Description
Pseudotrisaccharide, Verfahren zu deren Herstellung,Pseudotrisaccharides, processes for their production,
deren Verwendung als Arzneimittel und die Pseudotrisaccharide enthaltende Arzneimittel sowie deren Herstellung Die vorliegende Erfindung betrifft neue Aminoglykosid-Antibiotika vom Typ des Sisomicins, Verfahren zu ihrer Herstellung und ihre Verwendung, insbesondere als antibakteriell wirkende Arzneimittel.their use as medicaments and those containing pseudotrisaccharides Medicines and their production The present invention relates to new aminoglycoside antibiotics of the sisomicin type, process for their preparation and their use, in particular as antibacterial drugs.
Aminoglycoside antibiotics are important substances for effective Fight bacterial infections.
However, the occurrence of resistant germs reduces theirs in many cases broad applicability; furthermore, side effects such as ototoxicity and nephrotoxicity can occur appear. In some cases, derivatisation can overcome these disadvantages remove.
For example, very effective antibiotics are obtained if, in accordance with the applicant's earlier proposals, sisomicin one or more of the amino groups, but preferably the 1-position amino group, derivatized by reductive alkylation with aldehydes or by acylation. Compounds of comparable effectiveness derivatized in accordance with the OH groups in the 5-position, however, have not yet become known.
Surprisingly, however, it has now been found that pseudotrisaccharides, especially those with the basic structure of sisomicin, which have a substituted one at the 5-position Have urethane group, are new highly effective antibiotics.
The invention thus relates to compounds of the general formula (I): in which R1 is hydrogen or a methyl group, R2 is hydrogen or one of the groups R5, CO-R5, CO-NH-R5 or CO-O-R5 and R4 is CO-NH-R5 or CO-N (R5) 2, where R5 is an alkyl group with 1 to 7 carbon atoms, optionally substituted by one or more of the groups -OH, -NHR 'or -N (R3) 2, R' is an aryl group with 5 to 10 carbon atoms and the R3s independently of one another represent hydrogen or an alkyl group with 1 to 6 carbon atoms, as well as their pharmacologically acceptable salts.
Those compounds in which the radical R5 in the group R4 has a terminal amino or hydroxyl group are preferred. In this connection, radicals R5 of the formulas - (CH2) 2-OH-CH2-CHOH-CH2-NH2 and (CH2) 2 NH2-The compounds according to the invention can be prepared using the protective group chemistry known per se from earlier publications by the applicant . The preparation of the compounds according to the invention is based on sisomicin derivatives of the general formula (II): in which R1, R2 have the meaning given above, Y is hydrogen when R2 is a group CO-R5, CO-NHR5 or COOR5 and R6 is when R2 is hydrogen or a group R5.
R6 für eine Gruppe -CO-Q oder vorzugsweise -SR7 steht, wobei Q für eine der Gruppen CC13, CF3 oder O-C(CH3)3 steht, und R7 eine gegebenenfalls substituierte Phenyl-, Di-oder Triphenylmethylgruppe darstellt.R6 stands for a group -CO-Q or preferably -SR7, where Q stands for one of the groups CC13, CF3 or O-C (CH3) 3, and R7 is an optionally substituted one Represents phenyl, di or triphenylmethyl group.
Optionally substituted phenyl R7 includes in particular the Phenyl group itself or through one to three substituents from the series nitro, C1-C4 -Alkyl, C1-C4-alkoxy, C1-C4-alkoxycaronyl or phenyl or by 1 to 5 halogen atoms to understand substituted phenyl groups.
Die durch Acyl-bzw. Sulfenylgruppen geschützten Sisomicin-Derivate sind als Zwischenprodukte bei der Herstellung von Antibiotika an sich bekannt und werden beispielsweise in den Deutschen Offenlegungsschriften 2 994 659 (Eruopäische Patentanmeldung Nr. 80 103 193.1), 2 753 769, 2 921 973 (Europäische Patentanmeldung Nr.The by acyl or. Sulfenyl-protected sisomicin derivatives are known as intermediate products in the manufacture of antibiotics and are for example in the German Offenlegungsschriften 2 994 659 (European Patent application No. 80 103 193.1), 2 753 769, 2 921 973 (European patent application No.
They are obtained, for example, by using sisomicin or 3 "-N-desmethyl-3" -N-ethyl-sisomic in with o-nitrophenylsulfenic acid p-nitrophenyl ester in an inert Solvent, optionally with the addition of water, at temperatures between -30 and + 500C implemented in the presence of a base.
In this case, five moles of the sulfenic acid ester are used for one mole of the starting compound a. Other reagents with which protecting groups can be introduced are Tritylsulfenylchlorid, o-Nitrophenylsulfenylchlorid, 2,4-Dinitrophenylsulfenylchlorid, 2,4,5-trichlorophenylsulfenyl chloride, pentachlorophenylsulfenyl chloride, 2,4-dinitrophenylsulfenic acid pnitrophenyl ester, 2,4,5-trichlorophenylsulfenic acid pnitrophenyl ester, pentachlorophenylsulfenic acid pnitrophenyl ester, Di-t-butyl pyrocarbonate and trifluoroacetic anhydride or trichloroacetic anhydride.
Instead of pure sisomicin or pure 3 "-N-desmethyl-3" -N-ethyl-sisomicin It has been found to be much more economical to go out the interconnections II to produce directly from the fermentation broths of the aminoglycosides. One such The method is described in DOS 30 00 841.
For the selective introduction of the 5-O-substituents according to the invention a protective group must first be introduced on the 2 "OH group.
Almost all O-protective groups familiar to the person skilled in the art are used for this purpose such as acyl protecting groups, specifically Acetyl, tetrahydropyranyl or trimethylsilyl in question.
With regard to the following steps, a process described in DOS 2 840 907, in which the compound (II) is reacted with 1 mol of a silylating or acylating agent of the general formula (III) R10G (III), in which G. means a leaving group and R10 for one of the groups where R8 is hydrogen or an optionally substituted phenyl group and n1, n2, n3 and n4 independently of one another are integers from 0 to 5, Rg is R8, a C1-C5 alkoxy group or a C3 -C5 alkenyloxy group, n5, n6, n7, n8, n9, n10 and n11 represent numbers from 0 to 3 independently of one another and n12 means 0 or 1.
Suitable leaving groups G are for example in the silylation Halogen, especially chlorine, and perfluorobutylsulfonyloxy.
Die Reaktionstemperatur beträgt 0 - 800C, vorzugsweise 20 - 400C.The silylation preferably used according to the invention is in one inert solvents such as dichloromethane or chloroform in the presence of a catalyst, which at the same time represents the auxiliary base required to bind the acid released, carried out. For this, all organic bases that activate the Si-G bond are suitable. Preference is given to using equivalent amounts of imidazole as the catalyst base worked. Silylating agent and catalyst base can each be stoichiometric Amounts are reacted with the compound of formula (II), however, are considerable shorter reaction times achieved with a 2-5-fold excess of silylating agent. The reaction temperature is 0-800C, preferably 20-400C.
The intermediate compounds thus selectively protected at the 2 "-OH group are in the next step first with 1 mol of an acylating agent of the general Formula (IV) A - CO - X (IV) acylated in a manner known per se, where A stands for a leaving group known per se (see e.g.
Houben-Weyl XV, Bd. II, S. 1-364) und X für Halogen stehen. Houben-Weyl XV, Vol. II, pp. 1-364) and X is halogen.
The reaction with activated chloroformic acid esters is preferred such as p-nitrophenyl chloroformate in inert or weakly basic form Solvents (dichloromethane or pyridine) with a 1-10 fold, preferably 3-5 times the reagent excess in the temperature range of 0 - 80 ° C, preferably 10 - 300C carried out. When using chloroformic acid esters as acylating agents are these in stoichiometric amounts or in a 2-3-fold excess in one inert solvent such as dichloromethane in the presence of equivalent amounts of a organic auxiliary base, for example triethylamine, at 0-400C, preferably at 10-300C implemented.
The reaction times are significantly reduced by adding 10-20 mol% of a catalyst such as p-dimethylaminopyridine.
In a second step, the acyl derivative obtained in this way is in itself known way with a compound of the formulas (V) HN (R5) 2 or H2NRs (V) implemented, where amino groups contained in the radicals R5 are analogous to the compounds of the formula (II) must be blocked by protecting groups R6.
Then the protective groups R6 and the protective groups R10 split off in a manner known per se.
The sulfenyl protective groups can be split off with weak acids or with strong nucleophiles such as thiophenols, the cleavage of the remaining N-protective groups can with aqueous alkali or alkaline earth metal hydroxide or with Acids such as trifluoroacetic acid, perchloric acid or boron trifluoride etherate are carried out.
The cleavage of the protective group R10 cannot be done either with water miscible organic solvents such as dichloromethane with tetralkylammonium fluorides, e.g. tetrabutylammonium fluoride or either in dimethyl sulfoxide-water mixtures with inorganic fluorides such as potassium fluoride or with inorganic bases such as caustic soda take place, preferably at room temperature.
Bei R6 = Sulfenyl z.B.All protective groups are preferably split off in one reaction step. When R6 = sulfenyl e.g.
Methanol.preferably by 2-mercaptobenzothiazole at pH = 1 and then Leave to stand in aqueous / alcoholic solution at pH = 11-13. With R6 = COCHal3 through reaction with alkali at 400C for 1 hour or at 250C for 12 hours in alcohols such as methanol.
