mit einer Verbindung, welche die Mercaptogruppe enthält, zur Reaktion bringt.
Die Erfindung betrifft ein neues Verfahren zur Herstellung von Thiaminderivaten der Formel

Einige der Thiaminderivate der obigen Formel I sind als wichtige biochemische Verbindungen bekannt.
Beispielsweise ist Thiamin-O-diphosphat seit langem als ein Coenzym bekannt, welches als Cocarboxylase
bezeichnet wird. Thiamin-O-triphosphat ist als Verbindung mit einer hochenergetischen Phosphatbindung
bekannt, die in der Leber und der Musku-

latur der Säugetiere ebenso wie in Mikroorganismen
40 enthalten ist.
Bisher sind diese Thiaminderivate hergestellt worden durch Reaktion von Thiamin mit Polyphosphorsäure,
die aus Phosphorsäure erhalten wurde. Durch Erhitzen beider Ausgangsstoffe ergibt sich ein Gemisch
von Thiaminphosphaten, das Thiamin-O-monophosphat, Thiamin- O-diphosphat, Thiamin- O-triphosphat
und Thiamin-O-polyphosphat enthält. Jedes Phosphat kann aus dem Reaktionsgemisch isoliert werden.
Das bisherige Verfahren ist jedoch in wirtschaftlicher Hinsicht unzweckmäßig, weil die Ausbeuten der isolierten
Phosphate außerordentlich gering sind.
Es wurde nunmehr gefunden, daß man Thiaminderivate der obigen Formel I mit hohem Reinheitsgrad
und mit ausgezeichneten Ausbeuten dadurch herstellen kann, daß man eine Verbindung der Formel

haben.
Die erste Stufe des erfindungsgemäßen Verfahrens umfaßt die Reaktion eines Thiaminderivates der
obigen Formel II mit Chlorphosphorsäurediphenylester oder einem an den Phenylgruppen alkoxy-substituierten
Derivat davon in Gegenwart eines tertiären Amins, wobei ein Thiaminderivat der obigen
Formel III entsteht. Bei Ausführung der ersten Stufe wird ein Thiaminderivat der obigen Formel II in
einem organischen Lösungsmittel gelöst oder suspendiert, worauf ein tertiäres Amin zugegeben wird. Dann
) wird Wasser von dem erhaltenen Gemisch durch bekannte Verfahren abgezogen, um ein im wesentlichen
wasserfreies Gemisch zu bilden, zu dem hierauf ein Chlorphosphorsäurediphenylester oder -di-(alkoxyphenyl)-ester
hinzugefügt wird. Es tritt eine Reaktion der letzteren Verbindung mit dem Aminsalz des Thiaminderivates
der Formel II ein. Beispiele besonders geeigneter organischer Lösungsmittel sind niedrige
Alkanole, wie Methanol und Äthanol, Amide, wie Dimethylformamid und Formamid, cyclische Äther, wie
Dioxan, Dialkylsulfoxyde, wie Dimethylsulfoxyd und Diäthylsulfoxyd, und organische Basen, wie Pyridin.
Das in der ersten Stufe verwendete tertiäre Amin ist z. B. ein Trialkylamin, wie Triäthylamin, Tributylamin,
Tri-(isooctyl)-amin, Tri-(n-octyl)-amin, Tri-(isononyl)-amin und Ν,Ν-Didodecyl-N-n-butylamin, oder
ein Dialkyl-monoaryl-amin, wie Dimethylphenylamin
und Diäthylphenylamin. ^0
Der Chlorphosphorsäurediphenylester kann injeder Phenylgruppe mit einer oder mehreren Alkoxygruppen
mit 1 bis 5 Kohlenstoffatomen substituiert sein. > Beispiele für Chlorphosphorsäurereaktionsteilnehmer
^ sind Chlorphosphorsäurediphenylester und Chlorphosphorsäure-di-(p-methoxyphenyl)-ester.
Die Zeit und Temperatur der Reaktion sind keine kritischen Merkmale der Erfindung. Die Reaktion
wird vorzugsweise bei Zimmertemperatur während 1 bis 5 Stunden ausgeführt.
Nach Beendigung der Reaktion kann das aus der ersten Stufe erhaltene Thiaminderivat der obigen
Formel II leicht durch eine der üblichen Methoden abgetrennt werden, beispielsweise durch Entfernung
des Lösungsmittels aus dem Reaktionsgemisch, und das so erhaltene Produkt kann für die folgende zweite
Stufe des Verfahrens verwendet werden, gegebenenfalls nach weiterer Reinigung mit Hilfe bekannter Verfahren.
Die zweite Stufe des erfindungsgemäßen Verfahrens &>
umfaßt zwei Abschnitte, nämlich die Reaktion des Thiaminderivates der obigen Formel III mit einer
Phosphorverbindung der obigen Formel IV in Gegenwart eines tertiären Amins, und die Behandlung des
dabei erhaltenen Zwischenproduktes mit einer Ver- t5
bindung, welche eine Mercaptogruppe enthält, um die Verbindungen der obigen Formel I, zu erhalten.
Bei der Ausführung der obigen zweiten Stufe wird das in der ersten Stufe erhaltene Thiaminderivat in einem
geeigneten organischen Lösungsmittel gelöst oder suspendiert. Das erhaltene Gemisch wird dann mit
einer Phosphorverbindung der obigen Formel IV in Gegenwart eines tertiären Amins und eines organischen
Lösungsmittels behandelt, worauf sich eine Behandlung mit einer Verbindung, die eine Mercaptogruppe
enthält, anschließt.
Beispiele besonders geeigneter organischer Lösungsmittel sind die bereits bei der ersten Stufe erwähnten.
Phosphorverbindungen der Formel IV sind z. B. Phosphorsäure und Pyrophosphorsäure, 5'-Adenylsaure
und 5'-Uridylsäure.
Die eine Mercaptogruppe enthaltende Verbindung, die in dem letzten Abschnitt der zweiten Verfahrensstufe verwendet wird, kann eine Verbindung sein,
welche eine Mercaptogruppe (— SH) im Molekül enthält. Beispiele hierfür sind Schwefelwasserstoff; Alkylmercaptane,
wie Methylmercaptan und Äthylmercaptan; Arylmercaptane, wie Thiophenol und a-(oder
/J-)-Thionaphthol; Mercaptogruppen enthaltendeAminosäuren,
wie Cystein; und Thioglykolsäure.
Die Zeit und die Temperatur sind in beiden Abschnitten der zweiten Stufe nicht kritisch. Die Reaktion
wird vorzugsweise während 1 bis 24 Stunden bei Zimmertemperatur im ersten Abschnitt und 1 bis
4 Stunden bei Zimmertemperatur oder unter leichtem Erwärmen im zweiten Abschnitt durchgeführt.
Nach Beendigung der Reaktion kann das Endprodukt aus der zweiten Stufe aus dem Reaktionsgemisch
durch eine der bekannten Methoden gewonnen werden, beispielsweise durch Entfernung des Lösungsmittels
aus dem Reaktionsgemisch. Das Endprodukt kann weiterhin durch ein bekanntes Verfahren gereinigt
werden, wie durch die Entfernung von Verunreinigungen mit einem geeigneten Lösungsmittel, wie
Methanol oder Äthanol, oder durch Behandlung mit einem Ionenaustauscherharz.
Die Erfindung wird durch die folgenden Beispiele näher erläutert.
Thiamin- O-triphosphat
Zu einer Suspension von 5 g S-Benzoylthiamin-O-monophosphat
in Methanol-Äthanol werden 10 ml Tri-(n-octyi)-amin hinzugefügt. Das Gemisch wird
zur Trockne eingeengt, der Rückstand in wasserfreiem Dioxan gelöst und die Dioxanlösung' erneut zur
Trockne eingeengt. Das vorbeschriebene Verfahren wird dreimal wiederholt, wonach eine Spur Wasser
aus der Dioxanlösung entfernt wird.
Zu der wasserfreien Dioxanlösung werden 3 ml Chlorphosphorsäurediphenylester hinzugefügt und
nach Rühren wird die erhaltene Lösung 3 Stunden bei Zimmertemperatur stehengelassen. Das Lösungsmittel
wird dann aus dem Reaktionsgemisch durch Destillation unter vermindertem Druck entfernt, der
Rückstand mit wasserfreiem Äther gerührt und dann

die Lösung stehengelassen, wobei sich eine kristalline Substanz abscheidet, die durch Filtration zurückgewonnen
wird und 6 g P2-Diphenyl-S-benzoylthiamin-O-pyrophosphat
als weiße pulverförmige Substanz ergibt.
Analysenwerte für C31H32O9N4SP2:
Berechnet ... C 53,20, H 4,62, N 8,02, P 8,87%; gefunden .... C 51,50, H 5,19, N 8,67, P 8,53%.
Das so erhaltene P2-Diphenyl-S-benzoylthiamin-O-pyrophosphat
(6 g) wird in Dimethylformamid gelöst. Zu der Lösung wird eine Lösung von 3,6 g Pyrophosphorsäure in Dioxan, das Tri-(n-octyl)-amin
enthält, hinzugefügt, das Gemisch heftig geschüttelt und dann über Nacht bei Zimmertemperatur stehengelassen.
Das Lösungsmittel wird aus dem Reaktionsgemisch abdestilliert und der Rückstand mit
überschüssigem, wasserfreiem Äther behandelt, wobei man 5,3 g S- Benzoylthiamin - O - triphosphat als
weiße pulverförmige Substanz erhält. Das so erhaltene S - Benzoylthiamin - O - triphosphat (5 g) wird in
einem Gemisch von Methanol und Wasser (70:35) gelöst. 1 ml Thiophenol wird zu der Lösung hinzugefügt,
die dann mit Natriumhydroxyd auf einen pH von 6,5 gebracht wird. Die neutralisierte Lösung
wird auf einem Wasserbad auf 50° C erwärmt, 2 Stunden bei Zimmertemperatur stehengelassen und dann
eingeengt, wobei eine kristalline Substanz zurückbleibt, die in Form langer Nadeln abgeschieden
wird und unlöslich in Wasser und löslich in Äthanol ist. Die kristalline Substanz wird durch Filtration
entfernt. Die Mutterflüssigkeit wird gefriergetrocknet und ergibt 3,86 g pulverförmiges Thiamin-O-triphosphat
mit einem Reinheitsgrad von 75%.
1 g Thiamin-O-triphosphat wird der Behandlung mit einem Ionenaustauscherharz unterworfen und
ergibt 0,57 g einer reinen Substanz, die mit einer authentischen Probe durch Chromatographie identifiziert
wird.
Thiamin- O-diphosphat
S-Benzoylthiamin-O-diphosphat wird aus S-Benzoylthiamin
- O - monophosphat über P2 - Diphenyl-S - benzoylthiamin - O - pyrophosphat durch das
gleiche Verfahren wie im Beispiel 1 hergestellt,
Das so erhaltene S-Benzoylthiamin-O-diphosphat wird in das Thiamin-O-diphosphat mit einem Reinheitsgrad
von 90% in der gleichen Weise wie im Beispiel 1 übergeführt. Ausbeute: 55%%. Das so
erhaltene Produkt wird durch die authentische Probe durch Chromatographie identifiziert.
Beispiel 3 Thiamin- O-triphosphat
Zu einer Lösung von Bis - (thiamin - O - monophosphat)-disulfid
(1 Mol) und Tri-(n-octyl)-amin (4 Mol) in Dimethylformamid wird Diphenylphosphorochloridat
(6 Mol) hinzugefügt.
Das erhaltene Gemisch wird wie im Beispiel 1 behandelt und ergibt Bis - (P2 - diphenyl - thiamin-O
- pyrophosphat) - disulfid.
Das so erhaltene Bis - (P2 - diphenyl - thiamin-O-pyrophosphat)-disulfid
wird in Dimethylformamid gelöst und eine Lösung von Pyrophosphorsäure (8 Mol) in Dimethylformamid, das Tri - (n - octyl)-amin
enthält, hinzugefügt. Das Gemisch wird etwa 48 Stunden bei Zimmertemperatur gerührt. Das Reaktionsgemisch
wird filtriert, die Mutterflüssigkeit zur Trockne eingeengt und der Rückstand mit wasserfreiem
Äther behandelt, wodurch sich ein weißes Pulver ergibt, das in Wasser gelöst wird. Die wäßrige
Lösung wird wie im Beispiel 1 mit Cystein behandelt und ergibt Thiamin-O-triphosphat, das ebenso wie
im Beispiel 1 durch Chromatographie identifiziert wird.

Thiamin- O-triphosphat
S - Benzoylthiamin - O - triphosphat (5 g) wird hergestellt aus S-Benzoylthiamin-O-monophosphat
'5 (5 g) durch das gleiche Verfahren wie im Beispiel 1.
Eine Lösung des so erhaltenen S-Benzoylthiamin-
O-triphosphat in Methanol-Wasser (70: 35) wird durch
Hinzufügen von wasserfreiem Natriumhydroxyd auf einen pH von 6,5 neutralisiert, und Schwefelwasserstoff
wird 30 Minuten bei 40 bis 50° C durch die Lösung geleitet. Das Methanol wird dann durch
Destillation unter vermindertem Druck aus dem Reaktionsgemisch entfernt und der Rückstand in
20 ml Äther gerührt. Die wäßrige Schicht wird abgeschieden und durch Hinzufügen von 7 ml 80%iger
Ameisensäure auf einen pH von 2,7 gebracht. Zu der sauren Lösung werden 20 ml Äther hinzugefügt, das
Gemisch wird eine Weile gerührt, die wäßrige Schicht abgeschieden und dann gefriergetrocknet, wobei sich
rohes Thiamin-O-triphosphat als weiße pulverförmige kristalline Substanz ergibt. Das so erhaltene rohe
Produkt wird mit einem Ionenaustauscherharz behandelt und ergibt 1,8 g einer kristallinen Substanz
mit einer Reinheit von 85 bis 90%, die aus verdünntem Äthanol umkristallisiert wird und das im wesentlichen
reine kristalline Material ergibt.

Eine Lösung von S-Benzoylthiamin-O-triphosphat
wird aus S - Benzoylthiamin - O - monophosphat durch das gleiche Verfahren wie im Beispiel 4 hergestellt.
Der so erhaltenen Lösung werden 2,7 g /i-Thionaphthol
hinzugefügt, und das Gemisch wird 10 Minuten auf 50° C erwärmt. Die kristalline Substanz,
die in dem Reaktionsgemisch abgeschieden wird, wird durch Filtration entfernt und aus dem Filtrat
das Methanol entfernt. Die zurückbleibende Lösung wird in der gleichen Weise wie im Beispiel 4 behandelt
und ergibt 1,7 g Thiamin-O-triphosphat mit einer Reinheit von 85 bis 90%.
Thiamin- O-triphosphat
Das gleiche Verfahren wie im Beispiel 5 wird wiederholt mit der Abänderung, daß 1,6 g Thioglykolsäure
an Stelle von 2,7 g /Ϊ-Thionaphthol verwendet
werden, wobei man 1,8 g Thiamin-O-triphosphat mit einer Reinheit von 85 bis 80% erhält,
das mit dem des Beispiels 5 identisch ist.

Thiamin-adenosin-diphosphat
Thiamin-adenosin-diphosphat
P2 - Diphenyl - S - benzoylthiamin - O - pyrophosphat
wird aus S - Benzoylthiamin - O - monophosphat durch das gleiche Verfahren wie im Beispiel 1 hergestellt.
Zu einer Lösung des P2-Diphenyl-S-benzoylthiamin-O-pyrophosphat
(1 Mol), das wie oben beschrieben in Dimethylformamid, welches Tri-(n-octyl)-amin
enthält, erlangt wurde, wird eine Lösung von 5'-Adenylsäure (2 Mol) in Dimethylformamid, welches
Tri - (n - octyl) - amin enthält, hinzugefügt. Das Gemisch wird etwa 48 Stunden bei Zimmertemperatur
stehengelassen. Das Lösungsmittel wird aus dem Reaktionsgemisch durch Destillation entfernt und
der Rückstand mit Äther behandelt. Dies ergibt eine weiße gelartige Substanz, die wie im Beispiel 1 mit
Cystein debenzoyliert wird und Thiamin-adenosindiphosphat ergibt; Rf-Wert = 0,41 (n-Propanpl-O^nammoniumacetat3:
2;Rf-WertvonThiamin-O-monophosphat: 0,55 und Rf-Wert von P2 - Adenosinthiamin-O-monophosphat:
0,49).

Thiamin-uridin-diphosphat
Thiamin-uridin-diphosphat
P2 - Diphenyl - S - benzoylthiamin - O - pyrophosphat
wird aus S - Benzoylthiamin - O - monophosphat (12,5 g) durch das gleiche Verfahren wie in Beispiel 1
hergestellt.
Zu einer Lösung des in oben beschriebener Weise erlangten P2 - Diphenyl - S - benzoylthiamin - O - pyrophosphat
in Dimethylformamid, das Tributylamin enthält, wird eine Lösung von 17,5 g 5'-Uridylsäure
in Dimethylformamid, das Tributylamin enthält, hinzugefügt und das Gemisch 2 Stunden bei Zimmertemperatur
stehengelassen. Das Lösungsmittel wird aus dem Reaktionsgemisch durch Destillation entfernt,
der Rückstand in wäßrigem Methanol gelöst und die Lösung mit Cystein behandelt. Dies ergibt
30 g rohes P2 - Uridin - thiamin - O - diphosphat als weiße pulverförmige Substanz.
Die so erhaltene Substanz wird mit Dowex-1 (in der Chloridform; Warenzeichen eines Anionenaustauscherharzes,
hergestellt und vertrieben durch die Firma Dow Chemical Company) behandelt und ergibt 4,5 g reines Thiamin-uridindiphosphat als weiße
pulverförmige Substanz, die durch Elektrophorese charakterisiert wird.
