleren Körpergewichten von 70 kg) möglich, Mengen, die wegen des Fehlens der Toxizität der Verbindung gut verträglich sind.
- Die geringe Löslichkeit von Kreatin in Wasser (1 g in 75 ml) ist daher eine praktische Limitierung für die Möglichkeit, in einer spezifischen Diät die notwendigen Mengen von Kreatin sofort verfügbar zu machen.
- WO 94/02127, veröffentlicht am 3. Februar1 1994 offenbart die Verwendung von Kreatin, gegebenenfalls mit Aminosäuren oder anderen Komponenten kombiniert, zur Erhöhung der Muskelleistung in Säugern.
- Die vorliegende Erfindung stellt wasserlösliche stabile Salze von Kreatin der Formel I bereit, charakterisiert durch hohe Wasserlöslichkeit (3 bis 15 mal höher wie die von Kreatin selbst) sowie ein Verfahren für ihre Herstellung. Die Salze der Formel I werden durch Salzbildung von Kreatin mit den entsprechenden Säuren in wäßriger oder hydroalkoholischer konzentrierter Lösung oder in einem nicht mit Wasser mischbaren Lösungsmittel bei Temperaturen im Bereich von Raumtemperatur bis 50ºC, gegebenenfalls Konzentrieren der Lösungen und Filtrieren der kristallisierten Salze hergestellt. Entsprechend einer bevorzugten Ausführungsform werden die Salze der Formel I durch Umsetzung von Kreatin mit einem Überschuß organischer Säure in Ethylacetat hergestellt, bis das Salz vollständig gebildet wird, nachweisbar durch IR-Analyse, Abkühlen und Filtrieren. Das die überschüssige Säure enthaltende filtrierte Lösungsmittel wird recycliert und nach Auffüllen der Komponenten für eine weitere Reaktion verwendet.
- Die Salze werden durch IR, Schmelzpunkt, potentiometrische und HPLC-Assays charakterisiert.
- Tabelle 1 gibt die Löslichkeit der Salze I gemäß der Erfindung an.

- 39,45 g (0,18 Mol) Citronensäure-Monohydrat werden in 100 ml Ethylacetat suspendiert. 20 g (0,134 Mol) Kreatin-Monohydrat werden bei 20 bis 25ºC zu der gerührten Suspension gegeben, und die Mischung wird während 4 Stunden bei 25ºC gerührt. Nach IR- Kontrolle wird das Produkt filtriert und mit Ethylacetat gewaschen, dann im Ofen bei 50 bis 55ºC getrocknet, wobei 90% Salze erhalten werden, Smp.: 112-114ºC, 99,2% Titer.
- 14,9 g (0,1 Mol) Kreatin-Monohydrat werden zu einer Lösung von 11,6 g (0,1 Mol) Maleinsäure und 20 ml Wasser gegeben. Die so erhaltene Lösung wird konzentriert, auf 5ºC gekühlt und das Produkt wird filtriert und unter Vakuum bei 50ºC getrocknet, wobei 87% Salz erhalten werden, Smp.: 128-129ºC, 99,8% Titer.
Claims (4)
1. Salze von Kreatin der allgemeinen Formel I:
wobei A das Anion von Citronen-, Malein-, Fumar- oder Äpfelsäure bedeutet.
2. Verfahren zur Herstellung von Salzen nach Anspruch 1, umfassend:
a) gleichzeitiges Lösen von Kreatin und der organischen Säure in Wasser;
b) Konzentrieren der Lösung bis zur vollständigen Kristallisation und Trocknen des
erhaltenen Produkts.
3. Verfahren zur Herstellung von Salzen nach Anspruch 1, umfassend:
a) Reaktion von Kreatin mit der organischen Säure in einem nicht mit Wasser
mischbaren Lösungsmittel bei einer Temperatur von Raumtemperatur bis 50ºC;
b) Filtration und Trocknen des erhaltenen Produkts.
4. Verwendung von Salzen nach Anspruch 1 als Additiv in diätetischen und/oder
Lebensmittel-Zusammensetzungen.
