Description
Die Erfindung betrifft neue Ergolinderivate der Formel I,
worin
R₁ Alkyl mit 1 bis 4 Kohlenstoffatomen bedeutet, und
R₂ und R₃ unabhängig voneinander je Wasserstoff oder Alkyl mit 1 bis 4 Kohlenstoffatomen bedeuten,
R₂ und R₃ unabhängig voneinander je Wasserstoff oder Alkyl mit 1 bis 4 Kohlenstoffatomen bedeuten,
in freier Form oder in Form ihrer Additionssalze mit Säuren.
Wenn nicht anders angegeben, enthält eine Alkylgruppe vorzugsweise
2, insbesondere 1, Kohlenstoffatom.
Stehen R₂ und/oder R₃ für Alkyl, so enthält dieses vorzugsweise
1 bis 3 Kohlenstoffatome, und insbesondere 2 oder 1 Kohlenstoffatom.
R₂ und R₃ sind vorzugsweise identisch.
Erfindungsgemäß gelangt man zu den Verbindungen der Formel I,
indem man Verbindungen der Formel II,
worin R₁ obige Bedeutung besitzt, mit einem reaktionsfähigen,
funktionellen Derivat einer Säure
in der R₂ und R₃
obige Bedeutung haben, umsetzt.
Das Verfahren kann analog zu bekannten Methoden erfolgen. Als
reaktionsfähiges, funktionelles Derivat obiger Säure kann das
Chlorid oder Bromid verwendet werden. Das Verfahren wird zweckmäßig
in Lösung durchgeführt. Geeignete Lösungsmittel sind z. B.
Methylenchlorid oder Dioxan.
Im allgemeinen arbeitet man mit Vorteil bei einer Reaktionstemperatur,
die zwischen -10°C und etwa Raumtemperatur liegt.
Das Verfahren wird zweckmäßig in Gegenwart einer teritären Base
wie Triäthylamin, oder vorteilhaft in Gegenwart von Pyridin oder
2,6-Lutidine durchgeführt.
Die Verbindungen der Formel I können z. B. in freier Form als
Base, oder in Form ihrer Additionssalze mit Säuren vorliegen. Aus
den freien Basen lassen sich in bekannter Weise Säureadditionssalze
herstellen und umgekehrt. Geeignete Salze umfassen das
Hydrochlorid.
Die Ausgangsprodukte sind bekannt oder nach an sich bekannten
Methoden, beispielsweise wie in den Beispielen beschrieben, herstellbar.
Die Verbindungen der Formel I in freier Form oder in Form von
physiologisch verträglichen Additionssalzen mit Säuren (erfindungsgemäße
Verbindungen) zeichnen sich durch interessante
pharmakodynamische Eigenschaften aus. Sie können als Heilmittel
verwendet werden.
So können die erfindungsgemäßen Verbindungen aufgrund ihrer
dopaminergen Eigenschaften bei der Behandlung von Morbus
Parkinson und depressiven Zuständen Anwendung finden.
Die erfindungsgemäßen Verbindungen besitzen ebenfalls eine
Prolactin-sekretionshemmende Wirkung. Prolactin-sekretionshemmende
Verbindungen können z. B. zur Prophylaxe und Therapie von
physiologischer Lactation und Galactorrhoe Anwendung finden.
Die erfindungsgemäßen Verbindungen der Formel I, worin R₇ und R₈
für Methyl stehen, besitzen besonders ausgeprägte Prolactin-
sekretionshemmende Wirkung.
Diese Wirkung äußert sich durch eine Fertilitätshemmung und kann
durch folgenden Versuch gezeigt werden:
Die Verbindungen werden inseminierten Rattenweibchen am 5. Tage
post coitum subcutan appliziert und die Tiere am 12. Tage
autoptisch auf Trächtigkeit untersucht (Bestimmung nach
Shlesnyak, Acta Endocrin., Suppl. (1960) 51, 677).
In der nachstehenden Tabelle wird die ED₅₀, d. h., die Dosis, die
die Fertilität bei 50% der Tiere hemmt, aufgeführt. Verbindung I
ist die in Beispiel 1 der vorliegenden Anmeldung beschriebene
Verbindung 1,6-Dimethyl-8α-(N,N-dimethylsulfamoylamino)ergolin I
und Verbindung II ist die Vergleichsverbindung
6-Methyl-8α-(3-pyridylamino)ergolin.

Wie aus der Tabelle hervorgeht, besitzt die Verbindung I
gegenüber Verbindung II eine stark ausgeprägte Wirkung auf die
Prolaktinsekretionshemmung.
Die Erfindung betrifft auch Heilmittel, die eine Verbindung der
Formel I in freier Form oder in Form ihrer physiologisch verträglichen
Additionssalze mit Säuren enthalten. Diese Heilmittel,
beispielsweise eine Lösung oder eine Tablette, können nach bekannten
Methoden, unter Verwendung der üblichen Hilfs- und
Trägerstoffe, hergestellt werden.
Die nachfolgenden Beispiele erläutern die Erfindung. Die Temperaturangaben
erfolgen in Celsiusgraden und sind unkorrigiert.
2,55 g (10 mMol) 1,6-Dimethyl-8α-aminoergolin I
werden in 20 ml 2,6-Lutidin gelöst und innert
10 Minuten in ein gerührtes Gemisch von 3,58 g
(25 mMol) N,N-Dimethylsulfaminsäurechlorid,
40 ml Methylenchlorid und 10 ml 2,6-Lutidin eingetropft.
Nach 1 Stunde Rühren bei Raumtemperatur
trägt man noch 1,5 ml N,N-Dimethylsulfaminsäurechlorid
ein und rührt noch 2 Stunden bei Raumtemperatur.
Zur Aufarbeitung versetzt man bei 0°
mit 2N Ammoniak bis zur basischen Reaktion
und extrahiert mit einem Gemisch von 10% Methanol
in Methylenchlorid. Nach Trocknen über Natriumsulfat
hellt man die organische Phase mit
Aktivkohle auf und dampft am Rotationsverdampfer
ein. Der so erhaltene, grünliche Schaum wird
an 150 g Kieselgel chromatographiert, wobei die
Titelverbindung mit 2% Methanol in Methylenchlorid
als gelbliches Harz eluiert wird. Das
Hydrochlorid der Titelverbindung kristallisiert
aus Aethanol, Smp. 226-228°. [α] = -23°
(c = 0,3 in Pyridin).
Zu dem als Ausgangsverbindung benötigten 1,6-Dimethyl-
8α-amino-ergolin I gelangt man durch Hydrierung
von 1-Methyl-Δ 7,8-lysergsäuremethylester
nach Zugabe von Platinoxyd, Umsetzung des
so erhaltenen 1-Methyl-9,10-dihydro-isolysergsäure
I-methylesters (Smp. 150-151°) mit einem
Gemisch von Hydrazinhydrat und Hydrazin-dihydrochlorid
und Umwandlung des so entstandenen
1-Methyl-9,10-dihydro-isolysergsäure I-hydrazids
(Smp. 208-211°) nach Curtius.
Analog Beispiel 1 wird 1,6-Dimethyl-8α-aminoergolin I
mit N,N-Diäthylsulfaminsäurechlorid umgesetzt. Das
Methansulfomat schmilzt bei 199-201°; [α] = -25°
(c = 0,3 in Dimethylsulfoxid).
Claims (3)
1. Neue Ergolinderivate der Formel I,
worin
R₁ Alkyl mit 1 bis 4 Kohlenstoffatomen bedeutet und in der
R₂ und R₃ unabhängig voneinander je Wasserstoff oder Alkyl mit 1 bis 4 Kohlenstoffatomen bedeuten,
und ihre Säureadditionssalze.

R₂ und R₃ unabhängig voneinander je Wasserstoff oder Alkyl mit 1 bis 4 Kohlenstoffatomen bedeuten,
und ihre Säureadditionssalze.
2. Verfahren zur Herstellung von neuen Ergolinderivaten der
Formel I, worin R₁, R₂ und R₃ die im Anspruch 1 angegebene
Bedeutung besitzen, und ihrer Säureadditionssalze, dadurch
gekennzeichnet, daß man nach an sich bekannter Arbeitsweise
die Aminogruppe in Stellung 8 der Verbindungen der Formel II,
worin R₁ obige Bedeutung besitzt, mit einem reaktionsfähigen,
funktionellen Derivat einer Säure
in der R₂ und R₃ obige Bedeutung haben, umsetzt
und die so erhaltenen Ergolinderivate der Formel I in Form der
Basen oder von Säureadditionssalzen gewinnt.
3. Heilmittel, enthaltend ein Ergolinderivat der Formel I, worin
R₁, R₂ und R₃ die im Anspruch 1 angegebene Bedeutung besitzen,
in freier Form oder in Form eines physiologisch verträglichen
Säureadditionssalzes.
