Description
Die Erfindung betrifft ein Verfahren zur Herstellung von 9-Thiabicyclononanderivaten der allgemeinen
Formel
[4,2,1] nonan
in der X gleich oder verschieden sein kann und Brom-, Jod-, Fluor- oder Wasserstoffatome oder eine
Hydroxyl-, Alkoxy-, Hydroxyalkoxy-, Aryloxy-, halogensubstituierte Alkoxy-, Amino-, Aryl-, Alkyl- und
cycloalkylsubstituierte Amino-, halogensubstituierte Aryl-, Pyridinyl-, Cyano-, Isothiocyano-, Chlorisothiocyano-,
Acyloxy-, Arylmercapto-, halogensubstituierte Arylmercapto-, Acylmercapto-, Alkylthiocarbamylmercapto-,
Alkylcarbamylmercaptogruppe und t = 0, 1 oder 2 bedeutet, welches darin besteht,
daß man durch Umsetzung von Dichlor-9-thiabicyclononan oder dessen 9-Oxid oder 9,9-Dioxid der

Diese beiden Isomeren sind leicht ineinander überführbar. Schon allein durch Auflösen in einem
ionisierenden Lösungsmittel, auf Grund der bekannten Tendenz von /i-Chlorsulfiden, im Gleichgewicht
mit den entsprechenden Sulfoniumchloriden mit dreigliedrigem Ring zu sein, wie es von F u s ο η
und Mitarbeiter, J. Org. Chem., 11, 476, und von Marvel und Weil, J. Am. Chem. Soc, 76, 61,
beschrieben ist.

in einem inerten Lösungsmittel bei einer Temperatur von —40 bis etwa 1500C mit Verbindungen der allgemeinen
Formel MX, in der M ein Wasserstoffatom, ein Metallkation, ein Ammonium- oder alkylsubstituiertes
Ammoniumion bedeutet und X eine der oben angegebenen Bedeutungen hat, entweder unmittelbar
beide Chloratome oder diese nacheinander ersetzt und gegebenenfalls so erhaltene 9-Thiabicyclononanderivate
in an sich bekannter Weise zum 9-Oxid oder 9,9-Dioxid oxydiert.
Das ^,ß'-Dichlor^-thiabicyclononan wird durch
eine Ringbrückenbildungsreaktion hergestellt, wobei ein Schwefelchlorid, vorzugsweise Schwefeldichlorid,
mit einem Cyclooctadien, wie beispielsweise Cyclooctadien-(l,5), Cyclooctadien-(1,4) und Cyclooctadien-(l,3),
umgesetzt wird, was zur Bildung eines ^,//'-Dichlor^-thiabicyclononans führt. Die in ß- und
^'-Stellung zu der Sulfidbindung befindlichen Chloratome ergeben bei Behandlung mit nucleophilen
Reagenzien Austauschreaktionen mit den gewünschten Substituenten.
Es wird angenommen, daß das Produkt von Schwefeldichlorid mit Cyclooctadion-(1,5) hauptsächlich
ein Isomeres ist, wobei kleine Mengen eines anderen Isomeren ebenfalls vorhanden sind. Es
kommen daher zwei wahrscheinliche Strukturen in Betracht, wie sie durch die folgenden Formeln
gezeigt sind.
2,6-Dichlor-9-thiabicyclo [3,3,1 ] nonan
Demzufolge kann jede Austauschreaktion der Stammdichlorverbindung entweder ein 2,6-disubstituiertes
9-Thiabicyclo[3,3,l]nonan oder ein 2,5-disubstituiertes 9-Thiabicyclo[4,2,l]nonan oder beide
ergeben. Wegen dieser Uberführbarkeit der zwei Ringsysteme ineinander ist es schwierig, wenn nicht
unmöglich, irgendein Produkt dem [3,3,1]- oder dem [4,2,1}-System zuzuschreiben, und viele der
Rohprodukte zeigen, daß sie Gemische beider Isomeren sind, wenn auch ein Isomeres vorherrschen
kann.
Die 9-Oxide oder 9,9-Dioxide werden erfindungsgemäß durch Oxydation der entsprechenden Sulfide
hergestellt, ζ. B'. mit Wasserstoffperoxid, Peressigsäure, Perbenzoesäure, Perphthalsäure oder anderen
organischen Peroxidsäuren, Salpetersäure, Stickstoffdioxid oder Stickstofftetroxid, Permanganaten,
Chromsäure oder Dichromaten, Bromsäure oder Bromaten, unterchlorige Säure (oder Chlor + Wasser),
Ozon oder molekularem Sauerstoff (vorzugsweise unter Verwendung eines Katalysators, wie
beispielsweise Vanadiumoxid oder Stickstoffoxid). Die Sulfoxidgruppe wird auch leicht durch Einführung
von 1 Moläquivalent Chlor oder Brom in das Sulfid gebildet. Das erhaltene S-Dihalogenaddukt
wird dann hydrolysiert oder mit einer organischen Carbonsäure behandelt, um die beiden
Halogenatome durch ein Sauerstoffatom zu ersetzen, wobei auch Halogenwasserstoff gebildet wird.
Geeignete Lösungsmittel sind z. B. Alkohole, wie Methanol oder Äthanol, Ketone, wie Aceton oder
Methyläthylketon, aliphatische Säuren, wie Eisessig, Amide, wie Formamid oder Dimethylformamid,
Nitrile, wie Acetonitril, Nitrokohlenwasserstoffe, wie Nitrobenzol oder Nitromethan, Äther, wie Dioxan,
Tetrahydrofuran, Dimethoxyäthan oder Diäthyläther, tertiäre Amine, wie Pyridin, und andere polare
Lösungsmittel.
Wird die Umsetzung mit Ammoniak oder einem primären Amin durchgeführt, wobei 3 Mol hiervon
oder besser 1 MoI plus 1 oder 2 Mol eines Säureakzeptors (z. B. 2,6-Lutidin, Natriumcarbonat oder
Di-tert.-butylamin) je Mol Dichlor-9-thiabicyclononan
verwendet werden, so können neue poly-
cyclische heterocyclische Systeme des folgenden Typs gebildet werden, in denen die beiden Substituenten
X zusammenfallen:
Im allgemeinen haben die erfindungsgemäß erhältlichen Verbindungen fungicide Wirksamkeit,
wobei einige von ihnen überragend wirksam und den bisherigen Fungiciden in dieser Hinsicht weit
überlegen sind. Die erfindungsgemäßen Verbindungen sind auch als Herbicide, bakteriostatische Mittel,
Insekticide, Milbenbekämpfungsmittel, Nematocide und als chemische Zwischenprodukte wertvoll. Die
bifunktionellen Derivate sind zur Polymerisation brauchbar.
In den Beispielen bedeuten Teile Gewichtsteile, falls es nicht anders angegeben ist.
Beispiel 1
Dibrom-9-thiabicyclononan
Dibrom-9-thiabicyclononan
Zunächst wurden zur Herstellung von Dichlor-9-thiabicyclononan 1296 Teile Cyclooctadien-(1,5)
und 1236 Teile Schwefeldichlorid aus getrennten Gefäßen gleichzeitig zu 1500 Teilen Hexan unter
Rühren und Kühlen auf etwa 10 bis 20°C zugegeben. Nach beendeter Zugabe wurde das Gemisch
1 Stunde gerührt. Die Feststoffe wurden durch Filtrieren entfernt, und es wurde nach dem Trocknen
gefunden, daß sie 1295 Teile Produkt vom F. 100 bis 1010C enthielten. Weitere Fraktionen, insgesamt
zusätzlich 725 Teile, wurden durch teilweises Verdampfen und Kühlen gewonnen. Diese Fraktionen
schmolzen im Bereich von 89 bis 980C. Durch Umkristallisieren
aus Benzol—Heptan erhielt man eine farblose kristallisierte Festsubstanz vom F. 102 bis
103°C.
Eine Lösung von 21,1 Teilen des so erhaltenen Dichlor-9-thiabicyclononan in 100· VoI um teilen Eisessig
wurde mit gasförmigem Bromwasserstoff gesättigt und dann in einem verschlossenen Gefäß
12 Stunden stehengelassen. Die Lösung wurde dann in Wasser gegossen, was die Ausfällung des Produkts
bewirkte. Es wurden 26 Teile einer farblosen Festsubstanz vom F. 131 bis 134,5"C erhalten.

25 Teile Natriumjodid in 250 Volumteilen Aceton wurden zu einer Lösung von 10 Teilen Dichlor-9-thiabicyclononan
in 100 Volumteilen Aceton zugegeben. Nach 3 Stunden bei 25°C wurde die Lösung
eingeengt und zur Entfernung von Feststoffen filtriert. Die Feststoffe wurden mit Wasser salzfrei gewaschen,
wobei 14 Teile eines farblosen kristallinen Produkts zurückblieben, das nach Umkristallisieren aus Heptan
bei 139,5 bis 140°C schmolz.

42,2 Teile Dichlor-9-thiabicyclononan und 50 Teile Kaliumfluorid wurden in 400 Teilen Dimethylformamid
18 Stunden bei 12O0C erhitzt. Dann wurde die Lösung auf die Hälfte ihres Volumens eingeengt
und abgekühlt, um das Produkt auszufällen, das dann aus Benzol umkristallisiert wurde. Man erhielt
so 12 Teile farblose Festsubstanz vom F. 159 bis 166° C (Zersetzung).

Ein Gemisch aus 42,2 Teilen DichIor-9-thiabicyclononan,
200 Volumteilen 10%igem wäßrigem Natriumhydroxid und 200 Teilen Dimethoxyäthan wurde
3 Tage unter Rückfluß erhitzt. Das Gemisch wurde dann im Vakuum eingeengt, bis die Kristalle ausfielen.
Die Kristalle wurden abfiltriert und aus Äthanol umkristallisiert. Das Produkt (24 Teile) war eine
farblose Festsubstanz, die in Wasser mittelmäßig löslich war und bei 217 bis 218°C schmolz (gewisse
Ansätze schmolzen bei 249,5 bis 250°C, und es ist daher Dimorphismus anzunehmen).

Zu 8,7 Teilen Dihydroxy-9-thiabicyclononan und 100 Volumteilen Essigsäure wurden 13 Volumteile
30%iges Wasserstoffperoxid bei 25 bis 330C zugegeben. Dann wurde das Reaktionsgemisch langsam
auf 100°C erhitzt, abgekühlt und teilweise eingedampft, und die ausgefallenen Kristalle wurden aus
Methanol—Butylacetat umkristallisiert. Man erhielt so eine kristalline wasserlösliche Festsubstanz vom
F. >300°C.

Di-(2-hydroxyäthoxy)-9-thiabicyclononan
22,4 Teile Kaliumhydroxid und 500 Volumteile Äthylenglykol wurden unter Rückfluß erhitzt, um
Wasser abzutreiben und so das Kaliumsalz des Glykols zu bilden. Die Lösung wurde abgekühlt,
21,1 Teile Dichlor-9-thiabicyclononan wurden zugegeben, und es wurde dann wieder kurz unter
Rückfluß erhitzt. Nach dem Abkühlen wurde das Glykol bis 1000C (0,2 mm) abgestreift, dann Wasser
zugegeben, was die Abtrennung des organischen Produkts in Form eines viskosen Öls bewirkte.
Durch Bestimmung der Alkoholzahl wurde festgestellt, daß es das gewünschte Diol war.
Di-(2-hydroxyäthoxy)-9-thiabicyclononan-9,9-dioxid
24,3 Teile Dichlor-9-thiabicyclononan-9,9-dioxid, 22,4 Teile Kaliumhydroxid und 500 Volumteile Äthylenglykol
wurden 3 Stunden unter Rückfluß erhitzt. Das Glykol wurde dann unter 0,4 mm Vakuum
abgestreift, wobei ein trockener Rückstand verblieb, der dann in 500 Teilen Wasser gelöst wurde. Diese
Lösung schied beim Einengen Kaliumchlorid ab. Die ,letzten Spuren von Kaliumchlorid wurden als
Ausfällung durch Zugabe von Methanol und Salzsäure und anschließendes Filtrieren abgetrennt.
Durch Eindampfen des Filtrats und Waschen des Rückstands mit Aceton erhielt man 15,5 Teile
einer in Wasser löslichen, in Aceton unlöslichen farblosen kristallinen Festsubstanz vom F. 133 bis
134°C.

50 Teile Dichlor-9-thiabicyclononan wurden mit
300 Volumteilen Methanol einen Tag stehengelassen und dann 24 Stunden unter Rückfluß erhitzt und
dann der überschüssige Alkohol abdestilliert. Das zurückbleibende Produkt war ein bernsteinfarbener
Sirup vom Kp.i2142 bis 145 0C. Das Infrarotspektrum
zeigte das Vorhandensein von Methoxygruppen (-CH3- und C — O — C-Banden). Die
Chloranalyse zeigte, daß das Produkt ein Gemisch aus 26% Monomethoxy-monochlor- und 74% Dimethoxy-9-thiabicyclononan
war.

Ein Gemisch aus 10,5 Teilen Dichlor-9-thiabicyclononan
und 9,7 Volumteilen Anilin in 200 Teilen 1,2-Dimethoxyäthan wurde 16 Stunden unter Rückfluß
erhitzt. Zu dem teilweise kristallisierten Gemisch wurde überschüssiges alkoholisches Kaliumhydroxid
zugesetzt. Dann wurde kurz unter Rückfluß erhitzt, das Gemisch mit einem großen Überschuß Wasser
verdünnt, filtriert und die Feststoffe mit Wasser gewaschen. Die getrockneten Feststoffe (10 Teile)
schmolzen bei 225 bis 226,5 0C.

Eine Lösung von 211 Teilen Dichlor-9-thiabicyclononan
in 1000 Volumteilen 1,2-Dimethoxyäthan wurde mit 1000 Teilen einer 10%igen äthanolischen
Ammoniaklösung vermischt. Nach 4tägigem Stehen wurde das Gemisch mit 3000 Teilen 10%iger wäßriger
Natriumhydroxidlösung verdünnt und filtriert und die Feststoffe so lange mit Wasser gewaschen, bis
sie frei von starker Base waren. Nach dem Trocknen an der Luft erhielt man 70 Teile einer hochschmelzenden
Festsubstanz, die in Wasser unlöslich und in verdünnter Salzsäure löslich war.

zugegeben. Dann wurde 6 Stunden unter Rühren auf einem Dampfbad erhitzt. Das Reaktionsgemisch wurde zu Eiswasser zugesetzt, die organische
Schicht mit Salzsäure gewaschen, filtriert und unter Wasserstrahlvakuum eingedampft, wobei ein viskoser
Sirup zurückblieb.

und 200 Volumteilen Pyridin wurde 65 Stunden bei 90 bis 1000C erhitzt. Das Gemisch wurde
abgekühlt und die ausgefallene Festsubstanz durch Filtrieren isoliert. Das Produkt war eine farblose
mikrokristalline wasserlösliche Festsubstanz von Undefiniertem Schmelzpunkt, die eine korrekte Analyse
(Bestimmung von ionischem Chlorid durch Volhard-Titration) für das gewünschte Produkt zeigte.

Ein Gemisch aus 21,1 Teilen Dichlor-9-thiabicyclononan,
98 Teilen Trimethylamin und 200 Volumteilen 95%igem Äthanol wurde 20 Stunden bei
Zimmertemperatur gerührt. Die ausgefallenen Feststoffe wurden abfiltriert und an der Luft getrocknet.
Das Produkt war eine farblose wasserlösliche Festsubstanz von Undefiniertem Schmelzpunkt.

Salze abfiltriert, Wasser zu dem Filtrat gegeben, um das Produkt auszufällen, das abfiltriert, mit
Wasser gewaschen und an der Luft getrocknet wurde. Man erhielt so 103 Teile einer farblosen kristallinen
Festsubstanz vom F. 100 bis 1010C. Das Produkt schmolz nach Umkristallisieren aus Methanol bei
102 bis 103°C.

2 Stunden unter Rückfluß erhitzt. Dann wurde das Gemisch abgekühlt und mit Wasser verdünnt,
wobei ein Niederschlag erhalten wurde, der nach Abfiltrieren und Waschen mit zusätzlichen Mengen
Wasser bei 163 bis 164° C schmolz.

Nach 2stündigem Erhitzen unter Rückfluß mit 50%iger wäßriger Schwefelsäure war das Dinitril
in Dicarboxy^-thiabicyclononan übergeführt, einer hochschmelzenden, farblosen, in Wasser nahezu
unlöslichen und in wäßrigem Natriumhydroxid löslichen Festsubstanz mit einem Neutralisationsäquivalent von 165. Nach 5stündigem Erhitzen
unter Rückfluß des Dinitrils mit überschüssigem Kaliumhydroxid in Äthanol kristallisierte das Diamid
dieser Säure beim Abkühlen in Form einer farblosen Festsubstanz mit einem Schmelzpunkt oberhalb
2000C aus.

Ein Gemisch aus 38 Teilen Ammoniumthiocyanat, 42,2 Teilen Dichlor-9-thiabicyclononan und 400 Volumteilen
Methyläthylketon wurde 14 Stunden unter Rückfluß erhitzt. Dann wurde Wasser zugegeben,
was die Ausfällung von Feststoffen bewirkte. Diese Feststoffe wurden abfiltriert, und es wurde festgestellt,
daß sie aus 39 Teilen eines farblosen kristallinen Produkts vom F. 149°C bestanden.

Die Salze lösten sich, wobei ein viskoses organisches öl zurückblieb. Letzteres wurde von zurückbleibendem
Wasser und Keton unter 10 mm Vakuum bei 1000C befreit. Das Produkt war ein viskoses öl,
das sich beim Stehen verfestigte.

- Beispiel 22 (Phenylmercapto)-chlor-9-thiabicyclononanZu 13 Teilen Natriumthiophenolat in 100 Teilen Dimethylformamid wurden 21 Teile Dichlor-9-thiabicyclononan zugegeben. Nach einem Tag wurde das" ausgefallene Natriumchlorid durch Filtrieren entfernt. Das Filtrat wurde zu Wasser zugegeben, das Gemisch mit Benzol extrahiert und das Benzol mit Wasser gewaschen, über Magnesiumsulfat getrocknet und im Vakuum eingedampft, wobei ein gelbes öl zurückblieb, das beim weiteren Stehen teilweise kristallisierte und 8% hydrolysierbares Chlor enthielt, was zeigte, daß es das gewünschte Produkt in unreinem Zustand war.Beispiel 23 Di-(phenylmercapto)-9-thiabicyclononanZu 26 Teilen Natriumthiophenylat in 100 Teilen Dimethylformamid wurden 21 Teile Dichlor-9-thiabicyclononan zugegeben. Nach einem Tag wurde das Reaktionsgemisch wie im vorhergehenden Beispiel aufgearbeitet, wobei man ein öl und Kristalle erhielt. Beim teilweisen Eindampfen des Filtrats wurden 14 Teile eines farblosen festen Produkts vom F. 81,5 bis 85 "C erhalten.Analyse: C8Hi2S(SC6H4Cl).Berechnet ... S 22,5%;
gefunden ... S 21,7%.Beispiel 25 (p-Chlorphenylmercapto)-chlor-9-thiabicyclononanDie obige Arbeitsweise wurde unter Verwendung der Hälfte der Menge p-Chlorphenylmercaptid wiederholt, wobei ein gelbes öl erhalten wurde, das praktisch die korrekte Chloranalyse für (p-Chlorphenylmercapto)-chIor-9-thiabicyclononan zeigte.Beispiel 26Bis-(O,O-diäthoxythiophosphinyImercapto)-9-thiabicyclononan .Zu einer unter Rückfluß befindlichen Lösung von 10,45 Teilen Dichlor-9-thiabicyclononan in 300 Teilen Toluol wurden unter Rühren 22 Teile Natrium- Ο,Ο-diäthylphosphordithiolat zugegeben. Das Er hitzen unter Rückfluß wurde V2 Stunde fortgesetzt, dann wurde das Gemisch filtriert, das Filtrat wurde mit Wasser gewaschen und dann das Lösungsmittel verdampft. Die zurückbleibende Festsubstanz wurde aus Äthanol umkristallisiert, wobei man 22 g eines hellgelblichen Stoffes vom F. 54 bis 57° C erhielt.Analyse: Ci8Hi2O4P2Ss.Berechnet ... P 12,1%, S 31,4%; gefunden ... P 12,2%, S 31,4%.Beispiel 27Bis-(O,O-dimethoxythiophosphinylmercapto)-9-thiabicyclononanDie Arbeitsweise des vorhergehenden Beispiels wurde unter Verwendung von Natrium-O,O-dimethylphosphordithiolat an Stelle des Äthylesters wiederholt, wobei man das gewünschte Produkt als cremefarbige Festsubstanz vom F. 126 bis 128°C erhielt.Beispiel 28O,O-Diäthyl-chlor-9-thiabicyclononylphosphordithionatEine Lösung von 42,2 Teilen Dichlor-9-thiabicyclononan, 36 Teilen Natrium-O,O-diäthylphosphordithiolat und 21 Teilen Triäthylamin in 180 Teilen Benzol wurde 10 Stunden unter Rückfluß erhitzt, abgekühlt und zur Entfernung des Triäthylaminhydrochlorids filtriert. Das Benzol wurde verdampft, wobei man das Produkt als rötlichen Sirup erhielt.Analyse: Ci2H22O2ClPS3.

gefunden ... Cl 9,0%.Beispiel 29Bis-(N,N-dimethylthiocarbamylmercapto)-9-thiabicyclononanEin Gemisch aus 21,1 Teilen Dichlor-9-thiabicyclononan und 141 Teilen 20%igem wäßrigem Natriumdimethyldithiocarbamat wurde 1 Stunde unter Rühren bei 95°C erhitzt. Das Gemisch wurde dann filtriert, die Feststoffe wurden in heißem Benzol aufgenommen (etwas Salze verblieben ungelöst), filtriert, mit Hexan verdünnt und abgekühlt, wobei 19 Teile kristalline Festsubstanz vom F. 209 bis 2100C erhalten wurden.Analyse: C14H24N2S5.Berechnet ... Cl 0,0%, S 42,1%; gefunden ... Cl 0,0%, S 41,6%.o Patentanspruch:Verfahren zur Herstellung von 9-Thiabicyclononanderivaten der allgemeinen Formelin der X gleich oder verschieden sein können und Brom-, Jod-, Fluor- oder Wasserstoffatome oder eine Hydroxyl-, Alkoxy-, Hydroxyalkoxy-, Aryloxy-, halogensubstituierte Alkoxy-, Amino-, Aryl-, Alkyl- und cycloalkylsubstituierte Amino-, halogensubstituierte Aryl-, Pyridinyl-, Cyano-, Isothiocyano-, Chlorisothiocyano-, Acyloxy-, Arylmercapto-, halogensubstituierte Arylmercapto-, Acylmercapto-, Alkylthiocarbamylmer-capto-, Alkylcarbamylmercaptogruppe und t = 0, 1 oder 2 bedeutet, dadurch gekennzeichnet, daß man durch Umsetzung von Dichlor-9-thiabicyclononan oder dessen 9-Oxid oder 9,9-Dioxid der allgemeinen FormelClClin einem inerten Lösungsmittel bei einer Temperatur von —40 bis etwa 1500C mit Verbindungen der allgemeinen Formel MX, in der M ein Wasserstoffatom, ein Metallkation, ein Ammonium- oder alkylsubstituiertes Ammoniumion bedeutet und X eine der oben angegebenen Bedeutungen hat, entweder unmittelbar beide Chloratome oder diese nacheinander ersetzt, und gegebenenfalls so erhaltene 9-Thiobicyclonohanderivate in an sich bekannter Weise zum 9-Oxid oder 9,9-Dioxid oxydiert.
