Die vorliegende Erfindung betrifft gereinigte Heparin-Fraktionen und genauer Heparin-Fraktionen, die frei von Gruppen N-NQ sind, ein Verfahren zu ihrer Herstellung und Arzneimittel, die sie enthalten.The present invention relates to purified heparin fractions and more particularly to heparin fractions free from N-NQ groups, a process for their preparation and medicinal products containing them.
Die vorliegende Erfindung betrifft insbesondere Heparin- Fraktionen, die durch Nitrit-Depolyrnerisation erhalten wurden und die praktisch keine Nitroso-Verbindungen enthalten.The present invention relates in particular to heparin fractions obtained by nitrite depolymerization and which contain practically no nitroso compounds.
Es ist bekannt, daß Heparin-Fraktionen durch Nitrit-Depolymerisation erhalten werden, wie dies beispielsweise in den Patentanmeldungen EP-A-0 014 184 und EP-A-0 027 089 beschrieben wird. Verschiedene Heparin-Fraktionen, die durch Nitrit-Depolymerisation hergestellt wurden, sind Wirkstoffe in Arzneimitteln, die allgemein unter der Bezeichnung "Heparine mit niedrigem Molekulargewicht" oder "Heparine mit geringer Molekülmasse" allgemein verwendet werden. Bezüglich Einzelheiten zu Heparinen mit niedrigem Molekulargewicht, die als Arzneimittel verwendet werden, ihrer quantitativen Bestimmung und ihrem als Verhältnis Sulfatgruppen/Carboxylgruppen ausgedrückten Sulfatierungsgrad kann auf die Veröffentlichung "Héparines de faible masse moleculaire", Pharmeuropa 3 Nr. 3, Oktober 1991, S. 161-165, sowie auf die für das Europäische Arzneibuch (Pharmacopoea Europaea) vorgeschlagene Monographie "Heparina massae molecularis minoris", Februar 1993 (PA/PH/Exp. 3T (92) 93), verwiesen werden. So hergestellte und als Wirkstoffe in Arzneimitteln verwendete Heparine mit niedrigem Molekulargewicht werden in den unter den Nummern FR-A-2478646, EP-A-0037319, EP-A-0076279 und EP-A-0181259 veröffentlichten Patentanmeldungen oder Patenten beschrieben. Sie sind unter den internationalen Freinamen (INN) Nadroparin-Calcium, Dalteparin-Natrium und Reviparin-Natrium bekannt und stellen Wirkstoffe von im Handel befindlichen oder potentiellen Spezialitäten dar. Insbesondere Reviparin-Natrium bildet den Wirkstoff der in Deutschland im Handel befindlichen Spezialität CLIVARIN . Weitere Heparine mit niedrigem Molekulargewicht, die durch Nitrit-Depolymerisation erhalten wurden, liegen ferner in Spezialitäten vor, die der Ärzteschaft in Deutschland (MONO-EMBOLEX NM), in Österreich (SANDOPARIN und TROPARIN ) und in der Schweiz (SAN- DOPARINE ) zur verfügung stehen.It is known that heparin fractions are obtained by nitrite depolymerization, as described, for example, in patent applications EP-A-0 014 184 and EP-A-0 027 089. Dieser Lösung wird Natriumchlorid zugesetzt, bis eine Leitfähigkeit von etwa 20000 µS/cm erreicht wird. Danach wird der pH-Wert mit konzentrierter Salzsäure auf etwa 4 eingestellt, und der Lösung wird unter Rühren 1 Volumen absolutes Ethanol zugesetzt. Man läßt etwa 60 h absetzen und entfernt anschließend die überstehende wäßrig-alkoholische Lösung. Die in den Tabellen 1 und II angegebenen Ergebnisse zeigen, daß die Behandlung mit UV keinerlei Veränderung des Nadroparins hervorruft, da die verschiedenen physikochemischen und biologischen Eigenschaften der Probe 1, wie das Chromatogramm des Produkts, der Gehalt der verschiedenen Verunreinigungen, die Anti-Faktor-Xa-Aktivität und die Anti- Faktor-IIa-Aktivität sind mit einer Ausnahme mit den Eigenschaften der nicht mit UV-Strahlung behandelten Vergleichsprobe identisch sind. Die Reaktion der Depolymensation wird mit einem Kaliumiodid-Stärke-Papier verfolgt; die Reaktion ist beendet, wenn der Test negativ verläuft. Der pH-Wert der Reaktionslösung wird mit konzentriertem Natriumhydroxid auf 10 bis 10,5 eingestellt, und anschließend werden 2,5 g Natriumborhydrid zugesetzt. Man beläßt das Gemisch 15 h unter Rühren und stellt dann den pH-Wert mit konzentrierter Salzsäure auf 3,5 bis 4 ein, um den Überschuß an Borhydrid zu vernichten. Durch Zusatz von konzentriertem Natriumhydroxid wird der pH-Wert auf 7 eingestellt. Dann fällt man durch Zusatz von 2 Volumina Ethanol pro Volumen Lösung. Man läßt den Niederschlag absetzen und entfernt den Überstand. Man erhält so CY 222 in Form des Natriumsalzes. Man läßt den Niederschlag absetzen und entfernt den Überstand Der Niederschlag wird in gereinigtem Wasser so gelöst, daß eine Endkonzentration von 20 % (M/V), bezogen auf die Menge des anfänglich eingesetzten Heparin-Natriums, erhalten wird; die Leitfähigkeit der Lösung wird mit Natriumchlorid auf 30 bis 35 mS/cm eingestellt, und der pH-Wert wird mit konzentrierter Salzsäure oder konzentriertem Natriumhydroxid auf 7 bis 7,5 eingestellt. Man führt dann eine Fällung der Lösung mit 2 Volumina absolutem Ethanol durch und läßt absetzen. Man gewinnt den Niederschlag, wäscht mit Ethanol und trocknet ihn bei einer Temperatur von höchstens 60 ºC. Auf diese Weise wird reines CY 222 erhalten, das als Natriumsalz eines depolymerisierten Heparins, das durch Depolymerisation von Heparin aus der Darmschleimhaut von Schweinen mit salpetriger Säure erhalten wurde, definiert ist und folgende Eigenschaften aufweist:The solution obtained after treatment with UV radiation is purified on a chromatography column containing at least 2 l of anionic resin per kilogram of heparin sodium used.
