Abstract
1 Es werden Cephemverbindungen der allgemeinen Formel (I) beschrieben wobei R[hoch]1 eine Amino-subst.-thiazolylgruppe, die gegebenenfalls ein Halogenatom trägt, bedeutet; R[hoch]2 eine Niederalkylgruppe bedeutet; R[hoch]3 für eine Niederalkylgruppe steht; und R[hoch]4 eine veresterte Carboxylgruppe darstellt. Ferner werden pharmazeutisch akzeptable Salze der obigen Verbindungen beschrieben. Die Erfindung betrifft 7-[2-(2-subst.-Amino-thiazol-4-yl-2-alkoxyiminoacetamido]-3-alkylthio-3-cephem-4-carbonsäureester-Derivate sowie deren pharmazeutisch akzeptablen Salze.The invention relates to 7- [2- (2-substituted-amino-thiazol-4-yl-2-alkoxyiminoacetamido] -3-alkylthio-3-cephem-4-carboxylic acid ester derivatives and their pharmaceutically acceptable salts.

Bezüglich der vorliegenden Erfindung seien als Stand der Technik solche Verfahren erwähnt, bei denen Cephemverbindungen hergestellt werden, welche eine Methylthiogruppe, eine aromatische Thiogruppe oder eine heterocyclische Thiogruppe in der 3-Position aufweisen, wie sie beispielsweise in der JA-OS 51-48690; Helvetica Chimica Acta, Band 58, S. 2437 und S. 2450 (1975); der JA-OS 51-82292 und den JA-ASen 53-119888, 57-145883 und 53-103493 beschrieben sind. Unter diesen finden sich Verbindungen, welche, ähnlich wie die erfindungsgemäßen Verbindungen, Seitenketten in der 3- und 7-Position des Cephalosporins aufweisen. Demgemäß stellen Cephemverbindungen, welche beliebige Kombinationen von Alkylthiogruppen mit 2 oder mehr Kohlenstoffatomen in der 3-Position und Alkyloxyiminogruppen an der Seitenkette der 7-Position aufweisen, mit Ausnahme des Falls, daß beide Alkyleinheiten jeweils eine Methylgruppe darstellen, neue Verbindungen dar. Zur Überwindung der Nachteile des Standes der Technik wurden von den Erfindern umfangreiche Untersuchungen durchgeführt. Dabei wurde eine große Anzahl von Verbindungen hergestellt, welche an der Seitenkette der 7-Position 2-Amino-thiazol-4-yl-2-alkyloxyimino-2-acetamide aufweisen, und zwar unter Änderung der Anzahl der Kohlenstoffatome in der Alkylgruppe der Alkyloxyiminogruppe der 3-Cephemverbindungen sowie des Typs des Substituenten an der 3-Position. Ferner wurden verschiedene 4-Carbonsäureesterderivate hergestellt. Die erhaltenen Verbindungen wurden in einem Mikroorganismen-Test bewertet. Dabei wurde festgestellt, daß Verbindungen mit einer Kombination einer Alkylthiogruppe in der 3-Position und einer Alkylkette der Alkyloxyiminogruppe an der Seitenkette der 7-Position eine ausgezeichnete Wirksamkeit aufweisen.In order to overcome the drawbacks of the prior art, the inventors conducted extensive studies.
