Abstract
Description
Die Erfindung betrifft ein Verfahren zur Herstellung von Carbacyclinen. The invention relates to a process for the preparation of Carbacyclins.
The presently customary synthesis of carbacyclins is based on the available by multistep syntheses optically active 7 α-benzoyloxy-6 β-hydroxymethyl-2-oxabicyclo [3,3,0] octan-3-one (14).
Schema 1 zeigt den bisherigen Syntheseweg (EP 0 41 661):In a leading over several stages synthesis, the 2-oxa group is replaced by a methyl group and so optically active 3-benzoyloxy-α 2 β -hydroxy-7,7-ethylenedioxy-bicyclo [3,3,0] octane prepared (15) in the form of less stable β 2 -Carbaldehyds (16) is an essential starting material for carbacyclins. Scheme 1 shows the previous synthetic route (EP 0 41 661):
The described synthesis of carbacyclins or known Variants of this synthesis have the disadvantage that many Intermediates of synthesis at room temperature in the form of oils. These can only be done by consuming Clean chromatography. Furthermore, the treatment of viscous oils is difficult and their stability is usually worse than that similar crystalline compounds.
The object of the invention is thus the creation of a new Process for the preparation of carbacyclins using new intermediate compounds or the modification known Intermediates to crystalline products for to obtain the synthesis of carbacyclins. The new process should allow the intermediates in crystalline form in higher yields and with fewer by-products are obtained.

In place of microbiological saponification in stage d) can also occur a chemical reaction with racemate separation.
The main advantages of the method according to the invention consist in that almost all intermediates, at least but from bicyclooctanedione-carboxylic acid methyl ester mononeopentyl ketal (4) starting in as pure a form as possible and in crystalline state can be used. This is with some Intermediates, which usually occur as oils, right consuming. However, this approach has the advantage that also in the following stage the formation of crystals becomes more likely and also produces fewer by-products. In addition, crystalline products are easier to handle.
