Nicht vakzinierte Tiere dienten als Kontrolle. 35 Tage nach dem Challenge wurden die Tiere geschlachtet und in der Lunge die Anzahl adulter Würmer bestimmt; gleichzeitig wurde die Länge intakter männlicher und weiblicher Würmer gemessen. In der vakzinierten Gruppe wurde eine um 80% reduzierte Anzahl adulter Würmer gefunden. Nach 3-maligem Waschen wurden die Vertiefungen mit einer 1 : 200-Verdünnung spezifischer Infektions- und Kontrollseren beschickt und 1 Stunde bei Raumtemperatur inkubiert. Als Nachweisantikörper wurde nach 3-maligem Waschen Peroxidase-konjugierte, polyklonale Kaninchenantikörper gegen bovines IgG (Fc-Fragment spezifisch; Fa. Dianova) in einer Verdünnung von 1 : 10 000 verwendet; die Inkubation dauerte 30 min. Die Platten wurden anschließend 4 mal gewaschen und dann mit Substratlösung inkubiert (Zusammensetzung: 5 ml 10-fach konzentrierter ABTS-Puffer + 45 ml Reinstwasser + 1 Tablette ABTS (≘ 50 mg ABTS). Die Substratentwicklung erfolgte bei Raumtemperatur und wurde im ELISA-Reader alle 10 min bei 414 nm überwacht.
