Abstract
Description
Cyclohexylglycine is a common structural element biologically active substances. The present invention relates to a process for the preparation of cyclohexylglycine benzyl ester with high space-time yields.
Literature-known ways that z. B. in cyclohexylalanine to one have led suitable synthesis building block, due to the peculiarities of cyclohexylglycine not realize. The reason for this is the solubility and crystallization behavior ten of the salts of cyclohexylglycine or cyclohexylglycinben zylesters.
If the Ver described for cyclohexylalanine in WO 98/4761 drive to transfer to the cyclohexylglycine has been observed that the para-toluenesulfonate of the cyclohexylglycine benzyl ester extremely voluminous crystallized and for further processing was very poorly suited. At the same time, this salt is bad water soluble so that instead of cleaning the substance over a crystallization also a purification via the extraction in the aqueous phase was not possible.
The HCl salt of the cyclohexylglycine benzyl ester is in the Literature (e.g. WO 98/07697) as a starting product for others Implementations described.
There have been experiments with ethereal HCl and aqueous salt acid directly undertakes the product from Boc-cyclohexylglycine with benzyl alcohol. Both with concentrated HCl as well as with ethereal HCl solution, the results were inconclusive peaceful because the HCl salt of cyclohexylglycine forms, precipitates in the solvent mixture of benzyl alcohol and toluene and thus a further reaction to the ester is prevented.
The use of trifluoroacetic acid has the disadvantage that this due to the boiling point of 73 ° C together with water is deposited.
The task was therefore to develop a process which cyclohexylglycine benzyl in reasonable space-time yields makes ester with high purity accessible.
Such a method was surprisingly one-step the use of methanesulfonic acid starting from a cyclo hexylglycine derivative of formula I can be found.
The use of methanesulfonic acid in the acidic esterification of cyclohexylglycine to the cyclohexylglycine benzyl ester leads to a very simple process for providing the substance on a pilot plant scale with good space-time yields. The reason is the favorable solubility behavior of the salts formed between the methane sulfonic acid and the cyclohexylglycine or cyclohexylglycine benzyl ester. During the first salt formation, the solubility in the reaction mixture of the esterification is ensured, in the case of the second salt it is possible to prepare an aqueous solution which allows the product to be purified.

The reaction is carried out by dissolving cyclohexylglycine derivative of the formula I in a suitable mixture of benzyl alcohol and a water-entraining solvent, preferably toluene. Under these conditions, the formation of salt III with the methanesulfonic acid and the further esterification to the cyclohexylalycinester salt of the formula IV take place
The compounds of formula III and IV are new and can be made from the corresponding solutions are isolated.
The Cyclohexylglycinbenzylestersalz of formula IV can now by Extraction with water can be converted into an aqueous phase. By washing the aqueous phase with organic solvents, preferably ether, methyl tert-butyl ether and toluene separated organic impurities. Subsequent basic put the aqueous phase with subsequent extraction before added with solvents such as ether, methyl tert-butyl ether, Toluene and dichloromethane, after washing, gives the organic Phase a form of cyclo that is well suited for further processing hexylglycine benzyl ester II.
