Die Erfindung betrifft 2-Furfuryl-3-(naphthyl-l')-propionsäure der FormelThe invention relates to 2-furfuryl-3- (naphthyl-1 ') propionic acid of the formula
CH-COOHCH-COOH
und deren Natriumsalz.and their sodium salt.
The compounds of the above formula have cholesterol-lowering and bile secretion-increasing properties. These effects were trying in comparison T over the known Natriumbiphenylylbutyrat (Journal of the American Chemical Society, Vol. 79 [1957], p 3514) tested and the values for lowering cholesterol, lipid and Lipoproteinspiegels in the blood after administration of the substances tested. The experiments were carried out with rats in which hypercholesterolemia according to that of Garattini. and CoI. in "Experiencia", 1956, 9, p. 347, described method by intervening administration of 250 mg / kg p-isooctyl polyoxyethylene phenol. 20 hour old male rats. The compounds to be tested were administered to these rats intraperitoneally at a dose of 150 mg / kg of body weight. The rats were decapitated 18 hours after the injection. The cholesterol was then determined with the serum of the decapitated animals according to the method described by Zack and Z a 1 kis in American Journal of Chimical Pathology 1957, pp. 583 to 588. The lipids were determined according to the method described by D e 1 sa 1 in Bulletin de la Societe de Chimie Biologique, 26 (1944), p. 99, and the ß- lipoproteins according to Burstein, "Pathologie et Biologie" (La semaine des Hospitoux- Annales de Pathologie et Biologie, Paris) April 1958, p. 54.

Cholesterol is responsible for the formation of atheromas, which are caused by an accumulation of Form lipids and lipoproteins at certain points in the circulatory system. Since the invention Compounds the cholesterol and in some cases also the lipid and lipoprotein levels in the blood lower, they are thus useful as an agent against arteriosclerosis and are superior to the known substance in this regard.
The 2-furfuryl-3- (naphthyl-l ') - propionic acid and its sodium salt can be mixed or dissolved are used in solvents or conventional auxiliaries for the treatment of arteriosclerosis, the daily dosages for this by oral, rectal or parenteral route between 0.010 and 2 g lie.
Die Verbindungen der oben angegebenen Formel werden dadurch hergestellt, daß man in an sich bekannter Weise einen 2-Furfuryl-2-(naphthyl-l'-methyl)-malonsäurealkylester der allgemeinen FormelThe compounds of the formula given above are prepared by being known per se Way a 2-furfuryl-2- (naphthyl-l'-methyl) -malonic acid alkyl ester of the general formula

The reaction is preferably carried out in the presence of benzyl alcohol. The starting material Substituted alkyl malonate to be used can expediently by reacting a Sodium alcoholate with a 2- (naphthyl-r-methyl) -malonic acid alkyl ester or 2-furfuryl malonic acid alkyl ester and subsequent condensation of the resulting Sodium derivative with 1-chloromethyl or 1-bromomethylfuran or with 1-chloromethyl or 1-bromomethylnaphthalene can be obtained.
Beispiel
2-Furfuryl-3-(naphthyl-1 ^-propionsäureexample
2-furfuryl-3- (naphthyl-1 ^ -propionic acid
A mixture of 11 g of caustic potash in 860 cm 3 of benzyl alcohol and 38 g of 2-furfuryl-2- (naphthyl-l'-methyl) -malonic acid diethyl ester is refluxed for 15 hours. The reaction mixture is then distilled and collected the over at Kp. 15207-209 0 C fraction as a product. There are thus 19 g of a viscous liquid of Kp. 1 obtained 5207-209 C 0. The yield of 2-furfuryl-3- (naphthyl-1 ') propionic acid is 68% of theory.
When extracting with benzene instead of ether, the yield is 72% of theory.
The viscous liquid obtained in the distillation is covered with a layer of hexane and crystallized quickly turns into practically white crystals when scraping the vessel wall with a glass rod.
Die Säurezahl der Verbindung beträgt 198 (theoretisch 199).18 g of these crystals are dissolved in 18 cm 3 of lukewarm ethyl acetate, the solution is filtered, then wetted with 90 cm 3 of hexane and left to stand in a refrigerator. The oil which precipitates out is transformed into a white, crystalline mass of F. = 72 to 74 ° C. (heated platinum) after the crystallization has been triggered. The acid number of the compound is 198 (theoretical 199).
70,4 g 1-Chlormethylnaphthalin hinzu. Dann
wird das Ganze 24 Stunden unter Rückfluß erhitzt. The sodium salt of the above acid is prepared from this in the usual way by stoichiometric neutralization at room temperature with sodium carbonate. F. (in capillary tube) = 260 to 262 ° C.
The 2-furfuryl-2- (naphthyl-Γ - methyl) -malonic acid diethyl ester used above as starting material has been prepared as follows: 96 g of a sodium ethylate solution, which has been prepared by dissolving 9 g of sodium in 256 cm 3 of absolute ethyl alcohol, is obtained Furfurylmalonic acid diethyl ester is added, and the well-stirred mixture is brought to the boil for 5 minutes. It is then allowed to cool and 70.4 g of 1-chloromethylnaphthalene are added dropwise to the mixture from a dropping vessel. The whole is then refluxed for 24 hours.
The mixture is then extracted with benzene and the extract was distilled to give 105 g of a viscous, honey-yellow liquid from Kp.i, 5202 are obtained up to 203 0 C. The yield is 69% of theory.
