Inhibierung der Hydrolyse von Acetylsalicylsäure und d-Propoxyphenhydrochlorid
Die Erfindung betrifft Verfahren zur Stabilisierung von pharmazeutischen Mitteln, die d-Propoxyphenhydrochlorid
und Acetylsalicylsäure enthalten.
überraschenderweise wurde gefunden, daß die Hydrolyse
von Acetylsalicylsäure, die im folgenden als Aspirin bezeichnet wird, unter Bildung von freier Essigsäure
und Salicylsäure in pharmazeutischen Mitteln, die d-Propoxyphenhydrochlorid enthalten, durch Zumischen
eines Hydrochlorids einer Verbindung der allgemeinen Formel

Die Erfindung betrifft ferner ein gegen Hydrolyse stabilisiertes pharmazeutisches Mittel, das Acetylsalicylsäure,
d-Propoxyphenhydrochlorid und das Hydrochlorid einer natürlich vorkommenden Aminosäure enthält.
Die für die oben erläuterten erfindungsgemäßen Zwecke geeigneten Verbindungen umfassen die 23 natürlich vorkommenden
Aminosäuren, welche unter der populären Bezeichnung "essentielle Aminosäuren" bekannt sind. Die Bezeichnung
"essentiell" soll nicht lebensnotwendig als Gegensatz
zu nicht unbedingt erforderlich im Sinne menschlicher Ernährungsbedürfnisse heißen, sondern schließt Aminosäuren
beider Art ein und umfaßt folgende natürlich vorkommende Aminosäuren, die sowohl mit ihrem üblichen
als auch mit ihrem wissenschaftlichen Namen bezeichnet sind.

Im folgenden wird der Begriff " natürlich vorkommende
Aminosäuren" zur Bezeichnung der oben beschriebenen Verbindungsklasse verwendet.
Die für die erfindungsgemäßen Zwecke verwendeten Hydrochloride von natürlich vorkommenden Aminosäuren werden
durch Umsetzung solcher Aminosäuren mit Chlorwasserstoffsäure in wässriger oder äthanolischer Lösung hergestellt,
und durch Umkristallisieren nach bekannten Methoden gereinigt. Im Rahmen der Erfindung kommen sowohl die Dihydrochloride
als auch die Monohydrochloride der Diaminocarbonsäuren Arginin, Lysin und Citrullin in Betracht.

Natürlich vorkommende Aminosäuren, die in Form des Hydrochlorids für die erfindungsgemäßen Zwecke geeignet sind,
treten in der .Natur in der L-Form auf. Die DL-Aminosäureracematmischungen
sind synthetisch hergestellt worden und die D-Form ist ebenfalls bestätigt. Alle drei Formen
sind in Form ihrer Hydrochloridsalze verfügbar und als Hydrochloridsalze für die erfindungsgemäßen Zwecke geeignet.
Die L-Form ist als Handelsprodukt von den Formen natürlich vorkommender Aminosäurehydrochloride
am verbreitetsten und stellt die bevorzugte Ausführungsform dar, die hierin als Beispiel verwendet wird.
Bei der praktischen Anwendung der Erfindung wurde gefunden, daß die Menge des Aminosäurehydrochlorids,
die zur Inhibierung der Hydrolyse des Aspirins erforderlich ist, zu der Menge an d-Propoxyphenhydrochlorid, die in der
Aspirin/d-Propoxyphenhydrochlorid-Kombination vorhanden ist, in Beziehung steht. Eine wirksame Inhibierung der Hydrolyse
wird erzielt, wenn eine natürlich vorkommende Aminosäure als Hydrochlorid in das pharmazeutische Mittel in
einer Menge von etwa 5 bis etwa 130 Gewichtsprozent der vorhandenen Menge an d-Propoxyphenhydrochlorid eingemischt
wird. Vorzugsweise wird das Hydrochlorid der natürlich vorkommenden Aminosäure in einer Menge von etwa 5 bis etwa
50 % der vorhandenen Menge an d-Propoxyphenhydrochlorid angewandt. Das für die erfindungsgemäßen Zwecke vorteilhafte
natürlich vorkommende Aminosäurehydrochlorid kann mit den anderen Bestandteilen des pharmazeutischen Mittels
in beliebiger Reihenfolge vermischt werden und ergibt eine wirksame Inhibierung der Hydrolyse von Aspirin.
Vorzugsweise werden das natürlich vorkommende Aminosäurehydrochlorid und d-Propoxyphenhydrochlorid vor Vereinigung
mit Aspirin innig vermischt. Nach einer besonders

bevorzugten Ausführungsform der Erfindung werden ein natürlich
vorkommendes Aminosäurehydrochlorid und d-Propoxyphenhydrochlorid
innig vermischt und zusammen mit einem pharmazeutisch annehmbaren Lösungsmittel, in dem einer der Bestandteile
in erheblichem Maß löslich ist, zum Beispiel Wasser, Methanol, Äthanol, Isopropanol, Aceton, Methylethylketon,
Methylisobutylketon, Chloroform, Methylenchlorid, Trichloräthylen, Trichlorfluormethan, 1,2-Dichlor-l,!,2,2-tetrafluoräthan
und dergleichen, vorzugsweise Äthanol, granuliert, das Lösungsmittel durch Verdampfen entfernt und das getrocknete
Granulat mit dem Aspirin vereinigt.
Das Vermischen des Hydrochlorids einer natürlich vorkommenden Aminosäure, vorzugsweise in der L-Form, mit
der Kombination aus dem Aspirin und d-Propoxyphenhydrochlorid führt zu einer vielfachen Verminderung der Geschwindigkeit
und des Grads der Hydrolyse von Aspirin in Gegenwart von Feuchtigkeit.
Eine besonders vorteilhafte Nutzung eines natürlich vorkommenden Aminosäurehydrochlorids zur Inhibierung der
Hydrolyse von Aspirin wird durch Verwendung von L-Glutaminsäurehydrochlorid
anstelle der gesamten Menge des inerten Bestandteils Stärke in einem der oben genannten pharmazeutischen
Mittel erreicht, wodurch ein pharmazeutisches Mittel mit folgender Zusammensetzung erhalten wird: 40,0 % Aspirin;
11,4 % d-Propoxyphenhydrochlorid; 28,4 % Phenacetin; 5,7 % Coffein; 14,6 % L-Glutaminsäurehydrochlorid. Die
Hydrolysegeschwindigkeit von Aspirin in einem solchen Mittel ist im Vergleich zu dem unstabilisierten Mittel
um 70 bis 90 % vermindert.
Wenn L-Lysindihydrochlorid in einer Menge von etwa 18 % des d-Propoxyphenhydrochloridgehalts in einem

pharmazeutischen Mittel, das 52,4 % Aspirin, 10,5 %
d-Propoxyphenhydrochlorid und 37,1 % Kaolin enthält,
zugesetzt wird, wodurch, ein pharmazeutisches Mittel erhalten wird, in dem die natürlich vorkommende Aminosäure
als Hydrochlorid etwa 2 % der Gesamtmenge ausmacht, wird die Hydrolysegeschwindigkeit um mehr als
50 % vermindert.
In dem pharmazeutischen Mittel, das sich durch die Kombination aus Aspirin, d-Propoxyphenhydrochlorid
und dem Hydrochlorid einer natürlich vorkommenden Aminosäure auszeichnet, können weitere verträgliche
Wirkstoffe und pharmazeutisch annehmbare inerte Bestandteile vorhanden sein, oder das Mittel kann allein aus
dieser Kombination bestehen. In jedem Fall wird die Geschwindigkeit der Aspirinhydrolyse in Gegenwart des
natürlich vorkommenden Aminosäurehydrochlorids erheblich
verzögert.
Andere pharmazeutisch annehmbare wasserlösliche Salze von natürlich vorkommenden Aminosäuren, zum Beispiel
Phosphate, Sulfate und dergleichen können ebenfalls ^ mit Erfolg zur Hemmung der Hydrolyse von Aspirin in
pharmazeutischen Mitteln, die d-Propoxyphenhydrochlorid und Aspirin enthalten, angewandt werden.
Die neuen pharmazeutischen Mittel nach der Erfindung können in bekannter Weise in geeigneten Mengen zur Erzielung
von Dosierungseinheiten, welche die geeigneten Mengen der therapeutischen Mittel zur oralen Verabreichung
enthalten, in Gelatinekapseln abgefüllt oder zu Tabletten verpreßt werden.
Durch die folgenden Beispiele wird die Erfindung näher erläutert.

Pharmazeutische Mittel, die Aspirin und d-Propoxyphenhydrochlorid enthalten, werden entsprechend den folgenden
Rezepturen zubereitet und in Gelatinekapseln abgefüllt. Die gefüllten Kapseln werden vier Tage bei 50 Grad C gelagert.
Dann wird der Inhalt nach der Methode, die auf Seite 607 von N.F. XIII (1970) erläutert ist, auf freie
Salicylsäure analysiert. Die gefundene Salicylsäuremenge in % ist in Tabelle I angegeben.

Das d-Propoxyphenhydrochlorid und das L-Glutaminsäurehydrochlorid
werden 5 Minuten in einem Hobart-Mischer
miteinander vermischt. Die Pulvermischung wird mit 10 ml Äthylalkohol versetzt und der Mischer wird weitere
5 Minuten in Gang gehalten. Die angefeuchtete Masse wird aus dem Mischer entnommen, durch ein Sieb mit einer lichten
Maschenweite von 3,36 mm (No. 6 mesh, U.S.) geführt und auf Blechen ausgebreitet. Der Äthylalkohol wird aus
dem Granulat im Vakuum während zwei Stunden bei 25 Grad C entfernt. Das getrocknete Granulat wird durch ein Sieb
mit einer lichten Maschenweite von 1,19 mm (16 mesh, U.S.) getrieben und in einen Laboratoriumsbandmischer gegeben.
Dann werden die übrigen Bestandteile in den Mischer gegeben. Nach 15 Minuten langem Mischen wird in Gelatinekapseln
Größe 0 abgefüllt. Jede Kapsel enthält 570 mg.

Die Ergebnisse zeigen deutlich, daß die Vereinigung von L-Glutaminsäurehydrochlorid mit Aspirin und d-Propoxyphenhydrochlprid
zu einer 3- bis 10-fachen Verminderung der Geschwindigkeit der Aspirinhydrolyse, gemessen an
der Bildung freier Salicylsäure in dem pharmazeutischen Mittel, führt.

Die Werte in Tabelle II zeigen, daß alle drei Formen
von Glutaminsäurehydrochlorid die Hydrolyse von Aspirin
in Gegenwart von d-Propoxyphenhydrochlorid wirksam inhibieren, wobei die L-Form die stärkste Wirkung zeigt, wie durch Verminderung der Menge an freier Salicylsäure in
dem pharmazeutischen Mittel, in dem Glutaminsäurehydrochlorid als einer der Bestandteile enthalten ist, gemessen wird.

Pharmazeutische Mittel, die Aspirin, d-Propoxyphenhydrochlorid und L-Lysindihydrochlorid enthalten, werden entsprechend
den folgenden Rezepturen hergestellt und in Gelatinekapseln abgefüllt. Die gefüllten Kapseln werden 4 Tage bei 50 Grad C
gelagert und dann wird der Inhalt nach der Methode, die
auf Seite 607 von N.F.XIII (1970) angegeben ist, auf freie Salicylsäure analysiert. Die gefundene Salicylsäuremenge in % zeigt Tabelle III.

Jedes Ergebnis ist der Mittelwert von 6 getrennten Bestimmungen.
Die Ergebnisse zeigen, daß der Zusatz von L-Lysindihydrochlorid
zu einem pharmazeutischen Mittel aus Aspirin und d-Propoxyphenhydrochlorid eine beträchtliche
Wirkung auf die Geschwindigkeit der Aspirinhydrolyse, gemessen an der Bildung von freier Salicylsäure,
hat.

Claims (7)
1. Verfahren zum Inhibieren der Hydrolyse von Acetylsalicylsäure in einem pharmazeutischen Mittel,
das Acetylsalicylsäure und d-Propoxyphenhydrochlorid enthält, dadurch gekennzeichnet, daß man dem pharmazeutischen
Mittel das Hydrochlorid einer Verbindung der Formel
R-C-C-OH oder

zusetzt, worin
R ein Wasserstoffatom oder einen Methyl-, Isopropyl-,
Butyl-, Isobutyl-, sec.-Butyl-, Hydroxymethyl-, 1-Hydroxyäthyl-, Carboxymethyl-, Carboxyäthyl-,
Carboxy-(1-hydroxy)äthyl-, Aminobutyl-, Methy1-thioäthyl-,
Mercaptomethyl-, Carboxy-(2-aminoäthyl-dithiomethyl-,
p-Hydroxyphenyl-, Benzyl-, Indolylmethyl-, Guanidopropyl-, Imidazolylmethyl oder
üreidopropyl-Rest und
R. ein Wasserstoffatom oder eine Hydroxygruppe bedeutet.
2. Verfahren nach Anspruch 1, dadurch gekennzeichnet, daß man die Verbindung dem pharmazeutischen Mittel in
einer Menge von etwa 5 bis etwa 130 Gewichtsprozent der Menge an d-Propoxyphenhydrochlorid in dem pharmazeutischen

3. Verfahren nach Anspruch 1 oder 2, daß man
die Verbindung vor dem Einbringen in das pharmazeutische Mittel mit dem d-Propoxyphenhydrochlorid innig vermischt.
4. Verfahren nach Anspruch 1 oder 2, dadurch gekennzeichnet, daß man die Verbindung und das d-Propoxyphenhydrochlorid
unter Verwendung eines pharmazeutisch annehmbaren Lösungsmittels gemeinsam granuliert, das Lösungsmittel
daraus verdampft und das getrocknete Granulat in das pharmazeutische Mittel einbringt.
5. Verfahren nach Anspruch 1,2,3 oder 4, dadurch
gekennzeichnet, daß man als die Verbindung L-Glutaminsäurehydrochlorid
oder L-Lysindihydrochlorid verwendet.
6. Gegen Hydrolyse stabilisiertes pharmazeutisches Mittel, dadurch gekennzeichnet, daß es Acetylsalicy!säure,
d-Propoxyphenhydrochlorid und das Hydrochlorid einer natürlich vorkommenden Aminosäure enthält.
7. Pharmazeutisches Mittel nach Anspruch 6, dadurch gekennzeichnet, daß es als natürlich vorkommende Aminosäure L-Glutaminsäure oder L-Lysin enthält.
