Es wurde vorgeschlagen, daß die Aktivierung von IK-Kaliumkanäle eine entscheidende Rolle bei der Ca2+-Kaskade in T-Zellen spielt, die letztendlich zur Aktivierung der T-Zellen selber führt. Inhibitoren des IK-Kaliumkanals wurden daher bereits als Immunsuppressiva und als Ersatz für die immunsupressiv aktiven Stoffe FK506, Rapamycin und Cyclosporin A vorgeschlagen (Jensen et al., Cunent Drug Targets, 2001, 2: 401-422). in Abwesenheit markierter Peptide inkubiert. Die Dephosphorylierungsreaktionen
werden durch Zugabe der Peptide gestartet und dann für 10 min
bei 30°C
inkubiert. Die Beendigung der Reaktion sowie die Abtrennung freien
Phosphats von phosphorylierten Peptiden erfolgt wie in Liu et. al.
und Manalan und Klee (supra) beschrieben. Der Grad der Dephosphorylierung,
der in Abwesenheit der Testsubstanz gemessen wird, wird als 100%
Calcineurinaktivität
definiert, während
der Grad der Dephosphorylierung, der in Abwesenheit von Testsubstanz

Calcineurin-Antagonisten kann dann über die prozentuale Verringerung

Nach 7 h Vorinkubation der Zellen mit 1-EBIO (1 mM) ging die Aktivierung der IK-Kaliumkanäle durch ATP praktisch völlig verloren und es wurde eine plateau-ähnliche Depolarisierung beobachtet. Bei Vorinkubation der Zellen für 3 Tage mit 1-EBIO, ZOX und CZ (je 1 mM in DMSO) wurde praktisch keine Hyperpolarisierung der Zellen nach Zugabe mit ATP mehr beobachtet. Während in Kontrollexperimenten (Zugabe von ATP zu unbehandelten bzw. DMSO (0,1 %) behandelten Zellen) ein ΔVm von ca. -25 mV beobachtet wurde, wurde bei 3 d Vorbehandlung mit 1-EBIO, ZOX und CZ eine kaum messbare Hyperpolarisierung von -3 mM, -4 mV bzw. -5 mV gemessen. Darüber hinaus führte die Vorinkubation mit jedem der Kanal Aktivatoren zu einer deutlichen Depolarisierung des Ruhepotentials der Zellen (Kontrolle: -52 mV; 1-EBIO: -19mV; ZOX: -35 mV; CZ: -42mV). Die Desensitivierung die bei Langzeit-Inkubation mit IK-Kaliumkanal-Aktivatoren beobachtet wurde, wurde jedoch nicht bei Langzeitinkubation mit dem natürlichen Stimulans ATP beobachtet. Es wurde nun untersucht ob die Langzeitinkubation mit 1-EBIO einen Effekt auf die intrazelluläre Ca2+-Kaskade, die sowohl an der Proliferation und Differenzierung von Keratinozyten, aber auch an der Aktivierung von beispielsweise T-Zellen beteiligt ist, hat. Dazu wurden HaCaT-Zellen mit Fluoreszenzfarbstoffen über ihren membrangängigen AM Ester beladen. Stammlösungen von 0,9 M Calcium-Green-1 AM und Fura Red AM (Molecular Probes) wurden hergestellt indem 50 μg von jedem Farbstoff in 20 μl DMSO plus 20 μl Pluronic F-127 20% (Molecular Probes) gelöst wurden. Vor dem Beladen wurden die Zellen 3 mal mit HEPES-gepufferter Salzlösung (HBS) gewaschen. Die Zellkulturen wurden dann 30 min bei 37°C in 3 ml HBS zu dem 20 μl Stammlösung zugegeben wurde, inkubiert (Endkonzentration: 6 μM Calcium Green-1; 7 μM Fura Red) und anschließend gründlich gewaschen. Alle optischen Aufnahmen wurden in HBS (118 mM NaCl, 1,5 mM CaCl2, 1 mM MgCl2, 20 mM Na-Glukonat, 6 mM HEPES, 10 mM D-Glukose, pH 7,4) durchgeführt. Die Zellkulturschalen wurden auf einem Olympus BX50WI Mikroskop (Objektiv: Olympus LUMPlanFI 40xW) untersucht an dem ein konfokales Laser-scanning System (BioRad MRC 1024) und ein selbstgebautes photometrisches System angebracht waren. Die Proben wurden bei 48530 nm in 5 s Intervallen belichtet. Zwei Photodioden sammelten Fluoreszenzlicht, das von der Zellschicht emittiert wurde, gleichzeitig bei 530±30 nm und 660±50nm und produzierten Spannungssignale, die den Fluoreszenzemissionen proportional war. Die Signal wurden linear amplifiziert bevor sie digitalisiert wurden (DT2812 Data Translation). Die Daten wurden mittels einer modifizierten Version des Programms QTRAC wiedergegeben. - 4) Einer Kombination aus einem IK Kanal bevorzugt, insbesondere ausgewählt aus 1-EBIO, Chlorzoxazon, DCEBIO und Zoxazolamin und einem Calcineurin Antagonist, bevorzugt ausgewählt aus Cyclosporin A, Tacrolimus und Pimecrolimus.
- 5) Einer Kombination aus IK Kanal Inhibitor, bevorzugt ausgewählt aus Charybdotoxin, TRAM-34 und Clotrimazol und einem Calcineurin-Antagonist, bevorzugt ausgewählt aus Cyclosporin A, Tacrolimus und Pimecrolimus.
