Description
Die Erfindung betrifft Verbindungen der allgemeinen Formel (I)
worin
- a) X und Y die Gruppe CH bedeuten, wobei
R¹ für C₁- bis C₆-Alkyl, Phenyl, 2-Methoxyphenyl, 4-Methoxy phenyl, 2-Methoxycarbonylphenyl, 4-Methoxycarbonylphenyl oder N- Methylpiperidin-3-yl steht,
oder worin - b) X ein Stickstoffatom und Y die Gruppe CR² bedeutet, wobei
R¹ Phenyl, 2-Methoxyphenyl, 4-Methoxyphenyl, 2-Methoxycarbonyl phenyl, 4-Methoxycarbonylphenyl oder N-Methylpiperidin-3-yl be deutet,
R² für Wasserstoff, Methyl oder Phenyl steht,
und deren Salze mit physiologisch verträglichen Säuren und Basen.
Für die im Zusammenhang mit der vorliegenden Anmeldungsbe
schreibung genannten verschiedenen Substituenten bzw. Reste
gelten folgende Erläuterungen:
Beispiele für C₁- bis C₆-Alkylreste sind unverzweigte und ver
zweigte Kohlenwasserstoffreste mit ein bis sechs Kohlenstoff
atomen wie Methylreste, Ethylreste, n-Propylreste, iso-Propyl
reste, n-Butylreste, iso-Butylreste, 1-Methylpropylreste, tert.-
Butylreste, n-Pentylreste, 1-Methylbutylreste, 2-Methylbutyl
reste, 3-Methylbutylreste, 1,1-Dimethylpropylreste, 2,2-Di
methylpropylreste, 1,2-Dimethylpropylreste, 1-Ethylpropylreste,
n-Hexylreste, 1-Methylpentylreste, 2-Methylpentylreste, 3-Me
thylpentylreste, 4-Methylpentylreste, 1,1-Dimethylbutylreste,
2,2-Dimethylbutylreste, 3,3-Dimethylbutylreste, 1,2-Dimethyl
butylreste, 2,3-Dimethylbutylreste, 1,3-Dimethylbutylreste, 1-
Ethylbutylreste, 2-Ethylbutylreste, 1,1,2-Trimethylpropylreste,
1,2,2-Trimethylpropylreste, 1-Ethyl-2-methylpropylreste oder 1-
Ethyl-1-methylpropylreste.
Dabei sind Methylreste, Ethylreste, n-Propylreste, iso-Propyl
reste, n-Butylreste oder iso-Butylreste bevorzugt. Besonders
bevorzugt sind Ethylreste, iso-Propylreste und iso-Butylreste.
Sofern die erfindungsgemäßen Verbindungen in Salzform vorlie
gen, handelt es sich dabei um Salze mit physiologisch verträg
lichen anorganischen oder organischen Säuren und Basen. Bei
spiele für Salze mit physiologisch verträglichen Basen sind
Ammonium-, Natrium-, Kalium-, Lithium-, Magnesium- und Calcium
salze sowie Salze mit Ethanolamin, Triethanolamin, Morpholin
oder Piperidin.
Beispiele für Salze mit physiologisch verträglichen Säuren sind
Citrat-, Tartrat-, Acetat-, Fumarat-, Gluconat-, Glutamat-, Lactat-,
Malat-, Maleat-, Mesylat-, Succinat-, Carbonat-, Hydrogencarbo
nat-, Hydrogensulfat-, Phosphat-, Hydrogenphosphat-, Dihydro
genphosphat-, Chlorid- und Bromid-haltige Salze.
Die Synthese von Verbindungen der allgemeinen Formel (I) er
folgt in Analogie zu literaturbekannten Verfahren. Die Erfin
dung betrifft daher auch ein Verfahren zur Herstellung von
Verbindungen der allgemeinen Formel (I), das dadurch gekenn
zeichnet ist, daß man eine Verbindung der allgemeinen Formel
(II),
worin X und Y die vorstehend genannten Bedeutungen haben, mit
Halogenameisensäure-estern der allgemeinen Formel (III) um
setzt,
worin R¹ die vorstehend genannte Bedeutung hat und Hal die Be
deutung Fluor- oder Chloratome hat, gemäß dem folgenden Reak
tionsschema:
Als Base können sowohl organische, als auch anorganische Basen
verwendet werden, zum Beispiel tertiäre Amine, Pyridin, Natri
umacetat, Natrium- und Kaliumhydroxid, Natrium- und Kaliumcar
bonat, Natrium- und Kaliumhydrogencarbonat, Natrium- und Cal
ciumhydrid und elementares Natrium oder Kalium. Besonders be
vorzugt sind dabei Triethylamin, Natriumcarbonat und Natrium
hydrid (vergleiche Literatur: z. B. U. Petersen in Houben-Weyl,
Methoden der Organischen Chemie, Band E4, Kohlensäurederivate,
S. 142 ff, Georg Thieme Verlag, Stuttgart 1983 und dort zitier
te Literatur).
Als Lösungsmittel können je nach verwendeter Base Methylen
chlorid, Chloroform, Toluol, Essigsäureethylester, Aceton,
Tetrahydrofuran, Dimethylformamid, Pyridin oder Acetonitril
verwendet werden. Besonders bevorzugt sind dabei Methylenchlo
rid, Toluol, Aceton und Tetrahydrofuran.
Die Umsetzungen werden je nach Verbindung unter Eiskühlung, bei
Raumtemperatur oder beim Siedepunkt des jeweiligen Lösungsmit
tels durchgeführt. Alternativ setzt man eine Verbindung der
allgemeinen Formel (II) mit einem entsprechenden Kohlensäuredi
ester der allgemeinen Formel (IV) um,
worin R¹ die vorstehend genannte Bedeutung hat und R⁹ entweder
identisch mit R¹ ist oder eine geeignete Abgangsgruppe R⁹O⁻ bil
det, wie z. B. elektronegativ substituierte Phenoxy-Gruppen (z. B.
4-Nitro-, 2-Nitro-, 2,4-Dinitro-, 2,3,5-Trichlor-4-Acetyl
phenoxy) oder geeignete Hydroxylamine (z. B. 1-Hydroxypiperi
din, N-Hydroxy-succinimid, N-Hydroxy-phthalimid), gemäß dem
folgenden Reaktionsschema:
Die Umsetzung erfolgt bei ca. 100-150°C durch Zusammenschmel
zen der beiden Komponenten oder in einem hochsiedenden Lösungs
mittel wie Dimethylformamid oder Dimethylsulfoxid.
Die entsprechenden Ausgangsverbindungen mit der allgemeinen
Formel (II),
worin X und Y die vorstehend genannten Bedeutungen haben, wer
den in Anlehnung an verschiedene aus der Literatur bekannte Me
thoden hergestellt:

Verbindungen der allgemeinen Formel (III) und auch der allge
meinen Formel (IV) werden durch Umsetzung der entsprechenden
Alkohole mit Phosgen, mit Chlorameisensäuretrichlormethylester
oder mit Bis-(trichlormethyl)-carbonat erhalten. In Anlehnung
an literaturbekannte Methoden (vergleiche Literatur: z. B. G.
Heywang in Houben-Weyl, Methoden der Organischen Chemie, Band
E4, Kohlensäurederivate, S. 9 ff, Georg Thieme Verlag, Stutt
gart 1983 und dort zitierte Literatur; U. Petersen, dito, S. 64
ff) läßt man dabei das Phosgen direkt auf niedere Alkohole ein
wirken, oder man setzt beide Komponenten in einem inerten Lö
sungsmittel wie Methylenchlorid oder Diethylether mit einer
Base wie Triethylamin oder Pyridin um und erhält je nach
Stöchiometrie Verbindungen der allgemeinen Formel (III) bzw.
(IV).
Die Verbindungen der allgemeinen Formel (I) sind wertvolle
pharmazeutische Wirkstoffe. Wie die Anmelderin überraschender
weise festgestellt hat, zeichnen sich die erfindungsgemäßen
Verbindungen durch eine gute antithrombotische Wirksamkeit aus;
sie sind frei von unerwünschten Eigenschaften und somit sehr
gut verträglich. Diese Verbindungen sind ferner potente Inhi
bitoren bindegewebsabbauender Enzyme, insbesondere der Granulo
zytenelastase.
Aus dem Stand der Technik sind Verbindungen mit ähnlicher Struk
tur wie die erfindungsgemäßen Verbindungen bekannt (NL-PS
6702189; DE-OS 18 07 685; Bull. Korean Chem. Soc. 11, 7 (1990);
Reports Inst. Med. Dent. Eng. 8, 9 (1974); FR-PS 1460552). Von
diesen bekannten Verbindungen sind jedoch andere Wirkungen be
schrieben, als sie die erfindungsgemäßen Verbindungen aufwei
sen.
Ferner sind aus der Literatur folgende Arbeiten bekannt:
In Chem. Abst. 79, 146537 (1973) sind Phtalazinoncarbamate be
schrieben, die an der Stelle von R² in den erfindungsgemäßen
Verbindungen notwendigerweise eine carbamoylsubstituierte Me
thylgruppe tragen.
Brit. Pat. 1 365 806 beschreibt Phtalazinoncarbamate, die den
obengenannten sehr ähnlich sind, jedoch ein leicht unterschied
liches Substitutionsmuster am Benzolring aufweisen. Diese Ver
bindungen zeigen antithrombotische und antiatherosclerotische
Eigenschaften.
Die DE-OS 24 51 417 beschreibt Phtalazinone mit antithrombo
tischen und antiatherosclerotischen Eigenschaften. Die hier
beschriebenen Verbindungen tragen an der Stelle von R² in den
erfindungsgemäßen Verbindungen notwendigerweise eine Hydroxy
methylgruppe und weisen nicht die R¹-substituierte Carbamoyl
gruppe der erfindungsgemäßen Verbindungen auf.
Die DE-OS 29 32 259 beschreibt antithrombotische Phtalazinon
derivate, die an der Stelle von R² eine Hydroxymethylgruppe
tragen und am Stickstoffatom, das der Ringcarbonylgruppe be
nachbart ist, einen Phenylsubstituenten tragen.
Die Wirkungen der erfindungsgemäßen Verbindungen ermöglichen
deren Verwendung zur Prävention und Behandlung thromboembo
lischer Erkrankungen oder Zustände und ferner zur Prävention
und Behandlung von Krankheiten, die mit übermäßigem Bindege
websabbau, insbesondere des Gelenkknorpels, verbunden sind.
Beispiele für thromboembolische Erkrankungen, bei denen die An
wendung der erfindungsgemäßen Substanzen vorteilhaft ist, sind
tiefe und oberflächliche Venenthrombosen sowie arterielle Throm
bosen wie Herzinfarkt oder ischämische Gehirninsulte.
Als Krankheiten, die mit übermäßigem Bindegewebsabbau verbunden
sind und die sich mit den erfindungsgemäßen Verbindungen behan
deln lassen, sind vor allem Arthritiden und Arthrosen und fer
ner Muskeldystrophie und Lungenemphysem zu nennen. Ein weiteres
Krankheitsbild, das mit den erfindungsgemäßen Elastaseinhibi
toren erfolgreich behandelt werden kann, ist die disseminierte
intravasale Gerinnung (DIC), die beim septischen oder trauma
tischen Schock oder bei Verbrennungen auftreten kann.
In Untersuchungen zur antithrombotischen Wirkung im Modell der
Schwanzblutungszeit bei der Ratte in einem modifizierten Test
nach Diness (V. Diness et al., Thrombos. Haemostas. 55 (1986)
410-414) an Gruppen von 10 Ratten erwiesen sich erfindungsge
mäße Verbindungen bei oraler Gabe als stark wirksam.
Die Hemmung der granulozytären Elastase wurde in einem Enzym
hemmtest mit Elastase aus menschlichen Granulozyten bestimmt.
Hierbei wurde das spezifische Substrat L-Pyroglutaminyl-L-
prolyl-L-valin-p-nitroanilid verwendet (J. A. Kramps et al.,
Scand. J. Lab. Invest. 43 (1983) 427-432). Die IC₅₀-Werte (Kon
zentrationen zur 50% Hemmung) sind in Tabelle 1 für beispiel
hafte erfindungsgemäße Verbindungen dargestellt.

Gegenstand der Erfindung sind daher auch Arzneimittel zur Be
handlung von Mensch und Tier, bestehend aus oder enthaltend
eine oder mehrere Verbindungen der allgemeinen Formel (I), ge
gebenenfalls zusammen mit üblichen Träger- und Hilfsstoffen.
Die erfindungsgemäßen Verbindungen können in einer Vielzahl
pharmazeutischer Zubereitungsformen und Formulierungen appli
ziert werden, wie zum Beispiel Tabletten, Dragees, Kapseln,
Pillen, Granulate, flüssige, oral einzunehmende Zubereitungen,
Suppositorien, Lösungen, Suspensionen und Emulsionen, Pasten,
Salben, Gele, Cremes, Lotions, Pflaster, Injektionslösungen,
Puder, Sprays oder Aerosole, wobei allgemein übliche Träger-
und Hilfsstoffe verwendet werden können, die mit den erfin
dungsgemäßen Verbindungen verträglich sind.
Die erfindungsgemäßen Arzneimittel enthalten neben den Verbin
dungen der allgemeinen Formel (I) bevorzugt nichttoxische,
inerte pharmazeutisch geeignete Träger- und Hilfsstoffe.
Unter nicht-toxischen, inerten pharmazeutisch geeigneten Trä
ger- und Hilfsstoffen sind feste, halbfeste oder flüssige Ver
dünnungsmittel, Füllstoffe und Formulierungshilfsmittel jeder bekannten
Art zu verstehen.
Der oder die Wirkstoffe können gegebenenfalls
auch in mikrover
kapselter Form vorliegen.
Die genannten Formulierungsformen können auch Färbemittel, Kon
servierungsstoffe sowie geruchs- und geschmacksverbessernde Zu
sätze, zum Beispiel Pfefferminzöl und Eucalyptusöl und Süßmit
tel, zum Beispiel Saccharin enthalten.
Die erfindungsgemäßen Verbindungen sind in den oben aufgeführ
ten pharmazeutischen Zubereitungen vorzugsweise in einer Kon
zentration von etwa 0.1 bis 99.5, besonders bevorzugt von etwa
0.5 bis 95 Masse-% der Gesamtmischung enthalten.
Die oben aufgeführten pharmazeutischen Zubereitungen können
außer den erfindungsgemäßen Wirkstoffen auch weitere pharma
zeutische Wirkstoffe enthalten.
Die Herstellung der oben aufgeführten pharmazeutischen Zuberei
tungen erfolgt in üblicher Weise nach bekannten Methoden, zum
Beispiel durch Mischen der Wirkstoffe mit den Trägerstoffen.
Die vorliegenden Wirkstoffe bzw. pharmazeutischen Zuberei
tungen, die einen oder mehrere Wirkstoffe enthalten, können in
der Human- und Veterinärmedizin zur Verhütung, Besserung und/
oder Heilung thrombo-embolischer Krankheiten oder Zustände be
ziehungsweise von Krankheiten, in denen ein übermäßiger Binde
gewebsabbau von Bedeutung ist, eingesetzt werden.
Im allgemeinen erweist es sich als vorteilhaft, in der Human
medizin den oder die erfindungsgemäßen Wirkstoffe in Gesamt
dosen von etwa 1 bis etwa 2000 mg, vorzugsweise 5 bis 1000 mg,
täglich zu verwenden, wobei insbesondere ein bis vier Dosier
einheiten pro Tag zur Erzielung der gewünschten Ergebnisse
appliziert werden.
Es kann jedoch erforderlich sein, von den genannten Dosierungen
abzuweichen, und zwar in Abhängigkeit von der Art und dem Kör
pergewicht des zu behandelnden Objekts, der Art und der Schwere
der Erkrankung, der Art der Zubereitung und der Applikation des
Arzneimittels sowie dem Zeitraum bzw. Intervall, innerhalb wel
chem die Verabreichung erfolgt. So kann es in einigen Fällen
ausreichend sein, mit weniger als der oben genannten Wirkstoff
menge auszukommen, während in anderen Fällen die oben angeführ
te Menge Wirkstoff überschritten werden muß.
4.38 g (30 mmol) 1(2H)-Phthalazinon und 4.16 ml (30 mmol) Tri
ethylamin werden in 150 ml Dichlormethan gelöst und bei 0°C
eine Lösung von 3.33 ml (35 mmol) Chlorameisensäurephenylester
in 50 ml Dichlormethan zugetropft. Nach 12stg. Rühren bei
Raumtemperatur wird die erhaltene Lösung drei Mal mit jeweils
50 ml Wasser gewaschen, über Natriumsulfat getrocknet und das
Lösungsmittel entfernt. Der Rückstand kristallisiert nach
einiger Zeit vollständig durch. Alternativ wird bis zur voll
ständigen Kristallisation mit Diisopropylether verrührt. Die
anschließende Umkristallisation aus Ethanol ergibt die Titel
verbindung in 76%iger Ausbeute.

Analog der vorstehenden Arbeitsweise werden durch Umsetzung der
jeweiligen Ausgangsverbindungen (Isochinolin-1(2H)-on, bzw. 1(2H)-
Phthalazinon/Derivate mit den entsprechenden Chlorameisensäure
estern folgende Titelverbindungen erhalten. Die Kristallisation
bzw. Reinigung erfolgt durch Verrühren in einem geeigneten Lö
sungsmittel wie Petrolether, Diisopropylether, Diethylether,
durch Umkristallisation in Lösungsmitteln wie Cyclohexan, Toluol,
Xylol, Diisopropylether, tert-Butyl-methylether, Ethanol, Iso
propanol oder durch Säulenchromatographie:
Zu einer Lösung von 2.11 ml (17.5 mmol) Chlorameisensäuretri
chlormethylester ("Diphosgen") in 150 ml Dichlormethan wird bei
0°C eine Lösung von 4.04 ml (35 mmol) 3-Hydroxy-N-methylpipe
ridin und 4.85 ml (35 mmol) Triethylamin in 50 ml Dichlormethan
getropft und anschließend 3 Stunden bei Raumtemperatur gerührt.
Zur entstandenen farblosen Suspension werden unter Eiskühlung
4.35 g (30 mmol) festes Isochinolin-1(2H)-on ("Isocarbostyril")
gegeben, eine Lösung von 4.15 ml (30 mmol) Triethylamin in 50
ml Dichlormethan zugetropft und über Nacht bei Raumtemperatur
gerührt. Die nunmehr klare Lösung wird einmal mit 50 ml halb
gesättigter Natriumhydrogencarbonatlösung und dreimal mit
jeweils 50 ml Wasser gewaschen, über Natriumsulfat getrocknet
und zur Trockne gebracht. Der Rückstand wird mehrmals mit Pe
trolether verrührt, die vereinigten Filtrate werden vollständig
vom Lösungsmittel befreit und anschließend der Kristallisation
überlassen. Die Titelverbindung wird in 90%iger Ausbeute er
halten.

Wie in Beispiel 6 beschrieben, wird die folgende Titelverbin
dung durch Umsetzung von 1(2H)-Phthalazinon) mit dem entsprechen
den Alkohol und mit Chlorameisensäuretrichlormethylester ("Di
phosgen") erhalten. Die Kristallisation bzw. Reinigung erfolgt
durch Verrühren in einem geeigneten Lösungsmittel wie Petrol
ether, Diisopropylether, Diethylether, durch Umkristallisation
in Lösungsmitteln wie Cyclohexan, Toluol, Xylol, Diisopropyl
ether, tert-Butyl-methyl-ether, Ethanol, Isopropanol oder durch
Säulenchromatographie:
In eine Lösung von 5.72 g (20 mmol) Isochinolin-1(2H)-on-N-
carbonsäure-N′-methylpiperidin-3-yl-ester in 200 ml Diiso
propylether wird bei 0°C Chlorwasserstoff bis zur Sättigung
eingeleitet, die erhaltene Suspension noch 2 h bei Raumtempe
ratur gerührt, anschließend wird unter Stickstoff abgesaugt,
der Niederschlag mit Diisopropylether und mit Petrolether ge
waschen und im Vakuum über Phosphorpentoxid getrocknet. Die
sehr hygroskopische Titelverbindung wird in quantitativer Aus
beute erhalten.

1000 Tabletten stellt man aus den nachstehenden Verbindungen
auf die unten beschriebene Weise her. Eine Tablette enthält
dann als Wirkstoff 100 mg 1(2H)-Phthalazinon-N-carbonsäure
phenylester.

Die folgende Rezeptur ergibt eine 5% 1(2H)-Phthalazinon-N-
carbonsäurephenylester-Creme (Stoffangaben in Gew.-%):

Claims (3)
1. Verbindungen der allgemeinen Formel (I)
worin
- a) X und Y die Gruppe CH bedeuten, wobei
R¹ für C₁- bis C₆-Alkyl, Phenyl, 2-Methoxyphenyl, 4-Methoxy phenyl, 2-Methoxycarbonylphenyl, 4-Methoxycarbonylphenyl oder N- Methylpiperidin-3-yl steht,
oder worin - b) X ein Stickstoffatom und Y den Rest CR² bedeutet, wobei
R¹ Phenyl, 2-Methoxyphenyl, 4-Methoxyphenyl, 2-Methoxycar bonylphenyl, 4-Methoxycarbonylphenyl oder N-Methylpiperidin- 3-yl bedeutet,
R² für Wasserstoff, Methyl oder Phenyl steht,
und deren Salze mit physiologisch verträglichen Säuren und Basen.
2. Verfahren zur Herstellung von Verbindungen der allgemeinen
Formel (I), dadurch gekennzeichnet, daß man eine Verbindung
der allgemeinen Formel (II),
worin X und Y die Bedeutungen gemäß Anspruch 1 haben, in An
wesenheit einer Base mit den entsprechenden Halogenameisen
säureestern der allgemeinen Formel (III),
worin R¹ die Bedeutung gemäß Anspruch 1 hat, und Hal die Be
deutung Fluor- oder Chloratome hat, umsetzt.
3. Arzneimittel für die Verwendung bei Mensch und Tier, be
stehend aus oder enthaltend eine oder mehrere Verbindungen
gemäß Anspruch 1, sowie übliche Hilfs- und Zusatzstoffe.
