Description
Die Erfindung betrifft eine Mutante, die gegen einen Zellmembransynthese-Inhibitor resistent ist, und insbesondere eine gegen den Zellmembransynthese-Inhibitor resistente Mutante eines zur Gattung Bacillus gehörenden, extrazellulares Enzym erzeugenden Mikroorganismus, wie beispielsweise Cellulase-, Protease- oder Amylase-erzeugenden Mikroorganismus der Gattung Bacillus. Man kennt verschiedene Verfahren zur Erzeugung von alkalischer Cellulase. Nr. 1139 [Fukumori F., Kudo T., und Horikoshi K.; J. Gen. Dabei wurden von den Erfindern bereits früher verschiedene neue Mikroorganismen mit überlegener Fähigkeit zur Erzeugung von alkalischer Cellulase entdeckt, einschließlich Bacillus sp. KSM-635 (Ferm 8872). Die Entwicklung der neuen, oben erwähnten Mikroorganismen hat es möglich gemacht, extrazellulare Enzyme für die praktische Anwendung zu erzeugen. Bei diesem Sachverhalt haben die Erfinder umfangreiche Untersuchungen zur Verbesserung der Produktivität von extrazellularen Enzymen, insbesondere mittels Mutation, unternommen und haben dabei festgestellt, daß eine bemerkenswerte Produktivitätsverbesserung erhalten wird, indem man extrazellulares Enzym erzeugende Mikroorganismen, die zur Gattung Bacillus gehören, mit Resistenz gegenüber Zellmembransynthese-Inhibitor- Antibiotika ausstattet. Der Ausdruck "Inhibitor-resistent", wie in der vorliegenden Erfindung verwendet wird, bedeutet einen Zustand, bei dem ein Mikroorganismus die Fähigkeit hat, unter Bedingungen zu wachsen, bei denen ein Zellmembransynthese-Inhibitor mit einer Konzentration von mindestens der minimalen Hemmkonzentration (MIC) des Elternstammes anwesend ist. Um die Inhibitor-resistenten Stämme unter den Mutanten zu selektieren, wird daher zunächst der MIC des Elternstamms gemessen, eine Mutante wird in einem Medium kultiviert, das den Zellmembransynthese-Inhibitor mit einer Konzentration oberhalb der MIC enthält, und Kolonien, welche Zellen bilden, werden gesammelt.
