Description
The invention relates to the use of the ether-containing substances obtained in the manufacture of placenta African extracts Residues and a process for the preparation of a collagen-containing placenta extract.
Soluble collagen, which is a biological precursor to mature collagen, has been used in increasingly processed as an active ingredient component in cosmetic preparations. Such preparations are supposed to stimulate the metabolism of the skin and connective tissue cells through the percutaneous supply of biological agents revitalize and tighten withered, wrinkled skin by increasing the skin turgor. Is collagen a fibrous scleroprotein found in the bones and connective tissue of animals. Accordingly Collagen obtained by extraction from the connective tissue of mostly embryonic or young mammals. the Procurement of the raw material is expensive because of the quantity required and is usually also included associated with considerable difficulties, since it is as fresh as possible and free of unusable waste and other impurities must be.
From DE-OS 24 05 002 it is known, soluble native collagen from human tissue, in particular Placemen, to win. For this purpose, the cleaned and shredded human tissue is made gradually fractionated by first using an aqueous salt solution, the neutral salt-soluble tropocollagen and then the acid-soluble procollagen and another structured modification of the native collagen is extracted. However, this known method is because of the large number of process steps and of the existing placental constituents belonging to the most varied of substance classes in proportion laborious. In addition, human placents and placemas of animal origin are the starting material extremely uneconomical for the production of soluble collagens, not least because of their scarcity, especially since in this way the raw material for obtaining the more valuable placenta extract is lost.
The object of the invention is to provide an inexpensive raw material for the production of soluble collagens find that is available in sufficient quantity.
The invention is based on the knowledge that occurs in the manufacture of placenta African extract to use ethereal residues for the extraction of the soluble collagen.
Das Ausgangsmaterial zur Gewinnung von Placentafrischextrakten ist ausnahmslos tierischen Ursprungs, da entsprechendes Humangewebe in den erforderlichen Mengen in der Regel nicht zur Verfugung steht Zu Beginn des Fabrikationsprozesses werden die zuvorThe starting material for obtaining placenta African extracts is without exception of animal origin, as the corresponding human tissue is required Quantities usually not available At the beginning of the manufacturing process, the previously

After the aqueous components have been separated off, a residue consisting predominantly of solids remains, which contains a lot of ether and has hitherto been used in the coverings for destruction or further processing, for example. B. for fertilization purposes (see. Also DE-AS 11 42 373) was fed.
These organ residues, which were previously discarded as worthless, are now used according to the invention to obtain the soluble native collagen, whereby the long-term action of the ether has no influence on the nativity of the soluble collagens and their yield because of the extensive denaturation of foreign proteins and the complete removal of lipoids and fats could be improved. These organ residues are extremely inexpensive because they are produced in large quantities and their previous destruction has even resulted in considerable costs.
The invention also consists in a method for the production of a collagen-containing placenta extract, which is characterized in that the at from the production of placenta African extracts at 0-3 ° C, initially by washing with a buffered weak NaCl solution freed from some of the foreign proteins present and extracted by adding a sufficient amount of a neutral salt solution within 2-6 days that then the neutral salt-soluble collagen after separation of the remaining placenta residues is precipitated by salting out with a sufficient amount of common salt and that the precipitated soluble Collagen in a buffer solution containing a preservative made from placenta extract with a pH value below 7 is dissolved.
The collagen-containing placenta extract produced in this way is particularly durable and is preferred Used in the manufacture of skin care products as the neutral salt soluble precursor of native collagen because of its low inter- and intramolecular cross-linking, it is particularly beneficial for the biological Activation process is.
The invention is explained in more detail below on the basis of exemplary embodiments.
Nachdem das Hydrolysat nochmals 9 Stunden mit 100 ml 6n HCl unterBefore carrying out the preparative experiments to obtain soluble collagen from placental residues from the extract production, it was first necessary to prove that biologically active collagen was actually present in the starting material, and to determine the total collagen content (soluble and insoluble collagen). For this purpose, samples of an ether-containing placenta residue from manufacture were freeze-dried at the stage of obtaining the crude centrifugate, weighed precisely (Table 1) and each sample was heated to 102 ° C. for 24 hours with 4 ml of 6N HCl in a sealed test tube in order to close the sample hydrolyze. Gewinnung löslichen Kollagens aus Placentarückständen der Fabrikation von PlacentaextraktenRecovery of soluble collagen from placenta residues from the manufacture of placenta extracts
Nach Zentrifugieren (30 Minuten, 4000 rpm) wird der Rückstand nach Zusatz einer kleineren Menge Toluol320 kg placental residues, which are produced in the production of placenta African extract after separation of the crude extract incurred during centrifugation, are - if necessary after freeze-drying - crushed again and with 300 kg of ice water washed at 0 ° to + 3 ° C. After centrifugation (30 minutes, 4000 rpm) the Residue after adding a small amount of toluene
A.to prevent possible bacterial activity for 24 hours with 500 kg of a 0.01 molar sodium chloride solution of pH 7.4 (phosphate buffer: 152 g Na 2 HPO 4 and 18.5 g NaH 2 PO 4 in 701 water) at 0 Let stand to 3 ° C. The nz.ch hour centrifugation at 4000 rpm resulting residue and then centrifuged to collect the neutral salt soluble collagen for 4 days at 0 ° to + 3 molar 0 C with 0.15 sodium chloride solution at pH 7.1 is extracted (addition of toluene): A centrifugate .
For this centrifugate A, the neutral salt soluble collagen is precipitated by addition of solid sodium chloride to a concentration of 12% at +2 0 C and collected by centrifugation. The precipitated, neutral salt-soluble collagen can be purified by dialysis and, if necessary, repeated reprecipitation. Before a stable, preserved final solution of the neutral salt-soluble collagen is prepared, the collagen content is determined by a hydroxyproline determination as described below. The isolated, purified, partially salt-soluble collagen is dissolved in such an amount of 0.1 molar preserved citrate buffer solution that the collagen content is 4 g per liter. The collagen solution obtained in this way is centrifuged again before sterile filling.
I.The residue remaining in the recovery of centrifugate A becomes acid-soluble for isolation Collagen after addition of toluene with 100 kg citrate buffer of pH 3.7 (the buffer solution is obtained by dissolving 21 kg of citric acid and 4.4 kg of NaOH in 1000 kg Ice water prepared and fine-tuned to pH 3.7) extracted with stirring in the cold at + 2 ° C and centrifuged again after 2 to 5 days: centrifugate B I.
The residue of this centrifugation is again 2 0 C to + 4 ° C with 1000 liters of citrate buffer of the above composition taken off at + and then centrifuged also after several days: centrifugate B2.
The centrifugates B 1 and B 2 are each brought in itself for separating the collagen fraction by addition of sodium chloride to a concentration of 7% NaCl, while still at a temperature of + 2 is maintained ° to +3 0C. The resulting precipitate of each precipitation is collected by centrifugation or filtration in the cold and freed from salts by washing with ice water. The collagen fraction can be purified further by dialysis and / or falling over from a neutral or acidic medium. After purification, the collagen fraction is brought into a 0.1 molar sodium citrate buffer solution, with stirring, containing 0.2 to 0.5% preservatives, e.g. B. Phenonip, 1 to 5% Tween 80 and 0.3 to 2% propylene glycols. This solution is centrifuged or filtered again, if necessary, and stored at 2 ° C.
Before a long-life, preserved, acid-soluble collagen preparation is manufactured, one Sample of the collagen from centrifugate B 1 and B 2 to determine the collagen content, a hydroxyproline determination carried out:
A sample freeze-dried after centrifuging again had a weight of 2.13 g in the present case. 7.1 mg of this are hydrolyzed for 24 hours with 2 ml of 6N HCl at 102 ° C in a sealed test tube. Gehah of 11.2 γ corresponds to the test specimen thus contained 0.248 g collagen.

Placentaextrakt als PufferlösungPlacenta extract as a buffer solution
For example, a placenta extract containing 0.3 to 0.4% collagen with a pH of 4.0 can be obtained by stirring an amount of collagen characterized by hydroxyproline determination at +1 ° C. into a standardized placenta extract which has the following parameters :
