Description
Die Erfindung betrifft ein Verfahren zur Herstellung von hydrophilen makroporösen poly
meren Gelen, die in ihrer Struktur gleichzeitig kationische und anionische
funktionelle Gruppen oder dipolare Ionen enthalten.
Die Ionenaustauscher aus hydrophilen polymeren Gelen mit regulierbarer
Porosität, welche nach den CS-Patentanmeldungen PV-703-74, PV-704-74 und
PV-896-74 hergestellt werden, enthalten in ihrer Struktur entweder anionische
(Kationenaustauscher) oder kationische funktionelle Gruppen (Anionenaustau
scher). Es wurde nachgewiesen, daß diese Materialien durch ihre physikalischen
und chemischen Eigenschaften die bisher gebräuchlichen Kationen- und Anionen
austauscher mit Cellulose- oder Polydextranmatrix bei der Sorption und der
Chromatographie von verschiedenen Biopolymeren übertreffen.
Neben den gebräuchlichen Kationen- und Anionenaustauschern wurden aber schon
früher synthetische organische Polystyrol-Divinylbenzol-Ionenaustauscher, die
in ihrer Struktur gleichzeitig basische und saure funktionelle Gruppen ent
halten, hergestellt. Auf den Anionenaustauscher aus Polystyrol wurde durch
Ionenaustausch eine saure Komponente (z. B. Acrylsäure) sorbiert, welche nach
dem Durchdringen in das Körnchen des Anionenaustauschers in sein Netz ein
polymerisiert wurde. Auf ähnliche Weise kann man basische Komponenten an
Kationenaustauscher binden.
Die jetzige Entwicklung der Chromatographie von Diopolymeren weist auf die
große Bedeutung der amphoteren Ionenaustauscher hin, wenn ihre funktionellen
Gruppen an die die Penetration von Makromolekülen erlaubende Matrix, wie
z. B. Polydextran, gebunden werden (J. Porath, L. Fryklund: Nature 226 (1970)
1169; J. Porath, N. Formstedt: J. Chromatogr. 51 (1970) 479). In Hinsicht auf
die vorteilhaften chemischen und mechanischen Eigenschaften der hydrophilen
Hydroxyacrylat- und Hydroxymethacrylatgele, welche nach dem CS-Patent
1 48 828 und 1 50 819 hergestellt werden und die in mancher Hinsicht die Deri
vate vom Polysacchariden übertreffen, ist dieses synthetische polymere Gel
als Träger auch für amphotere oder dipolare ionogene Gruppen sehr geeignet.
Aus der DE-OS 20 22 781 sind ferner bereits schwach vernetzte, quellbare Hydro
gele bekannt, die auf der Basis von (Meth)acrylaten mit Dihydroxiverbindungen
zusammen mit weiteren Monomeren, die saure und basische Gruppen tragen, her
gestellt werden.
Das erfindungsgemäße Verfahren zur Herstellung von amphoteren hydrophilen
makroporösen Copolymeren wird durch die Ansprüche gekennzeichnet.
Die
Vernetzungsmittel können auch eine größere Anzahl von mischpolymerisierba
ren Acryloyl- oder Methycryloylgruppen enthalten.
Die radikalische Mischpolymerisation wird in Gegenwart von inerten organi
schen Lösungsmitteln, die aus der Gruppe von organischen Säuren, Alkoho
len, Aminen oder Nitrilen ausgewählt werden, in wäßriger Dispersionsphase
durchgeführt.
Amphotere hydrophile Ionenaustauschergele haben eine große Bedeutung für die
Entwicklung feiner Fraktionierungsmethoden. Sie werden zum Trennen der Gemische
von Eiweißstoffen und Nucleinsäuren schon bei der linearen Chromatographie
verwendet, ohne die Anwendungsmöglichkeit von Elutionsgradienten auszuschei
den. Durch die erreichten Resultate übertrifft die Separation in amphoteren
Ionenaustauschern das Trennen auf üblichen monofunktionellen Ionenaustauscher
derivaten (J. Porath: Chromatographic Methods in Fractionation of Encymes,
in Biotechnology and Bioengineering Symp. No. 3, (1972) 145-166).
Zu diesen Zwecken bisher verwendete Derivate der Polydextrane besitzen im
Vergleich zu hydrophilen Derivaten der Hydroxyacrylat- und Hydroxymethacrylat
gele eine wesentlich geringere chemische und mechanische Stabilität. Überdies
quellen die erfindungsgemäßen Ionenaustauscher nur sehr wenig auf; bei
höherem Durchfluß kommt es nicht zum Verstoßen der Säule, die mit diesen
Materialien gefüllt ist. Gele sind gegen mikrobielle Infektion beständig und
man kann sie im Bedarfsfalle durch Kochen sterilisieren.
Die angeführten Beispiele illustrieren das Herstellungsverfahren von amphoteren
Copolymeren nach dieser Erfindung.
34,8 Mol.-% 2-Hydroxyäthylmethyacrylat, 31,8 Mol.-% Aethylendimethacrylat,
16,7 Mol.-% Methacrylsäure und 16,7 Mol.-% N,N-Diäthylaminoäthylmethacrylat
wurden in wäßriger Dispersion in Gegenwart von Laurylalkohol und Cyclohexanol
durch Azoisobuttersäuredinitril ausgelöst, bei 70°C innerhalb von 13 Stunden
mischpolymerisiert. Nach dem Reaktionsende wurde das Polymerisationsgemisch
abfiltriert und mit Aethanol, Benzol, Aethanol und Wasser gewaschen. Durch
potentiometrische Titration wurden die Austauschkapazitäten der sauren und
basischen Funktionsgruppe, welche übereinstimmend waren, bestimmt; sie be
trugen 0,6 mäq/g.
35 Mol.-% Diäthylaminoäthylmethacrylat, 35 Mol.-% Sulfoäthylacrylat und
30 Mol.-% Aethylendimethacrylat wurden in wäßriger Dispersion in Gegenwart
von Laurylalkohol und Cyclohexanol durch Azoisobuttersäuredinitril ausgelöst,
bei 70°C innerhalb von 13 Stunden mischpolymerisiert. Nach dem Reaktions
ende wurde das Mischpolymerisationsgemisch abfiltriert und mit Aethanol,
Benzol, Aethanol und Wasser gewaschen.
25 Mol.-% Acrylsäure, 25 Mol.-% Dimethylaminoäthylmethacrylamid und 50 Mol.-%
Aethylendimethacrylat wurden in wäßriger Dispersion in Gegenwart von Lauryl
alkohol und Cyclohexanol durch Azoisobuttersäuredinitril ausgelöst, bei 70°C
innerhalb von 13 Stunden mischpolymerisiert. Nach dem Reaktionsende wurde das
Mischpolymerisationsgemisch abfiltriert und mit Aethanol, Benzol, Aethanol
und Wasser gewaschen.
Claims (2)
1. Verfahren zur Herstellung von hydrophilen makroporösen
Copolymeren von amphoterem Charakter, dadurch gekenn
zeichnet, daß man
- a) anionogene Monomeren aus der Gruppe der Sulfoalkyl(meth) acrylate, Sulfoalkyl(meth)acrylamide, Acryl- und Methacryl säure mit
- b) kationogenen Monomeren aus der Gruppe der Aminoalkyl(meth) acrylate, Aminoalkyl(meth)acrylamide oder deren quater nisierte Ammoniumverbindungen mit
- c) vernetzenden Monomeren aus der Gruppe der Dialkyl(meth) acrylate, Oligoglykol- und Polyglykoldi(meth)acrylate, Bis(meth)acrylamide und Divinylbenzol und
- d) gegebenenfalls mit Monomeren aus der Gruppe der Hydroxy (meth)acrylate in Gegenwart von inerten organischen Ver bindungen aus der Gruppe von Alkoholen, organischen Säuren, Aminen und Nitrilen in wäßriger Dispersions phase mischpolymerisiert.
2. Verwendung der nach Anspruch 1 erhältlichen Copolymeren
für die Isolierung und Trennung von biologischen Materia
lien.
